Characteristic neuropathology and plasticity in periventricular leukomalacia.

The brains of extremely low-birth-weight infants with periventricular leukomalacia, who survived for more than 30 days, were examined by means of neuropathologic and immunohistochemical methods. The characteristic neuropathology of the brain is comprised of spongy changes with astrogliosis, a widespread distribution (i.e., in the deep to intermediate white matter), and a diffuse distribution of associated recent lesions. Also, these lesions, both remote and recent, are located in the frontal to occipital lobes. Regarding the correlation between the lesions and transneuronal connecting fibers,

Neuropathology in mice expressing human alpha-synuclein.

The presynaptic protein alpha-synuclein is a prime suspect for contributing to Lewy pathology and clinical aspects of diseases, including Parkinson's disease, dementia with Lewy bodies, and a Lewy body variant of Alzheimer's disease. alpha-Synuclein accumulates in Lewy bodies and Lewy neurites, and two missense mutations (A53T and A30P) in the alpha-synuclein gene are genetically linked to rare familial forms of Parkinson's disease. Under control of mouse Thy1 regulatory sequences, expression of A53T mutant human alpha-synuclein in the nervous system of transgenic mice generated animals with n

Fronto-temporal dementia: nosology, neuropsychology, and neuropathology.

Fronto-temporal dementia is the commonest clinical manifestation of fronto-temporal lobar atrophy. It is associated with three types of underlying histology: microvacuolation or spongiosus (frontal lobe degeneration type), gliosis with or without inclusion bodies and swollen neurones (Pick-type), and motor neurone disease type. The clinical disorder presents in the presenium, is highly familial and is characterized by profound personality change, social incompetence and stereotypic ritualistic behavior. Language becomes attenuated, culminating in total mutism. Visuo-spatial skills are striking

Neuropathology, cell biology, and newer diagnostic methods.

Recent publications emphasized the correlation between molecular events in brain tumors and clinical outcome. Alterations in tumor-suppressor genes, growth factors, and enzyme systems have been previously described in gliomas; the relationships between these various changes and tumor grade, growth rate, and response to therapy are subjects of current investigations. Loss of genetic heterozygosity on chromosomes 10 and 17p, amplification and rearrangement of the epidermal growth factor receptor gene, express ion of its protein product, and changes in chromosomes 9, 19, and the enzyme system pro

Neuropathology of lubag (x-linked dystonia parkinsonism).

Lubag is an x-linked recessive dystonia parkinsonism that affects Filipino men originating principally from the Panay Island. Linkage analysis has confirmed the mode of inheritance and localized the disease gene to the proximal long arm of the x-chromosome. We studied the brain of a 34 year old Filipino man affected with lubag. He developed truncal dystonia at age 30, which subsequently generalized. With disease progression, he also presented with parkinsonism including, rigidity, bradykinesia, and impaired balance. His symptoms were largely unaffected by medication and, at age 34, he underwen

Dementia and neuropathology in Lewy body disease.

In order to elucidate the pathological differences between demented and nondemented patients with Lewy body disease, brains from 35 patients were clinicopathologically examined. In diffuse Lewy body disease, cortical lesions, including numerous Lewy bodies and senile changes, were found to be responsible for the dementia. In some of the cases with the brain-stem type of Lewy body disease, the dementia was attributed to an Alzheimer pathology, while in many cases the various combinations of degeneration in the subcortical nuclei, mainly the nucleus basalis of Meynert and locus ceruleus, played

Neuropathology of human prion diseases (spongiform encephalopathies).

The human prion diseases (spongiform encephalopathies) comprise Creutzfeldt-Jakob disease (CJD), Gerstmann-Sträussler-Scheinker syndrome (GSS), and kuru. Their clinical characteristics are progressive neurological illness with dementia and ataxia as the most prominent signs. The typical neuropathological changes are limited to the central nervous system; they consist of spongiform degeneration, amyloid plaques, astrocytic gliosis, and nerve cell loss. The aetiology of human spongiform encephalopathies remained unclear for many years, until in the 1960s they were finally recognized as transmiss

Case report: neuropathology of methyl bromide intoxication.

