Optimism Bias in the Design of Phase III Randomized Control Trials Evaluating PD-1/PD-L1 Targeting Monoclonal Antibodies.

BackgroundMany randomized control trials (RCTs) evaluating programmed death receptor-1 (PD-1)/programmed death ligand-1 (PD-L1) targeting monoclonal antibodies (mAbs) have been completed or are in progress. We examined hypothesized hazard ratios (HHRs) and observed hazard ratios (OHRs) from published RCTs evaluating these mAbs.MethodsPublications of RCTs evaluating at least one PD-1/PD-L1 targeting mAbs approved by the US Food and Drug Administration were identified through PubMed searches. The primary reports of RCTs were retrieved. Two investigators extracted HHR, OHR for the primary endpoin

Optimizing Cannabis Sample Preparation to Reduce Chloroform Usage: A Comparison of I-Optimal, D-Optimal, and A-Optimal Designs.

The aim of this study was to identify a safer solvent system to reduce toxic chloroform usage, consisting of a mixture of acetonitrile, methanol, and water, for the preparation of cannabis samples. A comparison among I-optimal, D-optimal, and A-optimal experimental designs for Charlotte's Angel and Hang Kra Rog Phu Phan cultivars was conducted. Results indicated that the I-optimal and D-optimal designs exhibited similar contour plot patterns and design spaces, which differed from those of the A-optimal design. The optimal solvent mixture that maximized both cannabidiol and ∆9-tetrahydrocannabi

Undetectable = Untransmittable (U = U) Awareness and HIV Treatment Optimism Among Men Who Have Sex with Men in Mumbai, India.

Little is known about Undetectable=Untransmittable (U = U) awareness among MSM in India. We analyzed baseline survey data from MSM (N = 1004) in an mHealth HIV testing trial in Mumbai and Thane, India. We assessed factors associated with U = U awareness and agreement (HIV treatment optimism) using multivariable Poisson regression. U = U awareness (29%) was associated with English survey completion, searching for sexual health information online, awareness of PrEP/PEP, and participation in MSM-focused HIV prevention programs (all P < 0.05). Overall, 50% of the participants agreed with HIV treat

Socioeconomic, Behavioural, and Social Health Correlates of Optimism and Pessimism in Older Men and Women: A Cross-Sectional Study.

BACKGROUND: Optimism is a disposition characterised by positive future expectancies, while pessimism is characterised by expecting the worst. High optimism and low pessimism promote the health of older adults and may potentiate full engagement in life. We identified socioeconomic, behavioural, and social factors associated with optimism and pessimism in older adults. METHODS: Participants included 10,146 community-dwelling, apparently healthy Australian adults aged 70 years and over from the ASPREE Longitudinal Study of Older Persons (ALSOP). Optimism and pessimism were measured using the rev

Optimism as predictor of patient-reported outcomes in patients with an implantable cardioverter defibrillator (data from the WEBCARE study).

OBJECTIVE: The implantable cardioverter defibrillator (ICD) is the treatment of choice for prevention of sudden cardiac death. However, a subgroup of ICD patients experiences psychological adjustment problems post implant. To date, positive psychological constructs (e.g. optimism) have been understudied in this population. Hence, we examined the association between optimism and anxiety, depression, and health status at 12-months post implant. METHODS: Patients (N=171) enrolled in the WEB-based distress management study for ICD patients were included in the analyses. Optimism and pessimism (LO

Optimism and depression: a new look at social support as a mediator among women at risk for breast cancer.

OBJECTIVE: Breast cancer risk is a chronic stressor associated with depression. Optimism is associated with lower levels of depression among breast cancer survivors. However, to our knowledge, no studies have explored the relationship between optimism and depression among women at risk for breast cancer. We hypothesized that women at risk for breast cancer who have higher levels of optimism would report lower levels of depression and that social support would mediate this relationship. METHOD: Participants (N = 199) with elevated distress were recruited from the community and completed self-r

Optimism, ambivalence, and opportunities: staff perspectives signaling a critical turn in patient-oriented research in forensic mental health care settings.

This qualitative descriptive study examines the views of 25 clinicians, clinical leaders, and senior hospital leaders about forensic patient-oriented research (POR) practices and the inclusion of patients as research partners in forensic mental health care environments. Data from semi-structured interviews were analyzed inductively using thematic analysis to pinpoint predominant themes. Perspectives on forensic POR materialized across three attitudinal domains: optimism, ambivalence, and opportunities. Findings underscore positive overtures toward including patient voices in research and conse

Parental and Adolescent Positive Affect and Optimism as Predictors of Postsurgical Mood and Functioning in Adolescents Undergoing Spinal Fusion Surgery.

