Enantioselective total synthesis and structural assignment of callyspongiolide.

We have elucidated the complete absolute configuration of callyspongiolide and unambiguously assigned its stereochemistry at the C-21 center through synthesis. Four stereoisomers of callyspongiolide were synthesized in a convergent and enantioselective manner. A late-stage Sonogashira coupling forges the diene-ynic side chain. Other notable reactions are Yonemitsu's variation of Yamaguchi macrolactonization to cyclize an alkynic seco acid, highly trans-selective Julia-Kocienski olefination, CBS reduction to set the C-21 stereocenter, and methyl cuprate addition to an unsaturated pyranone to in

Product in indole detection by Ehrlich's reagent.

Ehrlich's reagent (p-dimethylaminobenzaldehyde [DMAB, 1] in 95% EtOH with HCl as catalyst) was employed in spot tests of indoles, providing a diagnosis of, for example, liver diseases, hemolytic processes, occlusion of the common bile duct, and carcinoid syndrome. Although the reagent has been widely used for more than a century, it is not clear how many indole molecules react with a DMAB molecule and whether the reaction takes place at the α- or β-position of the indole molecule. Research here shows that the reaction of DMAB (1) with indole (2) in a 1:2 ratio gives β-bis(indolyl)methane (3).

Synthesis and optical properties of chlorin monomer, dimer and trimer on an amino nitrogen atom.

Naturally occurring chlorophyll-a was chemically modified to methyl 3-aminomethyl-pyropheophorbides-a including primary, secondary, and tertiary amines. Reductive amination of methyl pyropheophorbide-d possessing the 3-formyl group with ammonia efficiently gave a chlorin dimer covalently linked with CH2NHCH2 at the 3-position, which was transformed into a trimer through the substitution at the amino group. Conformational analyses by (1)H NMR spectroscopic observation and molecular modeling estimation indicated that the dimer and trimer were apt to form closely packed structures. Chlorin chromo

Trifluorosilane induced structural transitions in beta-lactoglobulin in sol and gel.

Fluoroalcohols such as 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) are strong inducers of protein secondary structure. Here, hydrolyzed (3,3,3-trifluoropropyl)trimethoxysilane (3F) is demonstrated to exhibit greater protein conformation inducing activity than HFIP, which is preserved when 3F is copolymerized in tetraethylorthosilicate (TEOS) sol-gels through protein molecular imprinting. Hydrolyzed 3F formed a pre-polymerization complex with the template protein, β-lactoglobulin, inducing distinct α-helical structures as evidenced by circular dichroism. Multiscale Elucidation of Taste Interactions in Osmanthus Green Tea: Bitter Masking via EGCG and Phenylethanoid Glycoside Interplay.

This study revealed the molecular mechanism by which Osmanthus fragrans masks the bitterness of summer-autumn green tea infusion. Taste dilution analysis identified catechins such as EGCG in green tea as the primary bitter compounds. Phenylethanoid glycosides (PhGs), such as verbascoside and salidroside in Osmanthus fragrans exhibited significant bitter-masking effects. Molecular docking identified TAS2R31 as the primary bitter receptor for EGCG, with verbascoside or salidroside competitively disrupting the EGCG-TAS2R31 binding. Molecular dynamics simulations showed that PhGs altered the TAS2R

Rationalization of the Membrane Permeability Differences in a Series of Analogue Cyclic Decapeptides.

Cyclization and selected backbone N-methylations are found to be often necessary but not sufficient conditions for peptidic drugs to have a good bioavailability. Thus, the design of cyclic peptides with good passive membrane permeability and good solubility remains a challenge. The backbone scaffold of a recently published series of cyclic decapeptides with six selected backbone N-methylations was designed to favor the adoption of a closed conformation with β-turns and four transannular hydrogen bonds. Although this conformation was indeed adopted by the peptides as determined by NMR measureme

Simulation Reveals the Chameleonic Behavior of Macrocycles.

Conformational analysis is central to the design of bioactive molecules. It is particularly challenging for macrocycles due to noncovalent transannular interactions, steric interactions, and ring strain that are often coupled. Herein, we simulated the conformations of five macrocycles designed to express a progression of increasing complexity in environment-dependent intramolecular interactions and verified the results against NMR measurements in chloroform and dimethyl sulfoxide. Molecular dynamics using an explicit solvent model, but not the Monte Carlo method with implicit solvation, handle

Recognition of cholinergic agonists by the muscarinic receptor. 1. Acetylcholine and other agonists with the NCCOCC backbone.

A theoretical model is used to deduce the pharmacologically active conformation of acetylcholine and other agonists interacting with the muscarinic receptor of the parasympathetic and central nervous systems. This is accomplished by replacing the usual dihedral angles tau 1 and tau 2, which define the conformations of cholinergic drugs, with two new geometric parameters more suitable for describing the muscarinic pharmacophore: a characteristic distance, [PQ], and a dihedral angle, PNOQ. Values for these parameters are determined by conformational analysis on semirigid muscarinic agonists usin

Crystal structure of Staudtienic acid, a diterpenoid from Staudtia kamerunensis Warb. (Myristicaceae).

This title compound, C20H26O2, was isolated from the benzene fraction of the stem bark of Staudtia kamerunensis Warb. (Myristicaceae) using column chromatography techniques over silica gel. The compound was fully characterized by single-crystal X-ray diffraction, one and two-dimensional NMR spectroscopy, IR and MS spectrometry. The compound has two fused cyclo-hexane rings attached to a benzene ring, with a carb-oxy-lic acid on C-4. This cyclo-hexene ring has a chair conformation while the other adopts a half-chair conformation. The benzene ring is substituted with a propenyl moiety. The struc

Structure of the K-turn U4 RNA: a combined NMR and SANS study.

