Differential engagement of ORAI1 and TRPC1 in the induction of vimentin expression by different stimuli.

The Ca2+ signal is essential in both hypoxia- and epidermal growth factor (EGF)-mediated epithelial to mesenchymal transition (EMT) in MDA-MB-468 breast cancer cells. This finding suggests that Ca2+-permeable ion channels participate in the induction of expression of some mesenchymal markers such as vimentin. However, the ion channels involved in vimentin expression induction have not been fully characterized. This work sought to define how differential modulation of the calcium signal effects the induction of vimentin and the Ca2+ influx pathways involved. We identified that the intracellular

Antithrombotic potential of blockers of store-operated calcium channels in platelets.

OBJECTIVE: Platelet Orai1 channels mediate store-operated Ca(2+) entry (SOCE), which is required for procoagulant activity and arterial thrombus formation. Pharmacological blockage of these channels may provide a novel way of antithrombotic therapy. Therefore, the thromboprotective effect of SOCE blockers directed against platelet Orai1 is determined. METHODS AND RESULTS: Candidate inhibitors were screened for their effects on SOCE in washed human platelets. Tested antagonists included the known compounds, SKF96365, 2-aminoethyl diphenylborate, and MRS1845 and the novel compounds, Synta66 and

A Novel Strategy for TNF-Alpha Production by 2-APB Induced Downregulated SOCE and Upregulated HSP70 in O. tsutsugamushi-Infected Human Macrophages.

Orientia (O.) tsutsugamushi-induced scrub typhus is endemic across many regions of Asia and the Western Pacific, where an estimated 1 million cases occur each year; the majority of patients infected with O. tsutsugamushi end up with a cytokine storm from a severe inflammatory response. Previous reports have indicated that blocking tumor necrosis factor (TNF)-α reduced cell injury from a cytokine storm. Since TNF-α production is known to be associated with intracellular Ca2+ elevation, we examined the effect of store-operated Ca2+ entry (SOCE) inhibitors on TNF-α production in O. tsutsugamushi-

Serum and glucocorticoid inducible kinase, metabolic syndrome, inflammation, and tumor growth.

Serum-and-glucocorticoid-inducible-kinase-1 (SGK1) is under regulation of several hormones, mediators and cell stressors. More specifically, SGK1 expression is particularly sensitive to glucocorticoids, mineralocorticoids, and TGFβ. Moreover, SGK1 expression is exquisitely sensitive to hypertonicity, hyperglycemia, and ischemia. SGK1 is activated by insulin and growth factors via phosphatidylinositol-3-kinase, 3-phosphoinositide dependent-kinase PDK1, and mTOR. SGK1 up-regulates the Na⁺/K⁺-ATPase, a variety of carriers (e.g. NCC, NKCC, NHE1, NHE3, SGLT1, several amino acid transporters) and ma

CRAC channels as targets for immunotherapy.

Ca²⁺ release-activated Ca²⁺ (CRAC) channels facilitate store-operated Ca²⁺ entry in both immune and nonimmune cells. They are crucial for the function of many immune cell types and strongly associated with the pathophysiology of immune-related disorders. Inherited null mutations in the genes encoding the CRAC channel, ORAI1, and its activator STIM1, are linked to inborn errors of immunity in patients, highlighting the essential role of these channels in immunity. Preclinical studies using knockout mice and CRAC channel inhibitors (CRACi) have further demonstrated their pivotal function in the

Combined diosmin and bisoprolol attenuate cobalt chloride-induced cardiotoxicity and endothelial dysfunction through modulating miR-143-3P/MAPK/MCP-1, ERK5/CXCR4, Orai-1/STIM-1 signaling pathways.

Источник пометил публикацию как отозванную

Even while accelerated cardiomyocyte apoptosis is one of the primary causes of cardiac damage, the underlying mechanism is still mostly unknown. In addition to examining potential protective effects of bisoprolol and diosmin against CoCl2-induced cardiac injury, the goal of this study was to identify potential mechanisms regulating the hypoxic cardiac damage caused by cobalt chloride (CoCl2). For a period of 21 days except Cocl2 14 days from the first day of the experiment, rats were split into the following groups: Normal control group, rats received vehicle only (2 ml/kg/day, p.o.), (Cocl2,

Store-operated CRAC channel inhibitors: opportunities and challenges.

