Microfluidic valves for wearable sweat analysis: a review.

Wearable sweat analysis can extend health monitoring from clinics to daily life. However, sweat is produced slowly and at changing rates, which makes sweat handling difficult. During collection and transport, sweat may remain in the device, flow backward, mix with older sweat, or become contaminated. The liquid that reaches the sensor may therefore differ from freshly secreted sweat. Microfluidic valves can control when sweat moves, where it flows, and how it reaches the sensing region. This review compares five valve classes relevant to wearable sweat systems. They are capillary, Tesla, mecha

Suspended Tissue Open Microfluidic Patterning (STOMP).

Free-standing tissue structures tethered between pillars are powerful mechanobiology tools for studying cell contraction. To model interfaces ubiquitous in natural tissues and upgrade existing single-region suspended constructs, we developed Suspended Tissue Open Microfluidic Patterning (STOMP), a method to create multiregional suspended tissues. STOMP uses open microfluidics and capillary pinning to pattern subregions within free-standing tissues, facilitating the study of complex tissue interfaces, such as diseased-healthy boundaries (e.g., fibrotic-healthy) and tissue-type interfaces (e.g.,

Microfluidic Nanobiosensor Detection of Botulinum Neurotoxin Serotype A.

Botulinum neurotoxin (BoNT) poses a significant threat to public health as the most potent toxin known to humans. BoNT intoxication can lead to botulism, a life-threatening disease that may result from ingesting food contaminated with biologically active BoNT. The gold standard method for BoNT detection, the mouse bioassay, is time-consuming and raises ethical concerns regarding the use of laboratory animals. Therefore, there is a critical need for alternative methods that are rapid and require minimal training for field use. In this study, with potential in-field use in mind, we developed a m

Inertial microfluidic mixer for biological CubeSat missions.

Nanosatellites of CubeSat type due to, i.a., minimized costs of space missions, as well as the potential large application area, have become a significant part of the space economy sector recently. The opportunity to apply miniaturized microsystem (MEMS) tools in satellite space missions further accelerates both the space and the MEMS markets, which in the coming years are considered to become inseparable. As a response to the aforementioned perspectives, this paper presents a microfluidic mixer system for biological research to be conducted onboard CubeSat nanosatellites. As a high complexity

Rapid On-Site Evaluation (ROSE): A Microfluidic Approach.

Rapid on-site evaluation (ROSE) increases the diagnostic accuracy of fine-needle aspiration (FNA) samples from cysts, a sack-like fluid-containing tissue that sometimes can be precancerous, but is highly dependent on the skills and availability of cytopathologists. We present a semiautomated sample preparation device for ROSE. The device consists of a smearing tool and a capillary-driven chamber that allow smearing and staining of an FNA sample in a single platform. Here, we show the capability of the device to prepare samples for ROSE, using a human pancreatic cancer cell line (PANC-1) and li

Microfluidic Gas Sensors: Detection Principle and Applications.

With the rapid growth of emerging point-of-use (POU)/point-of-care (POC) detection technologies, miniaturized sensors for the real-time detection of gases and airborne pathogens have become essential to fight pollution, emerging contaminants, and pandemics. However, the low-cost development of miniaturized gas sensors without compromising selectivity, sensitivity, and response time remains challenging. Microfluidics is a promising technology that has been exploited for decades to overcome such limitations, making it an excellent candidate for POU/POC. However, microfluidic-based gas sensors re

Closable Valves and Channels for Polymeric Microfluidic Devices.

This study explores three unique approaches for closing valves and channels within microfluidic systems, specifically multilayer, centrifugally driven polymeric devices. Precise control over the cessation of liquid movement is achieved through either the introduction of expanding polyurethane foam, the application of direct contact heating, or the redeposition of xerographic toner via chloroform solvation and evaporation. Each of these techniques modifies the substrate of the microdevice in a different way. All three are effective at closing a previously open fluidic pathway after a desired un

A microfluidic platform for the characterisation of membrane active antimicrobials.

The spread of bacterial resistance against conventional antibiotics generates a great need for the discovery of novel antimicrobials. Polypeptide antibiotics constitute a promising class of antimicrobial agents that favour attack on bacterial membranes. However, efficient measurement platforms for evaluating their mechanisms of action in a systematic manner are lacking. Here we report an integrated lab-on-a-chip multilayer microfluidic platform to quantify the membranolytic efficacy of such antibiotics. The platform is a biomimetic vesicle-based screening assay, which generates giant unilamell

Microfluidic devices for disease modeling in muscle tissue.

Microfluidic devices have advanced significantly in recent years and are a promising technology for the field of tissue engineering. Highly sophisticated microfabrication techniques have paved the way for the development of complex ex vivo models capable of incorporating and measuring the real-time response of multiple cell types interacting together in a single system. Muscle-on-a-chip technology has drastically improved and serves as a drug screening platform for many muscular diseases such as muscular dystrophy, tendinosis, fibromyalgia, mitochondrial myopathy, and myasthenia gravis. Microfluidic approach to genotyping human platelet antigens.

Centralised laboratories routinely determine blood types by serological and molecular methods. Current practices have limitations in terms of cost, time and accessibility. Miniaturised microfluidic platforms offer an alternative to conventional genotyping methods, since they consume fewer reagents, provide faster analysis and allow for complete integration and automation. As these 'lab-on-a-chip' devices have been used for bacterial and viral detection, the authors investigated blood group genotyping as a novel application of microfluidic technology. To demonstrate the feasibility of microflui

Distillation and detection of SO2 using a microfluidic chip.

