Portal Vein Pressure Estimation and Portal Hypertension Discrimination Based on Subharmonic Scattering of Ultrasound Contrast Agent Microbubbles.

OBJECTIVE: The acquisition of real-time portal vein pressure (PVP) is important for portal hypertension (PH) discrimination to monitor disease progress and select treatment options. To date, the PVP evaluation approaches are either invasive or noninvasive but with less stability and sensitivity. METHODS: We customized an open ultrasound scanner to explore in vitro and in vivo the ultrasound contrast agent SonoVue microbubbles' subharmonic characteristics with acoustic pressure and local ambient pressure, and obtained promising results of PVP measurements in canine models with induced PH by li

Lymphography using microbubbles and contrast-enhanced ultrasound (CEUS) before lymphatic supermicrosurgery for severe lymphorrhea of lower extremity.

Lymphaticovenous anastomosis (LVA) is an effective surgical treatment for lymphorrhea. However, the traditional indocyanine green (ICG) fluorescent lymphography mapping for lymphatic vessels has limitations, it can only depict the initial capillary lymphatic network localized in the dermis of the skin, which cannot visualize lymphatics deeper than 1.5 cm. A new mapping technique, microbubbles and contrast-enhanced ultrasound (CEUS) can address the problem. In one case of lymphocutaneous fistula, for the first time, we used microbubbles and CEUS technique for preoperative localization for LVAs.

Ultrasound-Induced Destruction of Nitric Oxide-Loaded Microbubbles in the Treatment of Thrombus and Ischemia-Reperfusion Injury.

Objectives: Early recanalization of large vessels in thromboembolism, such as myocardial infarction and ischemic stroke, is associated with improved clinical outcomes. Nitric oxide (NO), a biological gas signaling molecule, has been proven to protect against ischemia-reperfusion injury (IRI). However, the underlying mechanisms remain to be explored. This study investigated whether NO could mitigate IRI and the role of NO during acoustic cavitation. Methods: In vivo, thrombi in the iliac artery of rats were induced by 5% FeCl3. NO-loaded microbubbles (NO-MBs) and ultrasound (US) were used to tr

Ultrasound Molecular Imaging of Renal Cell Carcinoma: VEGFR targeted therapy monitored with VEGFR1 and FSHR targeted microbubbles.

Recent treatment developments for metastatic renal cell carcinoma offer combinations of immunotherapies or immunotherapy associated with tyrosine kinase inhibitors (TKI). There is currently no argument to choose one solution or another. Easy-to-use markers to assess longitudinal responses to TKI are necessary to determine when to switch to immunotherapies. These new markers will enable an earlier adaptation of therapeutic strategy in order to prevent tumor development, unnecessary toxicity and financial costs. This study evaluates the potential of ultrasound molecular imaging to track the resp

Effectiveness of a Layer-by-Layer Microbubbles-Based Delivery System for Applying Minoxidil to Enhance Hair Growth.

Minoxidil (Mx) is a conventional drug for treating androgenetic alopecia, preventing hair loss, and promoting hair growth. The solubility of Mx has been improved using chemical enhancement methods to increase its skin permeability over the long term. This study created a new ultrasound (US) contrast agent-albumin-shelled microbubbles (MBs) that absorb chitosan oligosaccharide lactate (COL) and Mx-and combined it with sonication by US energy in the water phase to enhance hair growth while shortening the treatment period. COL and Mx grafted with MBs (mean diameter of 1480 nm) were synthesized in

Preparation and characterization of luteinising-hormone releasing hormone nanoliposomal microbubbles specifically targeting ovarian cancer cells in vitro.

The aim of the present study was to prepare luteinizing-hormone releasing hormone (LHRH) nanoliposomal microbubbles specifically targeting ovarian cancer cells. The lyophilization/sonication method was used to prepare non-targeting nanoliposomal microbubbles (N-N-Mbs). Using the biotin-avidin bridge method, conjugated LHRH antibodies to N-N-Mbs generated LHRH nanoliposomal microbubbles (LHRH-N-Mbs) specifically targeting ovarian cancer cells. The morphology and physicochemical properties of the microbubbles was detected using an optical microscope and zeta detector. The binding affinity betwee

Mesenchymal stem cell transplantation enhancement in myocardial infarction rat model under ultrasound combined with nitric oxide microbubbles.

