Biology of the molybdenum cofactor.

The transition element molybdenum (Mo) is an essential micronutrient for plants where it is needed as a catalytically active metal during enzyme catalysis. Four plant enzymes depend on molybdenum: nitrate reductase, sulphite oxidase, xanthine dehydrogenase, and aldehyde oxidase. However, in order to gain biological activity and fulfil its function in enzymes, molybdenum has to be complexed by a pterin compound thus forming the molybdenum cofactor. In this article, the path of molybdenum from its uptake into the cell, via formation of the molybdenum cofactor and its storage, to the final modifi

Chromium(VI) reductase activity is associated with the cytoplasmic membrane of anaerobically grown Shewanella putrefaciens MR-1.

Shewanella putrefaciens MR-1 can reduce a diverse array of compounds under anaerobic conditions, including manganese and iron oxides, fumarate, nitrate, and many other compounds. These reductive processes are apparently linked to a complex electron transport system. Chromium (Cr) is a toxic and mutagenic metal and bacteria could potentially be utilized to immobilize Cr by reducing the soluble and bioavailable state, Cr(VI), to the insoluble and less bioavailable state, Cr(III). Formate-dependent Cr(VI) reductase activity was detected in anaerobically grown cells of S. putrefaciens MR-1, with h

Impacts of carbon source type on metabolic pathways and microbial synergy in the simultaneous anammox and endogenous denitrification process.

Simultaneous anammox and endogenous denitrification (SAED) process enables efficient nitrogen removal from low carbon-to-nitrogen wastewater, yet how carbon source type influences microbial synergy and system robustness remains unclear. This study evaluated the sludge characteristics, performance, and ecological traits of three SAED systems fed with acetate (HAc), propionate (HPr), and glucose (Glc) over 476 days. Results showed that the Glc-fed system achieved the highest and most stable nitrogen removal performance (95.0 % ± 2.4 %), significantly outperforming the HAc-fed (93.3 % ± 2.7 %) an

Foliar folic acid application enhances nitrogen assimilation and improves quality-related metabolites accumulation in Dendrobium officinale.

Folates are essential cofactors in one-carbon metabolism, yet their roles in coordinating nitrogen assimilation and the accumulation of quality-related metabolites in medicinal orchids remain poorly understood. Here, we profiled folate derivatives across tissues and stem developmental stages of Dendrobium officinale and evaluated how foliar nutrient treatments affect folate accumulation and nitrogen metabolism. Folate profiling revealed distinct tissue-specific patterns: leaves were the major sink for total and reduced folates, whereas flowers preferentially accumulated formylated folates. Fol

Tissue-specific cadmium accumulation and its effects on nitrogen metabolism in tobacco (Nicotiana tabaccum, Bureley v. Fb9).

Tobacco (Nicotiana Tabaccum, Bureley v. Fb9) seedlings were grown for 30 days on control medium, and then treated for seven days with different concentrations (0, 10, 20, 50 and 100 muM) of CdCl(2). Cadmium (Cd) was mostly accumulated in the leaves. However, nitrate reductase and nitrite reductase activities (NR, EC 1.6.1.6 and NiR, EC 1.7.7.1) were more inhibited by Cd stress in the roots than in leaves. Glutamine synthetase activity (GS, EC 6.3.1.2) was inhibited by Cd treatment in roots and leaves. In both organs, aminating activity of glutamate dehydrogenase (GDH, EC 1.4.1.2) and protease

Independence of nitrate and nitrite inhibition of Desulfovibrio vulgaris Hildenborough and use of nitrite as a substrate for growth.

Sulfate-reducing microbes, such as Desulfovibrio vulgaris Hildenborough, cause “souring” of petroleum reservoirs through produced sulfide and precipitate heavy metals, either as sulfides or by alteration of the metal reduction state. Thus, inhibitors of these microbes, including nitrate and nitrite ions, are studied in order to limit their impact. Nitrite is a potent inhibitor of sulfate reducers, and it has been suggested that nitrate does not inhibit these microbes directly but by reduction to nitrite, which serves as the ultimate inhibitor. Here we provide evidence that nitrate inhibition o

[Toxicologic and nutritional aspects of nitrates and nitrites].

