Fluorescence Microscopy Imaging of Light-Harvesting in Self-Assembled Nanofibers of Naphthalenediimides toward Perylenediimide Guests.

The self-assembly of a bichromophoric naphthalenediimide (NDI) into nanofibers showed efficient energy transfer (light-harvesting) to perylenediimide (PDI) molecules through a host-guest interaction, which can be visualized by microscopy using samples deposited on glass surfaces. In combination with atomic force microscopy, spectral, and polarization analyses, fluorescence imaging unveiled the inhomogeneity, packing defects, and relative spatial arrangement of PDI and NDI molecular units, which were found to affect the exciton mobility along the NDI nanofibers and the energy transfer efficienc

High Molecular Crowding in Repair Foci Surrounding DNA Breaks, Measured by Fluorescence Lifetime Imaging Microscopy.

The density of mammalian cells is determined primarily by the protein content. Local concentration of proteins in a cell is tightly controlled and varies between the cytoplasm, nucleoplasm, and nucleoli. We demonstrate that repair foci that are formed in response to DNA breaks are much more densely packed with proteins than the surrounding nucleoplasm. Using fluorescence lifetime imaging (FLIM), we demonstrated that the local concentration of all proteins (i.e., the residing and recruited ones) in double- and single-strand DNA repair foci can be even 2.2 times higher than that in the surroundi

In-depth 3D exploration of autosomal dominant polycystic kidney disease through light sheet fluorescence microscopy.

Autosomal Dominant Polycystic Kidney Disease (ADPKD) is the most prevalent genetic kidney disorder. Animal preclinical studies are one of the main tools to study this disease, often through either 2D histology imaging for high-resolution analysis or CT or MRI for full kidney segmentation. As an alternative to these modalities, we propose the use of Light Sheet Fluorescence Microscopy (LSFM) for high-resolution 3D imaging of healthy and ADPKD-induced mouse kidneys, enabling a detailed volumetric morphological analysis of the disease's effects. In a mouse ADPKD model, ex vivo imaging of the kidn

BigReg: an efficient registration pipeline for high-resolution X-ray and light-sheet fluorescence microscopy.

PURPOSE: We aim to propose a reliable registration pipeline tailored for multimodal mouse bone imaging using X-ray microscopy (XRM) and light-sheet fluorescence microscopy (LSFM). These imaging modalities have emerged as pivotal tools in preclinical research, particularly for studying bone remodeling diseases such as osteoporosis. Although multimodal registration enables micrometer-level structural correspondence and facilitates functional analysis, conventional landmark-, feature-, or intensity-based approaches are often infeasible due to inconsistent signal characteristics and significant mi

Supramolecular Complex of Cucurbit[7]uril with Diketopyrrolopyrole Dye: Fluorescence Boost, Biolabeling and Optical Microscopy.

New photostable and bright supramolecular complexes based on cucurbit[7]uril (CB7) host and diketopyrrolopyrole (DPP) guest dyes having two positively charged 4-(trimethylammonio)phenyl groups were prepared and characterized. The dye core displays large Stokes shift (in H2O, abs./emission max. 480/550 nm; ϵ~19 000, τfl>4 ns), strong binding with the host (~560 nM Kd) and a linker affording fluorescence detection of bioconjugates with antibody and nanobody. Combination of protein-functionalized DPP dye with CB7 improves photostability and affords up to 12-fold emission gain. Two-color confocal

Machine learning based classification of mitochondrial morphologies from fluorescence microscopy images of Toxoplasma gondii cysts.

The mitochondrion is intimately linked to energy and overall metabolism and therefore the morphology of mitochondrion can be very informative for inferring the metabolic state of cells. In this study we report an approach for automatic classification of mitochondrial morphologies using supervised machine learning to efficiently classify them from a large number of cells at a time. Fluorescence microscopy images of the chronic encysted form of parasite Toxoplasma gondii were used for this development. Manually classifying these morphologies from the hundreds of parasites within typical tissue c

Moxifloxacin-Based Extended Depth-of-Field Fluorescence Microscopy for Real-Time Conjunctival Goblet Cell Examination.

Conjunctival goblet cells (CGCs) are mucin-secreting cells in the eye and play essential roles for ocular surface homeostasis. Since various ocular surface pathologies are related to CGC dysfunction, CGC examination is important for the evaluation of ocular surface conditions. Recently we introduced moxifloxacin-based fluorescence microscopy (MBFM) for non-invasive CGC imaging. However, the imaging speed was up to 1 frame per second (fps) and needed to be improved for clinical applications. In this study, we developed a high-speed moxifloxacin-based, extended depth-of-field (EDOF) microscopy s

HDL particles incorporate into lipid bilayers - a combined AFM and single molecule fluorescence microscopy study.

The process, how lipids are removed from the circulation and transferred from high density lipoprotein (HDL) - a main carrier of cholesterol in the blood stream - to cells, is highly complex. HDL particles are captured from the blood stream by the scavenger receptor, class B, type I (SR-BI), the so-called HDL receptor. The details in subsequent lipid-transfer process, however, have not yet been completely understood. The transfer has been proposed to occur directly at the cell surface across an unstirred water layer, via a hydrophobic channel in the receptor, or after HDL endocytosis. The role

SNAP-tagged Chikungunya Virus Replicons Improve Visualisation of Non-Structural Protein 3 by Fluorescence Microscopy.