Aberrant Ca(2+) release-activated Ca(2+) (CRAC) channel activity has been implicated in a number of human disorders, including immunodeficiency, autoimmunity, occlusive vascular diseases and cancer, thus placing CRAC channels among the important targets for the treatment of these disorders. We briefly summarize herein the molecular basis and activation mechanism of CRAC channel and focus on discussing several pharmacological inhibitors of CRAC channels with respect to their biological activity, mechanisms of action and selectivity over other types of Ca(2+) channel in different types of cells.

Progressive Elevation of Store-Operated Calcium Entry-Associated Regulatory Factor (SARAF) and Calcium Pathway Dysregulation in Multiple Sclerosis.

Multiple Sclerosis (MS) is a chronic autoimmune disorder characterized by demyelination and neuronal damage in the central nervous system. Dysregulation of calcium homeostasis, particularly through the Store-Operated Calcium Entry-Associated Regulatory Factor (SARAF), has been implicated in MS pathogenesis. This study investigated SARAF, STIM1, and Orai1 expression patterns and their relationship to calcium homeostasis in 45 Bahraini MS patients and 45 matched healthy controls using ELISA and real-time PCR analyses. MS patients showed significantly elevated serum SARAF levels in both early (19

β-carotene alleviates PM2.5-induced pulmonary fibrosis via suppression of the Frizzled5-mediated Wnt5a/Ca2+pathway.

Exposure to fine particulate matter (PM2.5) is a major environmental risk factor for pulmonary fibrosis, yet the involvement of calcium dyshomeostasis and its upstream Wnt signaling regulators in this process remains incompletely understood. This study investigated whether the dietary antioxidant β-carotene (BC) alleviates PM2.5-induced pulmonary fibrosis by modulating the Wnt5a/Ca2+ pathway and, if so, to identify the receptor mediating this effect. We employed a C57BL/6 murine model of intratracheal PM2.5 instillation, alongside BEAS-2B human bronchial epithelial cells subjected to Fzd5 over

Whole Exome Sequencing Reveals Candidate Variants in Ion Channel Genes for Pelvic Muscle Dysfunction in Young Females with a Family History.

INTRODUCTION AND HYPOTHESIS: Pelvic floor dysfunction usually results in pelvic organ prolapse (POP) and/or urinary incontinence. In women, several factors, including pregnancy and vaginal delivery, can affect pelvic muscle conditions. The aim of the study was to perform a genetic analysis in young women with a family history of pelvic floor dysfunction to find potentially harmful variants or variants that increase the risk of developing pelvic floor disorders. METHODS: We employed whole exome sequencing to test ten young women with pelvic floor muscle dysfunction (along with their parents) a

Pathological Mechanism of a Constitutively Active Form of Stromal Interaction Molecule 1 in Skeletal Muscle.

Stromal interaction molecule 1 (STIM1) is the main protein that, along with Orai1, mediates store-operated Ca2+ entry (SOCE) in skeletal muscle. Abnormal SOCE due to mutations in STIM1 is one of the causes of human skeletal muscle diseases. STIM1-R304Q (a constitutively active form of STIM1) has been found in human patients with skeletal muscle phenotypes such as muscle weakness, myalgia, muscle stiffness, and contracture. However, the pathological mechanism(s) of STIM1-R304Q in skeletal muscle have not been well studied. To examine the pathological mechanism(s) of STIM1-R304Q in skeletal musc

Changes in expression of klotho affect physiological processes, diseases, and cancer.

Klotho (KL) encodes a single-pass transmembrane protein and is predominantly expressed in the kidney, parathyroid glands, and choroid plexus. Genetic studies on the KL gene have revealed that DNA hypermethylation is one of the major risk factors for aging, diseases, and cancer. Besides, KL exerts anti-inflammatory and anti-tumor effects by regulating signaling pathways and the expression of target genes. KL participates in modulation of the insulin/insulin-like growth factor-1 (IGF-1) signaling, which induces the growth hormone (GH) secretion. Accordingly, KL mutant mice display multiple aging

Loss of Calpain 3 dysregulates store-operated calcium entry and its exercise response in mice.

