Identification and mRNA expression of two 17β-hydroxysteroid dehydrogenase genes in the marine mussel Mytilus galloprovincialis following exposure to endocrine disrupting chemicals.

17β-Hydroxysteroid dehydrogenases (17β-HSDs) are multifunctional enzymes involved in the metabolism of steroids, fatty acids, retinoids and bile acid. In this study, two novel types of 17β-HSDs (named as MgHsd17b10 and MgHsd17b12) were cloned from Mytilus galloprovincialis by using rapid amplification of cDNA ends (RACE) approaches. Sequence analysis showed that MgHsd17b10 and MgHsd17b12 encoded a polypeptide of 259 and 325 amino acids, respectively. Phylogenetic analysis revealed that MgHsd17b10 and MgHsd17b12 were evolutionarily clustered with other invertebrate 17β-HSD type 10 and 17β-HSD t

Real time and non-destructive quantification of volatile organic compounds associated with spoilage bacteria in packed blue mussels (Mytilus edulis) using SIFT-MS.

Psychromonas arctica, Shewanella frigidimarina, and Pseudoalteromonas elyakovii have been identified and characterized as major spoilage bacteria in packed blue mussels (Mytilus edulis). This study aimed to analyze spoilage bacterial growth and to characterize the specific metabolic products of these spoilage bacteria. For this purpose, bacterial solutions were transferred onto irradiated mussel substrates and exposed to air or modified atmospheres (MAs) with the following compositions of (%CO2/O2/N2): (40/60/0) and (25/75/0). These packages were stored at 4 °C for 15 days, with quality monito

A lectin from the mussel Mytilus galloprovincialis has a highly novel primary structure and induces glycan-mediated cytotoxicity of globotriaosylceramide-expressing lymphoma cells.

A novel lectin structure was found for a 17-kDa α-D-galactose-binding lectin (termed "MytiLec") isolated from the Mediterranean mussel, Mytilus galloprovincialis. The complete primary structure of the lectin was determined by Edman degradation and mass spectrometric analysis. MytiLec was found to consist of 149 amino acids with a total molecular mass of 16,812.59 Da by Fourier transform-ion cyclotron resonance mass spectrometry, in good agreement with the calculated value of 16,823.22 Da. MytiLec had an N terminus of acetylthreonine and a primary structure that was highly novel in comparison w

Impact of climate change on the toxicity of bisphenol A in Mytilus galloprovincialis and assessment of phycoremediation using Nannochloropsis salina via a multi-biomarker strategy and modeling.

In the current study, the mussels Mytilus galloprovincialis, exposed to four varying temperatures (17, 20, 23, and 26 °C), were contaminated with 50 μg/L of bisphenol A both with and without Nannochloropsis salina. The toxicity evaluation is determined by quantifying various biomarkers related to oxidative stress, neurotoxicity, and cellular damage. The key findings indicate that the toxicity of bisphenol A is heightened by rising temperature. The impact of bisphenol A is most evident at 26 °C, leading to excessive production of reactive oxygen species, depletion of non-enzymatic antioxidants,

Effects of water temperature increase on gene expression, biochemical, and histopathological responses of the bivalve species Mytilus galloprovincialis to the antineoplastic drug 5-fluorouracil.

The present study aimed to assess the potential ecotoxicological effects of 5-fluorouracil (5-FU), an antineoplastic already found in aquatic ecosystems, towards the bivalve Mytilus galloprovincialis, considering actual and warming temperature scenarios. To this end, organisms were exposed for 28 days to increasing 5-FU concentrations (10, 100, and 500 ng/L) at control (17 ± 1.0 °C) and warming (21 ± 1.0 °C) temperatures. Transcriptomic of selected target genes, biochemical responses, and histopathological alterations were used to assess possible detrimental effects. Results showed an overall

The Mediterranean Mussel (Mytilus galloprovincialis) as Intermediate Host for the Anisakid Sulcascaris sulcata (Nematoda), a Pathogen Parasite of the Mediterranean Loggerhead Turtle (Caretta caretta).

