First Report of the Foliar Nematode Aphelenchoides ritzemabosi Infecting Chrysanthemum in Iran.

During a nematode survey on cut flowers in the Pakdasht Region, Tehran Province, Iran, a species of foliar nematode belonging to the genus Aphelenchoides Fischer was detected in leaves of 10- to 11-month-old, greenhouse-grown (26 to 28°C) chrysanthemum (Dendranthema grandiflorum Kitam., cv. Puja) plants. Chrysanthemum leaves appeared discolored and slightly deformed. Diseased plants comprised approximately 40% of all plants in the greenhouse and occurred in scattered clumps along irrigation paths. Spots and blackish brown, irregular, necrotic areas occupied 5 to 50% of the leaf surface. Sympto

Lack of galectin-3 involvement in murine intestinal nematode and schistosome infection.

Many parasitic helminths produce large quantities of glycosylated proteins, some if which are believed to be involved in the skewing towards the dominant Th2 response observed during helminth infection. Galectin-3 is a member of a family of lectin-binding proteins produced by many different types of immune cells, including macrophages. Galectin-3 recognizes the GalNAcbeta1-4GlcNAc (LDN) epitope present on many helminth antigens, including those of the schistosome eggs. Here we show that galectin-3 is not involved in the development of the Th2 response nor in schistosome granuloma formation. Ga

WSX-1: a key role in induction of chronic intestinal nematode infection.

Chronic infection by the gastrointestinal nematode Trichuris muris in susceptible AKR mice, which mount a Th1 response, is associated with IL-27p28 expression in the cecum. In contrast to wild-type mice, mice that lack the WSX-1/IL-27R gene fail to harbor a chronic infection, having significantly lower Th1 responses. The lower level of Ag-specific IFN-gamma-positive cells in WSX-1 knockout (KO) mice was found to be CD4(+) T cell specific, and the KO mice also had increased levels of IL-4-positive CD4(+) T cells. Polyclonal activation of mesenteric lymph node cells from naive WSX-1 KO or wild-t

Stat6 dependent goblet cell hyperplasia during intestinal nematode infection.

To identify the role of signal transducer and activator of transcription factor 6 (Stat6) in the development of intestinal goblet cell hyperplasia during nematode infection, we compared the number of goblet cells in Stat6 deficient (Stat6 -/-) mice with that generated in wild-type (Stat6 +/+) mice in Trichinella spiralis infection. The number of goblet cells significantly increased with infection in wild-type mice. However, Stat6 -/- failed to generate infection-induced goblet cell hyperplasia and a significantly lower number of goblet cells was observed in Stat6 -/- mice on days 14 and 21 pos

Suppression of food intake is linked to enteric inflammation in nematode-infected rats.

Our aim was to investigate the cause-effect relationship between intestinal inflammation induced by infection with enteric stages of Trichinella spiralis and decreased host food intake. A suppression of food intake in T. spiralis-infected rats occurred within the first 24 h postinfection (PI) and was maximized by day 6 PI. Food intake, cumulated over an 8-day PI period, decreased by 59% compared with uninfected animals. The anti-inflammatory glucocorticoid betamethasone 21-phosphate was orally administered to rats in their drinking water to suppress T. spiralis-induced jejunal inflammation. Wh

Tetracycline inhibits development of the infective-stage larvae of filarial nematodes in vitro.

In recent years, studies have linked tetracycline treatment of filaria-infected animals with reduced adult worm burdens and decreased levels of microfilaremia. These observations are believed to be attributable to clearance of Wolbachia, intracellular rickettsial-like organisms found within filarial tissues. Although maximal worm reductions were observed when treatment was initiated early in infection, it is not known whether tetracycline inhibits development of infective-stage larvae. To address this issue, we studied the effect of tetracycline on three different species of filarial nematodes

Cellular immune responses in mice infected with the intestinal nematode Trichuris muris.

CBA and B10.BR mice show variation in immune response to the intestinal nematode Trichuris muris. CBA mice develop strong resistance, eliminating worms from the intestine; B10BR mice are permissive and develop chronic infections. It is already known that resistance and permissiveness reflect differential T helper responses. The data reported here show that resistant CBA mice express good antigen-specific lymphocyte proliferative responses to infection, whereas cells from B10.BR mice are relatively anergic, although still responsive to Concanavalin A (ConA). The possibility that the altered pro

Increased intestinal muscle contractility and worm expulsion in nematode-infected mice.

Intestinal nematode infections are accompanied by mucosal inflammation and an increase in propulsive motor activity that may contribute to parasite eviction from the gut. To examine whether differences in worm expulsion correspond to the increased intestinal muscle contractility that accompanies nematode infection, we studied mice with genetically determined differences in their ability to expel the nematode parasite Trichinella spiralis. Specifically, we examined isometric contraction of longitudinal muscle, worm counts, and inflammation, as measured by myeloperoxidase activity, in two strain

Interleukin-9 is involved in host protective immunity to intestinal nematode infection.