The neuropathological findings in a man and his dog both of whom died after acute exposure to methyl bromide are presented. The dog had cerebral oedema, most marked in the depths of cortical sulci. The man survived 30 days after the initial illness and his brain contained well-defined symmetrical lesions in the mammillary bodies and inferior colliculi. He also had a peripheral neuropathy and lymphocytic thyroiditis. The pathology in the central nervous system has features resembling Wernicke's encephalopathology; the mechanism by which methyl bromide may produce such lesions is discussed.

Neuropathology of HIV infection: adults versus children.

The lower incidence of complicating opportunistic and reactivated latent infections in the CNS of children with HIV infection has resulted in a "cleaner" system, allowing better appreciation of the lesions associated with primary HIV brain infection. The most striking differences that we have seen in the CNS of children, when compared to adults with regard to primary HIV infection, have been the following: More florid inflammation and more frequent MGC in the children; more frequent localization of MGC in the cerebral cortex in children; more basophilic mineralization in the children; more per

Neuropathology of cerebral arteriovenous malformations in children.

Neuropathological findings in children who had died of cerebral arteriovenous malformation under 6 years of age were contrasted with those of children aged 6 to 15 years. In all subjects, the abnormalities were more marked in the shunting vessels and veins distal to the arteriovenous shunt than in the arteries. Fibrous thickening, calcification an adherent thrombus of vessel wall, and gliosis and haemosiderin in contiguous neural tissue were more common in the older than the younger children. Children less than 1 week old with vein of Galen malformations presented with congestive heart failure

Strümpell's familial spastic paraplegia: genetics and neuropathology.

Uncomplicated Strümpell's disease (Strümpell's familial spastic paraplegia) with a dominant mode of inheritance is recorded in six families. The neuropathological findings in two cases from these families are given, bringing the total of similar histologically documented reports in the literature to 11. It is concluded that, although exact classification and identification of the many different hereditary neurological degenerative diseases is not yet practicable, cases conforming to the picture described by Strümpell can be separated from larger general group of familial spastic paraplegias, s

Neuropathology of certain forms of mental retardation.

In presenting the neuropathology of asphyxia neonatorum and some related conditions, as illustrated in experiments with monkeys (12), I have tried to make six principal points. (i) Newborn monkeys need not be asphyxiated to the point of terminal apnea to suffer structural brain damage. Mental retardation has not been proved, but neither has it been excluded, in these monkeys. (ii) Asphyxia neonatorum requiring resuscitation of the offspring, which otherwise would die, is associated with a remarkably constant syndrome of bilaterally symmetrical, nonhemorrhagic lesions in thalamic and brain-stem

Chronological Diagnostic Algorithm Predicting Neuropathology in Parkinsonism.

OBJECTIVE: Pre-mortem diagnosis of parkinsonism is often challenging due to atypical presentations, overlapping syndromes, and co-pathologies. This study aimed to develop a machine learning-based algorithm predicting neuropathology in parkinsonism using chronological clinical presentations, which has previously been underexplored. METHODS: Clinical information was automatically abstracted from medical records of the Mayo Clinic Brain Bank using fine-tuned Generative Pre-trained Transformer 4 models. Patients who developed parkinsonism within 3 years of disease onset were included. Six machine

Role of CNTN6 in neurodevelopment and neuropathology.

Contactin 6 (CNTN6) is a recently discovered member of the contactins family, which belongs to a group of cell adhesion molecules. This review summarizes the current knowledge about the possible functions of CNTN6 in the organism and its manifestations in animal models and human diseases. Histological, cellular, and molecular studies in rodents have shown the involvement of this protein in neurite guidance, neural network development, and oligodendrocytogenesis. The expression levels in the cerebellum, hippocampus, and visual cortex of rodents vary depending on the period of neurodevelopment.

Neuropathology of Fatal Falls in Southwestern Ontario.

BACKGROUND: Fall from height is common in all age groups. In 2020 alone, over 6000 people in Canada died from fall-related injuries. Most of the published literature investigating fall-related injuries are often focused on fracture patterns, survival and recovery. Fatal falls are not well studied. The objective of this study is to characterize the demographics and craniocerebral and vertebrospinal injury patterns related to fatal falls within Southwestern Ontario. METHODS: A retrospective case review was conducted at the Department of Pathology, London Health Sciences Centre, for deaths attri
