The fungal kingdom includes at least 6 million eukaryotic species and is remarkable with respect to its profound impact on global health, biodiversity, ecology, agriculture, manufacturing, and biomedical research. Approximately 625 fungal species have been reported to infect vertebrates, 200 of which can be human associated, either as commensals and members of our microbiome or as pathogens that cause infectious diseases. These organisms pose a growing threat to human health with the global increase in the incidence of invasive fungal infections, prevalence of fungal allergy, and the evolution

Methionine (Met) is a nutritionally essential sulfur-containing amino acid (SAA) known for its preponderant role as initiator in protein synthesis. However, other functions for Met in plants are not well described. The implication of this SAA in oxidative stress tolerance has been recently reported, however the mode of action of Met is still poorly understood. Here, we analyzed the elicitor activity of Met in grapevine as well as its effect on Plasmopara viticola resistance. The results show that Met induces hydrogen peroxide (H2O2) generation, a key element in plant defense signaling, and upr

Global agricultural production relies heavily on chemical pesticides, leading to resistance, residues, and pollution, threatening sustainable agriculture. There is an urgent need to develop novel antimicrobial agents from low-toxicity, easily degradable natural products. Using formononetin as a lead compound, acylpiperidine-containing derivatives were synthesized and evaluated against one oomycete, seven plant pathogenic fungi, and four pathogenic bacteria. C25 exhibited superior inhibitory effects against Phytophthora capsici and Xanthomonas oryzaepv.oryzae, with EC50 values of 3.38 and 1.67

Pythiosis is a severe infectious disease caused by the oomycete Pythium insidiosum, affecting both humans and animals and often associated with poor therapeutic outcomes. Although triamcinolone acetonide has been used as an adjunctive therapy for equine pythiosis, its antimicrobial activity remains controversial. This study evaluated in vitro susceptibility of 15 clinical P. insidiosum isolates to triamcinolone acetonide, alone and in combination with azithromycin, itraconazole, or amorolfine hydrochloride, using zoospore-based broth microdilution assays. Triamcinolone acetonide and itraconazo

Non-histone deacetylation is a widespread and reversible post-translational modification (PTM) dynamically regulated by histone acetyltransferases (HATs) and histone deacetylases (HDACs). Although initially characterized as a chromatin-associated mechanism controlling transcription, recent discoveries demonstrate that HDAC-mediated non-histone deacetylation extends far beyond gene regulation, targeting diverse transcription factors and functional proteins across cellular pathways. Here, we summarize advances in Arabidopsis, rice, and wheat recently, highlighting how non-histone deacetylation m

Lysobacter enzymogenes is a beneficial soil bacterium renowned for its potent biocontrol properties, primarily attributed to antimicrobial secondary metabolites such as HSAF and WAP-8294A2, as well as specialized secretion systems. In this study, we investigate the post-transcriptional regulatory roles of the RNA chaperone Hfq in L. enzymogenes OH11 using an integrated multi-omics approach, combining transcriptomic, proteomic, and RNA-binding data. Our comprehensive analysis reveals that Hfq systemically regulates central metabolism and coordinates biocontrol-associated processes, including an

Polyadenylation of pre-mRNAs by poly(A) polymerase (PAPS) is a critical process in eukaryotic gene expression. As found in vertebrates, plant genomes encode several isoforms of canonical nuclear PAPS enzymes. In Arabidopsis thaliana these isoforms are functionally specialized, with PAPS1 affecting both organ growth and immune response, at least in part by the preferential polyadenylation of subsets of pre-mRNAs. Here, we demonstrate that the opposite effects of PAPS1 on leaf and flower growth reflect the different identities of these organs, and identify a role for PAPS1 in the elusive connect

The mRNA differential display technique was used to identify genes from Habanero pepper (Capsicum chinense Jacq.) seedlings whose expression is modified systemically by infection with the oomycete Phytophthora capsici L. Experiments with different oligonucleotide primer combinations revealed that no single gene was synthesised de novo. Instead, the quantitative accumulation of multiple transcripts was found. From these transcripts, levels of a nitrate reductase (Capsicum chinense nitrate reductase, CcNR), which has a high percentage of identity with other Solanaceae NRs, showed a consistent in

Methylxanthines, such as theobromine and caffeine, are alkaloids produced in a variety of widely consumed plant species including tea, coffee, yerba mate, and chocolate. While these compounds are favored by humans for their stimulating properties, plants are thought to produce them primarily for defense purposes. Cacao (Theobroma cacao; the chocolate tree), the source of edible cocoa and chocolate products, is an important crop in the tropics that suffers considerable yield losses due to diseases. Here, we report that a disease-tolerant cacao genotype predominantly accumulates caffeine in youn

Chitin is an essential polysaccharide of the fungal cell wall, critical for structural integrity, cell division and, in pathogenic fungi, virulence. As chitin is absent in both plant and mammalian systems, chitin synthases are considered attractive targets for the specific control of fungal pathogens. Yet despite decades of research, structural information on chitin synthases was lacking and inhibitors have failed to gain approval in the clinic. Current inhibitors are also ineffective against major agricultural pathogens such as Aspergillus and Fusarium species, largely due to the presence of