Murine studies have demonstrated that, as with other nematodes, infection with the intestinal nematode Trichinella spiralis is associated with a pronounced intestinal mastocytosis, eosinophilia and an elevation in serum levels of total IgE. Both interleukin (IL)-4 and IL-5 are clearly important in the generation of IgE responses and eosinophilia, respectively, but the control of mucosal mastocytosis in vivo is not as well defined. Mucosal mast cells appear to be particularly important with regard to T. spiralis infections as there is good evidence to suggest their involvement in expulsion of t

Neural integration of jejunal motility and ion transport in nematode-infected ferrets.

Jejunal motility measured manometrically and coupled fluctuations in electrogenic ion transport measured as transmural potential differences (PD) were simultaneously studied in ferrets infected with enteric stages of the parasitic nematode, Trichinella spiralis. Vagotomy in uninfected ferrets abolished jejunal motility clusters and associated PD oscillations. Conversely, in infected ferrets on days 8-12 postinfection (PI), vagotomy did not abolish jejunal motility and PD. Calculated motility indexes (MI) indicated that postvagotomy MI decreased to 12% of prevagotomy MI in uninfected ferrets, w

Carbon dioxide shapes parasite-host interactions in a human-infective nematode.

Skin-penetrating nematodes infect nearly one billion people worldwide. The developmentally arrested infective larvae (iL3s) seek out hosts, invade hosts via skin penetration, and resume development inside the host in a process called activation. Activated infective larvae (iL3as) traverse the host body, ending up as parasitic adults in the small intestine. Skin-penetrating nematodes respond to many chemosensory cues, but how chemosensation contributes to host seeking, intra-host development, and intra-host navigation - three crucial steps of the parasite-host interaction - remains poorly under

Human Infection with the Nematode Gongylonema sp.: A Case Report from Bulgaria.

We report a case of infection with the zoonotic nematode Gongylonema sp. in a 24-year-old woman from Varna, Bulgaria. Two days before seeking medical attention, the patient had noticed a filamentous mass in her buccal mucosa that migrated when touched with the tongue. She manually removed a thread-like worm from the mucosa near her lower lip. Albendazole was prescribed for three days. The patient had no history of travelling abroad or contact with animals. To our knowledge, this is the sixth case that has been reported in the country.

The generation of stable transgenic lines in the human-infective nematode Strongyloides stercoralis.

The skin-penetrating gastrointestinal parasitic nematode Strongyloides stercoralis causes strongyloidiasis, which is a neglected tropical disease that is associated with severe chronic illness and fatalities. Unlike other human-infective nematodes, S. stercoralis cycles through a single free-living generation and thus serves as a genetically tractable model organism for understanding the mechanisms that enable parasitism. Techniques such as CRISPR/Cas9-mediated mutagenesis and transgenesis are now routinely performed in S. stercoralis by introducing exogenous DNA into free-living adults and th

Genome-Wide Analysis of Sheep Artificially or Naturally Infected with Gastrointestinal Nematodes.

The anthelmintic resistance of gastrointestinal nematodes (GINs) poses a significant threat to sheep worldwide, but genomic selection can serve as an alternative to the use of chemical treatment as a solution for parasitic infection. The objective of this study is to conduct genome-wide association studies (GWASs) to identify single nucleotide polymorphisms (SNPs) in Rambouillet (RA) and Dorper × White Dorper (DWD) lambs associated with the biological response to a GIN infection. All lambs were genotyped with a medium-density genomic panel with 40,598 markers used for analysis. Separate GWASs

Botanicals against some important nematodal diseases: Ascariasis and hookworm infections.

Ascariasis and intestinal parasitic nematodes are the leading cause of mass mortality infecting many people across the globe. In light of the various deleterious side effects of modern chemical-based allopathic drugs, our preferences have currently shifted towards the use of traditional plant-based drugs or botanicals for treating diseases. The defensive propensities in the botanicals against parasites have probably evolved during their co-habitation with parasites, humans and plants in nature and hence their combative interference in one another's defensive mechanisms has occurred naturally u

First report about a cerebrospinal nematode infection in an alpaca (Vicugna pacos).

The number of New World camelids in European farms is rising and thus, the need for veterinary care towards these animals arises. However, veterinary care requires sophisticated knowledge on disease and pathogen occurrence within New World camelids. Here, an alpaca cria with neurological signs was admitted to the veterinary clinic. Although the animal was treated with antibiotics, vitamins and dexamethason, it refused to drink milk and the clinical status worsened. After euthanasia, necropsy and histopathological examination were carried out and revealed intracerebral nematode larvae. The morp
