Human onchocerciasis - commonly known as river blindness - is one of the most devastating yet neglected tropical diseases, leaving many millions in sub-Saharan Africa blind and/or with chronic disabilities. Attempts to eliminate onchocerciasis, primarily through the mass drug administration of ivermectin, remains challenging and has been heightened by the recent news that drug-resistant parasites are developing in some populations after years of drug treatment. Needed, and needed now, in the fight to eliminate onchocerciasis are new tools, such as preventive and therapeutic vaccines. This revi

During a hot Mediterranean summer, an expedition brought parasitologists from Brazil, France, Greece, Italy, and Serbia to a wooded area near Xanthi, Thrace, northeastern Greece, near the Turkish border, on the track of the vector of the little-known nematode Onchocerca lupi. The scientific purposes of the expedition blended then with stories of humans, animals, and parasites in this rural area.

In Latin America, onchocerciasis is targeted for elimination by 2012 through twice-yearly mass treatment of the eligible population with ivermectin. In Guatemala, two of the four historical endemic foci have demonstrated elimination of transmission, following World Health Organization guidelines. Using established guidelines ophthalmological, serological, and entomological evaluations were conducted in 2007-8 to determine the transmission status of onchocerciasis in the Huehuetenango focus. The prevalence of Onchocerca volvulus microfilariae in the anterior segment of the eye in 365 residents

A 3-year-old spayed female dog was admitted with a history of episodes of dyspnoea and coughing. Severe tracheal stenosis was found on radiography. The dog died during an episode of refractory dyspnoea. Necropsy revealed an obstruction of the thoracic part of the trachea because of a chronic granulomatous inflammation protruding into the tracheal lumen. Histological examination revealed nematodes, which were identified as Onchocerca sp. according to their morphological characteristics. In contrast to the common ocular manifestation in dogs, obstructive tracheitis caused by Onchocerca infection

Treatment of onchocerciasis with diethylcarbamazine (DEC) or ivermectin is associated with a posttreatment reaction characterized by fever, tachycardia, hypotension, lymphadenopathy, and pruritus. To investigate the role of the Wolbachia bacterial endosymbiont of Onchocerca volvulus in these reactions, serum samples collected before and after treatment with either anthelmintic were assessed for evidence of Wolbachia DNA. By use of real-time quantitative polymerase chain reaction, Wolbachia DNA was detected in both groups-with significantly higher levels in those who received DEC (P <.0001). In

Onchocerciasis remains an important public health problem throughout much of sub-Saharan Africa. Nigeria is the country whose population is most afflicted by onchocerciasis; however, little is known concerning the epidemiology of onchocerciasis in this country. Previous studies demonstrated that onchocerciasis in West Africa exists in two forms, which differ in their clinical and epidemiologic characteristics. This is believed to be due to the existence of 2 strains of Onchocerca volvulus, the causative agent of onchocerciasis. The O-150 polymerase chain reaction has been developed to differen

Ivermectin is not lethal to the adult worms of Onchocerca volvulus or to those of O. ochengi, a cattle parasite closely related to O. volvulus. Although ivermectin penetrates the nodules in which the adults of these nematodes live, it is not known what levels of the drug enter the worms. Adult male O. ochengi were incubated in [3H]ivermectin in a saturated solution of unlabelled ivermectin (11.44 microM), to measure uptake by the oral and transcuticular routes, and in [3H]inulin, to ascertain if oral ingestion occurs in vitro. Uptake of [3H]ivermectin was high [1040 disintegrations/min (d.p.m.

