Lipoyl dehydrogenase catalyzes reduction of nitrated DNA and protein adducts using dihydrolipoic acid or ubiquinol as the cofactor.

Inflamed tissues generate reactive nitrogen oxide species (RNO(x)), such as peroxynitrite (ONOO-)and nitryl chloride (NO2Cl), which lead to formation of nitrated DNA and protein adducts, including 8-nitroguanine (8NG), 8-nitroxanthine (8NX), and 3-nitrotyrosine (3NT). Once formed, the two nitrated DNA adducts are not stable in DNA and undergo spontaneous depurination. Nitration of protein tyrosine leads to inactivation of protein functions and 3NT has been detected in various disease states. We herein report that reduction of these nitro adducts to their corresponding amino analogues can be ca

The effect of oxygen concentration on the teratogenicity of salicylate, niridazole, cyclophosphamide, and phosphoramide mustard in rat embryos in vitro.

Comparisons were made of rat embryos cultured at 5% or 20% oxygen in the presence of salicylate (SAL), cyclophosphamide (CP), niridazole (NDZ), or phosphoramide mustard (PM). Multiple regression analyses were used to compare the effects of drug concentration, oxygen concentration, and the product of drug times oxygen concentration on malformation incidence, viability, and protein content of embryos cultured for 24 hours. Drug concentration significantly affected malformation incidence or severity and protein content (P less than 0.001) for the four drugs tested. Oxygen concentration significan

Mechanistic insights into the efficient degradation and detoxification of chloramphenicol by Clostridium pasteurianum DSM 525.

Chloramphenicol (CAP), a chlorinated nitroaromatic antibiotic, posed significant ecological and human health risks due to its persistence and toxicity. Although microbial degradation represented a promising remediation strategy, its practical application was often constrained by slow degradation kinetics and incomplete biotransformation. In this study, rapid and efficient CAP degradation by the Gram-positive anaerobe Clostridium pasteurianum DSM 525 was demonstrated. The strain achieved 83.6 % removal of 400 mg L-1 CAP within 8 h, and the degradation kinetics followed a first-order model (R2 ≥

The Medicinal Chemistry of 3-nitro-1,2,4-triazoles: Focus on Infectious Diseases.

Infectious diseases are among the leading causes of death worldwide, especially in developing countries. The historical lack of interest of the pharmaceutical industry in developing new drugs against many of these diseases, such as tuberculosis, leishmaniasis, Chagas disease, sleeping sickness, and fungal infections, has left millions of individuals dependent on old treatments that are often ineffective and present different adverse effects. In this sense, new substances against these diseases must be identified. A class of substances that has stood out in the search for new drugs against thes

A systematic review of the literature on mechanisms of 5-nitroimidazole resistance in Trichomonas vaginalis.

BACKGROUND: Trichomonas vaginalis is the most common non-viral sexually transmitted infection. 5-Nitroimidazoles [metronidazole (MTZ) and tinidazole (TDZ)] are FDA-approved treatments. To better understand treatment failure, we conducted a systematic review on mechanisms of 5-nitroimidazole resistance. METHODS: PubMed, ScienceDirect and EMBASE databases were searched using keywords Trichomonas vaginalis, trichomoniasis, 5-nitroimidazole, metronidazole, tinidazole and drug resistance. Non-English language articles and articles on other treatments were excluded. RESULTS: The search yielded 606

Peripheral glia have a pivotal role in the initial response to axon degeneration of peripheral sensory neurons in zebrafish.

Axon degeneration is a feature of many peripheral neuropathies. Understanding the organismal response to this degeneration may aid in identifying new therapeutic targets for treatment. Using a transgenic zebrafish line expressing a bacterial nitroreductase (Ntr)/mCherry fusion protein in the peripheral sensory neurons of the V, VII, IX, and X cranial nerves, we were able to induce and visualize the pathology of axon degeneration in vivo. Exposure of 4 days post fertilization Ntr larvae to the prodrug metronidazole (Met), which Ntr metabolizes into cytotoxic metabolites, resulted in dose-depend

A unique megaplasmid contributes to stress tolerance and pathogenicity of an emergent Salmonella enterica serovar Infantis strain.

Of all known Salmonella enterica serovars, S. Infantis is one of the most commonly isolated and has been recently emerging worldwide. To understand the recent emergence of S. Infantis in Israel, we performed extensive comparative analyses between pre-emergent and the clonal emergent S. Infantis populations. We demonstrate the fixation of adaptive mutations in the DNA gyrase (gyrA) and nitroreductase (nfsA) genes, conferring resistance to quinolones and nitrofurans, respectively, and the carriage of an emergent-specific plasmid, designated pESI. This self-transferred episome is a mosaic megapla

Changes in cecal microbial metabolism of rats induced by individual and a mixture of drinking water disinfection by-products.

Disinfection of drinking water has been one of the greatest public health successes. Numerous halogenated disinfection by-products (DBPs) occur and chronic ingestion has been associated with an increased risk for colorectal cancer in human populations. Because the intestinal microbiota can bioactivate xenobiotics, studies have been performed to examine the effects of individual DBPs on intestinal microbial metabolism. No studies have been conducted on a defined mixture of DBPs to determine if there is an enhancement of response to a mixture. Ten-week-old male Long-Evans rats were treated in th

In Vivo Phenotyping of Dopaminergic Neurodegeneration in Zebrafish Larvae Using Behavioral Analysis and High-Content Imaging.