OBJECTIVES: While most adolescents display a steady recovery trajectory following surgery, some develop chronic postsurgical pain (CPSP), which can significantly impact their functioning. Psychosocial factors are known to play a role in the recovery from pain following surgery, but positive psychosocial factors have received little attention in the literature. This study aimed to address this gap by investigating parental and adolescent positive affect and optimism as predictors of postsurgical pain recovery and positive outcomes in adolescents. METHODS: This study uses data collected as part

Unveiling the power of optimism: Exploring behavioral and neuromolecular correlates of alcohol seeking and drinking in rats with biased judgement.

Alcohol use disorder (AUD) is a common psychiatric condition with substantial global mortality. Despite extensive research into its pathophysiology, the cognitive predispositions driving alcohol dependence are less understood. This study explores whether biased cognition, specifically traits of optimism and pessimism, predicts susceptibility to alcohol-seeking behaviors using an animal model. Rats were initially tested for judgement bias through Ambiguous Cue Interpretation tests. Those identified as 'optimistic' or 'pessimistic' were further examined for their tendency to escalate alcohol int

Association of Optimism, Causal Thinking, and Karma Beliefs with PTSD and Depression 8 Years After the Tsunami in Sri Lanka.

In 2004, the Indian Ocean tsunami struck Asia and caused high mortality, displacement, and psychological trauma in Sri Lanka. Previous studies have found that optimism protects against PTSD and depression, whereas pessimistic causal thinking and karma beliefs are associated with depression soon after a trauma. The objective of this study is to examine whether these factors continue to be associated with psychological health among Sri Lankan coastal residents 8 years after the tsunami. A semi-structured survey was conducted in 5 coastal districts. Three hundred twenty-five participants had dire

Commentary: Optimism and guidance for improving treatment effects among children with callous-unemotional traits - reflections on Perlstein et al. (2023).

Callous-unemotional (CU) traits have increasingly received attention as a potential predictor and outcome of treatment for children with conduct problems. The results of Perlstein et al. (2023) offer the first meta-analytic evidence against the long-held belief that CU traits confer treatment resistance. The results also suggest that children with conduct problems and CU traits require something more or different to achieve treatment outcomes commensurate with their conduct problems-only peers. In this commentary, I reflect on how treatment adaptations for children with conduct problems and CU

Dispositional optimism, stress, post-traumatic stress disorder and post-traumatic growth in Greek general population facing the COVID-19 crisis.

The aim of this study is to explore psychological and mental health implications of social distancing and lockdown, in Greece in order to gain a better understanding of the COVID-19 crisis. For the purposes of this article, quantitative measures are used: the Greek versions of "Life Orientation Scale-Revised" (LOT-R) (Lyrakos, Damigos, Mavreas, Georgia, & Dimoliatis, 2010) to investigate the presence of dispositional optimism tendencies, "Impact of Event scale" (Mystakidou, Tsilika, Parpa, Galanos, & Vlahos, 2007) to assess the traumatic evaluation of the current events, "Perceived Stress Scal

The Impact of an Optimism Training Intervention on Biological Measures Associated With Cardiovascular Health: Data From a Randomized Controlled Trial.

OBJECTIVE: Positive psychological constructs, such as optimism, are associated with cardiovascular health, and changes in biological measures associated with heart health have been proposed as potential mediators of these relationships. In this analysis of data from a randomized controlled trial, we examined the impact of an optimism training intervention on biological measures associated with cardiac health in patients with coronary artery disease. METHODS: We analyzed data from an 8-week, randomized, controlled trial of a group-based optimism training intervention in 61 patients with corona

OBJECTIVE: Although not well-understood, dermatologic diseases studied in clinical trials often demonstrate substantial response to placebo. The study objective is to determine if optimism, public self-consciousness and other personality traits predict response to placebo or active treatment in a dermatology clinical trial. METHODS: A questionnaire was mailed to subjects previously enrolled in a two-center rosacea study who had been randomized to either a treatment or placebo gel. The questionnaire included the Revised Life Orientation Test (LOT-R), the Public Self-Consciousness Scale, and qu
