Platelet-derived microparticles provoke chronic lymphocytic leukemia malignancy through metabolic reprogramming.

BACKGROUND: It is well established that inflammation and platelets promote multiple processes of cancer malignancy. Recently, platelets have received attention for their role in carcinogenesis through the production of microvesicles or platelet-derived microparticles (PMPs), which transfer their biological content to cancer cells. We have previously characterized a new subpopulation of these microparticles (termed mito-microparticles), which package functional mitochondria. The potential of mitochondria transfer to cancer cells is particularly impactful as many aspects of mitochondrial biology

Metabolic reprogramming of Pseudomonas aeruginosa by phage-based quorum sensing modulation.

The Pseudomonas quinolone signal (PQS) is a multifunctional quorum sensing molecule of key importance to P. aeruginosa. Here, we report that the lytic Pseudomonas bacterial virus LUZ19 targets this population density-dependent signaling system by expressing quorum sensing targeting protein (Qst) early during infection. We demonstrate that Qst interacts with PqsD, a key host quinolone signal biosynthesis pathway enzyme, resulting in decreased levels of PQS and its precursor 2-heptyl-4(1H)-quinolone. The lack of a functional PqsD enzyme impairs LUZ19 infection but is restored by external supplem

Primary Aldosteronism: Metabolic Reprogramming and the Pathogenesis of Aldosterone-Producing Adenomas.

Aldosterone-producing adenomas (APAs) are characterized by aldosterone hypersecretion and deregulated adrenocortical cell growth. Increased energy consumption required to maintain cellular tumorigenic properties triggers metabolic alterations that shape the tumor microenvironment to acquire necessary nutrients, yet our knowledge of this adaptation in APAs is limited. Here, we investigated adrenocortical cell-intrinsic metabolism and the tumor immune microenvironment of APAs and their potential roles in mediating aldosterone production and growth of adrenocortical cells. Using multiple advanced

Muscle ERRγ mitigates Duchenne muscular dystrophy via metabolic and angiogenic reprogramming.

Treatment of Duchenne muscular dystrophy (DMD) by replacing mutant dystrophin or restoring dystrophin-associated glycoprotein complex (DAG) has been clinically challenging. Instead, identifying and targeting muscle pathways deregulated in DMD will provide new therapeutic avenues. We report that the expression of nuclear receptor estrogen-related receptor-γ (ERRγ), and its metabolic and angiogenic targets are down-regulated (50-85%) in skeletal muscles of mdx mice (DMD model) vs. wild-type mice. Corelatively, oxidative myofibers, muscle vasculature, and exercise tolerance (33%) are decreased in

Developmental origin of reproductive and metabolic dysfunctions: androgenic versus estrogenic reprogramming.

Polycystic ovary syndrome (PCOS) is one of the most common fertility disorders, affecting several million women worldwide. Women with PCOS manifest neuroendocrine, ovarian, and metabolic defects. A large number of animal models have evolved to understand the etiology of PCOS. These models provide support for the contributing role of excess steroids during development in programming the PCOS phenotype. However, considerable phenotypic variability is evident across animal models, depending on the quality of the steroid administered and the perinatal time of treatment relative to the developmenta

Decoding oxidative stress-driven metabolic reprogramming in skeletal degenerative diseases: Natural phytochemicals as precision metabolic modulators.

Skeletal degenerative diseases are prevalent chronic disorders associated with aging. Oxidative stress promotes metabolic reprogramming in bone, cartilage, and intervertebral disc cells; however, the underlying mechanisms remain incompletely understood. Plant-derived phytochemicals may suppress oxidative stress and regulate metabolic reprogramming, thereby offering potential therapeutic benefits. In this review, we summarize oxidative stress-induced alterations in mitochondrial function and glucose, lipid, and amino acid metabolism across major skeletal degenerative diseases and discuss the th

Pregnancy-induced metabolic reprogramming in skeletal muscle: a multi-omics interrogation of transcriptional and metabolic adaptations.

Pregnancy induces profound physiological adaptations to meet the dynamic nutritional demands of fetal development, including a deliberate reduction in maternal insulin sensitivity to ensure fetal glucose availability. However, excessive insulin resistance may precipitate gestational diabetes mellitus (GDM), increasing the risk of both obstetric complications and long-term metabolic disorders in mothers and offspring. Although the role of adipose tissue in pregnancy-associated metabolic adaptation has been extensively studied, the contribution of skeletal muscle remains poorly understood. Here,

Lipid metabolic reprogramming in Cancer radiation resistance: Mechanisms and therapeutic implications.

Currently, cancer treatment mainly includes surgery, radiotherapy, chemotherapy, immunotherapy, and targeted therapy, wherein radiotherapy is one of the core modalities. However, its efficacy is often limited by radiotherapy resistance in the tumour cells. In recent years, studies have found that tumour cells undergo lipid metabolic reprogramming to enhance lipid synthesis, uptake, and storage, thereby adapting to the energy and structural adjustment demands imposed by radiotherapy, gaining survival advantage and developing radiation resistance. This review mainly focused on the role of lipid

Macrophage Polarization and Metabolic Reprogramming in Endometriosis: Mechanisms and Therapeutic Implications.

