Anabolic steroid treatment increases myofiber damage in mdx mouse muscular dystrophy.

In order to study whether myofiber size is an important determinant of the severity of dystrophic injury, mdx and control mice were treated with an anabolic steroid, nandrolone decanoate, for 3 weeks. Treatment resulted in a population of significantly smaller fibers in both strains, and was accompanied by an increase in the proportionate area or the number of foci of dystrophic injury in mdx soleus (slow-twitch) or tibialis anterior plus extensor digitorum longus (fast-twitch) muscles, respectively. As well, serum creatine kinase activity was increased in steroid-treated mdx mice. Fiber centr

The molecular basis of skeletal muscle weakness in a mouse model of inflammatory myopathy.

OBJECTIVE: It is generally believed that muscle weakness in patients with polymyositis and dermatomyositis is due to autoimmune and inflammatory processes. However, it has been observed that there is a poor correlation between the suppression of inflammation and a recovery of muscle function in these patients. This study was undertaken to examine whether nonimmune mechanisms also contribute to muscle weakness. In particular, it has been suggested that an acquired deficiency of AMP deaminase 1 (AMPD1) may be responsible for muscle weakness in myositis. METHODS: We performed comprehensive funct

Fiber type and fiber size changes in selected thigh muscles six months after low thoracic spinal cord transection in adult cats: exercise effects.

The effects of low thoracic spinal cord transection on muscle weights, fiber type compositions and, fiber cross-sectional areas of selected hind limb muscles were studied. Adult cats were spinalized at T12 and maintained for approximately 6 months. Some spinalized cats were exercised on a treadmill 30 min/day, 5 days/week to determine the role of weight support in maintaining the muscle properties. Spinalization resulted in a significant decrease in the weights of most extensors, whereas the flexors or those that act as both flexors and extensors were maintained near control values. All muscle

Elevated maternal non-esterified fatty acid concentrations during late gestation are associated with altered skeletal muscle development and mitochondrial dynamics related-markers in calves.

BACKGROUND: Late gestation represents an important developmental window for fetal skeletal muscle formation, which is associated with postnatal body growth, metabolic health and mobility in offspring. Although elevated circulating non-esterified fatty acids (NEFA) in dairy cows have been extensively studied in relation to milk production, their associations with fetal and neonatal skeletal muscle development remain unclear. METHODS: Sixty healthy Holstein cows with similar body weight, parity and calving dates were enrolled in this study after dry-off, and retrospectively assigned to low NEFA

Increased Stretching of Mechanoreceptors in Superior Tarsal Muscle Reflexively Contracts Upper Trapezius Muscle as well as Levator Palpebrae Superioris and Occipitofrontalis Muscles as Eye-Eyelid-Eyebrow-Head Coordinated Movements: A Case Series.

To maintain a vertical visual field, fast-twitch fibers in the levator palpebrae superioris muscle (LPSM) stretch mechanoreceptors in the superior tarsal muscle (STM), which contracts the slow-twitch fibers in both the LPSM and the occipitofrontalis muscle (OFM). Exceeding the upgaze limit without head movement increases reflex contraction of the OFM to raise the eyebrows and pull the scalp backward while also causing involuntary contraction of the upper trapezius muscle (UTM) to extend the head backward, resulting in tension-type headaches (TTH) and neck pain. Due to aponeurosis disinsertion

Bioenergetic adaptation of individual human diaphragmatic myofibers to severe COPD.

To assess the effect of severe chronic obstructive pulmonary disease (COPD) on the ability of human diaphragmatic myofibers to aerobically generate ATP relative to ATP utilization, we obtained biopsy specimens of the costal diaphragm from seven patients with severe COPD (mean +/- SE; age 56 +/- 1 yr; forced expiratory volume in 1 s 23 +/- 2% predicted; residual volume 267 +/- 30% predicted) and seven age-matched control subjects. We categorized all fibers in these biopsies by using standard techniques, and we carried out the following quantitative histochemical measurements by microdensitometr

Phenotype-dependent functional and pharmacological properties of BK channels in skeletal muscle: effects of microgravity.

