Dental Age and Tooth Development in Orthodontic Patients with Agenesis of Permanent Teeth.

Objective. To compare the development of permanent teeth in a group of children with the congenitally missing permanent teeth (CMPT) and corresponding nonaffected group. Methods. The formation stages of all developing permanent teeth were determined on 345 panoramic radiographs (OPTs) by the method of Haavikko (1970), and dental age was calculated. The paired samples t-test was used to compare the differences between dental age (DA) and chronological age (CA) in those with CMPT and those not affected. Spearman test was used to evaluate the correlation between DA-CA and the number of missing te

Sema3A chemorepellant regulates the timing and patterning of dental nerves during development of incisor tooth germ.

Semaphorin 3A (Sema3A) axon repellant serves multiple developmental functions. Sema3A mRNAs are expressed in epithelial and mesenchymal components of the developing incisor in a dynamic manner. Here, we investigate the functions of Sema3A during development of incisors using Sema3A-deficient mice. We analyze histomorphogenesis and innervation of mandibular incisors using immunohistochemistry as well as computed tomography and thick tissue confocal imaging. Whereas no apparent disturbances in histomorphogenesis or hard tissue formation of Sema3A (-/-) incisors were observed, nerve fibers were p

Tooth development in a model reptile: functional and null generation teeth in the gecko Paroedura picta.

This paper describes tooth development in a basal squamate, Paroedura picta. Due to its reproductive strategy, mode of development and position within the reptiles, this gecko represents an excellent model organism for the study of reptile development. Here we document the dental pattern and development of non-functional (null generation) and functional generations of teeth during embryonic development. Tooth development is followed from initiation to cytodifferentiation and ankylosis, as the tooth germs develop from bud, through cap to bell stages. The fate of the single generation of non-fun

Formation of dermal skeletal and dental tissues in fish: a comparative and evolutionary approach.

Osteichthyan and chondrichthyan fish present an astonishing diversity of skeletal and dental tissues that are often difficult to classify into the standard textbook categories of bone, cartilage, dentine and enamel. To address the question of how the tissues of the dermal skeleton evolved from the ancestral situation and gave rise to the diversity actually encountered, we review previous data on the development of a number of dermal skeletal elements (odontodes, teeth and dermal denticles, cranial dermal bones, postcranial dermal plates and scutes, elasmoid and ganoid scales, and fin rays). A

Dental mineralization.

Extracellular matrix components and cell-derived microstructures are implicated in mineralization processes which occur in dental tissues. The respective role(s) of collagenic and non-collagenic matrix components are reviewed: phosphorylated and non-phosphorylated proteins, proteoglycans and phosphpholipids. Space-filling amphiphilic molecules seem to play an important role in the preorganization and oriented deposition of calcium phosphate on structures serving more or less as passive support in dentine as well as in enamel.

Immunocytochemical localization of choline-phospholipids in postnatal mouse molars.

This localization was studied using anti-choline phospholipid monoclonal IgM antibody (MC22-33F). By immunofluorescence, the apical portions of enamel-secreting ameloblasts and dentine-secreting odontoblasts were positively stained with MC22-33F. By immunoelectron microscopy, lysosomes in preodontoblasts, odontoblasts, preameloblasts and ameloblasts as well as matrix vesicles in dentine were strongly stained with MC22-33F. On the other hand, plasma membrane of these cells did not stain with MC22-33F. The frequency of positively reacting lysosomes in ameloblasts and odontoblasts and of matrix v

[Cysts and tumors of the jaw bone].

The new entities of cysts and odontogenic tumors of the second edition of WHO "Histological Typing of Odontogenic Tumors" (Kramer et al., 1992) are presented. The classification is based on tumor behavior. The benign epithelial odontogenic tumors are classified in relation to tooth development. The malignant odontogenic tumors are subdivided in carcinomas, sarcomas and carcinosarcomas, which are very rare. This international classification has the purpose to make rare cases comparable, so that more detailed knowledge of their characteristics can be achieved. Types of bone tumor that also occur

Case Report: Endodontic management of a fused maxillary lateral incisor and canine with split.

