Interruption of the cydB locus in Brucella abortus attenuates intracellular survival and virulence in the mouse model of infection.

Brucellosis is characterized by abortion in ruminants and a protracted undulant fever in humans, which often results in severe pathological manifestations. Scant information exists about the molecular mechanisms employed by Brucella abortus to combat host defenses or to persist and replicate within host cells. Transposon (Tn5) mutagenesis of B. abortus and the subsequent screening of mutants for sensitivity to killing in murine macrophages and in the mouse model led to the identification of mutants which were severely attenuated for intracellular survival. One group of mutants was interrupted

Evaluation of toxic effects of aeration and trichloroethylene oxidation on methanotrophic bacteria grown with different nitrogen sources.

In this study we evaluated specific and nonspecific toxic effects of aeration and trichloroethylene (TCE) oxidation on methanotrophic bacteria grown with different nitrogen sources (nitrate, ammonia, and molecular nitrogen). The specific toxic effects, exerted directly on soluble methane monooxygenase (sMMO), were evaluated by comparing changes in methane uptake rates and naphthalene oxidation rates following aeration and/or TCE oxidation. Nonspecific toxic effects, defined as general cellular damage, were examined by using a combination of epifluorescent cellular stains to measure viable cell

HLA molecules in autoimmune diseases.

The association of certain autoimmune diseases with HLA molecules is being refined through the use of sequence-specific oligonucleotide probes and amino acid sequencing, together with continuing elucidation of the functional features of HLA molecules derived from the milestone description by Bjorkman of the HLA molecular structure. The association of insulin-dependent diabetes mellitus and HLA began with weak associations of Class I antigens (B8 and B15) and progressed to Class II antigens (DR3 and DR4), then to subtypes of DR4 (Dw4, 10, and 14), and now to DQ molecules including the absence o

Developmental fates and N2-fixing efficiency of terminally-differentiated versus undifferentiated bacteroids from legume nodules.

Within legume root nodules, rhizobia differentiate into bacteroids, which reduce N2 into NH3 for secretion to the plant. Bacteroids may be swollen and terminally differentiated or non-swollen and can regenerate outside nodules. It is unclear why these different endosymbiotic lifestyles exist and whether they differ in symbiotic efficiency. Here, we compared N2 fixing bacteroids of the near isogenic strains Rhizobium leguminosarum bv. phaseoli 4292 (Rlp4292) and R. leguminosarum bv. viciae A34 (RlvA34), nodulating Phaseolus vulgaris (common bean) and Pisum sativum (pea), respectively. The large

Synergy of Rectangular Truncated Highly Reactive Facets of the Functional Heterometallic Oxo Cage for Enhanced Decomposition of Paraoxon.

Here, we synthesize a highly porous (17.2 nm, pore volume = 0.1753 cm3/g) infinite 3D coordination network of hexanuclear heterometallic mixed metallic oxo cage {MoTI5O2} in which the Ti and Mo centers are interconnected via edge and corner sharing MO6 polyhedra and demonstrate its performance to destroy the organophosphorus-based nerve simulants. The superior performance arises from a synergistic interplay of multiple pathways; defect-engineered facets create unsaturated active sites that facilitate reduction processes and stabilize oxygen vacancies, while ROS-active low-index facets promote

Decoding the adaptive strategies of versatile diazotrophs to multi-metal(loid) stress in mercury-mining impacted farmland soils.

Diazotrophs are crucial for Earth's nitrogen cycle via biological nitrogen fixation, while also modulating other elemental cycles and exhibiting bioremediation potential. However, their responses to co-occurring heavy metal(loid) (HM) contaminants in polluted soils remain poorly understood. Using combined nifH (encoding nitrogenase) amplicon and metagenomic sequencing, we characterized the taxonomic structure and metabolic potential of diazotrophic community across multi-HM contamination gradients in mercury-mining impacted farmlands (paddy vs. upland). Results identified selenium (upland soil

Rhizobia-mediated soybean rhizosphere and nodule endophytic microorganisms reduce bioavailability of Cd and Cu in soil.

Low-level bioavailable cadmium (Cd) and copper (Cu) in agricultural soils poses a severe threat to soil health and food safety; however, the mechanisms by which indigenous Cu-Cd tolerant rhizobia modulate plant-microbe-soil interactions remain poorly understood. In this field trial, the effects of two Cu-Cd tolerant strains, Sinorhizobium xinjiangense YN5 (RB) and Rhizobium pusense GF4 (RD), when inoculated individually and in combination (RC), were assessed with respect to soybean growth, heavy‑metal partitioning, rhizosphere and nodule endophytic microbiomes, and soil functional genes under

Pivotal role of nitric oxide in regulating beneficial plant-microbe interactions under both physiological and stress conditions.

Plant-microbe interactions involve a complex communication network, where microbes can serve as either beneficial partners or harmful pathogens. Nitric oxide (NO) is a crucial signaling molecule that regulates plant growth and development, while enhancing tolerance to both biotic and abiotic stresses. It does this by interacting directly with reactive oxygen species (ROS) and regulating antioxidant enzyme activity through post-translational modifications (PTMs), such as S-nitrosation and nitration. Additionally, NO affects transcription factors that regulate gene expression and modulates phyto

Burkholderia sp. induces functional nodules on the South African invasive legume Dipogon lignosus (Phaseoleae) in New Zealand soils.

