To analyze structural features of ω-Aga IVA, a gating modifier toxin from spider venom, we here investigated the NMR solution structure of ω-Aga IVA within DPC micelles. Under those conditions, the Cys-rich central region of ω-Aga IVA still retains the inhibitor Cys knot motif with three short antiparallel β-strands seen in water. However, 15N HSQC spectra of ω-Aga IVA within micelles revealed that there are radical changes to the toxin's C-terminal tail and several loops upon binding to micelles. The C-terminal tail of ω-Aga IVA appears to assume a β-turn like conformation within micelles, th

Phospholipase A2 (PLA2) is a peripheral membrane protein that can hydrolyze phospholipids to produce lysolipids and fatty acids. It has been found to play crucial roles in various cellular processes and is thought as a potential candidate for triggering drug release from liposomes for medical treatment. Here, we directly observed that PLA2 hydrolysis reaction can induce the formation of PLA2-binding domains at lipid bilayer interface and found that the formation was significantly influenced by the fluidity of the lipid bilayer. We prepared supported lipid bilayers (SLBs) with various molar rat

Aspirin and other non-steroidal anti-inflammatory drugs have a high affinity for phospholipid membranes, altering their structure and biophysical properties. Aspirin has been shown to partition into the lipid head groups, thereby increasing membrane fluidity. Cholesterol is another well known mediator of membrane fluidity, in turn increasing membrane stiffness. As well, cholesterol is believed to distribute unevenly within lipid membranes leading to the formation of lipid rafts or plaques. In many studies, aspirin has increased positive outcomes for patients with high cholesterol. We are inter

We demonstrate experimentally that anthrax toxin complexes rupture artificial lipid bilayer membranes when isolated from the blood of infected animals. When the solution pH is temporally acidified to mimic that process in endosomes, recombinant anthrax toxin forms an irreversibly bound complex, which also destabilizes membranes. The results suggest an alternative mechanism for the translocation of anthrax toxin into the cytoplasm.

We present a model system to demonstrate that the positioning of biomolecules (membrane proteins) in a nonnative, complex thin film environment can be regulated by the phase behavior of film components. Partial separation between an amphiphilic polymer and a lipid drives the protein to a fluid phase, mechanically more similar to a cellular bilayer.

The occurrence of oxidative stress has been proposed as a pathogenetic mechanism for melanocyte degeneration in vitiligo. In order to evaluate this possible correlation we focused on the lipid component of cell membranes. We observed in vitiligo melanocytes, through FACS methods, an increased median fluorescence intensity of rhodamine 123 and C11-BODIPY581/591 indicating a spontaneous higher production of reactive oxygen species (ROS) and membrane lipoperoxidation, associated with an altered pattern of cardiolipin (CL) distribution, defined on the basis of the fluorescence pattern after staini

Voltage-activated ion channels are essential for electrical signaling, yet the mechanism of voltage sensing remains under intense investigation. The voltage-sensor paddle is a crucial structural motif in voltage-activated potassium (K(v)) channels that has been proposed to move at the protein-lipid interface in response to changes in membrane voltage. Here we explore whether tarantula toxins like hanatoxin and SGTx1 inhibit K(v) channels by interacting with paddle motifs within the membrane. We find that these toxins can partition into membranes under physiologically relevant conditions, but t

Senile plaques in the cerebral parenchyma are a pathognomonic feature of Alzheimer's disease (AD) and are mainly composed of aggregated fibrillar amyloid beta (Abeta) proteins. The plaques are associated with neuronal degeneration, lipid membrane abnormalities, and chemical evidence of oxidative stress. The view that Abeta proteins cause these pathological changes has been challenged by suggestions that they have a protective function or that they are merely byproducts of the pathological process. This investigation was conducted to determine whether Abeta proteins promote or inhibit oxidative

Investigation was performed on 54 patients with angina pectoris. The patients were divided into three groups. Group 1 included patients with angina pectoris of functional classes I-II, group 2 consisted of the patients with angina pectoris classes III-IV, and group 3--unstable angina pectoris. All the patients underwent investigation of content and ratio of some fraction of the phospholipids and cholesterol metabolites in the thrombocyte membranes. Results of investigation showed damage of phospholipid metabolism of the thrombocyte membranes. This is manifested by increased in content of lysop

Thermal stress in living cells produces multiple changes that ultimately affect membrane structure and function. We report that two members of the family of small heat-shock proteins (sHsp) (alpha-crystallin and Synechocystis HSP17) have stabilizing effects on model membranes formed of synthetic and cyanobacterial lipids. In anionic membranes of dimyristoylphosphatidylglycerol and dimyristoylphosphatidylserine, both HSP17 and alpha-crystallin strongly stabilize the liquid-crystalline state. Evidence from infrared spectroscopy indicates that lipid/sHsp interactions are mediated by the polar hea

Considerable changes in the lipid phase of erythrocyte membranes were found in patients with paranoid schizophrenia. During exacerbation and remission of the disease the content of phosphatidylethanolamine decreases, while the amount of lysophosphatidylcholine decreases. These changes are accompanied by an increase in erythrocyte membrane microviscosity. Structural modification of the lipid phase in erythrocyte membranes in patients with schizophrenia is associated with intensification of lipid peroxidation.