OBJECTIVE: This study evaluated the effects of ultrasound combined with the homemade nitric oxide (NO) micro-bubble destruction on the in vitro proliferation, apoptosis, and migration of mesenchymal stem cells (MSCs). Furthermore, we studied whether or not irradiation of the NO micro-bubble combined with bone-marrow derived MSC infusion had a better effect on treating myocardial infarction. The possible mechanism of MSC delivery into the infarcted myocardium was also investigated. METHODS: The murine bone marrow-derived MSCs were isolated, cultured, irradiated, and combined with different con

Microbubbles manufactured using a tissue homogeniser: DoE-guided optimisation for a narrower size distribution and higher yield.

Microbubbles are clinically approved as ultrasound contrast agents because they oscillate in response to ultrasound (cavitation) and are increasingly explored for therapeutic applications. Microbubble diameter governs dynamic behaviour under ultrasound; therefore, a narrow size distribution is essential for predictable performance and optimal responsiveness. However, producing phospholipid-coated microbubbles with narrow size distributions using simple methods remains challenging. This study evaluated a bead-type tissue homogeniser as an alternative to probe sonication for generating DSPC-PEG4

Radiation enhancement using focussed ultrasound-stimulated microbubbles for head and neck cancer: A phase 1 clinical trial.

BACKGROUND AND PURPOSE: Preclinical research demonstrated that the exposure of microbubbles (intravascular gas microspheres) to focussed ultrasound within the targeted tumour upregulates pro-apoptotic pathways and enhances radiation-induced tumour cell death. This study aimed to assess the safety and efficacy of magnetic resonance (MR)-guided focussed ultrasound-stimulated microbubbles (MRgFUS-MB) for head and neck cancers (HN). MATERIALS AND METHODS: This prospective phase 1 clinical trial included patients with newly diagnosed or recurrent HN cancer (except nasopharynx malignancies) for who

[Experimental study on high-frequency subharmonic scattering characteristics of ultrasound contrast agent microbubbles under low ambient pressure].

Correlation between nonlinear subharmonic scattering of ultrasound contrast agent microbubbles and ambient pressure is expected to be used for local brain tissue pressure monitoring. Although high-frequency ultrasound has achieved high-resolution imaging of intracranial microvessels, the research on high-frequency subharmonic scattering characteristics of microbubbles is insufficient at present, which restricts the research progress of estimating local brain tissue pressure based on high-frequency subharmonic scattering of microbubbles. Therefore, under the excitation of 10 MHz high-frequency

Ultrasound Participated Optomechanical Frequency Comb Generation and Tunability in High-Q Microbubble Cavities.

Optomechanical frequency combs (OMFCs) are generated in high-Q microbubble cavities when an optomechanical system is simultaneously driven by ultrasound and laser light. The combs originate from nonlinear interactions between optomechanical modes and ultrasonically driven mechanical oscillations. The comb spacing and intensity are manipulated by the frequency and amplitude of the ultrasound, as well as by the optical pump power and frequency detuning. The combination of the OMFC and the optical spring effect further enhances the temperature-sensing sensitivity.

Ultrasound-targeted microbubble destruction enhances RSL3-induced ferroptosis in anaplastic thyroid carcinoma.

Anaplastic thyroid carcinoma (ATC) is a highly aggressive malignancy with limited effective therapies. Ferroptosis, a form of programmed cell death, is closely associated with thyroid cancer prognosis and represents a potential therapeutic target. This study evaluated the antitumor effects of ultrasound-targeted microbubble (MB) destruction (UTMD)-mediated RAS-selective lethal 3 (RSL3) delivery to induce ferroptosis in ATC. MBs loaded with ferroptosis inducer RSL3 were synthesized and intravenously administered in a nude mouse ATC model, followed by localized ultrasound irradiation to enable t
