Plasminogen activator-specific inhibitors produced by human monocytes/macrophages.

Human monocytes/macrophages produce plasminogen activator-specific inhibitors (PAIs) that form covalent complexes with urokinase-type plasminogen activator (uPA). We have characterized two functionally and antigenically related forms of PAIs produced by resting and phorbol myristate acetate (PMA)-treated U 937 cells: an Mr 40,000 form, presumably nonglycosylated, with a pI of 5.2, that is constitutively synthetized by these cells and that remains predominantly intracellular; a PMA-induced form of heterogeneous Mr (50,000-65,000) with a pI of 4.7, that is preferentially secreted; this PAI is gl

Monocytes in rheumatoid arthritis. Regulatory, effector, and phenotypic properties.

We reported that rheumatoid arthritis (RA) blood mononuclear cells (MNC) secreted less Ig and IgM rheumatoid factor (RF) than synovial cells. Since antibody elaboration is partly monocyte dependent, we compared regulatory, effector, and phenotypic properties of monocytes from 31 patients with RA with those of 21 normal subjects. RA IgG and IgM elaboration was less than normal. Monocyte, T or B cell numbers were comparable in RA and normal MNC and monocyte enriched/depleted preparations. RA and normal Ig production were monocyte dependent and this differed for IgG and IgM for RA and normals. Ig

IgE induces secretion of prostaglandin E2 by human monocytes.

IgE was isolated from a patient with the hyper IgE, recurrent infection syndrome by immunoadsorption on sepharose bound goat anti-human IgE. Addition of this IgE to a monolayer culture of human monocytes resulted in a dose-dependent increase in PGE2 secretion. The addition of F(ab')2 fraction of goat anti-human IgE in the presence of sub-stimulating doses of IgE markedly increased PGE2 secretion; whereas addition of F(ab')2 fragment of irrelevant goat IgG had no effect. Similar activation of monocytes which could be enhanced by anti IgE was observed in the presence of the patient's serum. No s

[Phagocytic activity of monocytes tin glomerulonephritis in complete remission].

Circulating immune complexes (CIC) account for a majority of GN. Their pathogenicity depends on size, molecular composition, glomerular hemodynamics and activity of phagocytes. The phagocytic function of peripheral monocytes was studied in 23 patients with non-systemic active GN and in 16 GN in complete remission. Phagocytic activity of peripheral blood monocytes was assessed in vitro, by calculating phagocytic index (IP: number of zymozan particles in each monocyte) and phagocytosis percentage (PP: number of phagocyting monocytes); the tests were carried out on autologous and heterologous ser

Function studies of monocytes from patients with cyclic neutropenia.

In vitro studies of peripheral blood monocytes from patients with cyclic neutropenia have shown them to be able to phagocytize and kill bacteria as efficiently as PMN's, but not monocytes, from normal controls at both low and high ratios of bacteria to leukocytes. Monocytes from these patients, as well as from normal controls, are able to respond to chemotactic stimuli in vitro. In vivo Rebuck skin-window studies revealed the mononuclear cell response to be normal in these patients, even in the complete absence of circulating and inflammatory exudate neutrophils. These findings suggest that pe

Monocytes Display Altered Inflammatory Profiles in Individuals with Diabetic Foot Ulcers Indicating Monocyte Dysfunction.

Diabetic foot ulcers (DFUs) often exhibit impaired healing. Macrophage imbalance driven by persistent inflammation is one contributing factor, with a predominance of inflammatory over anti-inflammatory macrophages in non-healing ulcers. Monocytes, precursors of macrophages, are skewed towards an inflammatory phenotype in diabetes. Here, we investigated whether monocyte inflammatory skewing is further exacerbated in individuals with DFUs and explored how it relates to an individual's metabolic (HbA1c and lipids) profile. Monocyte inflammatory profile was assessed by examining phenotypic markers

Monocyte recruitment and expression of monocyte chemoattractant protein-1 are developmentally regulated in remodeling bone in the mouse.

Tooth eruption is defined as the movement of a tooth from its site of development within the alveolar bone to its position of function in the oral cavity. It represents an excellent model to examine osseous metabolism as bone resorption and bone formation occur simultaneously and are spatially separated. Bone resorption occurs in the coronal (occlusal) area, whereas bone formation occurs in the basal area. Monocytes are thought to have a significant role in the regulation of osseous metabolism. The goal of this study was to examine the recruitment of monocytes to bone in C57BL/6J mice that are

Studies of binding and internalization of human recombinant monocyte chemotactic and activating factor (MCAF) by monocytic cells.