Sulcascaris sulcata (Anisakidae), a pathogenic nematode of sea turtles, may cause ulcerous gastritis with different degrees of severity. Previous studies demonstrated a high prevalence of infection in the Mediterranean loggerhead turtle (Caretta caretta), although no data on the potential intermediate hosts of this nematode has been published thus far from the Mediterranean basin. Here, using molecular analyses, we demonstrated that the cross sections of nematode larvae observed histologically in Mediterranean mussels (Mytilus galloprovincialis) collected from a farm along the Tyrrhenian coast

Elevated water temperatures enhance 2,4,6-trinitrotoluene induced stress response of carbonyl reductase in blue mussels (Mytilus edulis): Another environmental impact of climate change?

Toxic explosives leaking from submerged munitions appear as an emerging marine pollutant in recent years. The nitroaromatic compound 2,4,6-trinitrotoluene (TNT) is of particular interest in this context. TNT poses a threat to marine environments and organisms. Previous studies showed that TNT and its metabolites are incorporated in mussel tissue thereby causing adverse effects through cellular oxidative stress. Recently, the impact of TNT on mRNA expression of the enzyme carbonyl reductase (CR) in blue mussels was discovered. This enzyme plays a key part in the cellular antioxidative system. A

High-resolution LA-ICP-MS imaging combined with attention-enhanced residual network reveal cadmium-induced metal distribution heterogeneity and toxicity in Mytilus galloprovincialis.

Marine bivalves often face heavy metal stress, yet we still lack of high-resolution way to observe the metal pollution status in soft tissue. This study provides the first high-resolution spatial mapping of cadmium (Cd) and associated heavy metals in marine environmental monitor Mytilus galloprovincialis, revealing distinct organ- and cell-specific accumulation, with the digestive gland, especially digestive cells, being the primary site of Cd uptake. Cd co-localizes with several metals, including calcium, chromium, copper, and zinc, suggesting shared uptake pathways and potential synergistic

Immunoenhancement Function of the Novel Hexapeptide (LVVLGH) from Thick-Shelled Mussel (Mytilus coruscus) on Immunodeficient Mice by Activating the NF-κB/MAPK Pathway.

A novel hexapeptide LVVLGH (LH-6) from the thick-shelled mussel (Mytilus coruscus) demonstrated potent immune-enhancing effects in RAW264.7 cells in vitro, but its immunological activity in vivo is unclear. As a result, the present study was designed to investigate the in vivo effects of LH-6 on cyclophosphamide-induced immunodeficient mice. The results demonstrate that LH-6 promoted the growth and development of immunodeficient mice in a concentration-dependent manner, remarkably elevated the immune organ index, and relieved the pathological characteristics of the spleen and thymus. Okadaic acid as a major problem for the seafood safety (Mytilus galloprovincialis) and the dynamics of toxic phytoplankton in the Slovenian coastal sea (Gulf of Trieste, Adriatic Sea).

This article presents the first results on shellfish toxicity in the Slovenian sea (Gulf of Trieste, Adriatic Sea) since the analytical methods for the detection of biotoxins (PSP, ASP, DSP and other lipophilic toxins) in bivalve molluscs were included in the national monitoring program in 2013. In addition to toxins, the composition and abundance of toxic phytoplankton and general environmental characteristics of the seawater (surface temperature and salinity) were also monitored. During the 2014-2019 study period, only lipophilic toxins were detected (78 positive tests out of 446 runs), of w

The value of toxin profiles in the chemotaxonomic analysis of paralytic shellfish toxins in determining the relationship between British Alexandrium spp. and experimentally contaminated Mytilus sp.