A miniaturized distillation system is presented for separating sulfurous acid (H(2)SO(3)) into sulfur dioxide (SO(2)) and water (H(2)O). The major components of the proposed system include a microfluidic distillation chip, a power control module, and a carrier gas pressure control module. The microfluidic chip is patterned using a commercial CO(2) laser and comprises a serpentine channel, a heating zone, a buffer zone, a cooling zone, and a collection tank. In the proposed device, the H(2)SO(3) solution is injected into the microfluidic chip and is separated into SO(2) and H(2)O via an appropr

Electrochemical sensing in paper-based microfluidic devices.

This paper describes the fabrication and the performance of microfluidic paper-based electrochemical sensing devices (we call the microfluidic paper-based electrochemical devices, microPEDs). The microPEDs comprise paper-based microfluidic channels patterned by photolithography or wax printing, and electrodes screen-printed from conducting inks (e.g., carbon or Ag/AgCl). We demonstrated that the microPEDs are capable of quantifying the concentrations of various analytes (e.g., heavy-metal ions and glucose) in aqueous solutions. This low-cost analytical device should be useful for applications

FISH and chips: chromosomal analysis on microfluidic platforms.

Interphase fluorescence in situ hybridisation (FISH) is a sensitive diagnostic tool used for the detection of alterations in the genome on cell-by-cell basis. However, the cost-per-test and the technical complexity of current FISH protocols have slowed its widespread utilisation in clinical settings. For many cancers, the lack of a cost-effective and informative diagnostic method has compromised the quality of life for patients. We present the first demonstration of a microchip-based FISH protocol, coupled with a novel method to immobilise peripheral blood mononuclear cells inside microfluidic

Performance of sonication and microfluidization for liquid-liquid emulsification.

The purpose of this research was to evaluate and compare liquid-liquid emulsions (water-in-oil and oil-in-water) prepared using sonication and microfluidization. Liquid-liquid emulsions were characterized on the basis of emulsion droplet size determined using a laser-based particle size analyzer. An ultrasonic-driven benchtop sonicator and an air-driven microfluidizer were used for emulsification. Sonication generated emulsions through ultrasound-driven mechanical vibrations, which caused cavitation. The force associated with implosion of vapor bubbles caused emulsion size reduction and the fl

Integrated Antigen-DNA Co-Detection Using Digital Microfluidics for Improving Diagnostic Accuracy in POCT Analysis.

Simultaneous detection of protein and nucleic acid (NA) provides complementary information essential for accurate analysis of complex samples. Conventional approaches for the analysis of protein and NA typically rely on separate workflows and instruments, limiting the reliability of point-of-care testing. Here, we present a digital microfluidics-based dual-mode molecular assay for simultaneous detection of both protein and NA from the same microscale sample. The protein is captured by a gold nanoparticle-antibody conjugate, while NA is enriched by magnetic beads. Magnetic manipulation separate

Electrokinetic Nucleic Acid Amplification (E-NAAMP) Using Paper-PDMS Microfluidics and High-Frequency Joule Heating.

We present a novel paper-based nucleic acid amplification (NAA) technique using electrokinetic nucleic acid amplification (E-NAAMP). In E-NAAMP, a high radio frequency (RF) potential is applied across a conductive aqueous sample to induce an Ohmic current and drive the sample temperature to increase by Joule heating. Using this RF approach, we investigate the ability to induce E-NAAMP in pressurized paper-based microfluidic channels. We use the microfluidic pressure-in-paper (µPiP) method to encapsulate synthetic and natural fiber-based paper channels between thin sheets of polydimethylsiloxan

Size optimization of fire-extinguishing microcapsules fabricated via non-planar microfluidics and their performance study.

In confined spaces where early fire detection and suppression are particularly challenging, failures in fire prevention and control can lead to severe personal injury and property loss. In such scenarios, the structured encapsulation and controlled release of fire-extinguishing agents are especially critical. However, systematic studies on the structural design of extinguishing agents and their fire-suppression mechanisms remain notably insufficient. Based on non-planar microfluidic technology, this study designed a novel PDMS microfluidic device for the highly efficient preparation of thermal

Machine learning-driven single-cell phenotyping in size-controlled microenvironments via parallel deterministic droplet microfluidics.

Understanding how individual cells respond to distinct physical microenvironments is critical for mechanobiology, cell therapy, and tissue engineering. Current single-cell encapsulation methods are often limited by Poisson loading and fixed droplet sizes, preventing parallel generation of multiple, size-specific microenvironments and constraining high-resolution phenotypic analyses. Here, we present a droplet microfluidic platform that enables deterministic single-cell encapsulation within microgels of multiple sizes from a single precursor stream, achieved through parallelized flow-focusing c

Extrusion-printed silicone microarchitectures for geometry-controlled flow in lateral flow diagnostics and paper microfluidics.

Paper-based diagnostics such as lateral flow assays (LFAs) and microfluidic paper-based analytical devices (μPADs) have attracted considerable attention because of their low cost, portability, and ease of use. Currently, to enable fabrication of μPADs and improve LFA performance, hydrophobic blocks are patterned on paper substrates. However, fabrication of high-resolution hydrophobic barriers remains a major challenge. In this work, we developed a novel silicone extrudable ink for the fabrication of hydrophobic features on paper substrates. The ink was formulated using a vinyl-terminated polyd

Fast multi-resolution 3D printing of microfluidics: enabling 2 μm channels and ultra-compact mixers.

Microfluidic devices with ultra-fine features are critical for applications in biomedical diagnostics, chemical analysis, and lab-on-chip systems, but achieving high-resolution negative features with fast print times remains a significant challenge due to limitations in conventional 3D printing techniques. Motivated by the need for rapid fabrication of precise, compact microfluidic structures to enhance performance and miniaturization, we present an efficient multi-resolution 3D printing technique designed to fabricate microfluidic devices with exceptionally high-resolution negative features.
