Do neuroleptics impair learning in schizophrenic patients?

Dopamine (DA) has been shown to be involved in reward-related (incentive) learning but not stimulus-stimulus (s-s) associative learning. Schizophrenic individuals receive neuroleptics (DA receptor blockers) for therapy and therefore may have impaired incentive learning. To test this hypothesis, in experiment 1, schizophrenic outpatients receiving haloperidol or flupenthixol and matched controls were tested on tasks involving incentive or s-s learning. Patients were also given the Brief Psychiatric Rating Scale (BPRS). Results showed the patients to be significantly impaired in every task. Howe

Reconciling reinforcement learning models with behavioral extinction and renewal: implications for addiction, relapse, and problem gambling.

Because learned associations are quickly renewed following extinction, the extinction process must include processes other than unlearning. However, reinforcement learning models, such as the temporal difference reinforcement learning (TDRL) model, treat extinction as an unlearning of associated value and are thus unable to capture renewal. TDRL models are based on the hypothesis that dopamine carries a reward prediction error signal; these models predict reward by driving that reward error to zero. The authors construct a TDRL model that can accommodate extinction and renewal through two simp

Learning and generalization from reward and punishment in opioid addiction.

This study adapts a widely-used acquired equivalence paradigm to investigate how opioid-addicted individuals learn from positive and negative feedback, and how they generalize this learning. The opioid-addicted group consisted of 33 participants with a history of heroin dependency currently in a methadone maintenance program; the control group consisted of 32 healthy participants without a history of drug addiction. All participants performed a novel variant of the acquired equivalence task, where they learned to map some stimuli to correct outcomes in order to obtain reward, and to map other

Impaired dimensional selection but intact use of reward feedback during visual discrimination learning in Parkinson's disease.

It has been suggested that Parkinson's disease (PD) impairs the ability to learn on the basis of reward or reinforcing feedback i.e., by trial-and-error. In many learning tasks, particular 'dimensions' of stimulus information are relevant whilst others are irrelevant; therefore, efficient performance depends on identifying the dimensions of these 'compound' stimuli and selecting the relevant dimension for further processing. We investigated the ability of patients with PD, as well as patients with Huntington's disease and patients with frontal or temporal lobe lesions, to learn visual discrimi

Learning strategy differentially impacts memory connections in children and adults.

Even once children can accurately remember their experiences, they nevertheless struggle to use those memories in flexible new ways-as in when drawing inferences. However, it remains an open question as to whether the developmental differences observed during both memory formation and inference itself represent a fundamental limitation on children's learning mechanisms, or rather their deployment of suboptimal strategy. Here, 7-9-year-old children (N = 154) and young adults (N = 130) first formed strong memories for initial (AB) associations and then engaged in one of three learning strategies

Impairment in acquisition of conditioned fear in schizophrenia.

Individuals with schizophrenia show impairments in associative learning. One well-studied, quantifiable form of associative learning is Pavlovian fear conditioning. However, to date, studies of fear conditioning in schizophrenia have been inconclusive, possibly because they lacked sufficient power. To address this issue, we pooled data from four independent fear conditioning studies that included a total of 77 individuals with schizophrenia and 74 control subjects. Skin conductance responses (SCRs) to stimuli that were paired (the CS + ) or not paired (CS-) with an aversive, unconditioned stim

The Impact of Accented Input on Spanish-English Bilingual Children's Word Learning.

BACKGROUND: Bilingual children are frequently exposed to accented speech, yet it remains unclear how accent familiarity affects their ability to learn new words. This study examined Spanish-English bilingual children's (n = 46) novel word learning from speakers with familiar and unfamiliar accents and investigated individual differences related to learning from accented input. METHODS: Children completed an experimental word-learning task in which they learned novel word-object pairings produced by three speakers: a speaker of General American English, a Spanish-accented English speaker (fami

Drug-specific recovery of long-term memory and visual discrimination after anaesthetic-induced unconsciousness in rats.

BACKGROUND: Perioperative neurocognitive disorders represent a growing public health concern. To improve the translatability of preclinical findings, it is important to use similar research methods across species. In this study, we used a touchscreen-based cognitive testing system to train rats on the paired-associate learning (PAL) task, which assesses long-term visuospatial associative memory. The goal was to capture the time course of cognitive recovery after receiving mechanistically distinct anaesthetic drugs. METHODS: Adult Sprague-Dawley rats (eight males, eight females) were trained o

Cognitive Decline and Dementia in Chronic Widespread Pain: A Longitudinal Population-based Study.

BACKGROUND: Despite preliminary research suggesting an impact of chronic pain on cognition, the direct effects of chronic widespread pain (CWP) on cognition and its underlying mechanisms remain unclear. This study aims to investigate the effects of CWP on dementia and cognitive performance and explore its potential neurobiological mechanisms. METHODS: This was a population-based cohort study utilizing data from the UK Biobank, which enrolled 500,000 individuals aged 37 to 73 yr from 2006 to 2010, with brain imaging scans initiated in 2014. CWP was defined based on participants' self-reported

White Matter Microstructural Alterations Mediate the Association between Polygenic Risk for Alzheimer's Disease and Cognitive Performance in Clinically Asymptomatic Adults.

PURPOSE: Alzheimer's disease (AD) is a progressive neurodegenerative disorder influenced by genetic factors, with a long preclinical phase characterized by subtle alterations in brain microstructure. Although the apolipoprotein E ε4 allele is a well-established genetic risk factor, AD is increasingly recognized as a polygenic condition. However, how polygenic risk manifests in white matter (WM) microstructure and cognition remains unclear. This study aims to investigate the associations between AD polygenic risk scores (ADPRS), WM microstructure, and cognitive performance in clinically asympto

Computational assessment of memory function in kidney transplant recipients and donors.