We investigated the involvement of calcium-activated potassium channel (BK) in skeletal muscle phenotype determination and response to acetazolamide, a BK opener. The BKs of slow-twitching soleus (SOL) and fast-twitching flexor digitorum brevis (FDB) muscles of the rat were investigated by patch-clamp technique. The changes of BK properties following muscle disuse were investigated in the hindlimb-unloaded (HU) rat, an animal model of disuse/microgravity. Two functionally different BKs were found in skeletal muscle. The BK of FDB was sensitive to calcium and to acetazolamide, in contrast the B

Muscle-fiber transdifferentiation in an experimental model of respiratory chain myopathy.

INTRODUCTION: Skeletal muscle fiber composition and muscle energetics are not static and change in muscle disease. This study was performed to determine whether a mitochondrial myopathy is associated with adjustments in skeletal muscle fiber-type composition. METHODS: Ten rats were treated with zidovudine, an antiretroviral nucleoside reverse transcriptase inhibitor that induces a myopathy by interfering with mitochondrial functions. Soleus muscles were examined after 21 weeks of treatment. Ten untreated rats served as controls. RESULTS: Zidovudine induced a myopathy with mitochondrial DNA d

Analysis of histochemically and morphometrically in the anterior belly digastric muscle of osteopetrotic (op/op) mice.

The fibers of the anterior belly digastric muscle of mice, fed a granulated diet for various periods, have been studied histochemically and morphometrically. The diameters of the anterior belly digastric fibers in normal mice fed only a granulated diet were smaller than those in mice fed a solid diet. Differences in the succinate dehydrogenase (SDH) activity of muscle fibers between op/op and normal mice gradually appeared in the anterior belly digastric muscle and, by the age of 90 days, under-development of muscle fibers was observed in the mild-belly region of the anterior belly digastric m

Skeletal muscle sympathetic denervation disrupts the neuromuscular junction postterminal organization: A single-cell quantitative approach.

The sympathetic nervous system (SNS) regulates skeletal muscle motor innervation and stabilizes the NMJ in health, disease and aging. Previous studies using both chemical (6-hydroxydopamine, 6-OHDA) and microsurgically-induced sympathetic denervation examined the NMJ organization and transmission in the mouse; however, a detailed quantification of the postterminal on larger hindlimb muscles involved in gait mechanics and posture is lacking. The purpose of this study was to determine whether targets of the sympathetic neuron (SN) exhibiting different intrinsic composition such as the fast-twitc

FTO-dependent m6A regulates muscle fiber remodeling in an NFATC1-YTHDF2 dependent manner.

BACKGROUND: Adolescent idiopathic scoliosis (AIS) is characterized by low lean mass without vertebral deformity. The cause-and-effect relationship between scoliosis and paraspinal muscle imbalance has long puzzled researchers. Although FTO has been identified as a susceptibility gene for AIS, its potential role in the asymmetry of paraspinal muscles has not been fully elucidated. METHODS: We investigated the role of Fto in murine myoblast proliferation, migration, and myogenic differentiation. We examined its precise regulatory influence on murine muscle fiber remodeling in vitro and in vivo.

Fatigue rate of the external anal sphincter.

AIM: Studies of skeletal muscle show that fatigue rate corresponds to the proportion of fast-twitch and slow-twitch fibres that are present in the muscle. Limited work has been done on the fatigue rate of the external anal sphincter. We have prospectively studied fatigability of the external anal sphincter in women with faecal incontinence and women with normal bowel control. METHOD: Anorectal manometry was measured by a station-pull technique using a water-filled microballoon. Fatigue rate was calculated from anal pressure measurements taken every 0.1 s over a 20-s squeeze. RESULTS: Women w

Contractile properties of the human diaphragm in vivo.

The mechanical properties of the human diaphragm have been studied at fractional residual capacity in normal seated subjects with closed glottis. The transdiaphragmatic pressure (Pdi) developed in response to single shocks or to trains of stimuli at increasing frequency was approximately 3 times greater during bilateral than unilateral stimulation. During unilateral phrenic nerve stimulation the Pdi twitches increased as the interval (0-200 ms) of a preceding conditioning stimulus to the contralateral phrenic nerve was decreased suggesting that the two hemidiaphragms are mechanically coupled i

An immunohistochemical evaluation of fast twitch muscle formation induced by repeated administration of flavan 3-ols in mice.

