Mycobacterium avium causes atypical mycobacterial infection in humans and animals worldwide. M. avium comprises the subspecies avium (MAA), hominissuis (MAH), silvaticum (MAS) and paratuberculosis (MAP). The M. avium complex (MAC), comprising M. avium and M. intracellulare, causes opportunistic infections of humans. M. avium subsp. avium (MAA) mainly causes avian tuberculosis while subsp. hominissuis (MAH) mainly infects pig. Distinguishing between these two subspecies is essential to the effective control of these atypical mycobacterial infections and minimization of the resulting economic lo

The use of reference genes is commonly accepted as the most reliable approach to normalize qRT-PCR and to reduce possible errors in the quantification of gene expression. The aim of this study was to identify a set of reference genes suitable for gene expression analysis in the distal portion of small intestine and ileocecal valve in cattle. These sites of intestine are of interest in veterinary science as they are the main sites of inflammation caused by Mycobacterium avium subsp. paratuberculosis, agent of paratuberculosis. We employed ten PCR assays for commonly used reference genes belongi

Paratuberculosis is a chronic bowel disease of ruminants caused by Mycobacterium avium subsp. paratuberculosis (MAP). Its secretion through semen highlights the importance of paratuberculosis-free breeding bulls. The breeding and teaser bulls at three semen production units (SPUs) located in Punjab, Pakistan, were screened for the presence of antibodies against MAP. A total of 253 samples were collected from SPUs and a commercially available indirect screen ELISA (Is-ELISA) was applied. Is-ELISA detected antibodies in 20 (24.6%), 16 (22.8%), and 17 (16.6%) samples from SPU-I, SPU-II, and SPU-I

Mycobacterium avium subsp. hominissuis (MAH) causes pulmonary and disseminated infections in immunocompromised humans. In pigs, MAH commonly causes subclinical infections that lead to granulomatous lesions in the mesenteric lymph nodes, liver, spleen, and jejunum. MAH infections have been reported less commonly in other animal species and in birds. Here, we report a case of systemic mycobacteriosis caused by MAH in a sheep. The affected animal had a history of hypoproteinemia, weight loss, and fever. Gross pathology findings included serous fluid accumulation in the peritoneal and pleural cavi

BACKGROUND: The zoonotic potential of Mycobacterium avium ssp. paratuberculosis (MAP) has been debated for almost a century because of similarities between Johne's Disease (JD) in cattle and Crohn's disease (CD) in humans. Our objective was to evaluate scientific literature investigating the potential association between these two diseases (MAP and CD) and the presence of MAP in retail milk or dairy products using a qualitative systematic review.
METHOD: The search strategy included 19 bibliographic databases, 8 conference proceedings, reference lists of 15 articles and contacting 28 topic-re

Cultural and serological studies have provided limited, often conflicting, evidence of a role for mycobacteria in the pathogenesis of Crohn's disease. Interest has focussed on Mycobacterium paratuberculosis, previously considered to be common in the environment with no major role as a human pathogen. Whether a specific serum antibody response to mycobacteria occurs in Crohn's disease or ulcerative colitis was investigated. Sera from patients with Crohn's disease (n = 38), ulcerative colitis (n = 15), and a healthy control population (n = 30) were assayed in an enzyme linked immunosorbent assay

BACKGROUND & OBJECTIVES: Of the three major genotypes of Mycobacterium avium subspecies paratuberculosis (MAP), 'Bison type' is most prevalent genotype in the domestic livestock species of the country, and has also been recovered from patients suffering from Crohn's disease. Recently, a new assay based on IS1311 locus 2 PCR- restriction endonuclease analysis (REA) was designed to distinguish between 'Indian Bison type' and non-Indian genotypes. The present study investigated discriminatory potential of this new assay while screening of a panel of MAP isolates of diverse genotypes and from diff

Paratuberculosis (Johne's disease), a chronic granulomatous enteritis caused by Mycobacterium avium subspecies paratuberculosis (MAP), is difficult to diagnose in the subclinical stage(s) because of intermittent shedding and nonspecific immune responses. Given their stability and potential as biomarkers, exosomal microRNAs (miRNAs) were investigated in this study to identify serum-derived candidates associated with Paratuberculosis-associated disease stages in goats. Exosomes were isolated from the serum of three groups: Control (ELISA- and PCR-negative), ENPP (ELISA-negative but PCR-positive)

BACKGROUND: Mycobacterium avium subspecies paratuberculosis (MAP) is the bacterial pathogen which causes Johne’s disease (JD) in animals and is reported to be significantly associated with Crohn’s disease (CD) in humans. MAP is the causative bacterial pathogen of Johne’s disease in infected animals, including ruminants. Sub-clinically and clinically infected animals shed large numbers of MAP onto pastures. Previous studies have detected MAP in pasture areas deposited by both sub-clinically and clinically infected animals with Johne’s Disease and in areas outside pastures spread by non-farmed a

Paratuberculosis is a chronic disease that poses a threat to the global dairy industry. In the absence of effective treatments and ideal vaccines, screening and culling strategies are the primary means of control. Enzyme-linked immunosorbent assay is the most commonly used method for paratuberculosis screening, and its results have been widely used to estimate the prevalence of paratuberculosis. However, no comprehensive meta-analysis has evaluated the detection performance of ELISA for paratuberculosis in cattle. This systematic review and meta-analysis aimed to fill this gap. We conducted a

