Therapeutic intervention with sinomenine and irisin to preserve cardiac function after ischaemia-reperfusion injury.

Despite advancements in reperfusion therapy, myocardial ischaemia-reperfusion (IR) injury remains a major clinical challenge. This study investigated whether a novel dual-target pre-conditioning strategy using sinomenine and irisin could enhance myocardial resistance against IR injury. Ninety male Sprague-Dawley rats were utilized. Protocol 1 evaluated cardioprotection by assigning rats to Sham, IR, sinomenine, irisin or a combination of both agents, with pharmacological pre-treatments administered for 7 days prior to surgery. To model IR injury in vivo, animals underwent 30 min surgical ligat

Sinomenine-glycyrrhizic acid self-assembly enhanced the anti-inflammatory effect of sinomenine in the treatment of rheumatoid arthritis.

Rheumatoid arthritis (RA) is a common chronic systemic autoimmune disease that causes cartilage and bone damage in multiple joints, ultimately leading to disability. There is an urgent need to develop multidimensional strategies to treat RA. Sinomenine (SIN) has the distinctive pharmacological activity in treating RA, but its broader clinical application is limited by its exceedingly short half-life and adverse digestive tract effects. To overcome this obstacle, a self-assembled nanohydrogel (S-G hydrogel) was designed and produced with sinomenine (SIN) and glycyrrhizic acid (GA) without carri

Systematic Structure-Activity Relationship Study of Nalfurafine Analogues toward Development of Potentially Nonaddictive Pain Management Treatments.

Despite the availability of numerous pain medications, the current array of Food and Drug Administration-approved options falls short in adequately addressing pain states for numerous patients and consequently worsens the opioid crisis. Thus, it is imperative for basic research to develop novel and nonaddictive pain medications. Toward addressing this clinical goal, nalfurafine (NLF) was chosen as a lead and its structure-activity relationship (SAR) systematically studied through design, syntheses, and in vivo characterization of 24 analogues. Two analogues, 21 and 23, showed longer durations

Decrease of morphine-CPP by sinomenine via mediation of tyrosine hydroxylase, NMDA receptor subunit 2B and opioid receptor in the zebrafish brain.

To study the effects of sinomenine on conditioned place preference (CPP) zebrafish induced by morphine and expression levels of intracephalic tyrosine hydroxylase (TH), NMDA receptor subunit 2B (NR2B), μ-opioid receptor (zfmor) and δ-opioid receptors (zfdor1 and zfdor2), morphine (40mg/kg) was administrated to zebrafish and the effect of CPP was detected in these zebrafish treated with sinomenine. The expression of TH and NR2B was detected by immunohistochemistry; and the mRNA expression of opioid receptors zfmor, zfdor1 and zfdor2 in the zebrafish brain was assayed by RT-qPCR. In the CPP test

Naloxegol for the treatment of opioid-induced constipation.

With increasing chronic opioid use, opioid-induced constipation (OIC) is a rapidly increasing clinical challenge. Naloxegol, an orally administered, peripherally-acting, µ-opioid receptor antagonist, was developed for the treatment of OIC. This drug profile summarizes published information and presentations at meetings on the effects of naloxegol in OIC. In animal studies, naloxegol was able to inhibit gastrointestinal opioid effects while preserving central analgesic actions and human pharmacodynamic studies were in agreement with such mode of action. Phase II and Phase III studies in patient

Different types of opioid receptors involved in the suppression of LH secretion in lactating sheep.

To learn the involvement of endogenous opioid peptides (EOP) in the regulation of reproductive activity in ruminants, the effects of different opioid antagonists on luteinizing hormone (LH) secretion were determined in sheep during the early stage of lactation. The opioid receptor antagonists: naloxone (all types of receptors, n=5), naloxonazine (μ receptor, n=5), GNTI- (κ receptor, n=5), naltrindole (δ receptor, n=5) or the vehicle (control, n=5) were infused intracerebroventricularly in a series of five 30-min infusions (60μg/60μl) at 30-min intervals. The period of the experiment included t

The influence of opioid antagonists on the discriminative stimulus effects of ethanol.

