Skin aging: molecular pathology, dermal remodelling and the imaging revolution.

Skin is a multifunctional organ but, alongside every other organ system, is subject to both intrinsic (chronological) and extrinsic (environmental) aging, resulting in a loss of functional capacity. Cutaneous aging manifests as an observable change in the external appearance of the skin, the major accelerator of the aging process being our interactions with our environment, such as chronic exposure to solar irradiation (UV, IR or visible wavelengths of light). The aim of this contribution, therefore, was to provide a review of the pathological mechanisms which may play roles in the development

Molecular imaging of brown adipose tissue in health and disease.

PURPOSE: Brown adipose tissue (BAT) has transformed from an interfering tissue in oncological (18)F-fluorodeoxyglucose (FDG) positron emission tomography (PET) to an independent imaging research field. This review takes the perspective from the imaging methodology on which human BAT research has come to rely on heavily. METHODS: This review analyses relevant PubMed-indexed publications that discuss molecular imaging methods of BAT. In addition, reported links between BAT and human diseases such as obesity are discussed, and the possibilities for imaging in these fields are highlighted. Radiop

Clinical molecular imaging with PET agents other than 18F-FDG.

Molecular imaging represents an extremely important advancement in patient care and numerous agents have been developed and are being studied. Hundreds of molecular imaging agents currently exist for use in multiple modalities such as magnetic resonance imaging (MRI), computed tomography (CT), ultrasound, optical imaging, general nuclear medicine and positron emission tomography (PET). In PET, only a very small percentage of these agents are being used clinically in humans and more show promise for clinical use in the short-term future. The purpose of this paper is to present a brief overview

Molecular imaging for efficacy of pharmacologic intervention in myocardial remodeling.

OBJECTIVES: Using molecular imaging techniques, we examined interstitial alterations during postmyocardial infarction (MI) remodeling and assessed the efficacy of antiangiotensin and antimineralocorticoid intervention, alone and in combination. BACKGROUND: The antagonists of the renin-angiotensin-aldosterone axis restrict myocardial fibrosis and cardiac remodeling after MI and contribute to improved survival. Radionuclide imaging with technetium-99m-labeled Cy5.5 RGD imaging peptide (CRIP) targets myofibroblasts and indirectly allows monitoring of the extent of collagen deposition post-MI. M

Molecular imaging of innate immune cell function in transplant rejection.

BACKGROUND: Clinical detection of transplant rejection by repeated endomyocardial biopsy requires catheterization and entails risks. Recently developed molecular and cellular imaging techniques that visualize macrophage host responses could provide a noninvasive alternative. Yet, which macrophage functions may provide useful markers for detecting parenchymal rejection remains uncertain. METHODS AND RESULTS: We transplanted isografts from B6 mice and allografts from Balb/c mice heterotopically into B6 recipients. In this allograft across major histocompatability barriers, the transplanted hear

Use of adrenomedullin derivatives for molecular imaging of pulmonary circulation.

UNLABELLED: Currently, there is no low-molecular-weight agent for imaging of the pulmonary circulation. Adrenomedullin (AM) is a peptide predominantly cleared by the pulmonary circulation through specific endothelial receptors. We developed human AM derivatives radiolabeled with 99mTc and evaluated their biodistribution, plasma kinetics, and utility as pulmonary vascular imaging agents. METHODS: Two derivatives radiolabeled with 99mTc were evaluated: the natural cyclic form of the peptide, to which the chelator diethylenetriaminepentaacetic acid was added (C-DTPA-AM), and the linear form, whi

Potential applications of conventional and molecular imaging to biodefense research.

Imaging methods that visualize the structure and function of the living body are widely used in patient care and biomedical research, but their full potential has not yet been applied to the study and treatment of the severe illnesses caused by pathogens of biodefense concern. "Conventional" imaging techniques (e.g., radiography, computed tomography, ultrasound, or magnetic resonance imaging) delineate anatomic changes in tissues, whereas "molecular" methods employ magnetic resonance, positron emission tomography, single-photon emission computed tomography, or optical (fluorescence or biolumin

Molecular Mapping of Atherosclerotic Plaques with MALDI Mass Spectrometry Imaging.

Atherosclerosis develops within heterogeneous arterial plaques. Here, even closely neighbouring regions can differ substantially in biological activity and potential vulnerability to rupture and thrombosis. Through matrix-assisted laser desorption/ionisation mass spectrometry (MALDI-MS) and especially MALDI imaging, these differences within the plaque can be studied while also preserving tissue architecture. With MALDI analysis, prepared tissue sections are converted into spatial molecular maps that show the differences in plaque chemistry across the arterial wall. These maps can be used to li

Enhanced Imaging in Bladder Cancer: Fluorescence Cystoscopy and Molecular Diagnostics.

BACKGROUND/OBJECTIVES: Bladder cancer remains one of the most frequently diagnosed malignancies worldwide and is characterized by high recurrence rates requiring long-term surveillance. Conventional white-light cystoscopy (WLC) remains the standard diagnostic method; however, it may fail to detect flat lesions such as carcinoma in situ or small papillary tumors. In recent years, enhanced imaging techniques, including fluorescence cystoscopy and autofluorescence-based systems, have been introduced to improve diagnostic accuracy. The aim of this study is to evaluate the usefulness of fluorescenc

Molecular Imaging in Pancreatic Cancer: Current Applications and Future Perspectives.

Pancreatic cancer ranks among the most lethal malignancies, characterized by a five-year survival rate of approximately 10%. This dismal prognosis is largely attributable to diagnoses occurring at advanced stages and the inherent limitations of conventional imaging modalities in detecting early lesions, identifying metastases, and assessing tumor heterogeneity. Consequently, there is a critical need for non-invasive imaging techniques capable of visualizing pancreatic cancer lesions to enable accurate diagnosis, risk assessment, and the development of personalized treatment strategies. Improved Nondestructive Ultrasound Molecular Imaging With Lightweight Convolutional Neural Network.

Ultrasound molecular imaging (USMI) is an imaging approach that utilizes targeted microbubbles (MBs) to highlight biomarkers of disease. While differential targeted enhancement (DTE) is the current state-of-the-art for USMI, its reliance on destructive pulses hinders real-time clinical application. We have developed a neural network-based nondestructive USMI, validated in vivo using a transgenic mouse model of spontaneous breast cancer. To enhance training efficacy despite a limited animal number (N=14), we utilized several augmentation strategies including the use of several targeted MB types

Histologic correlates of background parenchymal uptake on molecular breast imaging.

BACKGROUND: Molecular breast imaging (MBI) has shown utility as a supplement to mammography in women with dense breasts. Background parenchymal uptake (BPU), which describes the level of Tc-99 m sestamibi in fibroglandular tissue on MBI, varies among women with similar density. Higher BPU is associated with greater breast cancer risk. The goal of this work is to investigate the histological correlations of BPU that may explain this association with risk. METHODS: In healthy women with dense tissue on mammography, we prospectively biopsied regions of fibroglandular tissue exhibiting one of the

PURPOSE: Lymph node(LN) dissection is part of most oncologic resections. Intraoperatively identifying a positive LN(+ LN), that harbors malignant cells, can be challenging. We hypothesized that intraoperative molecular imaging(IMI) using a cancer-targeted fluorescent prober can identify + LNs. This study aimed to develop a preclinical model of a + LN and test it using an activatable cathepsin-based enzymatic probe, VGT-309. PROCEDURES: In the first model, we used peripheral blood mononuclear cells (PBMC), representing the lymphocytic composition of the LN, mixed with different concentrations