Chikungunya virus (CHIKV), a mosquito-borne alphavirus, causes febrile disease, muscle and joint pain, which can become chronic in some individuals. The non-structural protein 3 (nsP3) plays essential roles during infection, but a complete understanding of its function is lacking. Here we used a microscopy-based approach to image CHIKV nsP3 inside human cells. The SNAP system consists of a self-labelling enzyme tag, which catalyses the covalent linking of exogenously supplemented synthetic ligands. Genetic insertion of this tag resulted in viable replicons and specific labelling while preservi

In vivo fluorescence confocal microscopy: indocyanine green enhances the contrast of epidermal and dermal structures.

In recent years, in vivo skin imaging devices have been successfully implemented in skin research as well as in clinical routine. Of particular importance is the use of reflectance confocal microscopy (RCM) and fluorescence confocal microscopy (FCM) that enable visualization of the tissue with a resolution comparable to histology. A newly developed commercially available multi-laser device in which both technologies are integrated now offers the possibility to directly compare RCM with FCM. The fluorophore indocyanine green (ICG) was intradermally injected into healthy forearm skin of 10 volun

Innervation and vasculature of human sweat glands: an immunohistochemistry-laser scanning confocal fluorescence microscopy study.

Secreting tubules, nerves fibers, and blood vessels in human sweat glands (SGs) were fluorescently stained by immunohistochemical and lectin methods for examination with a laser scanning confocal microscope (LSCM). Using these techniques, the three-dimensional distribution of up to three substances within a single specimen was investigated by collecting a series of optical sections for each of three fluorophores. Each SG received several nerve fibers. These branched into delicate bands of one or more axons that ran longitudinal to the sweat tubule then encircled the tubule. A heavy complement

Cathodoluminescence-emitting lipid droplets in rat testis: a study by analytical color fluorescence electron microscopy.

Cathodoluminescence (CL) from lipid droplets (LDs) in the rat testis was examined by analytical color fluorescence electron microscopy. The results show that: (1) the CL at wavelengths of 320 nm (CL320) and 450 nm (CL450) is derived from cholesterol esters and a mixture of lipids including vitamin A esters, respectively; (2) CL320 in the LDs of Leydig cells sharply decreases on postnatal day 21, while CL320 and CL450 in the LDs of Sertoli cells begin to be detectable; (3) the CL450-emitting LDs in seminiferous tubules, whose distributional patterns display cyclic changes during the spermatogen

Artificial intelligence-enabled cytopathological interpretation of fluorescence microscopy images via a multimodal large language model.

Quantitative analysis of fluorescence microscopy images remains a significant challenge in cytopathological assessments, as subjective interpretation and inter-observer variability hinder reproducibility and standardization of classifications. In this study, an artificial intelligence-assisted analytical framework utilizing a multimodal large language model (MLLM) was employed to classify cytotoxic responses in acridine orange/propidium iodide-stained MCF-7 cells subjected to doxorubicin treatment for 24 h. A total of 500 fluorescence images, collected from five experimental groups including a

Label-free metabolic imaging of Pseudomonas aeruginosa infection using two-photon fluorescence lifetime imaging microscopy.

In this study we have applied high-spatial and temporal label-free imaging of individual live multidrug-resistant bacteria and bacteria-infected cells and animal tissue using two-photon fluorescence lifetime imaging microscopy (2p-FLIM). 2p-FLIM can identify and quantify fluorescence intensity and lifetimes among bacteria, infected cells, and tissues. We have implemented 2p-FLIM in combination with phasor plot analysis for quantifying molecular differences of NAD(P)H intensities and lifetimes for fast, sensitive, high-resolution, non-destructive imaging of live bacteria and bacteria-infected c

Acute lymphoblastic leukemia cancer diagnosis in children and adults using transforming blood fluorescence microscopy imaging.

Leukemia is a highly aggressive kind of cancer that may impact the bone marrow. The most fatal type acute lymphoblastic leukemia (ALL), is characterized by the excessive growth of immature white blood cells in the bone marrow. For diagnostic purposes, hematologists and experts use a state-of-the-art microscope fitted with a high-powered magnifying lens to analyze blood and bone marrow samples. Experts attribute the rapid progress to the presence of adolescent white blood cells, not fully developed ones. A good treatment for ALL, no matter where it comes from, includes chemotherapy, medication

Dual-ligand fluorescence microscopy enables chronological and spatial histological assignment of distinct amyloid-β deposits.

Different types of deposits comprised of amyloid-β (Aβ) peptides are one of the pathological hallmarks of Alzheimer's disease (AD) and novel methods that enable identification of a diversity of Aβ deposits during the AD continuum are essential for understanding the role of these aggregates during the pathogenesis. Herein, different combinations of five fluorescent thiophene-based ligands were used for detection of Aβ deposits in brain tissue sections from transgenic mouse models with aggregated Aβ pathology, as well as brain tissue sections from patients affected by sporadic or dominantly inhe

Preparing Planarian Cells for High-Content Fluorescence Microscopy Using RNA in Situ Hybridization and Immunocytochemistry.

High-content fluorescence microscopy combines the efficiency of high-throughput techniques with the ability to extract quantitative information from biological systems. Here we describe a modular collection of assays adapted for fixed planarian cells that enable multiplexed measurements of biomarkers in microwell plates. These include protocols for RNA fluorescent in situ hybridization (RNA FISH) as well as immunocytochemical protocols for quantifying proliferating cells targeting phosphorylated histone H3 as well as 5-bromo-2'-deoxyuridine (BrdU) incorporated into the nuclear DNA. The assays
