BACKGROUND: Periodontal disease, an inflammatory condition initiated by the build-up of plaque on the tooth surface, is a common problem seen within veterinary practices. There are an increasing number of studies which indicate distinctive microbial profiles associated with healthy gingiva and periodontal disease. Most have been based on analysis of samples from populations of mixed breed dogs collected at a specific point in time. A study investigating the development of periodontal disease in Yorkshire terriers provided an opportunity to investigate the concurrent changes in the oral microbi

Oral microbiome dysbiosis has been proposed as a potential contributing factor to rising rates of diabetes in pregnancy, with oral health previously associated with an increased risk of numerous chronic diseases and complications in pregnancy, including gestational diabetes mellitus (GDM). However, whilst most studies examining the relationship between GDM and the oral microbiome identify significant differences, these differences are highly variable between studies. Additionally, no previous research has examined the oral microbiome of women with pre-existing type 2 diabetes mellitus (T2DM),

SUMMARYHaemophilus ducreyi is a Gram-negative coccobacillus that forms a distinct lineage with Aggregatibacter pleuropneumoniae and Mannheimia haemolytica within the Pasteurellaceae. H. ducreyi causes chancroid, which is characterized by painful genital ulcers (GU) and inguinal lymphadenitis, and facilitates the transmission of HIV. Although once thought to be exclusively sexually transmitted, H. ducreyi is now recognized as a major cause of non-sexually transmitted cutaneous ulcers (CU) on the lower legs of children who live in yaws-endemic areas. Due to the impact of chancroid on global heal

BACKGROUND: Celiac Disease (CeD) is an autoimmune disorder triggered by gluten intake in genetically susceptible individuals. Highest risk individuals are homozygous for the Human Leucocyte Antigen (HLA) DQ2.5 haplotype or DQ2.5/DQ2.2 heterozygous. Both the HLA-DQ2-positive high genetic risk individuals and those that have developed the disease have altered intestinal microbiota, but it remains unclear whether these alterations are a cause or a consequence of CeD.
OBJECTIVE: To investigate a potential bidirectional causality between gut microbiota (GM) and CeD in HLA-DQ2 high genetic risk ind

Identification and analysis of the antimicrobial resistance of Pasteurella aerogenes (P. aerogenes) isolated from poultry. For susceptibility testing in accordance with the CLSI, plasmids were extracted via alkaline lysis and transferred by CaCl2 treatment. Genomic DNA of a representative P. aerogenes isolate was subjected to whole genome sequencing. CCCP was utilized to determine whether SF190908 contains an efflux pump. The blaVEB gene was ligated with the pET-28 plasmid and transferred to Escherichia coli to verify it as an ESBL gene. SF190908 isolated from poultry was identified as P. aero

Haemophilus parasuis causes Glässer's disease and pneumonia in pigs. Indirect hemagglutination (IHA) is typically used to serotype this bacterium, distinguishing 15 serovars with some nontypeable isolates. The capsule loci of the 15 reference strains have been annotated, and significant genetic variation was identified between serovars, with the exception of serovars 5 and 12. A capsule locus and in silico serovar were identified for all but two nontypeable isolates in our collection of >200 isolates. Here, we describe the development of a multiplex PCR, based on variation within the capsule l

The frequently occurring DNA uptake sequence (DUS), recognized as a 10-bp repeat, is required for efficient genetic transformation in the human pathogens Neisseria meningitidis and Neisseria gonorrhoeae. Genome scanning for DUS occurrences in three different species of Neisseria demonstrated that 76% of the nearly 2,000 neisserial DUS were found to have two semiconserved base pairs extending from the 5' end of DUS to constitute a 12-mer repeat. Plasmids containing sequential variants of the neisserial DUS were tested for their ability to transform N. meningitidis and N. gonorrhoeae, and the 12

Pooled sera from patients with chancroid contain antibodies to a Haemophilus ducreyi antigen with an approximate molecular weight of 28,000 (28K). Rabbit polyclonal serum that reacts to a 28K protein can be used to detect H. ducreyi in clinical samples. A monoclonal antibody, designated 5C9, bound to a 28K outer membrane protein and to 35 of 35 H. ducreyi isolates with diverse geographic origins and did not bind to many species of the families Pasteurellaceae, Neisseriaceae, and Enterobacteriaceae or to Corynebacterium and Candida species strains. A 5C9-reactive phage was recovered from a geno

