PINK1/Parkin-Mediated Mitophagy Partially Protects against Inorganic Arsenic-Induced Hepatic Macrophage Polarization in Acute Arsenic-Exposed Mice.

Inorganic arsenic is a well-known environmental toxicant and carcinogen, and there is overwhelming evidence for an association between this metalloid poisoning and hepatic diseases. However, the biological mechanism involved is not well characterized. In the present study, we probed how inorganic arsenic modulates the hepatic polarization of macrophages, as well as roles of PTEN-induced kinase 1 (PINK1)/Parkin-mediated mitophagy participates in regulating the metalloid-mediated macrophage polarization. Our results indicate that acute arsenic exposure induced macrophage polarization with up-reg

Heavy metal poisoning: management of intoxication and antidotes.

Of the known elements, nearly 80% are either metals or metalloids. The highly reactive nature of most metals result in their forming complexes with other compounds such oxygen, sulfide and chloride. Although this reactivity is the primary means by which they are toxic, many metals, in trace amounts, are vital to normal physiological processes; examples include iron in oxygen transport, manganese and selenium in antioxidant defense and zinc in metabolism. With these essential metals toxicity occurs when concentrations are either too low or too high. For some metals there are no physiological co

Proposal of novel Predicted No Effect Concentrations (PNEC) for metals in freshwater using Species Sensitivity Distribution for different taxonomic groups.

Water pollution by metals and metalloids promotes toxic effects to aquatic biota especially in mining regions. Environmental legislation applied to protect aquatic life from the toxicity of metals relies on the definition of protective values (PVs) for each compound. Among methods used to define PVs, Species Sensitivity Distribution (SSD) curves enable the derivation of the Predicted No Effect concentration (PNEC). In this context, this is one of the first studies to propose the construction of acute and chronic split SSD curves built separately for three groups of freshwater organisms (algae,

Trachemys dorbigni as a metal(loid) bioindicator: a study in rural and urban areas.

Metals and metalloids are persistent environmental pollutants with the potential for bioaccumulation, posing significant health risks, including genotoxicity. These contaminants are prevalent in industrial and agricultural runoff. This study utilizes Trachemys dorbigni, an aquatic reptile, as a bioindicator to assess environmental contamination by metals and metalloids in both rural and urban settings in Pelotas, Rio Grande do Sul, Brazil. We captured specimens using pit-type traps with barriers (fyke nets), dividing them into two groups: 15 from a rural area and 15 from an urban area. Each an

Oxidative activation of the human carcinogen chromate by arsenite: a model for synergistic metal activation leading to oxidative DNA damage.

Human exposure to toxic metals and metalloids in the environment seldom occurs from a single pure compound. Most environmental exposure profiles are heterogeneous with co-exposure occurring coincident with multiple toxic metal species. This co-exposure to metals and metalloids in complex mixtures can result in a synergistic, additive or even depletive toxic response. The complexity of interactions presented by metal mixtures presents a need for convenient and sensitive methods to determine potential toxic responses from such co-exposure. We have studied the reaction between the two commonly as

Retrospective study of methylmercury and other metal(loid)s in Madagascar unpolished rice (Oryza sativa L.).

The rice ingestion rate in Madagascar is among the highest globally; however studies concerning metal(loid) concentrations in Madagascar rice are lacking. For Madagascar unpolished rice (n = 51 landraces), levels of toxic elements (e.g., total mercury, methylmercury, arsenic and cadmium) as well as essential micronutrients (e.g., zinc and selenium) were uniformly low, indicating potentially both positive and negative health effects. Aside from manganese (Wilcoxon rank sum, p < 0.01), no significant differences in concentrations for all trace elements were observed between rice with red bran (n

[Heavy metals in the enviroment: Guillain-Barre like syndrome].

Guillain-Barré syndrome is an entity of diverse etiology, characterized by acute, symmetric, ascending and progressive muscle weakness, being one of the most frequent acquired polyneuropathies in childhood. Neuropathies produced by heavy metals, mercury and lead, and metalloids, such as arsenic, organophosphorus pesticides and carbon tetrachloride, should be considered among the differential diagnoses. We present a 14-year-old patient with a presumptive diagnosis of Guillain-Barré syndrome without response to conventional treatment with gamma globulin. Considering other etiologies, heavy metal

Production of volatile derivatives of metal(loid)s by microflora involved in anaerobic digestion of sewage sludge.