Recombinant human monocyte chemotactic and activating factor (MCAF) was iodinated and specific binding sites for this cytokine were detected on human peripheral blood monocytes, the monocytic leukemia cell line THP-1, and on PMA-differentiated HL60 and U937 cell lines. The binding sites were specific for MCAF since other polypeptide cytokines and the chemotactic peptide formyl-methionyl-leucyl-phenylalanine (fMLP) failed to compete for 125I-rhMCAF binding. Steady-state binding experiments at 4 degrees C revealed the presence of 13,000 and 18,000 receptor sites/cell on monocytes and THP-1 cells

A monocyte disorder in siblings with chronic candidiasis. A combined abnormality of monocyte mobility and phagocytosis-killing ability.

Immunologic studies were performed on siblings (a 9-year-old boy and an 11-year-old girl) with chronic candidiasis since infancy and showed defective monocyte functions. In vivo migration and in vitro mobility of the monocytes were impaired. In addition, they had defective phagocytosis-killing ability against Candida albicans. There was no factor(s) to inhibit the monocyte functions in the patients' serum, and their mononuclear cells (75% to 80% lymphocytes) did not secrete such an inhibitory factor(s) in vitro. The serum IgG, IgA, IgM, and C3 levels were normal. Delayed hypersensitivity respo

ADAM17-Mediated Reduction in CD14++CD16+ Monocytes ex vivo and Reduction in Intermediate Monocytes With Immune Paresis in Acute Pancreatitis and Acute Alcoholic Hepatitis.

Impaired immune responses and increased susceptibility to infection characterize acute inflammatory conditions such as pancreatitis and alcoholic hepatitis and are major causes of morbidity and mortality. However, the mechanisms that drive this apparent immune paresis remain poorly understood. Monocytes mediate host responses to damage and pathogens in health and disease, and three subsets of monocytes have been defined based on CD14 and CD16 expression. We sought to determine the changes in monocyte subsets in acute pancreatitis (AP) and acute alcoholic hepatitis (AAH), together with function

CD14+ CD16+ monocytes rather than CD14+ CD51/61+ monocytes are a potential cytological marker of circulating osteoclast precursors in multiple myeloma. A preliminary study.

INTRODUCTION: Osteolytic bone destruction is a major clinical problem in multiple myeloma patients. Osteoclasts can differentiate in vitro from bone marrow-resident monocyte progenitors, such as common monocyte progenitors, as well as circulating monocytes. Various types of monocytes, including osteoclast precursors, appear to circulate systemically. METHODS: We investigated the possibility of demonstrating, by in vitro differentiation and flow cytometry, a circulating osteoclast precursor population in multiple myeloma (MM) patients by studying the distribution of CD14(+/++) CD11b(+) CD51/61

Monocytic papulosis: A clonal mature monocyte-derived dendritic cell histiocytopathy of potentially diverse monocyte-derived dendritic cell phenotypes as a reflection of underlying chronic myeloproliferative disease.

Clonal histiocytopathy involving non-Langerhans mature, monocyte-derived dendritic cells is a poorly characterized cutaneous manifestation of chronic myeloproliferative diseases. We describe the clinical and histopathological features of a well-differentiated histiocytopathy composed of mature, CD1a-negative, S100-negative, langerin-negative monocyte-derived dendritic cells, proposing it as a potential dermatologic marker for acute and/or chronic myeloproliferative diseases. This case series includes two cases and a review of eight others from the Weill Cornell Medicine Dermatopathology databa

CD71+ Red Blood Cells Mediate Increased Erythrophagocytosis and Reduce Monocyte Number in a Monocyte Suspension Assay.