Limb-Girdle Muscular Dystrophy R1/2A (LGMD R1/2A) is caused by mutations in the CAPN3 gene encoding Calpain 3, a skeletal-muscle specific, Ca2+-dependent protease. Localization of Calpain 3 within the triad suggests it contributes to Ca2+ homeostasis. Through live-cell Ca2+ measurements, muscle mechanics, immunofluorescence, and electron microscopy (EM) in Capn3 deficient (C3KO) and wild-type (WT) mice, we determined whether loss of Calpain 3 altered Store-Operated Calcium Entry (SOCE) activity. Direct Ca2+ influx measurements revealed loss of Capn3 elicits elevated resting SOCE and increased

The ryanodine receptor agonist 4-chloro-3-ethylphenol blocks ORAI store-operated channels.

BACKGROUND: Depletion of the Ca(2+) store by ryanodine receptor (RyR) agonists induces store-operated Ca(2+) entry (SOCE). 4-Chloro-3-ethylphenol (4-CEP) and 4-chloro-m-cresol (4-CmC) are RyR agonists commonly used as research tools and diagnostic reagents for malignant hyperthermia. Here, we investigated the effects of 4-CEP and its analogues on SOCE. EXPERIMENTAL APPROACH: SOCE and ORAI1-3 currents were recorded by Ca(2+) imaging and whole-cell patch recordings in rat L6 myoblasts and in HEK293 cells overexpressing STIM1/ORAI1-3. KEY RESULTS: 4-CEP induced a significant release of Ca(2+) i

Orai1 mediates ICRAC and SOCE in human placental extravillous trophoblasts and contributes to cell viability, migration, and invasion.

A healthy pregnancy depends on proper placental development, which requires tightly regulated calcium (Ca2+) signaling. In non-excitable cells, store-operated Ca2+ entry (SOCE) is mediated by Ca2+ release-activated Ca2+ (CRAC) channels, composed of Orai1, Orai2 and Orai3 subunits and activated by stromal interaction molecules (STIM1 and STIM2). In this study, we investigated the potential role of CRAC channels in extravillous trophoblast (EVT) function. Using the human placenta-derived EVT cell line HTR8/SVneo, we combined CRISPR/Cas9, immunocytochemistry, confocal microscopy, whole-cell patch

Specific Inhibition of Orai1-mediated Calcium Signalling Resolves Inflammation and Clears Bacteria in an Acute Respiratory Distress Syndrome Model.

Rationale: Acute respiratory distress syndrome (ARDS) has an unacceptably high mortality rate (35%) and is without effective therapy. Orai1 is a Ca2+ channel involved in store-operated Ca2+ entry (SOCE), a process that exquisitely regulates inflammation. Orai1 is considered a druggable target, but no Orai1-specific inhibitors exist to date. Objectives: To evaluate whether ELD607, a first-in-class Orai1 antagonist, can treat ARDS caused by bacterial pneumonia in preclinical models. Methods: ELD607 pharmacology was evaluated in HEK293T cells and freshly isolated immune cells from patients with A

Orai1 Contributes to Platelet-Activating Factor (PAF)-Induced Contraction in Association with Voltage-Dependent Ca2+ Channel-Mediated Mechanisms in Guinea Pig Urinary Bladder Smooth Muscle.

Platelet-activating factor (PAF) is known to induce smooth muscle contraction; however, its mechanisms in urinary bladder smooth muscle (UBSM) remain unclear. We investigated the involvement of Orai1 in PAF-induced contraction in guinea pig UBSM. PAF (10-6 M)-induced contraction was markedly inhibited by the Orai1 inhibitor Synta66 (10-5 M) and the L-type voltage-dependent Ca2+ channel (VDCC) inhibitor verapamil (10-5 M). Similarly, under Ca2+-free conditions with store depletion induced by cyclopiazonic acid (3 × 10-5 M), Ca2+-induced contraction was suppressed by Synta66 and verapamil. In co
