Liposomal amphotericin B is more effective in polymorphic lesions of post kala-azar dermal leishmaniasis.

BACKGROUND: Post kala-azar dermal leishmaniasis (PKDL) is thought to be the reservoir of infection for visceral leishmaniasis in South Asia. The development of strategies for the diagnosis and treatment of PKDL are important for the implementation of the visceral leishmaniasis elimination program. AIMS: Liposomal amphotericin B (L-AMB) has been an overwhelming success in the treatment of visceral leishmaniasis. However, the empirical three-week regimen of L-AMB proposed for PKDL was shown to be inadequate, especially in the macular variant. This study aimed to delineate response of the differ

Clinical and parasitological impact of short-term treatment using miltefosine and allopurinol monotherapy or combination therapy in canine visceral leishmaniasis.

Canine visceral leishmaniasis is an endemic zoonosis in Brazil. Dogs are the main hosts in urban environments. The treatment has gained popularity since the Brazilian government authorized miltefosine for canine treatment. The aim of this study was to investigate the clinical and parasitological impact of short-term treatment with miltefosine and allopurinol, alone and in combination. We evaluated the ability of pharmacotherapy to reduce clinical signs of disease, antibody levels using the indirect fluorescence antibody test (IFAT) and skin parasite load via qPCR after 28 days of treatment. Neurocysticercosis, IgG immunoglobulins, and nitric oxide.

This study evaluated the role of parasite load and nitric oxide on IgG levels in neurocysticercosis. Total serum IgG, IgG antibodies specific for cysticercus antigens, and nitric oxide were compared between 85 neurocysticercosis patients, 65 with solitary cysts and 20 with multiple cysts, and 13 normal healthy controls. Sixty-six percent of patients were seropositive for cysticercus IgG antibodies. Among seropositive patients, IgG levels did not differ between those infected with multiple or solitary cysts whose serum nitric oxide levels were low (<40 nmol/ml). Among seropositive solitary cyst

Heterogeneity of humoral immune response to Leishmania tropica in an experimental model.

Humoral (antibody) response is an important part of immunity against pathogens. Despite the clear role of cell-mediated immune response in protection against leishmaniasis, the role of humoral responses is challenging. There is very limited data regarding humoral immune response against Leishmania tropica which is the causative agent of human cutaneous leishmaniasis in many parts of the world. Here, we have compared pathogenicity and antibody response against six Iranian Leishmania tropica isolates in BALB/c mice. A Leishmania major isolate was used for comparison. The parasites were injected

[Hematologic-nutritional study in children predisposed to infection with high-load of Trichuris trichiura].

A coproparasitological survey is carried out in 3 family doctors' home-offices in City of Havana Province following the Kato-Katz technique (3 samples from each person); we found 15 children predisposed to high-load infection by Trichuris trichiura. This group was compared to a group of 20 children infected with a low parasite load, and to another group of 20 children with no parasite infection. A study of nutritional anemia was carried out to all of them including: hemoglobin, hematocrit, ferritin, serum iron, vitamin B12, and serum and erythrocyte folates. No significant differences were fou

Correlation of Parasite Burden, kDNA Integration, Autoreactive Antibodies, and Cytokine Pattern in the Pathophysiology of Chagas Disease.

Chagas disease (CD), caused by the protozoan Trypanosoma cruzi (T. cruzi), is the main parasitic disease in the Western Hemisphere. Unfortunately, its physiopathology is not completely understood, and cardiomegaly development is hard to predict. Trying to explain tissue lesion and the fact that only a percentage of the infected individuals develops clinical manifestations, a variety of mechanisms have been suggested as the provokers of CD, such as parasite persistence and autoimmune responses. However, holistic analysis of how parasite and host-related elements may connect to each other and in

Comparative evaluation of lesion development, tissue damage, and cytokine expression in golden hamsters (Mesocricetus auratus) infected by inocula with different Leishmania (Viannia) braziliensis concentrations.

The golden hamster (Mesocricetus auratus) is a susceptible model to Leishmania (Viannia) spp.; however, available studies employ different infection protocols, which account for clinical and pathological presentation differences. Herein, L. (V.) braziliensis preparations were standardized to contain 10(4), 10(5), or 10(6) parasites to determine an optimal inoculum that ensured cutaneous lesions without causing a disseminated infection in hamsters. Lesion development was followed for 105 days by size measurements, and skin, draining lymph node, spleen, and sera were investigated to check parasi

The dynamic of Trypanosoma cruzi transmissibility in field-caught Mepraia spinolai (Hemiptera: Reduviidae) of contrasting seasons: A developmental stage-dependent study.

A competent vector can harbor a pathogen and infect a high number of hosts. Scarce information is available on how triatomine vectors accomplish this task in nature, particularly without knowing the number of parasites they excrete. We study the dynamics of Trypanosoma cruzi transmissibility in the triatomine Mepraia spinolai collected in summer (December 2018) and winter (July 2019) of an austral endemic area. The insects received two feedings in the laboratory, the day after arrival and 45 days after the first feeding. Excreta samples were collected, DNA was extracted, and parasite loads wer

Assessing the therapeutic potential of silicon dioxide nanoparticles in acute and chronic toxoplasmosis in BALB/c mice.

