Implanted microelectrode arrays in reinnervated muscles allow separation of neural drives from transferred polyfunctional nerves.

Targeted muscle reinnervation surgery reroutes residual nerve signals into spare muscles, enabling the recovery of neural information through electromyography (EMG). However, EMG signals are often overlapping, making the interpretation of limb functions complicated. Regenerative peripheral nerve interfaces surgically partition the nerve into individual fascicles that reinnervate specific muscle grafts, isolating distinct neural sources for precise control and interpretation of EMG signals. Here we combine targeted muscle reinnervation surgery of polyvalent nerves with a high-density microelect

Composite additive manufacturing for suspended microelectrode arrays: Advancing oriented myocardial tissue culturing and electrophysiological sensing.

Microelectrode arrays (MEAs) have ushered in a new era of in vitro drug screening and cardiotoxicity evaluation. However, the morphological constraints of two-dimensional (2D) planar culture and the mechanical rigidity of conventional electrodes hinder the formation of myocardial tissues that closely resemble native physiological conditions and limit the accuracy of drug efficacy analysis based on electrophysiological signals. Here, we present a flexible MEA platform enabled by a composite additive manufacturing approach, with key steps including melt electrowriting of microfibers, electrostat

Low-intensity pulsed ultrasound stimulation (LIPUS) modulates microglial activation following intracortical microelectrode implantation.

Microglia are important players in surveillance and repair of the brain. Implanting an electrode into the cortex activates microglia, produces an inflammatory cascade, triggers the foreign body response, and opens the blood-brain barrier. These changes can impede intracortical brain-computer interfaces performance. Using two-photon imaging of implanted microelectrodes, we test the hypothesis that low-intensity pulsed ultrasound stimulation can reduce microglia-mediated neuroinflammation following the implantation of microelectrodes. In the first week of treatment, we found that low-intensity p

Revealing Low Amplitude Signals of Neuroendocrine Cells through Disordered Silicon Nanowires-Based Microelectrode Array.

Today, the key methodology to study in vitro or in vivo electrical activity in a population of electrogenic cells, under physiological or pathological conditions, is by using microelectrode array (MEA). While significant efforts have been devoted to develop nanostructured MEAs for improving the electrophysiological investigation in neurons and cardiomyocytes, data on the recording of the electrical activity from neuroendocrine cells with MEA technology are scarce owing to their weaker electrical signals. Disordered silicon nanowires (SiNWs) for developing a MEA that, combined with a customized

Microelectrode implants, inflammatory response and long-lasting effects on NADPH diaphorase neurons in the rat frontal cortex.

At present, one of the main therapeutic challenges comprises the development of technologies to improve the life quality of people suffering from different types of body paralysis, through the reestablishment of sensory and motor functions. In this regard, brain-machine interfaces (BMI) offer hope to effectively mitigate body paralysis through the control of paralyzed body parts by brain activity. Invasive BMI use chronic multielectrode implants to record neural activity directly from the brain tissue. While such invasive devices provide the highest amount of usable neural activity for BMI con

Multi-channel intraneural vagus nerve recordings with a novel high-density carbon fiber microelectrode array.

Autonomic nerves convey essential neural signals that regulate vital body functions. Recording clearly distinctive physiological neural signals from autonomic nerves will help develop new treatments for restoring regulatory functions. However, this is very challenging due to the small nature of autonomic nerves and the low-amplitude signals from their small axons. We developed a multi-channel, high-density, intraneural carbon fiber microelectrode array (CFMA) with ultra-small electrodes (8-9 µm in diameter, 150-250 µm in length) for recording physiological action potentials from small autonomi

A method for electrophysiological characterization of hamster retinal ganglion cells using a high-density CMOS microelectrode array.

Knowledge of neuronal cell types in the mammalian retina is important for the understanding of human retinal disease and the advancement of sight-restoring technology, such as retinal prosthetic devices. A somewhat less utilized animal model for retinal research is the hamster, which has a visual system that is characterized by an area centralis and a wide visual field with a broad binocular component. The hamster retina is optimally suited for recording on the microelectrode array (MEA), because it intrinsically lies flat on the MEA surface and yields robust, large-amplitude signals. However,

Formation of supported lipid bilayers on indium tin oxide for dynamically-patterned membrane-functionalized microelectrode arrays.

Supported lipid bilayers (SLBs) are ideal platforms for the study of membrane proteins and function. Assembly of functional SLBs in an array format would lead to a breakthrough in high-throughput screening of membrane-associated processes, e.g., drugs binding to transmembrane proteins. We report the formation of SLBs from the rupture of anionic vesicles in the presence of Ca(2+) ions on ITO-coated surfaces and characterise the assembly and SLB properties. Furthermore, the formation, manipulation and regeneration of SLBs adsorbed on ITO microelectrode array spots using an electric potential swi

Microelectrode-guided deep brain stimulation for Tourette syndrome: within-subject comparison of different stimulation sites.

BACKGROUND: As medical therapy for Tourette syndrome (TS) is ineffective in a small subset of patients, surgical interventions, including deep brain stimulation at various sites, have been developed in recent years. CASE DESCRIPTION: We present the case of a 40-year-old woman with TS whose severe tics had caused unilateral blindness. Despite trials of more than 40 medications, her symptoms improved significantly only after placement of bilateral deep brain stimulators in the anterior inferior internal capsule. However, symptomatic improvement was not complete, and her electrode connections ev

Five topographically organized fields in the somatosensory cortex of the flying fox: microelectrode maps, myeloarchitecture, and cortical modules.