This review synthesizes evidence on macrophage roles, polarization (M1/M2), and immunometabolic/signaling pathways in endometriosis, outlining therapeutic implications. We searched PubMed, Cochrane Library, ClinicalTrials.gov, Scopus, Embase, and Google Scholar. Available evidence demonstrates increased macrophage accumulation and substantial heterogeneity across the eutopic endometrium, peritoneal fluid, and established endometriotic lesions. Rather than undergoing a uniform shift from an M1 to an M2 phenotype, endometriosis-associated macrophages acquire overlapping inflammatory, immunoregul

Metabolic reprogramming in differentiated thyroid cancer progression: A tumor metabolomics analysis.

BACKGROUND: Differentiated thyroid cancers exhibit biologic and clinical heterogeneity not fully explained by conventional staging. Metabolic reprogramming is a hallmark of cancer, and its contribution to differentiated thyroid cancer progression is an ongoing area of study. Metabolomics-the comprehensive analysis of metabolites within biologic systems-may identify stage-specific metabolic signatures. METHODS: Fifty-five differentiated thyroid cancer specimens underwent untargeted metabolomic profiling using liquid chromatography-mass spectrometry. Stage was assigned per American Joint Commit

SASH1 Inhibits Gastric Cancer Progression by Regulating YAP1-Mediated Metabolic Reprogramming.

Sterile alpha motif and SH3 domain-containing 1 (SASH1), a tumor suppressor, is involved in multiple biological processes in cancer cells. However, the potential role and mechanism of SASH1 in regulating glucose metabolism of gastric cancer (GC) remains unknown. SASH1 and Yes-associated protein 1 (YAP1) expressions in GC tissues were investigated based on The Cancer Genome Atlas (TCGA) database. Reverse transcription-quantitative polymerase chain reaction (RT-qPCR) and Western blot were used to analyze the expressions of the SASH1 and YAP1 in GC tissues and cell lines. Metabolic reprogramming of microglia: Effects on development, disease, and therapeutic potential.

Microglia, the immune sentinels of the central nervous system, play vital roles in maintaining neural homeostasis and mediating responses to injury and disease. Their functions, including synaptic pruning to neuroinflammation, are tightly linked to their metabolic state. Emerging evidence suggests that metabolic reprogramming is a key driver of microglial activation, functional transitions, and interactions with neurons and other glial cells. This review summarizes current findings on the developmental origins, region-specific adaptations, and metabolic plasticity of microglia. We review lipid

[Advances in Lactate Metabolic Reprogramming in Non-small Cell Lung Cancer].

Non-small cell lung cancer (NSCLC) is characterized by high incidence and mortality, with a low five-year survival rate. Lactate metabolism plays a central role in the metabolic reprogramming of NSCLC. Beyond serving as the end-product of glycolysis, lactate accumulates in the tumor microenvironment (TME), contributing to acidification, and can also enter the tricarboxylic acid cycle to participate in energy metabolism. Moreover, the G protein-coupled receptor 81 (GPR81)/phosphoinositide 3-kinase (PI3K)/mammalian target of rapamycin (mTOR) signaling axis induces the expression of immune checkp

Lipid metabolic reprogramming of CD8+ T cells in the tumor microenvironment.

Metabolic reprogramming within the tumor microenvironment is a critical driver of CD8+ T cell dysfunction that limits the efficacy of cancer immunotherapy. While glucose and amino acid deprivation are well-characterized, lipid metabolic rewiring has emerged as a fundamental determinant of T cell fate. This review systematically examines the mechanisms by which the tumor microenvironment disrupts CD8+ T cell lipid metabolism to promote functional exhaustion and ferroptosis. We first discuss how local stressors such as hypoxia and acidosis alongside systemic host factors including obesity and hy

Therapeutic strategies of metabolic reprogramming in non-small cell lung carcinoma.

BACKGROUND: Non-small cell lung carcinoma (NSCLC) remains a leading cause of cancer-related mortality worldwide, with existing therapies frequently hindered by drug resistance and immunosuppression. Metabolic reprogramming (glycolysis, lipid metabolism, and amino acid metabolism) has emerged as a core hallmark driving NSCLC progression, tumor microenvironment (TME) remodeling, and treatment failure, transcending the classical Warburg effect to involve intricate cross-talk between cancer cells and stromal components. DISCUSSION: This review systematically synthesizes the latest insights into t

Reprogramming immunity at the metabolic-epidermal interface in obesity-associated psoriasis.

Obesity and psoriasis are chronic inflammatory disorders, now recognized to be interconnected, in which metabolic overload drives immune dysregulation and therapeutic resistance. Excess adiposity converts adipose tissue into an inflammatory organ that releases adipokines and cytokine-like mediators, reprogramming keratinocytes and immune cells to sustain cytokine-driven inflammatory circuits in the skin. Excess nutrients and lipotoxic stress impair mitochondrial function, enhance glycolysis, and induce epigenetic remodeling in myeloid and epithelial lineages, generating metabolic memory that p
