New magnetic resonance spectroscopy biomarker for monitoring neurodegenerative diseases: animal models.

Creatine kinase (CK) plays a central role in energy transfer in cells with high-energy demands, and the enzyme is rather susceptible to oxidative inactivation. The aim of the present study was to investigate whether the rate constant of forward CK reaction (k(for)) is a suitable indicator of alterations in cerebral energy metabolism. We monitored k(for) in the rat brain non-invasively by in vivo phosphorus ((31)P) magnetic resonance spectroscopy (MRS). To alter energy metabolism, we applied following experimental models: Huntington's disease, diabetes mellitus, chronic alcohol intoxication and

Differential diagnosis between cerebral tuberculosis and neurocysticercosis by magnetic resonance spectroscopy.

Single enhancing brain lesions (SELs), mostly as a result of neurocysticercosis or tuberculosis, are a common cause of seizures. Ten patients with SELs caused by neurocysticercosis (n=6) or tuberculosis (n=4) were examined by proton magnetic resonance spectroscopy. Tuberculomas had a high peak of lipids, more choline, and less N-acetylaspartate and creatine. The choline/creatine ratio was greater than 1 in all tuberculomas but in none of the cysticerci. Magnetic resonance spectroscopy differentiates SELs caused by cysticercosis or tuberculosis and may avoid brain biopsies or unnecessary antitu

Measurement of human brain dexfenfluramine concentration by 19F magnetic resonance spectroscopy.

OBJECTIVE: The goals of this study were to quantitate the brain concentration of the anorectic drug dexfenfluramine (DF) in human subjects receiving clinical doses of DF and to determine whether human brain DF concentrations approach those reported to cause irreversible neurochemical changes in animals. Each subject's brain DF concentration was measured several times over an extended period of DF treatment to determine whether drug accumulation in the brain would plateau or continue to increase throughout the treatment period. DESIGN: Fluorine magnetic resonance spectroscopy (19F-MRS) was use

Proton magnetic resonance spectroscopy pattern of progressive multifocal leukoencephalopathy in AIDS.

The objective was to determine whether the use of intermediate echo times (135 ms) in proton magnetic resonance spectroscopy (1H-MRS) detects a homogenous pattern in progressive multifocal leukoencephalopathy (PML) in HIV-1 infected people, and to confirm the results of previous studies. Six patients infected with HIV-1, with PML established by biopsy, and six healthy age and sex matched volunteers were evaluated to define their spectroscopic pattern. 1H-MRS spectra performed at 1.5 T were obtained with the STEAM sequence: TE/TM/TR, 20 ms/13.7 ms/2000 ms; 2500 Hz, size 2048 points, 256 acquisi

Metabolic abnormalities in developmental dyslexia detected by 1H magnetic resonance spectroscopy.

BACKGROUND: Neurological and physiological deficits have been reported in the brain in developmental dyslexia. The temporoparietal cortex has been directly implicated in dyslexic dysfunction, and substantial indirect evidence suggests that the cerebellum is also implicated. We wanted to find out whether the neurological and physiological deficits manifested as biochemical changes in the brain. METHODS: We obtained localised proton magnetic resonance spectra bilaterally from the temporo-parietal cortex and cerebellum of 14 well-defined dyslexic men and 15 control men of similar age. FINDINGS:

Use of localized proton nuclear magnetic resonance spectroscopy in Canavan's disease.

Canavan's disease is characterized by megalencephaly, leukodystrophy and early motor and mental retardation. On computerized tomography and magnetic resonance imaging, severe changes compatible with white matter disease due to demyelination is observed. It has been demonstrated that urinary N-acetylaspartate levels are increased because of a deficiency of aspartoacylase (N-acyl-L-aspartate aminohydrolase) in these patients. In this study, with the use of proton nuclear magnetic resonance spectroscopy, we were able to demonstrate elevated levels of N-acetylaspartate compared to choline and crea

Detection of apoptotic cell death by proton nuclear magnetic resonance spectroscopy.