Although phytoplankton is ubiquitous in the world's oceans some species can produce compounds that cause damaging effects in other organisms. These include the toxins responsible for paralytic shellfish poisoning, which, in UK waters, are produced by dinoflagellates from the Alexandrium genus. Within Great Britain (GB) a monitoring programme exists to detect this harmful genus as well as the Paralytic Shellfish Poisoning (PSP) toxins in the flesh of shellfish from classified production areas. The techniques used for toxin analysis allow for detailed analysis of the toxin profiles present in co

Experimental study on the bioaccumulation of oxytetracycline and oxolinic acid by the blue mussel (Mytilus edulis). An evaluation of its ability to bio-monitor antibiotics in the marine environment.

The ability of blue mussel (Mytilus edulis) to act as a potential antibiotic bioindicator in marine waters was experimentally tested by the study of the kinetics of two veterinary antibiotics (oxolinic acid: OA and oxytetracycline: OTC). Antibiotic uptake was fast in the soft parts of the mussels. OA was quickly eliminated while OTC was released more slowly (half-life in viscera=3.9 days). OA and OTC were preferentially accumulated in gills and in viscera, respectively. Bio-accumulation factors were low (maximum: 2 for OTC in viscera) in accordance with the low K(ow)s. It was assumed that the

New insights into car tire rubber particle toxicity: chemical composition and ecotoxicity assessment of leachate on gamete quality of the Mediterranean mussel Mytilus galloprovincialis.

Thousands of tire rubber particles (TPs) enter the marine environment every year, contributing to microplastic pollution. The toxicity of TPs can be related to the particles themselves or chemical additives, which can leach into seawater and potentially affect marine organisms. The current study presents new insights into TPs' impact on marine organisms' reproductive processes. The leachates of end-of-life TPs and their adverse effects on gamete quality were evaluated by analysing the chemical compositions of seawater leachates and several gamete physiological parameters, taking the mussel Myt

High amount of dinophysistoxin-3 in Mytilus chilensis collected in Seno de Reloncaví, chile, during massive human intoxication associated with outbreak of Vibrio parahaemolyticus.

This study describes the detection of high amount of 7-O-acyl-derivative dinophysistoxin-1 (Dinophysistoxin-3) in filter bivalves collected on February 2005 in the Seno de Reloncaví, Puerto Montt City, Southern Chile, in the same period of time where an intoxication episode was associated with the presence of Vibrio parahaemolyticus in shellfish. The Diarrhetic Shellfish Poisoning (DSP) mouse bioassay of mussel extract samples, performed as described for regulatory testing, were negative to DSP toxins. Therefore, the same mussel samples collected from 8 places of Seno de Reloncaví were then an

Species identification based on a semi-diagnostic marker: Evaluation of a simple conchological test for distinguishing blue mussels Mytilus edulis L. and M. trossulus Gould.

Cryptic and hybridizing species may lack diagnostic taxonomic characters leaving researchers with semi-diagnostic ones. Identification based on such characters is probabilistic, the probability of correct identification depending on the species composition in a mixed population. Here we test the possibilities of applying a semi-diagnostic conchological character for distinguishing two cryptic species of blue mussels, Mytilus edulis and M. trossulus. These ecologically, stratigraphically and economically important molluscs co-occur and hybridize in many areas of the North Atlantic and the neigh

Comparative study of immune responses in the deep-sea hydrothermal vent mussel Bathymodiolus azoricus and the shallow-water mussel Mytilus galloprovincialis challenged with Vibrio bacteria.

The deep-sea hydrothermal vent mussel Bathymodiolus azoricus and the continental European coast Mytilus galloprovincialis are two bivalves species living in highly distinct marine habitats. Mussels are filter-feeding animals that may accumulate rapidly bacteria from the environment. Contact with microorganism is thus inevitable during feeding processes where gill tissues assume a strategic importance at the interface between the external milieu and the internal body cavities promoting interactions with potential pathogens during normal filtration and a constant challenge to their immune system
