This study analyzes the results of clinical trials of treatments with melatonin conducted in children, mostly focused on sleep disorders of different origin. Melatonin is beneficial not only in the treatment of dyssomnias, especially delayed sleep phase syndrome, but also on sleep disorders present in children with attention-deficit hyperactivity, autism spectrum disorders, and, in general, in all sleep disturbances associated with mental, neurologic, or other medical disorders. Sedative properties of melatonin have been used in diagnostic situations requiring sedation or as a premedicant in c

AIMS: Melatonin is a hormone synthesized principally in the pineal gland that has been classically associated with endocrine actions. However, several lines of evidence suggest that melatonin plays a role in pain modulation. This paper reviews the available evidence on melatonin's analgesic effects in animals and human beings.
MAIN METHODS: A medline search was performed using the terms "melatonin", "inflammatory pain", "neuropathic pain", "functional pain", "rats", "mice", "human", "receptors", "opioid" and "free radicals" in combinations.
KEY FINDINGS: The antinociceptive effect of melaton

Melatonin, the pineal gland hormone and a strong antioxidant, has long been known, particularly in animal-experiment based research and the wool-producing industry, to be a potent regulatory neuroendocrine substance in relation to hair growth, hair color and hair cycle, depending on light periods, seasonal rhythms, environmental factors and reproductive rhythms. Nevertheless, the biological mechanisms of this extremely versatile hormone, especially with regard to human hair follicles, are not fully understood. In recent years, however, essential knowledge has been gained on the melatoninergic

Morphological changes that take place in the epiphysis in the course of age involution are inseparably associated with a drastic decrease in the synthesis of melatonin (MT) - the main hormone of the pineal gland that coordinates biorhythms of the body at all system levels. The pineal gland is not the only organ able producing MT Multiple research have proved that the extrapineal MT is also widely present in human and animal organism. In this paper we present the results of our own research upon whose analysis we can base a hypothesis that MT and serotonin can play an important role in the mech

Aging is associated with a decline in immune function (immunosenescence), a condition known to correlate with increased incidence of cancer as well as infectious and degenerative diseases. Innate, cellular and humoral immunity all exhibit increased deterioration with age. Circulating melatonin decreases with age, and in recent years much interest has been focused on its immunomodulatory effect. Melatonin stimulates the production of progenitor cells for granulocytes and macrophages. It also stimulates the production of natural killer cells and CD4+ cells and inhibits CD8+ cells. The production

Melatonin is the time-keeping molecule of the organism. The production by the pineal organ is responsible for the diurnal and annual rhythms of plasma melatonin content. This contributes to synchronizing behavioural, biochemical and physiological processes to the environmental variations in photoperiod and temperature. Conservation and diversity characterize the melatonin system in vertebrates: conservation because its nocturnal pattern of production as well as its synchronizing properties are a constant; diversity because the modalities of its biosynthesis and modes of action have been profou

The number of patients with well-controlled hypertension is alarmingly low worldwide and new approaches to treatment of increased blood pressure (BP) are being sought. Melatonin has a role in blood pressure regulation. The nighttime production of melatonin is found to be reduced in hypertensive individuals. Administration of melatonin decreased BP in several animal models of hypertension, in healthy men and women, and in patients with arterial hypertension. Most promising results were achieved in patients with non-dipping nighttime pressure, in which the circadian rhythm of BP variation is dis

This paper reviews the possible participation of melatonin in the process of human reproduction. The results of several studies have shown the clear correlation between melatonin and gonadotropins and/or sexual steroids, which suggest that melatonin may be involved in the sexual maturation, ovulation or menopause. Decreased secretion of melatonin which coexists with increased fertility in the summer is specific for women living on the north hemisphere. Moreover, abnormal levels of melatonin in the blood are associated with several disorders of the hypothalamus-pituitary-gonads axis activity, i

Melatonin effects are discussed by reviewing results from mice with intact or disrupted melatonin signaling. Melatonin, the neuroendocrine hand of the clock produced in the pineal gland during night, acts upon two receptor subtypes. Melatonin receptors are found in the suprachiasmatic nuclei (SCN), hypophysial pars tuberalis (PT) and adrenal gland. In SCN, melatonin interacts with PACAP, a neuropeptide of the retinohypothalamic tract. Moreover, melatonin acts on the SCN to modulate the activity of the sympathetic nervous system. Melatonin is not required to maintain rhythmic clock gene express

Melatonin and its major metabolite, 6-sulphatoxymelatonin, can routinely be measured by RIA or ELISA. Plasma, serum or saliva samples maybe used for melatonin measurement and urine for the metabolite. Melatonin is a hormone produced by the pineal gland witha clear circadian rhythm giving low levels by day and a peak during the night in normally entrained individuals. The timing of sample collection for measurement is therefore crucial. Melatonin levels are primarily assessed to determine the timing of an individual's internal body clock and therefore indicate any misalignment to the 24 h day.