Drug discovery research in neurodegeneration is constrained by the high cost and low throughput of traditional mammalian models. This bottleneck is particularly observed in Parkinson's disease research, where rigorous and scalable dopaminergic (DA) neurodegeneration studies remain slow and resource intensive. To address this gap, we present a standardized, high-throughput phenotyping pipeline using a transgenic zebrafish model expressing nitroreductase in DA neurons to study DA neuron loss within five days post-fertilization. Zebrafish offer key advantages for translational neuroscience, inclu

Genotoxicity profile of fexinidazole--a drug candidate in clinical development for human African trypanomiasis (sleeping sickness).

The parasitic disease human African trypanomiasis (HAT), also known as sleeping sickness, is a highly neglected fatal condition endemic in sub-Saharan Africa, which is poorly treated with medicines that are toxic, no longer effective or very difficult to administer. New, safe, effective and easy-to-use treatments are urgently needed. Many nitroimidazoles possess antibacterial and antiprotozoal activity and examples such as tinidazole are used to treat trichomoniasis and guardiasis, but concerns about toxicity including genotoxicity limit their usefulness. Fexinidazole, a 2-substituted 5-nitroi

X-ray structure of kijd3, a key enzyme involved in the biosynthesis of D-kijanose.

D-kijanose is an unusual nitrosugar found attached to the antibiotic kijanimicin. Ten enzymes are required for its production in Actinomadura kijaniata, a soil-dwelling actinomycete. The focus of this investigation is on the protein encoded by the kijd3 gene and hereafter referred to as KijD3. On the basis of amino acid sequence analyses, KijD3 has been proposed to be an FAD-dependent oxidoreductase, which catalyzes the sixth step in d-kijanose biosynthesis by converting dTDP-3-amino-2,3,6-trideoxy-4-keto-3-methyl-d-glucose into its C-3' nitro derivative. This putative activity, however, has n

Optimization of the Biocatalysis for D-DIBOA Synthesis Using a Quick and Sensitive New Spectrophotometric Quantification Method.

D-DIBOA (4-hydroxy-(2H)-1,4-benzoxazin-3-(4H)-one) is an allelopathic-derived compound with interesting herbicidal, fungicidal, and insecticide properties whose production has been successfully achieved by biocatalysis using a genetically engineered Escherichia coli strain. However, improvement and scaling-up of this process are hampered by the current methodology for D-DIBOA quantification, which is based on high-performance liquid chromatographic (HPLC), a time-consuming technique that requires expensive equipment and the use of environmentally unsafe solvents. In this work, we established a

Dual activation of azoreductase SaCinD by Copper(II) via upregulated expression and activity enhancement.

Copper can accelerate microbial detoxification of dyes, yet the precise impact of Cu(II) on azoreductase expression and function remains unclear. Here, a Cu(II)-dependent expression of gene SaCinD, encoding azoreductase, was identified from azo-reducing Staphylococcus aureus LZ-01. Flow cytometry and fluorescence microscopy analysis revealed that the copper regulator SaCopR manipulated the gene expression of SaCinD. Moreover, the Km and Vmax values of SaCinD towards Acid Red were 30.1 μM and 1.2 μM/min/mg, with the presence of Cu(II) leading to a 1.5-fold increase in Vmax. ITC analysis confirm

NADPH-depletion micelles sensitizing apoptotic and ferroptotic cancer therapy under hypoxia.

The heterogeneous characteristics of cancer determine that monotherapeutic strategies is difficult to deal with various sub-populations of cancer cells, leading to tumor recrudescence and dissemination. The combination of reactive oxygen species (ROS) mediated apoptosis in photodynamic therapy (PDT) and lipid peroxides induced ferroptosis exhibits remarkable potential in overcoming tumor heterogeneity-driven therapeutic resistance. However, the intracellular antioxidant systems involve the glutathione (GSH), reduced thioredoxin (Trx(SH)2), and ubiquinol (CoQ10H2) can reduce the potency of inte

Transgenic zebrafish as a model for investigating diabetic peripheral neuropathy: investigation of the role of insulin signaling.

Diabetic peripheral neuropathy (DPN), a complication of diabetes mellitus (DM), is a neurodegenerative disorder that results from hyperglycemic damage and deficient insulin receptor (IR) signaling in peripheral nerves, triggered by failure of insulin production and insulin resistance. IR signaling plays an important role in nutrient metabolism and synaptic formation and maintenance in peripheral neurons. Although several animal models of DPN have been developed to identify new drug candidates using cytotoxic reagents, nutrient-rich diets, and genetic manipulations, a model showing beneficial e

Nanomicelles for GLUT1-targeting hepatocellular carcinoma therapy based on NADPH depletion.

Hepatocellular carcinoma (HCC) is a malignant tumor leading cancer-associated high mortality worldwide. Unfortunately, the most commonly used drug therapeutics not only lack of target ability and efficiency, but also exhibit severe systemic toxicity to normal tissues. Thus, effective and targeted nanodrug of HCC therapy is emerging as a more important issue. Here, we design and develop the novel nanomicelles, namely Mannose-polyethylene glycol 600-Nitroimidazole (Man-NIT). This micelle compound with high purity comprise two parts, which can self-assemble into nanoscale micelle. The outer shell