The toxic action of nitrates and nitrites has often been summarized by methemoglobinemia and nitrosation of amines inducing carcinogenic effects; these two impacts are spectacular, but they hide varying more or less insidious actions recently studied and perceived. In the NO3/NO2 couple, it is accepted that especially nitrite ion lays down a toxicological problem, probably because of its particular chemical reactivity: nitrite ion, which is the base of nitrous acid HNO2 (pKa = 3.36) can react with many functional groups from dietary or endogenous origin; it is also a reducing agent, only oxida

Sediment-isolated Comamonas terrigena strain HJ-2: a novel nitrate-dependent ferrous-oxidizing bacterium with multifunction on pollutant transformation.

Microbially driven Fe(II) oxidation is vital for Fe-cycling processes. In the present study, a novel strain of nitrate-dependent Fe-oxidizing bacteria (FOB) was isolated from the riparian zone sediment of the Hanjiang River, China. It was identified as Comamonas terrigena strain HJ-2. The strain HJ-2 oxidized 2.80 mmol l-1 Fe(II) within 144 h to form Fe(III)/Fe(II) complex on the cell surface using 1.63 mmol l-1 nitrate as an electron acceptor. The formed nitrite from nitrate reduction chemically oxidized Fe(II). Surprisingly, this strain also reduced nitrilotriacetic iron to form 0.5 mmol l-1

Biochemical metabolism of young plants of Ucuúba (Virola surinamensis) in the presence of cadmium.

Virola surinamensis is a forest species widely distributed in the estuaries of the Amazon. These ecosystems are susceptible to contamination by Cadmium (Cd), indicating that the plant has strategies for tolerating this metal. The aim of this study was to assess the nitrogen and carbon metabolism of young plants of Ucuúba (Virola surinamensis) in the presence of cadmium with the perspective of the phytoremediation of contaminated environments. The used experimental design was a completely randomized design with five Cd concentrations (0, 15, 30, 45, and 60 mg L- 1), for 60 days. In general, Cd

The role of alternative oxidase in the maintenance of cellular redox balance under hypoxia via participation in nitric oxide turnover.

Alternative oxidase (AOX) regulates the level of reactive oxygen species and nitric oxide (NO) in plants. While it alleviates NO formation under normoxic conditions, there are several indications that in conditions of low oxygen, such as during seed germination before radicle protrusion, in meristematic stem cells, and in flooded roots, AOX can be involved in the production of NO from nitrite. Whereas the first reports considered this role as indirect, more evidence has since accumulated that AOX can act as a nitrite:NO reductase. Such activity of the structurally similar di-iron proteins in b

Overcoming urea and nitrate inhibition of biological nitrogen fixation in Azotobacter vinelandii versus natural tolerance in Gluconacetobacter diazotrophicus.

Nitrogen is often a limiting nutrient for agricultural crops. Most microbes also require an external fixed-nitrogen source for optimal growth. Even for microbes capable of biological nitrogen fixation (BNF), it is generally the case that they will regulate their metabolism to prioritize assimilation of fixed-nitrogen. Microbes employ various strategies and pathways to take advantage of available fixed-nitrogen compounds found in their natural environments. Azotobacter vinelandii is a model microbe for the study of BNF. Due to the high energetic cost of the process, BNF in A. vinelandii is repr

Anaerobic activation of the entire denitrification pathway in Pseudomonas aeruginosa requires Anr, an analog of Fnr.

The Pseudomonas aeruginosa gene anr, which encodes a structural and functional analog of the anaerobic regulator Fnr in Escherichia coli, was mapped to the SpeI fragment R, which is at about 59 min on the genomic map of P. aeruginosa PAO1. Wild-type P. aeruginosa PAO1 grew under anaerobic conditions with nitrate, nitrite, and nitrous oxide as alternative electron acceptors. An anr deletion mutant, PAO6261, was constructed. It was unable to grow with these alternative electron acceptors; however, its ability to denitrify was restored upon the introduction of the wild-type anr gene. In addition,

Nitrous oxide reduction by two partial denitrifying bacteria requires denitrification intermediates that cannot be respired.