BACKGROUND: Bovine coronavirus (BCV) is associated with respiratory infections in cattle of all ages; however, a temporal study to evaluate the effect of BCV immunity on virus shedding and bovine respiratory disease (BRD) incidence in pre-weaned beef calves has not been reported. Thus, we report here a prospective study in three herds of crossbred beef calves (n = 817) with endemic BCV. Serial blood samples for measurement of serum anti-BCV antibody titers and nasal swabs for detection of BCV and other common viral and bacterial BRD pathogens were collected from all calves or subsets of calves

Pott's puffy tumour (PPT) is a rare condition defined by subperiosteal abscess formation following osteomyelitis of the frontal bone. Complications can be severe and involve intracranial progression. Here, we present a rare case of a PPT due to an Aggregatibacter aphrophilus infection of odontogenic origin in a young adult patient with a history of bilateral cheilognathopalatoschisis. Due to the unusual location of the abscess, the diagnosis was initially missed, leading to a delay of more than 1 month until adequate treatment. Odontogenic treatment was initiated as source control, external dra

BACKGROUND: Aggregatibacter aphrophilus (A. aphrophilus) is a rare cause of infective endocarditis (IE), but is a recognized cause of culture-negative IE. The risk of developing IE is increased in patients with valvular disease or prosthetic valves. To our knowledge, A. aphrophilus has never previously been reported to cause lumbar facet joint arthritis in combination with IE.
CASE PRESENTATION: We present the first case where facet joint arthritis was the presenting symptom for culture-negative A. aphrophilus native valve IE in a patient with no prior cardiac disease. A 58-year-old Japanese

INTRODUCTION: Septic arthritis by Aggregatibacter aphrophilus is an uncommon entity, so it's important to diagnose it early, to avoid serious consequences. In adults, the knee is the most affected site.
METHODS: We present a case of a 17-year-old boy, with pain in the left knee since one month ago. It shows an increase in local temperature, erythema and movement inability. A nuclear magnetic resonance (NMR) was performed and empirical treatment was given with cephalothin and clindamycin. A knee arthrotomy was realized, and intramedullary secretion and bone tissue was sent to study.

This work aimed to investigate some aspects related to the pathogenicity of Lechiguana, a bovine fibroproliferative lesion characterized by rapid collagen accumulation. Light and transmission electron microscopy and in situ hybridization studies were performed in order to elucidate the fibrogenic activity of this lesion. The characterization of fibroblastic plasticity in the lesion was done by immunohistochemical study for alpha-smooth-muscle cell actin. The ovoid-shaped cells presented positive reaction for alpha-smooth-muscle cell actin in their cytoplasm and, at the electron-microscopic lev

The blaOXA-1 gene encodes an oxacillin-hydrolyzing beta-lactamase of extended-spectrum beta-lactamase (ESBL)-producing microorganisms. The blaOXA-1 gene is found in the resistomes of some Enterobacteriaceae, Morganellaceae, Pasteurellaceae, Moraxellaceae, Aeromonadaceae, Pseudomonadaceae, Yersiniaceae, and Vibrionaceae. Most ESBL detection methods, including those to detect OXA-1-producing microorganisms, are time-consuming, and require specialized equipment and qualified personnel. Here, we report a new CRISPR(Clustered Regularly Interspaced Short Palindromic Repeats)/Cas12a-based detection a

Zoonotic diseases continue to rise globally, yet no existing genomic tool integrates virulence, antimicrobial resistance (AMR), and mobile genetic elements to predict zoonotic potential. Here, we present Zoonoticus, a machine learning-based model that classifies bacterial strains as zoonotic or non-zoonotic using whole-genome data. The model was developed using a curated reference database of 37,291 genes across 40 functional categories, encompassing virulence factors, AMR genes, and integrative and conjugative elements (ICEs). More than 5000 bacterial genomes from zoonotic (Brucella abortus,

BACKGROUND: The causal relationship between breast cancer (BC) and the oral microbiome remains unclear. In this case-control study, using two-sample Mendelian randomization (MR), we thoroughly explored the relationship between the oral microbiome and BC in the East Asian population.
METHODS: Genetic summary data related to oral microbiota and BC were collected from genome-wide association studies involving participants of East Asian descent. MR estimates were generated by conducting various analyses. Sequencing data from a case-control study were used to verify the validity of these findings.

BACKGROUND: Chronic Obstructive Pulmonary Disease (COPD) is an important comorbidity in patients living with human immunodeficiency virus (HIV). Previous bacterial microbiome studies have shown increased abundance of specific bacterium, like Tropheryma whipplei, and no overall community differences. However, the host response to the lung microbiome is unknown in patients infected with HIV.
METHODS: Two bronchial brush samples were obtained from 21 HIV-infected patients. One brush was used for bacterial microbiome analysis using the Illumina MiSeqTM platform, while the other was used to evalua