Flavan-3-ols (FL) are poorly bioavailable astringent polyphenols that induce hyperactivation of the sympathetic nervous system. The aim of this study was to investigate the effects of repeated oral administration of FL on mice hindlimb skeletal muscle using immunohistochemical techniques. C57BL/6J male mice were orally administered 50 mg/kg of FL for a period of 2 weeks, and bromideoxyuridine (BrdU) was administered intraperitoneally 3 days prior to the dissection. The soleus and extensor digitorum longus (EDL) were excised and prepared for frozen sections. Myosin heavy chain (MHC) antibodies

Leiomodin-3-deficient mice display nemaline myopathy with fast-myofiber atrophy.

Nemaline myopathy (NM) is one of the most common forms of congenital myopathy, and affects either fast myofibers, slow myofibers, or both. However, an animal model for congenital myopathy with fast-myofiber-specific atrophy is not available. Furthermore, mutations in the leiomodin-3 (LMOD3) gene have recently been identified in a group of individuals with NM. However, it is not clear how loss of LMOD3 leads to NM. Here, we report a mouse mutant in which the piggyBac (PB) transposon is inserted into the Lmod3 gene and disrupts its expression. Lmod3(PB/PB) mice show severe muscle weakness and po

The effects of sustained exercise and hypoxia upon oxygen tensions in the red muscle of rainbow trout.

Teleost fish possess discrete blocks of oxidative red muscle (RM) and glycolytic white muscle, whereas tetrapod skeletal muscles are mixed oxidative/glycolytic. It has been suggested that the anatomy of RM in teleost fish could lead to higher intramuscular O2 partial pressures (PO2) than in mammalian skeletal muscles. This study provides the first direct experimental support for this suggestion by using novel optical fibre sensors to discover a mean (+/- S.E.M., N=6) normoxic steady-state red muscle PO2 (PrmO2) of 61+/-10 mmHg (1 mmHg=133.3 Pa) in free-swimming rainbow trout Oncorhynchus mykis

Morphologic changes in the vastus medialis muscle in patients with osteoarthritis of the knee.

OBJECTIVE: To investigate the frequency of structural changes in the vastus medialis muscle in patients with osteoarthritis (OA) of the knee. METHODS: Specimens of vastus medialis muscle from 78 patients with end-stage OA of the knee undergoing total joint arthroplasty were examined histopathologically. Morphologic changes were assessed in relation to clinical features that might have contributed to muscle injury. RESULTS: All muscle specimens exhibited atrophy of type 2 fibers. In 32% of the patients, atrophy of type 1 fibers was also noted. Fiber type grouping of type 1 fiber in 15% of the

Glycogen synthesis from lactate in a chronically active muscle.

In response to neural overactivity (pseudomyotonia), gastrocnemius muscle fibers from C57Bl/6Jdy2J/dy2J mice have different metabolic profiles compared with normal mice. A population of fibers in the fast-twitch superficial region of the dy2J gastrocnemius stores unusually high amounts of glycogen, leading to an increased glycogen storage in the whole muscle. The dy2J muscle also contains twice as much lactate as normal muscle. A [14C]lactate intraperitoneal injection leads to preferential 14C incorporation into glycogen in the dy2J muscle compared with normal muscle. To determine whether skel

Mechanical properties of red and white swimming muscles as a function of the position along the body of the eel Anguilla anguilla.

The way in which muscles power steady swimming depends on a number of factors, including fibre type and recruitment, muscle strain, stimulation pattern and intensity, and the intrinsic mechanical properties of the muscle fibres. For a number of undulatory swimming fish species, in vivo studies have shown that muscles at different positions along the body are stimulated during different phases of the strain cycle. Moreover, some intrinsic contractile properties of the muscles have been found to vary according to their position along the body. We report the first results on the mechanical proper