Adverse reactions from donor-recipient sex-mismatched transfusions have been reported in the field of transfusion medicine. Immature CD71+ red blood cells (CD71+ RBCs) may influence these reactions through an immunomodulatory mechanism potentially linked to enriched intracellular reactive oxygen species (ROS). This study was conducted to investigate the effect of CD71+ RBCs on erythrophagocytosis, a common in vitro test for hemolysis risk in incompatible blood transfusions. Enriched CD71+ RBCs and CD71- RBCs were exposed to allogenic peripheral blood mononuclear cells (PBMCs). CD71+ RBCs were

Uptake and nuclear translocation of lactoferrin by porcine monocytes: impact of iron binding, protease activity, and monocyte activation.

Lactoferrin (LF) is a glycoprotein found in neutrophils, milk, and various mammalian secretions that plays a crucial role in host defense by modulating the immune response. Previous studies have shown that LF is taken up by human monocytes and can be present in their nucleus. However, it is unclear whether the iron saturation levels or the protease activity of LF are involved in this uptake and nuclear translocation. In addition, the activation of monocytes might influence these processes. The present study investigated the uptake and nuclear translocation of bovine LF (bLF) and porcine LF (pL

Antibodies to neutrophil cytoplasmic antigens induce monocyte chemoattractant protein-1 secretion from human monocytes.

Antibodies to neutrophil cytoplasmic antigens (ANCA) have been found in the serum samples of patients with a number of vasculitides (e.g., Wegener's granulomatosis, small vessel vasculitis, and idiopathic necrotizing and cresentic glomerulonephritis). Although detection of ANCA in serum samples has proven to be useful diagnostically and in selected activity of disease monitoring situations, the pathogenetic role of ANCA in vasculitis remains ill-defined. We sought to determine whether purified ANCA promotes the secretion of monocyte chemoattractant protein-1 (MCP-1) from isolated human periphe

Receptor-activated calcium influx in human monocytes exposed to monocyte chemotactic protein-1 and related cytokines.

Stimulation of human monocytes with monocyte chemotactic protein-1 (MCP-1) resulted in an increase of [Ca2+]i. The [Ca2+]i rise was dependent on external Ca2+, could be reconstituted by the addition of external Ca2+ and was blocked by Ni2+. Agonist-stimulated Ca2+ influx was demonstrated directly by the use of Mn2+: in the presence of extracellular Mn2+, MCP-1 and FMLP stimulated a dose-dependent quench in fluorescence of Fura-2-loaded monocytes. This quench was the result of stimulation of Mn2+ influx and was blocked by Ni2+ and by the Ca2+ channel inhibitor SC38249. Pretreatment of monocytes

Monocyte-mediated augmentation of human natural killer cell activity: conditions, monocyte and effector cell characteristics.

The characteristics of the effector cells and monocytes, and conditions required for the monocyte-mediated augmentation of human natural killer (NK) cell activity were investigated. Enriched null cell populations were further fractionated by Percoll centrifugation and used as effector cells. The LGL-enriched fraction was less susceptible than either the unfractionated cells or the other Percoll fractions to the monocyte augmentation when mixed with monocytes in the chromium-release assay and when precultured with monocytes for 12 hr, retrieved by carbonyl iron treatment, and tested for NK acti

Serological characterization of human monocytes for HLA, B-lymphocyte, granulocyte, and monocyte-associated antigens by cytotoxicity testing.

Highly enriched preparations of monocytes, B and T lymphocytes, and granulocytes from 18 normal donors were serotyped in parallel in a complement-dependent cytotoxicity assay using allogeneic and heterologous antisera defining three independent tissue antigen systems. HLA and B-lymphocyte tissue antigens were detected on human monocytes although granulocyte antigens were absent. By cytotoxicity testing the presence of Ia-like antigens on monocytes was significantly diminished compared to the autologous B-lymphocyte population and has important implications in B-lymphocyte serology. The study i

Granulocyte-derived Resistin Inhibits Monocyte Maturation and Promotes an Immunosuppressive Phenotype in Chronic Myelomonocytic Leukemia Monocytes.

Chronic myelomonocytic leukemia (CMML) is a hematological neoplasm characterized by expansion of malignant monocytes. However, as a stem cell neoplasm, the disease clone comprises most blood lineages, including granulocytes. To investigate their pathogenic contribution, we performed extensive phenotypic, transcriptomic and functional characterization of CMML granulocytes. These cells exhibited reduced granularity and impaired maturation, the latter of which is associated with poor prognosis. Functionally, CMML granulocytes displayed reduced phagocytic capacity and impaired NETosis, together wi
