A qualitative real-time PCR-based method for the detection of the Brazil nut (Bertholletia excelsa) component in food is described. The method consists of DNA isolation by chaotropic SPE and the subsequent PCR with Brazil nut-specific primers and a TaqMan fluorescent probe. The primers and the probe are targeted to the gene encoding for the 11s globulin of the Brazil nut. The method was positive for eight Brazil nut samples from the market and negative for all other tested plant and meat materials used in the food industry (36 samples). The intrinsic LOD of the method was 10 pg Brazil nut DNA.

UNLABELLED: NUT carcinoma is a rare malignant neoplasm characterised by recurrent NUTM1 rearrangements, initially reported in the midline. Recently, 10 cases of cutaneous NUT carcinoma with adnexal differentiation harbouring BRD3::NUTM1, NSD3::NUTM1, BRD4::NUTM1 or BRD3::NUTM2B fusions have been reported. Accordingly, 'NUT adnexal carcinoma' (NAC) has been introduced as a provisional tumour entity to the fifth edition of the WHO Classification of Skin Tumours.
AIMS: We report the histopathological, molecular genetic and epigenetic features of seven additional cases of NAC.

PURPOSE: This study aimed to provide a detailed clinicopathological, molecular genetics, and immune landscape analysis of a large cohort of primary thoracic NUT carcinomas to identify diagnostic challenges, uncover potential therapeutic targets, and define the spatial heterogeneity of the tumor immune microenvironment (TIME).

Bouveret syndrome, a rare complication of cholelithiasis (1%-3%), leads to high mortality (12%-30%). It occurs when a gallstone passes through a cholecystoduodenal fistula, causing gastric outlet obstruction. We report a 66-year-old woman with abdominal pain, nausea, and vomiting. Nuclear protein in testis (NUT) carcinoma is a rare and highly aggressive cancer, characterized by rearrangements involving the NUT gene located on chromosome 15q14. In this report, we present the case of a 52-year-old female diagnosed with primary parotid NUT carcinoma. Despite undergoing surgery, adjuvant chemotherapy, and incomplete regional radiotherapy, the patient succumbed to the disease after an overall survival duration of 7 months. We retrospectively discuss patient clinical and pathological features, as well as therapeutic approaches of NUT carcinoma of the head and neck.

NUT midline carcinoma is a rare malignancy most commonly seen in adolescents and young adults. The disease presents most often in the lung or head and neck area but can be seen occasionally elsewhere. The diagnosis can be difficult and requires a high degree of suspicion with demonstration of the classic fusion rearrangement mutation of the NUTM1 gene with one of a variety of partners by immunohistochemistry, fluorescent in situ hybridization, or genomic analysis. BACKGROUND: Sinonasal NUT carcinoma is an extremely rare, lethal malignancy with limited literature.

An 18-year-old man presented with sudden vision loss in his left eye. Magnetic resonance imaging revealed a tumor that had invaded the left optic nerve, originating from the left posterior ethmoid sinus. Immunohistochemical analyses identified positive staining for NUT protein in the nuclei of tumor cells. AIMS: NUT midline carcinoma (NMC) is a rare undifferentiated and aggressive carcinoma that locates characteristically to the midline of the head and neck, and mediastinum. NMC is characterized by chromosomal rearrangements of the gene NUT, at 15q14. The BRD4 gene on 19q13 is the most common translocation partner forming a fusion oncogene, BRD4-NUT.