Gases released from anaerobic wastewater treatment facilities contain considerable amounts of volatile methyl and hydride derivatives of metals and metalloids, such as arsine (AsH(3)), monomethylarsine, dimethylarsine, trimethylarsine, trimethylbismuth (TMBi), elemental mercury (Hg(0)), trimethylstibine, dimethyltellurium, and tetramethyltin. Most of these compounds could be shown to be produced by pure cultures of microorganisms which are representatives of the anaerobic sewage sludge microflora, i.e., methanogenic archaea (Methanobacterium formicicum, Methanosarcina barkeri, Methanobacterium

A comparison of the rates of methylation of mercury(II) species in aquatic media by various organotin and organosilicon moieties.

Metals can be methylated in environmental aqueous media by a variety of organotin and organosilicon compounds. Main group metals and metalloids were surveyed for the identification of species that can either donate or accept methyl groups. The methylation of mercury(II) by trimethyltin cation was found to be a bimolecular reaction, the reaction rate of which decreased with increasing chloride concentration. Kinetic investigations using NMR techniques showed that the most important pairs of reactants were (CH3)3Sn+ + HgCl2, (CH3)3SnCl + HgCl2, and (CH3)3SnCl + HgCl-3. Sodium 2,2,3,3-tetradeuter

The unicellular fungal tool RhoTox for risk assessments in groundwater systems.

The recent inclusion of yeasts in environmental monitoring recognizes their ecological significance and sensitivity to toxicants. Here we present a robust and simple two-step toxicity assay and demonstrate the sensitivity of an ubiquitous groundwater yeast, Rhodotorula minuta, to a range of metals and metalloids. The test species was sensitive to copper with a 24h EC50 of 35µg Cu/L, followed in order of decreasing sensitivity by zinc, chromium (VI) and arsenic (EC50 4.40mg As (III)/L). The strain demonstrated an unexpected tolerance to chromium (VI), having an EC50 value (3.45mg Cr (VI)/L) sim

Ecotoxicity assessment of primary producers in metal mining areas: biological indicators for ecosystem restoration.

Abandoned mines represent a considerable risk to ecosystems surrounding former exploitation sites. In metal mining, the exposure of waste significantly increases the mobility and bioavailability of potentially toxic elements (PTEs), affecting adjacent soils and organisms. Assessing the toxicity of mining waste involves challenges related to selecting appropriate bioassays and those recommended by current environmental regulations. Considering the expected increase in metal and metalloid extraction to supply critical raw materials, improving our understanding of the advantages and limitations o

Chemistry, Pharmacology, and Toxicology of Monoisoamyl Dimercaptosuccinic Acid: A Chelating Agent for Chronic Metal Poisoning.

Arsenic, a metalloid, is known to cause deleterious effects in various body organs, particularly the liver, urinary bladder, and brain, and these effects are primarily mediated through oxidative stress. Chelation therapy has been considered one of the promising medical treatments for arsenic poisoning. Meso 2,3- dimercaptosuccinic acid (DMSA) has been recognized as one of the most effective chelating drugs to treat arsenic poisoning. However, the drug is compromised with a number of shortcomings, including the inability to treat chronic arsenic poisoning due to its extracellular distribution.

Synthesis of bis-spiro piperidines using nano γ-alumina supported Sb(v) under ultrasonic irradiation at room temperature conditions.

Group VA metalloid ion Lewis acids, Sb(v) was identified as a highly potent catalyst for the one-pot three-component synthesis of bis-spiro piperidine derivatives. The reaction was performed using amines, formaldehyde, and dimedone under ultrasonic irradiation at room temperature. The strong acidic property of the nano γ-alumina supported antimony(v) chloride plays a key role in accelerating the rate of the reaction and initiates the reaction smoothly. The heterogeneous nanocatalyst was fully characterized by FT-IR spectroscopy, XRD, EDS, TGA, FESEM, TEM, and BET techniques. Also, the structur

It's not all about PFAS: Metal(loid)s are associated with physiological and metabolic changes in a native frog from conservation wetlands.