Cells undergoing apoptosis (programmed cell death) display profound morphologic and biochemical changes in the nucleus, cytoplasm, and plasma membrane. We have shown a direct temporal relationship between the onset of apoptosis in Jurkat T-cell lymphoblast cultures and a greater than two-fold increase in the signal intensity of the methylene resonance (at 1.3 ppm) as observed by proton nuclear magnetic resonance spectroscopy (1H NMR). The increase in the methylene resonance intensity was seen when apoptosis was induced by serum deprivation, glucocorticoid, and doxorubicin treatment but not in

Comparative Analysis of Magnetic Resonance Spectroscopy and Post-contrast Magnetic Resonance Imaging in the Evaluation of Intracranial Ring-Enhancing Lesions.

Background Intracranial ring-enhancing lesions are diagnostically heterogeneous because infective, neoplastic, demyelinating, and vascular etiologies may show overlapping conventional magnetic resonance imaging (MRI) appearances. This study evaluated and compared the diagnostic performance of post-contrast MRI and magnetic resonance spectroscopy (MRS) in the etiological characterization of intracranial ring-enhancing lesions. Materials and methods This hospital-based prospective observational, non-interventional study included 45 patients with intracranial ring-enhancing lesions. All patients

Magnetic resonance imaging, magnetic resonance spectroscopy, and facial dysmorphism in a case of Lowe syndrome with novel OCRL1 gene mutation.

Lowe syndrome is a multisystem disorder characterized by anomalies of the eye, the nervous system, and the kidney. It is an uncommon, X-linked disease. Bilateral cataract and severe hypotonia are present at birth. Psychomotor retardation is evident in childhood, while renal complications arise in adolescence. The mutation of the gene OCRL1 localized at Xq26.1 is responsible for the disease. The authors report on a 12-year-old male with mental retardation, facial dysmorphism as prominent forehead, long and slender-shaped face, prominent eyebrows, epicanthus, microphthalmia, low-posterior set ea

Maturation of limbic regions in Asperger syndrome: a preliminary study using proton magnetic resonance spectroscopy and structural magnetic resonance imaging.

People with autistic spectrum disorders (ASD, including Asperger syndrome) may have developmental abnormalities in the amygdala-hippocampal complex (AHC). However, in vivo, age-related comparisons of both volume and neuronal integrity of the AHC have not yet been carried out in people with Asperger syndrome (AS) versus controls. We compared structure and metabolic activity of the right AHC of 22 individuals with AS and 22 healthy controls aged 10-50 years and examined the effects of age between groups. We used structural magnetic resonance imaging (sMRI) to measure the volume of the AHC, and m

Brain abscess and glioblastoma identified by combined proton magnetic resonance spectroscopy and diffusion-weighted magnetic resonance imaging--two case reports.

The differential diagnosis between brain abscesses and necrotic tumors such as glioblastomas is sometimes difficult to establish by conventional computed tomography and magnetic resonance imaging. Combined proton magnetic resonance spectroscopy (1H-MRS) and diffusion-weighted magnetic resonance imaging (DWI) were used to establish the preoperative diagnosis of brain abscess and glioblastoma. DWI visualized the brain abscess as a homogeneous hyperintense lesion and 1H-MRS revealed the presence of acetate, lactate, and amino acids and the absence of the normal brain components. DWI sometimes sho

Metabolite Levels Measured Using Magnetic Resonance Spectroscopy in Pediatric Methotrexate-Induced Leukoencephalopathy.

BACKGROUND: Methotrexate (MTX)-induced leukoencephalopathy is a significant neurological complication in pediatric acute lymphoblastic leukemia (ALL) treatment. Magnetic resonance spectroscopy (MRS) is a noninvasive method to assess metabolic changes associated with neurotoxicity. This study aimed to evaluate the diagnostic and prognostic value of MRS-derived metabolite levels, particularly N-acetylaspartate and N-acetylaspartylglutamate (tNAA), in MTX-induced leukoencephalopathy. PROCEDURE: We retrospectively analyzed 89 pediatric patients with ALL who underwent brain magnetic resonance imag

Exploring nuclear magnetic resonance spectroscopy for the analysis of dried blood spots.

Dried blood spots (DBS) offer a practical and relatively non-invasive method for sample collection. Here, we evaluate the feasibility of applying 1H NMR spectroscopy to metabolomic analysis of DBS. Various solvent suppression techniques and extraction protocols were tested using aqueous and methanolic solvents. Methanol was found to provide heightened metabolite recovery with fewer interfering macromolecular signals compared to aqueous buffers, supporting its use as a preferred extraction solvent. Additional experimental variables, such as extraction time and the number of DBS punches, were op