In the savanna areas of tropical Africa, cattle are frequently infected with the filaria Onchocerca ochengi. This parasite is closely related to Onchocerca volvulus, the causative agent of human onchocerciasis (river blindness), and is capable of developing in the same vector, Simulium damnosum s.l. In North Cameroon, where both O. ochengi and O. volvulus are endemic, we carried out a field study (reported in this and 2 following papers) to examine to which extent the transmission of the 2 parasite species overlap and what influence this has on the epidemiology of human onchocerciasis. In this

We assessed the transmission potential and the mean infected and infective biting density of S. ochraceum in an onchocerciasis endemic community in the southern Mexican state of Chiapas. In addition, the impact of first treatment of ivermectin during the peak of natural infection of host-seeking S. ochraceum populations was also evaluated. Monthly entomological sampling was carried out during the year before treatment to assess the seasonal biting behavior of parous host-seeking S. ochraceum females, and either infected (with whatever stage of O. volvulus) or infective females (with only third

Parasitic nematodes do not multiply in definitive hosts, but they do molt, grow and mature for a certain period after infection, after which they devote their energies almost entirely to egg production. In this review, Sara Lustigman describes key metabolic enzymes that are essential to the development of the larval stages of Onchocerca volvulus in the host, making them potential therapeutic targets.

Eukaryotic transcription factors can be identified and classified according to conserved DNA-binding motifs and conserved regulatory domains. The functional and structural analysis, and the conservation of these genes among metazoans emphasizes the importance of these DNA-binding proteins in development and differentiation. In order to identify genes with common DNA-binding motifs in the genome of Onchocerca volvulus we have cloned a zinc finger encoding gene of the structure C2-H2. Using the Caenorhabditis elegans sex determining tra-1 gene as a probe we have identified two tra-1 related gene

A probable case of vertical transmission of Onchocerca volvulus microfilariae in Dass Bauchi State, Nigeria is presented. A 28-week-old male child delivered by a 39-year-old Fulani woman suffering from onchocerciasis was positive for O. volvulus microfilariae based on skin snip examinations. The child had general body pruritic rash and intermittent fever. Although the child's skin snip on re-examination five weeks later was also positive, the fever had gradually subsided.

"Wild" Simulium bovis females were collected after they had taken blood on a bait oxen with skin microfilariae (mf) of Onchocerca dukei and Onchocerca ochengi. Flies ingested O. dukei mf in high numbers (13.1 mf per blood fed fly) but rarely mf of O. ochengi (3 mf in 60 flies). In 32 flies dissected 14 to 17 hours after the blood meal, a total of 130 O. dukei mf but no of O. ochengi had migrated to the thorax. Six to 9 days after the blood meal, a total of 203 third stage larvae (L3) were found in 61 surviving flies. Invasive larvae from the head measured 738.0 x 17.8 microns and resembled tho

In order to identify Onchocerca volvulus antigens that could be considered as either diagnostic and/or immunoprophylactic, mouse monoclonal antibodies were produced against O. volvulus soluble antigens. Three were selected on the basis of their staining patterns in an indirect fluorescent antibody assay carried out on cryosections of adult O. volvulus. The first monoclonal antibody (K1-159) recognized a cuticular antigen which appeared in IFA to be restricted to the genus Onchocerca. However, neither Western blotting, nor the immunoprecipitation experiments performed on radiolabelled O. volvul

A cloned sequence, pOvs134, was isolated from a genomic library prepared from Onchocerca volvulus of savanna origin in the plasmid pUC9. pOvs134 hybridizes to all the geographic isolates of O. volvulus tested from both the New and the Old World, but not to the species Onchocerca gibsoni, Onchocerca gutturosa, Onchocerca ochengi, Onchocerca cervicalis, the filarial parasites Brugia malayi, or Dirofilaria immitis, nor to human or simuliid DNA. As little as 250 pg of DNA can be detected on a dot blot hybridization, suggesting that pOvs134 is sensitive enough to detect a single third stage larva.

The fine structures of male and female O. volvulus are described. The sexual dimorphism and the age dependent modifications are mentioned in particular. The cuticle of the male worm consists of more layers than that of the female. The epicuticle of the male is wrinkled, whereas the epicuticle of the female forms protuberances. The outer membrane of the hypodermis is folded to form lamellae, which are associated with a layer of mitochondria. The female worms have only small somatic muscle cells. The intestinal cells are filled with concentric spherules, and the intestinal lumen is reduced. Vari