K-turn motifs are universal RNA structural elements providing a binding platform for proteins in several cellular contexts. Their characteristic is a sharp kink in the phosphate backbone that puts the two helical stems of the protein-bound RNA at an angle of 60°. However, to date no high-resolution structure of a naked K-turn motif is available. Here, we present the first structural investigation at atomic resolution of an unbound K-turn RNA (the spliceosomal U4-Kt RNA) by a combination of NMR and small-angle neutron scattering data. With this study, we wish to address the question whether the

Characterization of the Conformations of Amyloid Beta 42 in Solution That May Mediate Its Initial Hydrophobic Aggregation.

Intrinsically disordered peptides, such as amyloid β42 (Aβ42), lack a well-defined structure in solution. Aβ42 can undergo abnormal aggregation and amyloidogenesis in the brain, forming fibrillar plaques, a hallmark of Alzheimer's disease. The insoluble fibrillar forms of Aβ42 exhibit well-defined, cross β-sheet structures at the molecular level and are less toxic than the soluble, intermediate disordered oligomeric forms. However, the mechanism of initial interaction of monomers and subsequent oligomerization is not well understood. The structural disorder of Aβ42 adds to the challenges of de

Conformations and Structures of 3-Oxopentanenitrile and 4-Methyl-3-oxopentanenitrile Revealed by Rotational Spectroscopy.

The conformations and molecular structures of two cyano-substituted ketones, namely 4-methyl-3-oxopentanenitrile (MPN) and 3-oxopentanenitrile (PN), have been investigated by using supersonic jet Fourier transform microwave (FTMW) spectroscopy. The rotational spectrum of a single conformation was measured and assigned for 4-methyl-3-oxopentanenitrile (MPN-I) and 3-oxopentanenitrile (PN-I), respectively. Additional measurements were conducted for 13C and 15N singly substituted isotopologues of conformer PN-I in natural abundance. The comprehensive rotational spectroscopic investigations yield a

Thermal Behavior and Local Structural Organization in Curcumin Polymorphs' Bulk Phase: A Molecular Dynamics Simulation Analysis.

Curcumin (CUR), a bioactive compound with known polymorphism, exhibits distinct conformational and thermophysical properties across its three crystalline forms. In this study, we employ molecular dynamics simulations to investigate the thermal behavior, local structural organization, and polymorph-specific stability of CUR in the bulk phase. We first evaluate four widely used classical force fields (OPLS-AA, CGENFF, GAFF2, and GROMOS) against experimental melting points, densities, and conformational preferences, identifying OPLS-AA as the most suitable one. Through targeted reparametrization

Charge-Regulated Conformational Transitions Govern Nonmonotonic Pinch-Off Dynamics.

While molecular conformational changes significantly influence drop pinch-off, the governing mechanisms by which conformational evolution reshapes capillary-driven breakup remain elusive. Here, we isolate the role of conformation using charge regulation to drive conformational transitions in weak polyelectrolytes. We demonstrate that dissociation-driven chain expansion and counterion-mediated screening compete to generate nonmonotonic pinch-off dynamics. Furthermore, a slip-dominated intermediate thinning regime is identified, which is suppressed by concentration-dependent topological constrai

Comparison of the Self-Assembly and Conformations of Glucose- and Galactose-Based Glycopyranosides in Dilute Aqueous Solution.

A series of glycolipids (glycopyranosides) was prepared by coupling glucosamine or galactosamine (at the C2 position) with myristic (tetradecanoic) or palmitic (hexadecanoic) acid. The conformation and self-assembly were examined in aqueous solutions containing 10% methanol. Circular dichroism and FTIR spectroscopy reveal notable differences in the chiral ordering and conformation comparing analogues with glucose and galactose "headgroups". A glucose derivative (to palmitic acid) was also prepared with a different (C1-) substitution position of the lipid chain, and this was found to significan

The Allosteric Revolution: From Static Structures to Conformational Ensembles and Next-Generation Therapeutics.

The rapid emergence of breakthrough allosteric drugs, together with increasing emphasis on the molecular mechanisms of disease, underscores the critical importance of understanding biological processes at the molecular level. Among these, few concepts are as fundamental as the allosteric behavior of proteins. The importance of allostery was recognized decades ago; however, at the time proteins were viewed as static molecular structures rather than dynamic conformational ensembles that continuously interconvert among states with distinct free energies. The realization that proteins exist as dyn

Molecular orientation and stability of poly(3-hexylthiophene) nanogratings affected by the fabricated solvent vapor.

During the nanoimprinting lithography (NIL) process, the role of solvent vapor in fabricating the pattern structure and inducing the molecular alignment of nanoimprinted polymer film has been attracting significant attention. We demonstrate here that the molecular orientation and thermal stability of poly(3-hexylthiophene) (P3HT) nanograting film can be affected obviously by the fabricated solvent vapor. A solvent-vapor nanoimprinting lithography (SV-NIL) technique based on a polydimethylsiloxane (PDMS) template is employed to fabricate a P3HT nanograting structure film successfully and solven

Polyethylene Glycol Impacts Conformation and Dynamics of Escherichia coli Prolyl-tRNA Synthetase Via Crowding and Confinement Effects.

Polyethylene glycol (PEG) is a flexible, nontoxic polymer commonly used in biological and medical research, and it is generally regarded as biologically inert. PEG molecules of variable sizes are also used as crowding agents to mimic intracellular environments. A recent study with PEG crowders revealed decreased catalytic activity of Escherichia coli prolyl-tRNA synthetase (Ec ProRS), where the smaller molecular weight PEGs had the maximum impact. The molecular mechanism of the crowding effects of PEGs is not clearly understood. PEG may impact protein conformation and dynamics, thus its functi