Toxoplasmosis, an infection caused by the obligate intracellular parasite Toxoplasma gondii, represents a significant global health concern, particularly for immunocompromised individuals. This study aimed to evaluate the therapeutic effects of silicon dioxide nanoparticles (SiO2-NPs) against both acute (T. gondii RH strain) and chronic (T. gondii PRU strain) infections in BALB/c mice. In the acute infection model, mice (n = 40) were infected with 104 T. gondii tachyzoites, while the chronic infection model (n = 40) involved the injection of 50 active cysts. Mice were treated with SiO2-NPs or

Eco-epidemiological Insights into Psammolestes arthuri: Trypanosoma cruzi Infection and Feeding Behavior in Casanare, Colombia.

Chagas disease, caused by the protozoan parasite Trypanosoma cruzi, affects millions of people and is primarily transmitted by triatomine bugs. Although Psammolestes arthuri has been traditionally classified as a secondary vector due to its predominantly ornithophilic feeding habits, recent findings suggest that its ecological and behavioral traits may enhance its epidemiological relevance. We hypothesized that P. arthuri exhibits natural infection rates, flexible feeding behavior with frequent mammalian contacts, and spatial variation in parasite load that challenge its conventional classific

Host heterogeneity affects both parasite transmission to and fitness on subsequent hosts.

Infectious disease dynamics depend on the speed, number and fitness of parasites transmitting from infected hosts ('donors') to parasite-naive 'recipients'. Donor heterogeneity likely affects these three parameters, and may arise from variation between donors in traits including: (i) infection load, (ii) resistance, (iii) stage of infection, and (iv) previous experience of transmission. We used the Trinidadian guppy, Poecilia reticulata, and a directly transmitted monogenean ectoparasite, Gyrodactylus turnbulli, to experimentally explore how these sources of donor heterogeneity affect the thre

Enhanced apoptosis and inflammation allied with autophagic and apoptotic Leishmania amastigotes in the seemingly undamaged ear skin of clinically affected dogs with canine visceral Leishmaniasis.

Programmed cell death plays a relevant role in the pathogenesis of visceral Leishmaniasis. Apoptosis selects suitable parasites, regulating parasite density, whereas autophagy eliminates pathogens. This study aimed to assess the inflammation and apoptosis in inflammatory cells and presents a unique description of the presence of autophagic and apoptotic Leishmania amastigotes in naturally Leishmania-infected dogs. Fragments from seemingly undamaged ear skin of sixteen Leishmania-infected dogs and seven uninfected dogs were evaluated through histomorphometry, ultrastructural, immunohistochemica

Severity of Vessel Color Changes and Macular and Peripheral Whitening in Malarial Retinopathy Are Associated with Higher Total Body and Sequestered Parasite Burdens.

Two-thirds of children with cerebral malaria (CM) exhibit retinopathy characterized by whitening, vessel color changes, and/or hemorrhages. The pathogenesis of malarial retinopathy is not fully understood. This study aimed to assess the relationship between malarial retinopathy and the severity of its components (macular whitening, retinal hemorrhages, and vessel color changes) with the total, circulating, or sequestered parasite load in children with CM. Total parasite burden was estimated by measuring plasma levels of Plasmodium falciparum histidine-rich protein 2 (PfHRP2), while the sequest

Population dynamics of Schistosoma mansoni in mice repeatedly exposed to infection.

Studies of host resistance to parasite infection are usually based on experimental designs involving a primary infection and subsequent challenge exposure, resistance being recorded as the percentage reduction in parasite establishment in challenged hosts when compared with that in uninfected animals. Few studies have focused on the dynamic nature of helminth establishment and mortality (and their presumed dependency on the rate of current exposure and past experiences of infection) in hosts repeatedly exposed to low levels of infection. Here, we report the results of population studies on the

Transcriptional Analysis of Human Skin Lesions Identifies Tryptophan-2,3-Deoxygenase as a Restriction Factor for Cutaneous Leishmania.

Disease manifestation after infection with cutaneous Leishmania species is the result of a complex interplay of diverse factors, including the immune status of the host, the infecting parasite species, or the parasite load at the lesion site. Understanding how these factors impact on the pathology of cutaneous leishmaniasis (CL) may provide new targets to manage the infection and improve clinical outcome. We quantified the relative expression of 170 genes involved in a diverse range of biological processes, in the skin biopsies from patients afflicted with CL caused by infection with either L.

Dietary glycemic and energy load differentially modulates Schistosoma mansoni-induced granulomatous inflammation and response to antiparasitic chemotherapy.

The impact of diet composition and energy content on schistosomiasis evolution and treatment efficacy is still controversial. This study compared the impact of sucrose-rich diet and intermittent fasting on Schistosoma mansoni infection and praziquantel (PZQ)-based chemotherapy response in mice. BALB/c mice were infected with S. mansoni and followed for 15 weeks. The animals were randomized into nine groups receiving high glycemic load (high-sucrose diet - HSD), low caloric load (standard chow alternate-day fasting - ADF), and standard chow ad libitum (AL).