Coincidently with the increase of lipid concentration in the culture medium, there was increase of lipid peroxide (LPO) and decrease of membrane fluidity (MF) in the smooth muscle cell (SMC). Animal experiment also showed that hyperlipidemia might damage SMC of the arterial wall and produce a similar result as described. Actually, there was a significant negative correlation between high LPO content and low MF. EM examination of SMC revealed a defeatured surface with many micropinocytic vesicles and fat droplets in the cytoplasm, and additionally atherosclerosis (AS) of various degree was obse

The influence of dietary copper on lipid composition, phospholipid fatty acid and protein profiles and fluidity of the erythrocyte membranes of rats is reported. In general Cu deficiency in rats induced some changes in the phospholipid-fatty acid profile of erythrocyte membranes when compared with Cu-adequate animals. Stearic (18:0) and docosadienoic (22:2n-3) acids contents, for example, were significantly increased (P < 0.001) while oleic (18:1n-9) and linolenic (18:3n-3) acid contents were significantly depressed (P < 0.001) as a result of Cu deficiency. Moreover the cholesterol:phospholipi

The influence of membrane potential on alpha-latrotoxin insertion into bilayer lipid membranes (BLM) has been investigated. It was found that positive potentials cis to toxin application stimulated the formation of channels in the bilayer. A two-step model of latrotoxin/membrane interaction is put forward to explain these data. In the first step, latrotoxin irreversibly binds to the bilayer without forming conductive structures. The second step of the process represents rapid insertion of the protein molecule into the bilayer with the formation of the conducting channel. We imagine the driving

The electrical capacitance of lipid membranes may increase by up to 50% on exposure to ionizing radiation. This is the consequence of lipid peroxidation induced by primary or secondary radicals of water radiolysis in the presence of oxygen. The polar products of this process give rise to an increase of the dielectric constant of the membrane. This in turn leads to the increase of membrane capacitance observed. An important consequence of this phenomenon is the reduction of the inner energy barrier between the two membrane/water interfaces influencing the movement of charged particles across th

Lipid peroxidation disrupts membrane integrity and causes structural and functional alterations in ischemic tissues. Taurine and ketamine are putative ischemic protectants that affect Ca2+ influx. Here we report the influence of these compounds on lipid peroxidation in subcellular fractions, isolated cells and intact tissue from bovine retinas. P2 membrane fractions and isolated cells were exposed to the lipid peroxidation inducers cadmium chloride (200 microM) or L-ascorbic acid (1 mM) in the presence of 0-50 mM taurine, 0-10 mM ketamine, 1 mM kynurenic acid or 1 mM dextromethorphan. Certain ultraviolet radiation-induced effects in skin may result from primary photochemical alterations in cell membranes. We have studied isolated erythrocyte membranes in order to determine the UV-fluence and wavelength dependence for protein alterations and lipid oxidation. Protein crosslinking was detected as high molecular weight protein (greater than 200,000 DA) on polyacrylamide/agarose gel electrophoresis. Spectrin decreased more rapidly than the other membrane proteins upon exposure to lambda = 250-380 nm radiation. Nitrogen-purging inhibited the UV-induced decrease in spectrin by 60%

Membrane fluidity and lipid composition of rat testis in experimental bilateral cryptorchidism were studied. Membrane fluidity measured by fluorescence anisotropy of diphenylhexatriene decreased on the 3rd day of intraabdominal fixation, at an early stage of cryptorchidism. Lipid composition of testicular membrane was changed drastically by the treatment, and time-dependent changes were observed during the course of the experiment. Cholesterol content increased significantly, but phospholipid content was not affected by the treatment. The ratio of cholesterol to phospholipids increased signifi

Asymmetric ion channels are formed in a bimolecular lipid membrane by beta-latrotoxin (LT) introduced to one (cis) side of the membrane. LT-specific antibodies added to the opposite (trans) side of the membrane block the current through the LT channels when a negative potential is applied to the cis side, no blockade is observed at positive potentials. LT-specific antibodies do not block the channel current when added to the cis compartment after removal of LT. LT-unspecific immunoglobulins have no influence on LT channel conductance.
