Optimised untargeted metabolomics workflow for human urinary extracellular vesicles.

Extracellular vesicles (EVs) have been a key focus in biomarker discovery, with urinary EVs (uEVs), primarily derived from cells of the urogenital tract, providing valuable insights into kidney and urinary tract health and disease. However, progress in uEV-based metabolomics remains limited by variability in EV isolation and extraction approaches. Here, we systematically evaluated and optimised experimental conditions for untargeted metabolite profiling of human uEVs. We compared three different EV isolation methods, namely precipitation, size-exclusion chromatography, and pH-adjustment with r

Metabolomics-Based Selection of Biostimulant and Biocontrol Microbial Consortia.

Microbial biostimulants and microbial plant protection products overlap in biological function, creating both R&D opportunities and regulatory challenges. In particular, multi-strain bacterial consortia may simultaneously affect nutrient mobilisation and abiotic stress tolerance, induce resistance, and demonstrate direct antagonism against phytopathogens. This multifunctionality complicates early product development because strain identity alone is sometimes insufficient in predicting product function, efficacy, or the most appropriate regulatory and claims strategy. Here, we used non-targeted

Benchmarking untargeted metabolomics data quality with allopurinol-induced perturbations.

INTRODUCTION: We present a simple test to assess whether a metabolomics dataset is fit-for-purpose. Current qualitycontrol approaches do not directly evaluate the ability to recover biologically meaningful perturbations. OBJECTIVES: To evaluate whether known drug-induced metabolic perturbations can serve as internal benchmarks fordataset quality. METHODS: In a study (the TROMBOLOME study, unrelated to allopurinol therapy), 1,000 serum samples were analyzedwith one targeted and two untargeted metabo lomics panels. Samples were classified as allopurinol-positive (N=19)using detection of allopu

Human Extracellular Vesicles, Metabolites, and Metabolomics: A Comprehensive Review.

Metabolomics is a powerful systems-level approach and has the potential to serve as an important part for understanding the biochemical pathways and metabolic phenotypes in physiological and pathological states. Extracellular vesicles (EVs), which include apoptotic bodies, microvesicles, and exosomes, have emerged alongside metabolomics as active mediators of intercellular communication, transporting diverse cargo that mirrors the cellular origin and metabolic status of their parent cell. EVs are enriched with lipids, proteins, nucleic acids, and biologically active metabolites involve in sign

Application of Metabolomics in Defence Responses of Brassica Crops.

Brassica crops, encompassing globally important vegetables and oilseeds, face severe threats from diverse biotic and abiotic stresses. Plant secondary metabolites constitute the chemical foundation of defence, and metabolomics has emerged as an effective systems biology tool for comprehensively dissecting stress-induced metabolic changes. Recent progress in applying metabolomics to elucidate defence mechanisms in Brassica crops is systematically synthesised here. Major stresses confronting Brassica crop production and the metabolic basis of plant defence are first outlined. Current analytical

Urinary metabolomics profile in children with ureteropelvic junction obstruction.

INTRODUCTION: Fetal hydronephrosis (HN) is detected in about 0.25-1% of fetuses, where ureteropelvic junction obstruction (UPJO) is the most commonly found congenital urinary tract anomaly, remaining the leading cause of kidney failure in infants and children. Since it is a spectral disease, UPJO represents a particularly challenging management. Severe UPJO must be treated surgically to avoid impairment of renal function, but children with non-obstructive hydronephrosis can be treated conservatively. There has been controversy regarding the indication for surgical intervention in asymptomatic

Metabolomics: Potential non-protein biomarker candidates of Parkinson's disease.

Despite the growing number of patients with neurodegenerative diseases, especially Parkinson's disease, and the associated increasing pressure on global healthcare systems, there is still a lack of reliable laboratory methods that allow for the rapid and highly accurate diagnosis of these diseases at a pre-symptomatic or early stage, i.e., before irreversible pathomorphological and functional changes occur. Furthermore, current treatment strategies remain limited to the management of symptoms and fail to address the underlying causes of Parkinson's disease. This diagnostic and therapeutic gap

Tandem metabolic reaction-based sensors unlock in vivo metabolomics.

Mimicking metabolic pathways on electrodes enables in vivo metabolite monitoring for decoding metabolism. Conventional in vivo sensors cannot accommodate underlying complex reactions involving multiple enzymes and cofactors, addressing only a fraction of enzymatic reactions for few metabolites. We devised a single-wall-carbon-nanotube-electrode architecture supporting tandem metabolic pathway-like reactions linkable to oxidoreductase-based electrochemical analysis, making a vast majority of metabolites detectable in vivo. This architecture robustly integrates cofactors, self-mediates reactions

Extracellular Vesicle Metabolomics Holds Promise for Adult Axon Regeneration.

Extracellular vesicles (EVs) are bilayer lipid membrane particles that are released by every cell type. These secretions are further classified as exosomes, ectosomes, and microvesicles. They contain biomolecules (RNAs, proteins, metabolites, and lipids) with the ability to modulate various biological processes and have been shown to play a role in intercellular communication and cellular rejuvenation. Various studies suggest exosomes and/or microvesicles as a potential platform for drug delivery. EVs may deliver lipids and nucleotides directly to an injury site in an axon, promoting growth co