The aim of the present study was to investigate the role of different endogenous opioid systems in the expression of ethanol's discriminative stimulus effects in a two-lever operant drug discrimination paradigm. Wistar rats trained to make differential responses following the administration of ethanol (1 g/kg, i.p.) or saline. The correct response (fixed-ratio schedule; FR10) resulted in the presentation of food. Once rats had acquired the discrimination an ethanol dose-response test was conducted. The effects of opioid antagonists on the discrimination were assessed by administering the mu-op

Opiates and the anorexia of uremia.

Uremia results in a decrease in food intake. In the present study we investigated whether opiates known to stimulate feeding would alter food intake in rats made uremic by 1 and 5/6 nephrectomy. Morphine increased food intake in sham nephrectomized rats, but failed to alter food intake in uremic animals and depressed the ingestion of rat chow in a group of weight restricted rats. Butorphanol tartrate increased feeding in sham and uremic animals but did not alter intake in the weight restricted group. Higher doses of butorphanol were needed to stimulate feeding in the uremic rats compared to th

A double-blind comparison of intramuscular pethidine and nalbuphine in labour.

A double-blind, between-patient comparison of intramuscular pethidine 100 mg and nalbuphine 20 mg for the relief of pain during labour in 80 patients is described. Severity of pain was assessed before and after treatment by subjective pain scores and visual analogue scales. Neither of these methods showed a significant difference between the treatments. Nalbuphine was associated with less maternal nausea and vomiting than pethidine, but this possible advantage was somewhat offset by a tendency of the drug to produce more maternal sedation and dizziness. The mean umbilical vein/maternal vein ra

Sublingual buprenorphine used postoperatively: clinical observations and preliminary pharmacokinetic analysis.

1 Buprenorphine is a long-acting opiate analgesic. This study was designed to investigate the pharmacokinetics of this drug when given by the sublingual route to ten postoperative patients. Plasma levels of buprenorphine were measured by a specific radioimmunoassay. 2 Plasma levels of the drug following sublingual administration of 0.4 mg showed an apparent delay in absorption and then rose slowly to reach low but significant levels by 3 h. There was considerable variation in the time at which peak levels were achieved. The average systemic availability of the drug by this route was estimated

Effects of Immobilon and Revivon on the cardiovascular system and acid base status of the Göttingen minipig.

Two groups of Göttingen minipigs, aged about five months, were used in these experiments. In the first group of five animals etorphine and acepromazine were administered intramuscularly. In the second group of five animals, atropine was administered in addition. Over a period of 30 min the effects of the drug combination on heart rate, blood pressure, cardiac output, arterial oxygen and carbon dioxide tensions and pH were monitored. Atropine caused a tachycardia rather than a bradycardia and an increase of cardiac output rather than a decrease. Diprenorphine reversed some of the effects of eto

Haemodynamic changes following buprenorphine and morphine.

Twenty-five women were investigated on the day after lower abdominal surgery in a single-blind non-cross-over trial to assess and compare the haemodynamic effects of intravenous injections of buprenorphine 0.3 mg and morphine 7.5 mg. Arterial blood pressure was measured by sphygmomanometry and cardiac output by thoracic impedance cardiography. Arterial blood pressure was significantly reduced following both drugs (p < 0.05), although the mean decrease in systolic arterial pressure was less than 8 mmHg. However, in one patient in each group the decrease was more than 20 mmHg. Cardiac output dec

Carcinomas arising in multilocular thymic cysts of the neck: a clinicopathological study of three cases.

BACKGROUND: The high prevalence rate of malaria due to the proliferation of drug-resistant strains of Plasmodium falciparum could mitigate the realization of sustainable development goals in Nigeria, necessitating research on the discovery of novel chemotherapies against resistant strains of the parasite. However, Solanum anomalum with ethnopharmacological claims could foster this breakthrough, prompting an investigation on the antiplasmodial and hepatoprotective activities of the plant's fruit, semi-purification and characterization of its constituents, as well as in silico studies on its com
