Chemoautotrophic growth of hydrogen-uptake-positive strains of Rhizobium japonicum.

Recently reported research from this laboratory has demonstrated the autotrophic growth of certain hydrogen-uptake-positive strains of Rhizobium japonicum and defined minimal conditions for such growth. Ribulose 1,5-bisphosphate carboxylase has been detected in autotrophically growing cells, but at low specific activity. Moreover, growth rates were low, and growth ceased at low cell densities. We report here improved autotrophic growth rates of R. japonicum SR through the use of a modified mineral salts/vitamins medium and a programmed increase in oxygen tension as autotrophic growth proceeds.

Ambient N2 Reduction to NH3 Electrocatalyzed by Nitrogen-Doped Carbon-Supported CoP Nanomaterials.

At present, more than 80% of ammonia is obtained by the Haber-Bosch process with harsh reaction conditions and high energy consumption. Considering these energy consumption and gas emission problems, it is important to design an environmentally friendly and low-carbon ammonia synthesis method under mild conditions to enhance nitrogen fixation. Ammonia synthesis via an electrocatalytic nitrogen reduction reaction (eNRR) is deemed a promising alternative, but stable and efficient electrocatalysts are highly desired. Herein, nitrogen-doped carbon-supported CoP nanomaterials (CoP@NC) are successfu

Intramolecular internal electric field and activated n-π* electronic transition co-promoted photocatalysis activity of carbon nitride: Mechanism and toxicity evaluation.

Graphitic carbon nitride (CN) exhibits enormous potential in addressing the global energy crisis and environmental issues, while poor charge behavior and insufficient light harvesting capability significantly impede its practical applications. Herein, CN with strong internal electric field and activated n-π* electron transitions was synthesized through one-pot thermal copolymerization of urea and 4,4-Bis(diethylphosphonomethyl)biphenyl. The integrated donor-acceptor structure induces powerful internal electric field, enhancing exciton dissociation efficiency and accelerating charge carrier tra

Electrostatic Confinement of Plasma Electrons by Water Microdroplets Enables Dinitrogen Oxidation.

Nonthermal plasma (NTP) offers exceptional theoretical energy efficiency for nitrogen fixation, yet realizing this potential in practice remains a formidable challenge due to rapid energy dissipation, particularly through low-energy electrons that typically lose their energy as heat. Conventional plasma modulation largely relies on mass-transfer-dominated frameworks, lacking mechanisms to selectively channel these electrons into productive chemical pathways. Here we demonstrate that positively charged water microdroplets constitute active electrostatic confinement interfaces to harvest and re-

Gain-of-Function Engineering of the NifEN Scaffold Into a Self-Assembling Nitrogenase.

Biological nitrogen fixation by Mo-nitrogenase is a complex process requiring coordinated electron transfer through specialized metal clusters. NifEN, an α2β2 heterotetrameric homolog of the nitrogenase catalytic component NifDK, naturally serves as a maturation scaffold but exhibits only trace catalytic activity due to its "imperfect" cluster environments. In this study, we systematically engineered NifEN to reconstruct the structural features of NifDK, specifically targeting the P→M cluster electron relay and the active-site M-cluster environment. Specifically, we developed a series of varia

Functional divergence and symbiotic significance of nitrate reductase isoforms in Medicago truncatula.

Nitrate reductase (NR) is a key enzyme in nitrate assimilation, yet its function within nodules remains poorly understood. In Medicago truncatula, 3 NR genes, MtNR1, MtNR2, and MtNR3, exhibit distinct evolutionary origins and regulatory features. Phylogenetic analyses indicate that NR3-type genes, originated from a duplication of NR1 within the inverted repeat-lacking clade (IRLC) legumes, have lost the conserved phosphorylation sites critical for post-translational regulation. To assess the functional significance of these isoforms, we characterized single and double nr mutants obtained throu

Thaumarchaeotal signature gene distribution in sediments of the northern South China Sea: an indicator of the metabolic intersection of the marine carbon, nitrogen, and phosphorus cycles?

Thaumarchaeota are abundant and active in marine waters, where they contribute to aerobic ammonia oxidation and light-independent carbon fixation. The ecological function of thaumarchaeota in marine sediments, however, has rarely been investigated, even though marine sediments constitute the majority of the Earth's surface. Thaumarchaeota in the upper layer of sediments may contribute significantly to the reservoir of nitrogen oxides in ocean waters and thus to productivity, including the assimilation of carbon. We tested this hypothesis in the northern South China Sea (nSCS), a section of a l

Isolation of rhizobia from Ontario soils that are effective at fixing nitrogen with common bean (Phaseolus vulgaris).

UNLABELLED: Common bean (Phaseolus vulgaris) is an important crop in Canada and globally. Like other legumes, common bean establishes symbiotic interactions with nitrogen-fixing bacteria called rhizobia. However, nitrogen fixation by rhizobia in association with common bean is often suboptimal, constraining its productivity and necessitating the application of nitrogen fertilizer. To support the development of high-performing, locally adapted rhizobial inoculants for Ontario common bean growers, we isolated 216 common bean-nodulating rhizobia from southern Ontario soils using a nodule trapping

Nitrogen addition level is more influential to aboveground productivity than nitrogen addition frequency in an alpine grassland.