The South African invasive legume Dipogon lignosus (Phaseoleae) produces nodules with both determinate and indeterminate characteristics in New Zealand (NZ) soils. Ten bacterial isolates produced functional nodules on D. lignosus. The 16S ribosomal RNA (rRNA) gene sequences identified one isolate as Bradyrhizobium sp., one isolate as Rhizobium sp. and eight isolates as Burkholderia sp. The Bradyrhizobium sp. and Rhizobium sp. 16S rRNA sequences were identical to those of strains previously isolated from crop plants and may have originated from inocula used on crops. Both 16S rRNA and DNA recom

Batch cultivation of Methylosinus trichosporium OB3B: IV. Production of hydrogen-driven soluble or particulate methane monooxygenase activity.

Batch culture conditions were established for the formation of H(2)-driven whole-cell soluble or particulate methane monooxygenase (sMMO or pMMO) activity in the obligate methanotroph, Methylosinus trichosporum Ob3b, to expand its potential uses in groundwater bioremediation and the production of specific chemicals. Addition of either Ni and H(2) to a nitrate-containing minimal salts growth medium or Ni and Mo to a nitrate-lacking growth medium (induces a nitrogenase that generates intracellular H(2)) markedly enhanced both the hydrogenase and the accompanying washed-cell H(2)-driven MMO activ

Insights into soil microbial assemblages and nitrogen cycling function responses to conventional and biodegradable microplastics.

Biodegradable microplastics (MPs) are proposed as sustainable alternatives to conventional MPs, yet their distinct effects on soil microbial communities and ecological functions remain insufficiently understood. This study compares the impacts of biodegradable polylactic acid (PLA) and conventional polyvinyl chloride (PVC) MPs on soil microbial assemblages and nitrogen cycling. Fluorescein diacetate hydrolase (FDAse) activity was temporarily stimulated by 2 % (w/w) PLA and PVC MPs, while 7 % (w/w) PVC MPs initially inhibited FDAse activity before promoting it. PLA MPs (2 % and 7 %, w/w) dramat

Hydrogenase genes from Rhizobium leguminosarum bv. viciae are controlled by the nitrogen fixation regulatory protein nifA.

Rhizobium leguminosarum bv. viciae expresses an uptake hydrogenase in symbiosis with peas (Pisum sativum) but, unlike all other characterized hydrogen-oxidizing bacteria, cannot express it in free-living conditions. The hydrogenase-specific transcriptional activator gene hoxA described in other species was shown to have been inactivated in R. leguminosarum by accumulation of frameshift and deletion mutations. Symbiotic transcription of hydrogenase structural genes hupSL originates from a -24/-12 type promoter (hupSp). A regulatory region located in the -173 to -88 region was essential for prom

In situ transcriptomic analysis of the globally important keystone N2-fixing taxon Crocosphaera watsonii.

The diazotrophic cyanobacterium Crocosphaera watsonii supplies fixed nitrogen (N) to N-depleted surface waters of the tropical oceans, but the factors that determine its distribution and contribution to global N(2) fixation are not well constrained for natural populations. Despite the heterogeneity of the marine environment, the genome of C. watsonii is highly conserved in nucleotide sequence in contrast to sympatric planktonic cyanobacteria. We applied a whole assemblage shotgun transcript sequencing approach to samples collected from a bloom of C. watsonii observed in the South Pacific to un

Insights into the Versatile Sulfur Metabolism of Sulfurovum sp. MH2-6 Isolated from Deep-Sea Hydrothermal Vent Environments.

In deep-sea hydrothermal ecosystems, inorganic sulfur compounds serve as key energy sources for microbes through oxidation, reduction, or disproportionation reactions. However, to date, the bacteria that disproportionate sulfur remain poorly understood. Here, we characterized the physiological and metabolic characteristics of Sulfurovum sp. MH2-6, which was isolated from hydrothermal sediments of the South Mid-Atlantic Ridge. Based on the results of 16S rRNA gene sequence, average nucleotide identity, and DNA-DNA hybridization value, strain MH2-6 belonged to the same species as Sulfurovum mang

Transcriptional landscape of Atractylodes chinensis contracting root rot.

Atractylodes chinensis is among the most economically valuable medicinal plants, serving as the primary source of Atractylodis rhizoma. Its pharmacological activities in treating digestive and urinary disorders have been officially recognized in both the Chinese and Japanese Pharmacopoeias. Whereas, the natural scarcity of Atractylodes chinensis necessitates enhanced cultivation strategies, particularly targeting root rot resistance to ameliorate crop yield and sustainable production. This study implemented controlled cultivation of Atractylodes chinensis and collected rhizome samples exhibiti

Cyanophage lysis reshapes nitrogen cycling and microbiome composition in diazotrophic cyanobacterium Aphanizomenon flos-aquae.

Although it is presumed that viruses play a significant role in nutrient cycling and bacterial community dynamics, this has only rarely been addressed in studies of diazotrophic cyanobacteria. In this study, we therefore examined how cyanophage infection affects the expression of nitrogen (N) cycling genes, N₂ fixation rates, and population structure in the diazotrophic cyanobacterium Aphanizomenon flos-aquae, a prevalent bloom-forming species in temperate brackish and freshwater ecosystems. We also assessed the influence of A. flos-aquae lysis on co-occurring bacterial assemblages throughout