Denitrification is a form of anaerobic respiration wherein nitrate (NO3-) is sequentially reduced via nitrite (NO2-), nitric oxide, and nitrous oxide (N2O) to dinitrogen gas (N2) by four reductase enzymes. Partial denitrifying bacteria possess only one or some of these four reductases and use them as independent respiratory modules. However, it is unclear if partial denitrifiers sense and respond to denitrification intermediates outside of their reductase repertoire. Here, we tested the denitrifying capabilities of two purple nonsulfur bacteria, Rhodopseudomonas palustris CGA0092 and Rhodobact

A multi-omics insight on the interplay between iron deficiency and N forms in tomato.

INTRODUCTION: Nitrogen (N) and iron (Fe) are involved in several biochemical processes in living organisms, and their limited bioavailability is a strong constraint for plant growth and yield. This work investigated the interplay between Fe and N nutritional pathways in tomato plants kept under N and Fe deficiency and then resupplied with Fe and N (as nitrate, ammonium, or urea) through a physiological, metabolomics and gene expression study. RESULTS: After 24 hours of Fe resupply, the Fe concentration in Fe-deficient roots was dependent on the applied N form (following the pattern: nitrate >

Metabolism of reactive nitrogen species in pea plants under abiotic stress conditions.

Nitric oxide (*NO) is a key signaling molecule in different physiological processes of animals and plants. However, little is known about the metabolism of endogenous *NO and other reactive nitrogen species (RNS) in plants under abiotic stress conditions. Using pea plants exposed to six different abiotic stress conditions (high light intensity, low and high temperature, continuous light, continuous dark and mechanical wounding), several key components of the metabolism of RNS including the content of *NO, S-nitrosothiols (RSNOs) and nitrite plus nitrate, the enzyme activities of l-arginine-dep

Lower photorespiration in elevated CO2 reduces leaf N concentrations in mature Eucalyptus trees in the field.

Rising atmospheric CO2 concentrations is expected to stimulate photosynthesis and carbohydrate production, while inhibiting photorespiration. By contrast, nitrogen (N) concentrations in leaves generally tend to decline under elevated CO2 (eCO2 ), which may reduce the magnitude of photosynthetic enhancement. We tested two hypotheses as to why leaf N is reduced under eCO2 : (a) A "dilution effect" caused by increased concentration of leaf carbohydrates; and (b) inhibited nitrate assimilation caused by reduced supply of reductant from photorespiration under eCO2 . This second hypothesis is fully

Characterization and genomics identification of key genes involved in denitrification-DNRA-nitrification pathway of plant growth-promoting rhizobacteria (Serratia marcescens OK482790).

BACKGROUND: A wide variety of microorganisms, including bacteria, live in the rhizosphere zone of plants and have an impact on plant development both favorably and adversely. The beneficial outcome is due to the presence of rhizobacteria that promote plant growth (PGPR). RESULTS: In this study, a bacterial strain was isolated from lupin rhizosphere and identified genetically as Serratia marcescens (OK482790). Several biochemically and genetically characteristics were confirmed in vitro and in vivo to determine the OK482790 strain ability to be PGPR. The in vitro results revealed production of

Nitric oxide and hydrogen sulfide modulate the NADPH-generating enzymatic system in higher plants.

Nitric oxide (NO) and hydrogen sulfide (H2S) are two key molecules in plant cells that participate, directly or indirectly, as regulators of protein functions through derived post-translational modifications, mainly tyrosine nitration, S-nitrosation, and persulfidation. These post-translational modifications allow the participation of both NO and H2S signal molecules in a wide range of cellular processes either physiological or under stressful circumstances. NADPH participates in cellular redox status and it is a key cofactor necessary for cell growth and development. It is involved in signifi

Impact of mixed carbon sources on microbial function and network structure in hypersaline denitrification.

This study evaluated denitrification systems using two carbon sources: glucose alone (G), and a mixture of glucose, methanol, and sodium acetate (GMS), treating synthetic saline wastewater to prove the superior operational efficiency of the mixed carbon sources. Both systems showed excellent nitrogen removal under 80 g-NaCl L-1, but the glucose-based system exhibited unstable nitrate removal at a low C/N ratio of 3. Microbial community composition was distinct between the systems. The GMS system showed the highest organic removal due to the abundance of Marinobacter spp., and the prediction of