BACKGROUND: Dental fusion, characterized by the joining of two neighboring tooth germs during odontogenesis, is an uncommon condition affecting approximately 0.1% of the permanent dentition. The unusual anatomy of the pulp chamber and the unpredictable root canal system pose significant challenges during endodontic treatment. CASE REPORT: A 47-year-old female patient presented with a 6-month history of spontaneous nocturnal pain in the upper left anterior region. Clinical examination revealed a solitary tooth occupying the positions of teeth 22 and 23; the tooth had a buccal groove and severe

Msx1 mutations: how do they cause tooth agenesis?

Mutations in the transcription factors PAX9 and MSX1 cause selective tooth agenesis in humans. In tooth bud mesenchyme of mice, both proteins are required for the expression of Bmp4, which is the key signaling factor for progression to the next step of tooth development. We have previously shown that Pax9 can transactivate a 2.4-kb Bmp4 promoter construct, and that most tooth-agenesis-causing PAX9 mutations impair DNA binding and Bmp4 promoter activation. We also found that Msx1 by itself represses transcription from this proximal Bmp4 promoter, and that, in combination with Pax9, it acts as a

Exclusion of coding region mutations in MSX1, PAX9 and AXIN2 in eight patients with severe oligodontia phenotype.

PURPOSE: This paper describes the screening of eight patients with severe oligodontia for PAX9 and AXIN2 mutations. SUBJECTS AND METHODS: Anamnestic data and a panoramic radiograph were collected to study the phenotype of eight patients with oligodontia and their first-degree relatives. A blood sample was taken for a mutational screening for PAX9 and AXIN2 mutations. RESULTS: No mutations were discovered, but a unique nucleotide change in a conserved 5' flanking region of PAX9 was revealed. Earlier screening of the same patients for MSX1 mutations also had a negative outcome. CONCLUSIONS: C

Sensory nerve regulates progenitor cells via FGF-SHH axis in tooth root morphogenesis.

Nerves play important roles in organ development and tissue homeostasis. Stem/progenitor cells differentiate into different cell lineages responsible for building the craniofacial organs. The mechanism by which nerves regulate stem/progenitor cell behavior in organ morphogenesis has not yet been comprehensively explored. Here, we use tooth root development in mouse as a model to investigate how sensory nerves regulate organogenesis. We show that sensory nerve fibers are enriched in the dental papilla at the initiation of tooth root development. Through single cell RNA-sequencing analysis of th

Regenerative endodontic treatment of dens in dente in maxillary lateral incisor with immature root: a case report.

Dens invaginatus (DI) is a developmental anomaly as a result of a deepening or invagination of the enamel organ into the dental papilla during tooth development. In addition, DI is a malformation with varying anatomical features, which poses numerous challenges to treatment. Endodontic treatment of dens in dente is one of the most complica-ted cases of DI. Herein, an immature lateral incisor that employed regenerative endodontic treatment was presented. The mentioned tooth was diagnosed with DI, pulp necrosis, and chronic apical periodontitis. Hence, a favorable prognosis has been shown by a 2

Localization of RELM-β/FIZZ2 Is Associated with Cementum Formation.

Resistin-like molecule-β/found in inflammatory zone 2 (RELM-β/FIZZ2) is a cysteine-rich secretory protein that is localized in the epithelium of the gastrointestinal tract and lung alveoli. Previous reports have suggested that this protein regulates glucose metabolism and inflammation. In the present study, to analyze the involvement of RELM-β/FIZZ2 in tooth development, we immunohistochemically examined the localization of RELM-β/FIZZ2 in tooth germs of embryonic days (E) 15-20 and postnatal days (P) 7-42 rats. RELM-β/FIZZ2 was hardly detected in the tooth germ at the bud (E15) stage. However

The Molecular Circuit Regulating Tooth Development in Crocodilians.

Alligators have robust regenerative potential for tooth renewal. In contrast, extant mammals can either renew their teeth once (diphyodont dentition, as found in humans) or not at all (monophyodont dentition, present in mice). Previously, the authors used multiple mitotic labeling to map putative stem cells in alligator dental laminae, which contain quiescent odontogenic progenitors. The authors demonstrated that alligator tooth cycle initiation is related to β-catenin/Wnt pathway activity in the dental lamina bulge. However, the molecular circuitry underlying the developmental progression of