BACKGROUND: Paratuberculosis is debilitating chronic enteritis usually characterized by diarrhea, decreased milk production, and progressive cachexia. Mycobacterium avium subspecies paratuberculosis (MAP) causes significant economic losses by affecting dairy herds globally. Development of protective vaccines is considered as one of the most effective controlling measures for MAP infections. In the current study, hydrophilic parts of MAP2191 and FAP-P proteins as two vaccine candidates were analyzed using immunoinformatics approaches.
METHODS: After selecting the most hydrophilic parts of MAP2

The clinical presentation of paratuberculosis in small ruminants is unthriftiness (poor body condition); severe diarrhea is not a common clinical sign. In the USA, goats are primarily infected with bovine strains of paratuberculosis and sheep are primarily infected with ovine strains. Because ovine strains cannot be easily cultured, confirmation of a diagnosis is best done by polymerase chain reaction on tissue or fecal samples. Control programs must be tailored to the business objectives of the herd/flock owner and primarily involved changes in herd management, with diagnostic testing used st

Paratuberculosis is a regional chronic enteritis caused by Mycobacterium avium subsp. paratuberculosis (MAP). The first complete description of this disease was made in 1895, but previous reports on clinical cases compatible with this entity can already be found in the literature of early 1800 s. Its obvious similarities with tuberculosis gave it the name of paratuberculosis, although it was clearly recognized it was not the same entity as that caused by mammal tuberculosis mycobacteria. Eradication has been considered the ideal goal for control of paratuberculosis, but the lack of efficient d

Paratuberculosis, which is also known as Johne's disease, is a chronic, progressive enteric disease of ruminants caused by infection with Mycobacterium paratuberculosis. Cattle become infected with M. paratuberculosis as calves but often do not develop clinical signs until 2 to 5 yr of age. The clinical disease is characterized by chronic or intermittent diarrhea, emaciation, and death. Although animals with clinical disease are often culled from the herd, animals with subclinical paratuberculosis may cause economic losses because of reduced milk production and poor reproductive performance. The cell mediated immune response and ability of immune cells to migrate to the site of infection are both key aspects of protection against many pathogens. Mycobacterium avium subsp. paratuberculosis (MAP) is an intracellular pathogen and the causative agent of paratuberculosis, a chronic wasting disease of ruminants. Current commercial vaccines for paratuberculosis reduce the occurrence of clinical disease but not all animals are protected from infection. Therefore, there is a need to understand the immune responses triggered by these vaccines at the site of infection, in circulating immune

A singular pathogen has been killing animals, contaminating food and causing an array of human diseases. Mycobacterium avium subspecies paratuberculosis (MAP) is the cause of a fatal enteric infectious disease called Johne's (Yo'-nees), a disorder mostly studied in ruminant animals. MAP is globally impacting animal health and imparting significant economic burden to animal agriculture. Confounding the management of Johne's disease is that animals are typically infected as calves and while commonly not manifesting clinical disease for years, they shed MAP in their milk and feces in the interval

Cell-mediated immune responses to Mycobacterium avium subsp. paratuberculosis (MAP) are regulated by various types of T lymphocytes. The aim of this study was to quantitate T cell subsets in the mid-ileum of cows naturally infected with MAP to identify differences during different stages of infection, and to determine whether these subsets could be used as predictors of disease state. Immunofluorescent labeling of T cell subsets and macrophages was performed on frozen mid-ileal tissue sections archived from naturally infected dairy cows in either subclinical or clinical disease status, and non

Mycobacterium avium subspecies paratuberculosis (MAP) is the causative agent of paratuberculosis (ParaTB) also known as Johne's disease (JD) in ruminants, which is characterized by chronic intestinal inflammation. A similar counterpart has been observed in the form of Crohn's disease in humans. The present study is the first trail in goats to understand the peripheral cellular immune responses following experimental MAP infection and vaccination. Fifteen apparently healthy male kids (3-6 months old) of Barbari breed were included in this study. In the experimental study, 5 kids were infected w

Mycobacterium avium ssp. paratuberculosis is the etiologic agent of Johne's disease in ruminants and is hypothesized to be an infectious cause of Crohn's disease, as well as some other human diseases. Due to key knowledge gaps, the potential public health impact of M. paratuberculosis is unknown. This scoping review aims to identify and characterised the evidence on potential sources and vehicles of M. paratuberculosis exposure for humans to better understand how exposure is likely to occur. Evidence from 255 primary research papers is summarized; most examined the prevalence or concentration

Between 2001 and 2004, 14 Austrian free-ranging red deer (Cervus elaphus hippelaphus) infected by Mycobacterium avium species were observed. Eight of the cases were from different geographical regions, and six originated from the same hunting area. The affected animals had signs of diarrhea, severe weight loss, and emaciation. On post-mortem examination, lymphadenitis associated with grossly enlarged mesenteric lymph nodes as well as multiple caseous or purulent nodular lesions in the thickened wall of the intestines were present in all animals. In 10 cases M. avium subsp. avium and in four ca