Urban wetlands provide essential habitats for wildlife but face increasing contamination from anthropogenic sources, including but not limited to per- and polyfluoroalkyl substances (PFAS) and metal(loid)s. Accumulation of these contaminants are known to impair organism biological functions. This study quantified PFAS and metal(loid) concentrations in liver tissue of motorbike frogs (Ranoidea moorei) along a contamination gradient of conservation significant urban wetlands in Perth, Western Australia, and assessed relationships of liver concentrations with whole-organism health metrics and met

Circulating metals, leukocyte microRNAs and microRNA networks: A profiling and functional analysis in Chinese adults.

BACKGROUND: Metals in the human body represent both environmental exposure and nutritional status. Little is known about the miRNA signature in relation to circulating metals in humans. OBJECTIVES: To characterize metal-associated miRNAs in leukocytes, individually and collectively as networks. METHODS: In a panel of 160 Chinese adults, we measured 23 metals/metalloids in plasma, and sequenced miRNAs and mRNAs in leukocytes. We used linear regression to model the associations between ln-transformed metal concentrations and normalized miRNA levels adjusting for potential confounders. We infer

[ELEMENTAL STATUS OF PATIENTS WITH VARIOUS FORMS OF VITILIGO].

Vitiligo is a multifactorial disease in which, in each specific case of its manifestation, different mechanisms of its pathogenesis and different levels of melanin formation in the skin can be involved. Skin is one of the most metabolically active organs. Carrying out a number of vital functions (barrier, protective, respiratory, excretory, metabolic, immune, etc.), it needs microelementss. Of the 92 naturally occurring chemical elements, 81 are found in the human body. Lack of the vital elements, leads to the emergence of diseases, which are based on deficiency, excess or imbalance of micro-

Metal(loid) contamination in seafood products.

Seafood products are important sources of proteins, polyunsaturated lipids and phospholipids, and also of numerous micronutrients (vitamins and minerals). However, they may also present chemical contaminants that can constitute a health risk and that must be considered when evaluating the risk/benefit associated with consumption of this group of foods. Toxic metals and metalloids in seafood, such as mercury (Hg), cadmium (Cd), arsenic (As), and lead (Pb), are subjected to legislative control in order to provide the consumer with safe seafood. This review provides an exhaustive survey of the oc

Human risk assessment of heavy metals: principles and applications.

Humans are exposed to a number of "heavy metals" such as cadmium, mercury and its organic form methylmercury, uranium, lead, and other metals as wel as metalloids, such as arsenic, in the environment, workplace, food, and water supply. Exposure to these metals may result in adverse health effects, and national and international health agencies have methodologies to set health-based guidance values with the aim to protect the human population. This chapter introduces the general principles of chemical risk assessment, the common four steps of chemical risk assessment: hazard identification, haz

Targeting selenium metabolism and selenoproteins: novel avenues for drug discovery.

Selenoproteins play a wide range of roles in metabolism and oxidative stress defense and are produced by organisms in all three domains of life. Recent evidence has been presented that metal based cancer drugs target the selenol nucleophile of the active site selenocysteine in thioredoxin reductase isoenzymes. Other metals and metalloids, such as tin, arsenic and gold, have also recently been shown to form stable complexes with hydrogen selenide, a required precursor for the synthesis of selenoproteins in all biological organisms. Moreover these metal based compounds have been shown to inhibit

Regiospecific Allenylation of Ketones with Propargyltrichlorosilane.

The inherent difficulty in eliciting regio-control over propargyl-allenyl isomerization of the corresponding metal/metalloid species has limited their utility as nucleophiles in organic synthesis. Consequently, synthetically versatile α-allenyl tertiary alcohols are generally synthesized in two steps from ketones (e.g., Crabbé homologation of propargylic alcohols) rather than by the direct addition of allenylation reagents to ketones. We have developed a regiospecific allenylation of ketones with isomerically pure propargyltrichlorosilane as an operationally simple, general method to prepare r