BACKGROUND: Impaired memory function is a frequent yet understudied symptom in kidney transplant recipients. In part, this knowledge gap reflects the lack of scalable, sensitive, and low-burden tools for quantifying memory function in large clinical populations. The aim of this study is to evaluate a brief, remote memory assessment and to examine memory function and its clinical correlates in kidney transplant recipients compared with healthy controls. METHODS: In this cross-sectional study, we demonstrate a remote, minimally burdensome memory screener, the Seattle-Groningen Memory Assessment

The Impact of Childhood Attention Hyperactivity Disorder on Cognitive Function in Adult Obsessive-Compulsive Disorder Patients: A Comprehensive Study.

Studies evaluating cognitive functions in patients with obsessive-compulsive disorder (OCD) have yielded inconsistent results in various cognitive domains. One key reason for contradictory findings is the frequent occurrence of comorbid psychiatric disorders in OCD. Attention deficit hyperactivity disorder (ADHD), especially in childhood, is one of the most common comorbidities in OCD. The aim of the current study was to identify potential differences in cognitive functions caused by childhood ADHD in adult OCD patients while minimizing the effects of symptoms that frequently accompany OCD, su

Assessing dynamic brain activity during verbal associative learning using MEG/fMRI co-processing.

BACKGROUND: The abilities of individual neuroimaging methods to resolve spatial and temporal contributions of brain regions during cognitive processes are limited. Co-processing of functional magnetic resonance imaging (fMRI) and magnetoencephalography (MEG) may overcome some of the limitations by utilizing Multiple Sparse Priors (MSP) in a Bayesian framework that takes advantage of the temporal resolution of MEG and spatial resolution of fMRI. METHODS: 24 healthy participants were recruited to perform a paired-associate verbal learning task during fMRI and MEG scans. FMRI data were processed

T2 heterogeneity as an in vivo marker of microstructural integrity in medial temporal lobe subfields in ageing and mild cognitive impairment.

A better understanding of early brain changes that precede loss of independence in diseases like Alzheimer's disease (AD) is critical for development of disease-modifying therapies. Quantitative MRI, such as T2 relaxometry, can identify microstructural changes relevant to early stages of pathology. Recent evidence suggests heterogeneity of T2 may be a more informative MRI measure of early pathology than absolute T2. Here we test whether T2 markers of brain integrity precede the volume changes we know are present in established AD and whether such changes are most marked in medial temporal lobe

Cognitive performance after stroke--the Framingham Heart Study.

BACKGROUND: Individuals with a high risk of stroke are also more prone to cognitive impairment perhaps because of concomitant vascular risk factors. In addition, clinical stroke increases the risk of subsequent dementia. Nevertheless, the relationship between clinical stroke and subsequent cognitive function in initially nondemented individuals remains less clear as most prior studies examined case series without controls. AIMS: To specify among nondemented individuals the cognitive domains affected by clinical stroke, independently of vascular risk factors and prestroke cognition. METHODS:

Double-blind randomized controlled study comparing short-term efficacy of bifrontal and bitemporal electroconvulsive therapy in acute mania.

BACKGROUND: Bifrontal electrode placement is as efficacious as bitemporal placement during electroconvulsive therapy (ECT) in depression but is associated with fewer cognitive adverse effects. There are no studies comparing these techniques in acute mania. This study compared the short-term efficacy and adverse effects of bifrontal and bitemporal ECT in the treatment of acute mania. METHOD: Thirty-six DSM-IV mania inpatients referred for ECT were recruited for study. They were randomized to receive bifrontal (BFECT; n = 17) or bitemporal (BTECT; n = 19) ECT. None of the subjects were on mood

Impaired emotional learning and reduced amygdala size in schizophrenia: a 3-month follow-up.

Individuals with schizophrenia have difficulties in emotional information processing. A relationship between behavioral variables of emotional processing and structural amygdala alterations in schizophrenia has been proposed but not shown, yet. Morphological studies of amygdala size in schizophrenia have yielded inconsistent results. The current study assessed paired associates learning of emotional and neutral faces in 16 subjects with schizophrenia during acute episode and in relative remission after 3 months. Sixteen matched controls were studied for comparison. Subjects also underwent stru

Do individuals with Williams syndrome have bizarre semantics? Evidence for lexical organization using an on-line task.

Williams syndrome, a neurodevelopmental disorder, has attracted a great deal of debate concerning the purported intactness of language in the face of other serious cognitive deficits. As more in-depth studies of specific aspects of WS language have emerged, the notion of a preserved language module has been seriously challenged. Although WS vocabulary scores are often impressive, several investigators have claimed the WS semantics are aberrant. All studies hitherto have been based on off-line experiments which necessarily involve metalinguistic processes. This clearly affects the performance o

Neuropsychological deficits in adults with dyslexia.

Adults with severe dyslexia were compared with age-, sex-, IQ- and SES-matched controls on a neuropsychological and neuromotor test battery, and a contrast group who had recovered from dyslexia was also included. The severely dyslexic group was substantially impaired on tests of verbal fluency and learning, as well as on non-verbal temporal order judgements. These test scores were strong predictors of the degree of reading impairment, as was the rate of repetitive movement of the right hand and foot. The results suggest that adult dyslexia is not 'isolated', but is one expression of a widespre