The crayfish plague is caused by the oomycete Aphanomyces astaci with North American crayfish (for example Pacifastacus leniusculus and Procambarus clarkii) serving as carriers and vectors for this pathogen. This poses a constant threat to native crayfish in Europe, Asia, South America and Australia, which all are highly susceptible to this pathogen. In this study we now show how the symbiotic balance between the pathogen and its host are maintained at the molecular level. The host factors involved in this balance between the pathogen, A. astaci and the host, P. leniusculus, are one glycine-ri

Crayfish plague disease, caused by the oomycete pathogen Aphanomyces astaci represents one of the greatest risks for the biodiversity of the freshwater crayfish. This data article covers the de novo transcriptome assembly and annotation data of the noble crayfish and the marbled crayfish challenged with Ap. astaci. Following the controlled infection experiment (Francesconi et al. in Front Ecol Evol, 2021, https://doi.org/10.3389/fevo.2021.647037 ), we conducted a differential gene expression analysis described in (Boštjančić et al. in BMC Genom, 2022, https://doi.org/10.1186/s12864-022-08571-z

Several isolates of an unknown oomycete resembling the genus Aphanomyces were obtained into laboratory culture from samples of noble crayfish (Astacus astacus) in 2016-2017. The crayfish were kept in cages in connection with a study on an eventually persistent crayfish plague infection in a small Finnish lake, following an acute episode of the disease in 2010. Despite the close resemblance of the isolates to the causative agent of crayfish plague, Aphanomyces astaci, and the positive results obtained in OIE recommended A. astaci-specific ITS-based conventional PCR and qPCR molecular assays, th

The pathogenic oomycete Aphanomyces astaci, transmitted mainly by invasive North American crayfish, causes the crayfish plague, a disease mostly lethal for native European crayfish. Due to its decimating effects on native crayfish populations in the last century, A. astaci has been listed among the 100 worst invasive species. Importantly, detecting the pathogen in endangered native crayfish populations before a disease outbreak would provide a starting point in the development of effective control measures. However, current A. astaci-detection protocols either rely on degradation-prone eDNA is

Mechanistically, nonhost resistance of Arabidopsis thaliana against the oomycete Phytophthora infestans is not well understood. Besides PEN2 and PEN3, which contribute to penetration resistance, no further components have been identified so far. In an ethylmethane sulphonate-mutant screen, we mutagenized pen2-1 and screened for mutants with an altered response to infection by P. infestans. One of the mutants obtained, enhanced response to Phytophthora infestans6 (erp6), was analyzed. Whole-genome sequencing of erp6 revealed a single nucleotide polymorphism in the coding region of the kinase do

Two possible means of transmission of crayfish plague were investigated: via fish (as vectors), and via crayfish (as hosts or vectors when dead). The crayfish transmission experiments focussed on both the viability of the fungus in dead crayfish when kept in simulated field conditions, and on the treatments which kill viable forms of Aphanomyces astaci within the recently dead host (cadaver). It was found that A. astaci remains viable for 5 d, and possibly longer in crayfish kept in water at 21 degrees C after dying of crayfish plague. Heating (boiling for 1 min) was the quickest way of decont

Lagenidium giganteum, a facultative parasite of mosquito larvae, cannot synthesize sterols, and requires an exogenous source of these lipids in order to enter its reproductive cycle. This parasite grows vegetatively in the absence of sterols, but requires cholesterol or structurally related compounds to produce motile zoospores, which are the only stage capable of infecting mosquitoes. Sterols structurally related to cholesterol and some steryl esters inhibited the activity of L. giganteum phospholipase A2 (PLA2), an enzyme that hydrolyzes fatty acids from the sn-2 position of glycerophospholi

Microbial cellulose (MC) is a natural biopolymer distinguished by its chemical purity, nanofibrillar architecture, eco-friendliness, biodegradability, and mechanical properties. These distinctive features of MC have expanded the potential applications across diverse fields. However, its transition from laboratory-scale production to industrial manufacturing remains constrained by its low productivity, high production costs, genetic instability of the cellulose-producing strains, accumulation of inhibitory by-products, and inadequate oxygen supply during the fermentation process. These limitati

Bacterial wilt caused by Ralstonia solanacearum is a destructive soil-borne disease affecting numerous crops worldwide. Although synthetic pesticides and antibiotics are commonly used for disease management, their excessive application has raised concerns regarding environmental pollution, human health risks, and the emergence of antibiotic-resistant pathogens. As a sustainable alternative, rhizosphere microorganisms have gained attention as potential biocontrol agents. In our previous study, strain JCK-8401 exhibited strong antibacterial activity against R. solanacearum. In the present study,

The Phosphorus (III) derivatives, named Phosphonates, include congeners with properties as fungicides that are effective in controlling Oomycetes. Examples are organic compounds like Fosetyl-Al [Aluminium tris-(ethylphosphonate)] and salts formed with the anion of phosphonic acid [(OH)2HPO] and Potassium, Sodium and Ammonium cations. According to IUPAC, the correct nomenclature for these compounds is "phosphonates", but in common language and scientific literature they are often named "phosphites", creating ambiguity. The European legislation restricts the use of phosphonates, with the ban for