Melatonin, the chief secretory product of the vertebrate pineal gland is also known to occur in numerous photoautotrophic organisms. The indoleamine is suspected to act as a transducer of photoperiodic information and/or to participate in antioxidative protection. In higher plants and other photoautotrophic organisms, contradictory results for melatonin content for samples from the same species show that further improvement of methods for reliable quantification is required. In the present study, melatonin was quantified in tomatoes, ginger and the marine green macroalga, Ulva lactuca, after e

Melatonin is a neuroendocrine hormone secreted by the pineal gland to transduce the body's circadian rhythms. An internal 24 hour time keeping system (biological clock) regulated by melatonin, controls the sleep-wake cycle. Melatonin production is a highly conserved evolutionary phenomenon. The indole hormone is synthesized in the pinealocytes derived from photoreceptors. Altered patterns and/or levels of melatonin secretion have been reported to coincide with sleep disorders, jetlag, depression, stress, reproductive activities, some forms of cancer and immunological disorders. Lately, the phy

Melatonin (N-acetyl-5-methoxytryptamine), the chief secretory product of the pineal gland in the brain, is well known for its functional versatility. In hundreds of investigations, melatonin has been documented as a direct free radical scavenger and an indirect antioxidant, as well as an important immunomodulatory agent. The radical scavenging ability of melatonin is believed to work via electron donation to detoxify a variety of reactive oxygen and nitrogen species, including the highly toxic hydroxyl radical. It has long been recognized that the damaging effects of ionizing radiation are bro

The present paper reviews the findings accumulated on the role of pineal gland and its hormone - melatonin - in regulation of the hypothalamo-neurohypophysial system activity. Effects of modified photoperiod, pinealectomy or treatment with melatonin on the vasopressin and oxytocin biosynthesis and/or secretion have been described. Taken together, the in vivo and in vitro data suggest that the effect of melatonin on the vasopressin and oxytocin secretion depends on this pineal hormone concentration and experimental conditions.

Melatonin (MLT) is the main hormone released from the pineal gland and has proved to have physiological antitumor activity. MLT has been shown to exert anticancer activity through several biological mechanisms: antiproliferative action, stimulation of anticancer immunity, modulation of oncogene expression, and anti-inflammatory, anti-oxidant and anti-angiogenic effects. Several experimental studies have shown that MLT may inhibit cancer cell growth, and preliminary clinical studies seem to confirm its anticancer property in humans. In addition, MLT may have other biological effects, which coul

The production of melatonin by the pineal gland and its functions are considered, and then its possible uses in the treatment of children. Institutionalized children, and those with severe learning disorders, often have irregular sleep-wake patterns, and there is evidence that melatonin can result in improvement to the benefit of both the child and the carers. The affected children can become less irritable, calmer, happier, and content. Also they may socialize better and become more attentive, with an improvement in their cognitive abilities. Another group of children who are likely to suffer

The relationship between the pineal gland and aging has been assumed already nearly a century ago. Recently, melatonin was considered by some authors as a "wonder drug." The present paper tries to summarize the relationship between melatonin and aging in three points. 1. Decline of melatonin production during aging. 2. The role of the pineal gland in the regulation of the ovarian cycle in aged females. 3. The antioxydant effect of melatonin and aging. The age-related decline of pineal melatonin production is due to the degenerative changes of the neural structures (serotonergic and noradrenerg

Many mammalian species from temperate latitudes exhibit seasonal variations in breeding activity which are controlled by the annual photoperiodic cycle. Photoperiodic information is conveyed through several neural relays from the retina to the pineal gland where the light signal is translated into a daily cycle of melatonin secretion: high at night, low in the day. The length of the nocturnal secretion of melatonin reflects the duration of the night and it regulates the pulsatile secretion of gonadotropin-releasing hormone (GnRH) from the hypothalamus. Changes in GnRH release induce correspond