Five somatosensory fields were defined in the grey-headed flying fox by using microelectrode mapping procedures. These fields are: the primary somatosensory area, SI or area 3b; a field caudal to area 3b, area 1/2; the second somatosensory area, SII; the parietal ventral area, PV; and the ventral somatosensory area, VS. A large number of closely spaced electrode penetrations recording multiunit activity revealed that each of these fields had a complete somatotopic representation. Microelectrode maps of somatosensory fields were related to architecture in cortex that had been flattened, cut par

Evidence for an acid pH in rat renal inner medulla: paired measurements with liquid ion-exchange microelectrodes on collecting ducts and vasa recta.

We reevaluated the pH in the renal medulla in rats. pH of the vasa recta blood was about 1 pH unit acidic in comparison to the pH of renal artery blood. During furosemide-induced diuresis pH of vasa recta blood increased whereas pH of collecting duct urine further decreased. The acidic pH in the rat renal inner medulla during antidiuresis raises important questions about the source of H+ in inner medulla.

In Vivo Real-Time Detection of Phenylephrine in Mice Using Boron-Doped Diamond Microelectrodes with Simultaneous Video-Based Assessment of Pupil Size.

Precise quantification of ocular pharmacokinetics remains a critical bottleneck in ophthalmic drug development, primarily constrained by the inability to monitor intraocular concentrations in real time. Current methods typically rely on destructive sampling, which precludes longitudinal assessment in individual subjects. Here, we report an electrochemical platform for time-resolved quantification of phenylephrine in the anterior chamber of living mice using needle-shaped boron-doped diamond microelectrodes. An optimized multistep potential protocol mitigated biofouling and enabled stable monit

Oxidative stress in cancer prone xeroderma pigmentosum fibroblasts. Real-time and single cell monitoring of superoxide and nitric oxide production with microelectrodes.

Sun exposure is clearly implicated in premature skin ageing and neoplastic development. These features are exacerbated in patients with Xeroderma pigmentosum (XP), a hereditary disease associated at the cellular level with DNA repair defects and a low catalase activity. The implications of oxidative stress in the defects and cancer proneness of XP skin cells (keratinocytes, fibroblasts) are multiple and remain unclear. They were investigated here at the level of a single fibroblast by an electrochemical method based on microelectrodes we have developed previously. These microelectrodes permit

Customizable 3D Microelectrode Arrays Enabled by Two-Photon Polymerization and Liquid Metal Integration for Neural Interface Applications.

Conventional silicon-based microfabrication with both 2D and 3D geometries has enabled increasingly sophisticated neural microelectrode arrays; however, the inherent rigidity of silicon fundamentally limits both the mechanical compliance necessary for chronic implantation in soft neural tissues and the geometric customizability achievable through microfabrication processes. Here, we present a silicon-free strategy in which soft polymeric 3D neural microelectrode arrays are fabricated via customizable two-photon polymerization (2PP) and readily functionalized as electrodes by simple injection o

Flexible Au-Pt-vertical graphene neural microelectrode for the rapid detection of steady-state and transient dopamine in rats.

Dopamine (DA) can serve as a biomarker to evaluate emotional and cognitive disorders; however, the correlation between transient and steady-state DA levels in the brain remains unclear. In this study, gold and platinum co-doped vertical graphene (AuPt-VG) is deposited onto the surface of carbon fibers (CFs) to prepare flexible neural microwires for DA detection. The diameter of the AuPt-VG/CFs is approximately 8.5 ± 0.5 μm. When using approximately thirty microwires, a detection limit of 7.6 nM is achieved in fast-scan cyclic voltammetry (FSCV). Owing to its excellent biocompatibility, antipol

Bio-Inspired Electrochemical Detection of Nitric Oxide Promoted by Coordinating the Histamine-Iron Phthalocyanine Catalytic Center on Microelectrode.

Biomimetic structures to fabricate bioelectronic interfaces that allow sensors to electrically communicate with electrodes have potential applications in the development of biosensors. Herein, inspired by the structure feature of nitric oxide (NO) sensory protein, we constructed a biomimetically catalytic center, the histamine coordinated iron phthalocyanine (FePc), for efficient and sensitive detection of NO. In specific, NO is recognized by axial tethered FePc, and the oxidative signal of NO on FePc is converted into output signal through electrocatalytic oxidation. Based on the fabricated c

Technical Applications of Microelectrode Array and Patch Clamp Recordings on Human Induced Pluripotent Stem Cell-Derived Cardiomyocytes.

Drug-induced cardiotoxicity is the leading cause of drug attrition and withdrawal from the market. Therefore, using appropriate preclinical cardiac safety assessment models is a critical step during drug development. Currently, cardiac safety assessment is still highly dependent on animal studies. However, animal models are plagued by poor translational specificity to humans due to species-specific differences, particularly in terms of cardiac electrophysiological characteristics. Thus, there is an urgent need to develop a reliable, efficient, and human-based model for preclinical cardiac safe

A reduced-graphene oxide-modified microelectrode for a repeatable detection of antipsychotic clozapine using microliters-volumes of whole blood.

Antipsychotic clozapine is the most effective medication currently available for schizophrenia. However, clozapine is dramatically underutilized due to its harsh side effects that are not effectively monitored. By continuously monitoring clozapine blood levels, such as use of an implantable glucometer, which has transformed diabetes management, the treatment can be optimized and side effects will be minimized. Currently, none of the methods for clozapine detection show the ability to repeatedly measure clozapine in whole blood without pretreatment steps. Here we propose using a microelectrode