AIMS: The combined effects of climate change and human activities have greatly increased atmospheric nitrogen deposition globally, with profound impacts on plant growth and grassland productivity. This study investigates the mechanisms governing how plant aboveground productivity responds to nitrogen addition with varied addition levels and frequencies in Bayanbulak alpine grassland. METHODS: A field experiment was conducted with five N addition levels (0, 5, 10, 15, 20 g·m-2) and two frequencies (low and high). Soil properties, plant aboveground biomass and nutrient content were analyzed usi

Graphitic carbon nitride alleviates cadmium toxicity to soybeans through nitrogen supply.

Graphitic carbon nitride (g-C3N4) is a promising candidate for heavy metal remediation, primarily composed of carbon (C) and nitrogen (N). It has been demonstrated that g-C3N4 adjusts rhizosphere physicochemical conditions, especially N conditions, alleviating the absorption and accumulation of Cadmium (Cd) by soybeans. However, the mechanisms by which g-C3N4 induces N alterations to mitigates plant uptake of Cd remain unclear. This study investigated the impact of g-C3N4-mediated changes in N conditions on the accumulation of Cd by soybeans using pot experiments. It also explored the microbio

Reduced N2 fixation in the tropical Atlantic Ocean during the warm late Pliocene.

Water column denitrification, a major oceanic nitrogen (N) sink, decreased under past warmer-than-present climates, but the response of N2 fixation, the main oceanic N source, remains unresolved. Here we address this gap using foraminifera-bound N isotope records from the tropical Atlantic spanning the warm late Pliocene, a geological analog for projected 21st century warming. Our records show that tropical North Atlantic N2 fixation was reduced during the late Pliocene and had increased markedly by the late Pleistocene, likely due to an increase in water column denitrification, a process that

Contrasting nitrogen-use strategies and photosynthetic carbon-water trade-offs in Pinus tabulaeformis plantations under different forest management modes.

INTRODUCTION: Forest management practices can alter canopy light environments and competitive intensities, thereby reshaping leaf functional traits and photosynthetic physiology. However, how different management modes coordinate leaf morphology, carbon gain, water use, and nitrogen utilization in coniferous plantations remains unclear. METHODS: A field experiment was conducted in a Pinus tabulaeformis plantation under four management modes: structure-based forest management (SM), randomized forest management (RM), close-to-nature forest management (NM), and an unmanaged control (CK). Leaf mo

Reactions of HNO with heme proteins: new routes to HNO-heme complexes and insight into physiological effects.

The formation and interconversion of nitrogen oxides has been of interest in numerous contexts for decades. Early studies focused on gas-phase reactions, particularly with regard to industrial and atmospheric environments, and on nitrogen fixation. Additionally, investigation of the coordination chemistry of nitric oxide (NO) with hemoglobin dates back nearly a century. With the discovery in the early 1980s that NO is biosynthesized as a molecular signaling agent, the literature has been focused on the biological effects of nitrogen oxides, but the original concerns remain relevant. For instan

Glycerol-Based Cryopreservation of Sinorhizobium/Ensifer Strains from Sweet Acacia (Vachellia farnesiana [L.] Wight & Arn.).

The genus Sinorhizobium (Ensifer) exhibits remarkable metabolic versatility and adaptive capacity, establishing effective nitrogen-fixing symbioses with diverse legume crops, including the pioneer shrub Vachellia farnesiana. This association is crucial for ecological restoration in arid and nutrient-deficient Mexican dryland ecosystems and for the remediation of contaminated sites. However, the prolonged preservation of specialized native strains through continuous serial subculturing risks genomic instability and the loss of extrachromosomal replicons harboring essential nod, nif, and heavy m

Frankobactin Metallophores Produced by Nitrogen-Fixing Frankia Actinobacteria Function in Toxic Metal Sequestration.

A series of new metallophores, referred to as frankobactins, were extracted from cultures of the symbiotic and nitrogen-fixing actinobacterium Frankia sp. CH37. Structure elucidation revealed a 2-hydroxyphenyl-substituted oxazoline core and a chain composed of five proteinogenic and nonproteinogenic amino acids, suggesting nonribosomal peptide synthesis as the biosynthetic origin. By whole-genome sequencing, bioinformatic analysis, and comparison with other Frankia strains, the genetic locus responsible for the biosynthesis was detected. Spectrophotometric titration of frankobactin with Fe(III

[Electrooptical properties of soil nitrogen-fixing bacterium Azospirillum brasilense: effect of copper ions].

The effects of copper ions on the uptake of some essential metals in the biomass and the electrooptical properties of cell suspensions of the nitrogen-fixing soil bacterium Azospirillum brasilense sp. 245 were studied. Copper cations were shown to be effectively taken up by the cell biomass from the culture medium. The addition of copper ions increased the rate of uptake of some other metals present in the culture medium. This was accompanied by changes in the electrooptical characteristics of cell suspension as measured within the orienting electric field frequency range of 10 to 10,000 kHz.

Assessment of nitrification potential in ground water using short term, single-well injection experiments.

Nitrification was measured within a sand and gravel aquifer on Cape Cod, MA, using a series of single-well injection tests. The aquifer contained a wastewater-derived contaminant plume, the core of which was anoxic and contained ammonium. The study was conducted near the downgradient end of the ammonium zone, which was characterized by inversely trending vertical gradients of oxygen (270 to 0 microM) and ammonium (19 to 625 microM) and appeared to be a potentially active zone for nitrification. The tests were conducted by injecting a tracer solution (ambient ground water + added constituents)
