Experimental tooth movement under light orthodontic forces: rates of tooth movement and changes of the periodontium.

AIM: To investigate light forces for experimental tooth movement. METHOD: Light orthodontic forces of 1.2, 3.6, 6.5, and 10 g force (gf) were applied for 14 days to move rat molars, and the effects of the forces on the rate of tooth movement and changes of the periodontium were examined. RESULTS: In the early period, despite the different levels of force used in each group, there were no significant differences in tooth displacement. From hour 56 to day 14, the tooth displacement in the 1.2 gf group was significantly smaller than that in the other groups and the rate was nearly constant. The

Strain relaxation of fibroblasts in the marginal periodontium is the common trigger for alveolar bone resorption: a novel hypothesis.

In summary, the present commentary proposes a hypothesis that alveolar bone remodeling and bone loss in periodontitis, periodontal surgery, and in orthodontic tooth movement is triggered by a common "strain relaxation" signaling pathway of gingival and periodontal fibroblasts. The abrupt splitting, degradation, or relaxation of collagen fibers in the marginal periodontium produces a "strain relaxation" signal in the local fibroblasts which reside on these fibers, activating an ECM-integrin-cytoskeleton pathway. A cascade of cellular reactions which lead to osteoclastic bone resorption starting

Effects of inflammatory periodontal disease ('broken mouth') on the ultrastructure of collagen fibrils in the sheep incisor periodontium.

The ultrastructure of the matrix of the sheep central incisor periodontium showing clinical signs of severe periodontitis was analysed quantitatively. The distribution of collagen fibril diameters in the lower dental pad changed from a bimodal distribution seen in healthy periodontia to a unimodal distribution. Collagen fibrils with an abnormal morphology were seen in the connective tissue adjacent to the crest of the alveolar bone. These results suggest that the deepening periodontal pocket resulting from inflammation removes the major area of support for the tooth and abnormal loads are appl

DLP 3D-printed biomimetic periodontium composite with xenogenic treated dentin matrix modulates periodontal regeneration and inflammatory response.

Xenogeneic treated dentin matrix (xTDM) roots serve as a promising donor source for tooth replantation. Nevertheless, the regeneration of the alveolar bone-periodontal ligament (PDL)-cementum complex persists as a challenging task owing to tissue heterogeneity, limited vascularization, and immune-associated complications. In this study, a biomimetic tubular periodontium was fabricated through digital light processing (DLP)-based 3D bioprinting around columnar xTDM roots. The bio-ink was composed of silk fibroin methacrylate (SilMA), dental follicle stem cells (DFCs), and transforming growth fa

Treponema denticola increases MMP-2 expression and activation in the periodontium via reversible DNA and histone modifications.

Host-derived matrix metalloproteinases (MMPs) and bacterial proteases mediate destruction of extracellular matrices and supporting alveolar bone in periodontitis. The Treponema denticola dentilisin protease induces MMP-2 expression and activation in periodontal ligament (PDL) cells, and dentilisin-mediated activation of pro-MMP-2 is required for cellular fibronectin degradation. Here, we report that T. denticola regulates MMP-2 expression through epigenetic modifications in the periodontium. PDL cells were treated with epigenetic enzyme inhibitors before or after T. denticola challenge. Fibron

Local application of prostaglandin E2 reduces trap, calcitonin receptor and metalloproteinase-2 immunoreactivity in the rat periodontium.

It has been shown that prostaglandin E2 (PGE2) locally released adjacent to the mandible over a 20-day period increases alveolar bone area, in part, due to a reduction in the percentage of eroded surface. To determine the effect of PGE2 on alveolar bone resorption, left mandibles from 24 Lewis rats were treated over a 20-day period with a local application of PGE2 (0.1, 0.05 or 0.025 mg/day) or placebo. The right side served as the non-treated matched control. Tissue sections were stained for tartrate resistant acid phosphatase (TRAP) calcitonin receptor (CTR) and metalloproteinase-2 (MMP-2).

[The marginal periodontium of impacted upper canines. Evaluation following various methods of surgical approach and orthodontic procedures].

The marginal periodontium of 29 impacted upper canines and several aspects of their treatment have been studied. The situation has been analysed after treatment according to 3 methods of surgery and orthodontics: a) excision and active eruption, b) excision and passive eruption and c) replaced flap and active eruption. Many aspects regarding the choice and course of treatment depend primarily on individual data of the patient and on the location of impaction. With surgical exposure by excision some mucogingival problems seem to appear, especially for vestibular impactions. In those cases the s

Reduced Periodontal Support for Lower Central Incisor - A 3D Finite Element Analysis of Compressive Stress in the Periodontium.

BACKGROUND: The aim of this study was to assess the stress concentration in simulated periodontal alveolar bone containing healthy teeth with and without attachment loss. METHODS: Six 3-D models of a lower central incisor were created simulating the teeth structure, cancellous and cortical bone and periodontal ligament. Each model presented a 1mm increasing distance between cement-enamel junction (CEJ) and alveolar bone crest (ABC) (1 to 6mm). A 100N, 45-degree load was applied to the buccal face of the lower central incisor. The effects of Minimum Principal Stress (MPS) on lamina dura (LD) a

Effects of acute pain and strain of the periodontium due to orthodontic separation on the occlusal tactile acuity of healthy individuals.

OBJECTIVES: The aim of this study was to assess whether pain and strain of the periodontal ligament (PDL), induced by orthodontic separation, alter the somatosensory ability to perceive small thicknesses between occluding teeth (occlusal tactile acuity, OTA). METHODS: The OTA was tested at baseline (T0), using 9 aluminum foils (range 8-72 μm), randomly placed between the molar teeth, and 1 sham test (without foil), asking the participants whether they felt the foil between their teeth. Afterwards, orthodontic separators were placed, and subjects were randomly assigned to one of the two experi

The effect of partial damage to the enamel-related periodontium combined with root resection on eruption of the rat incisor eruption.

Previous work has indicated that the enamel-related periodontium (ERP) has a role in the eruptive process of the rat lower incisor. By combining partial damage of this tissue with resection of the odontogenic organ, we examined the effect of the damage on subsequent incisor eruption. The connective tissue of the enamel-related periodontium was regenerated in less than 2 weeks, showing morphology close to normal. The injured part of the enamel organ was neither regenerated nor repaired, and a cement-like tissue, continuous with the true acellular cement, was formed on the denuded enamel. Before

Do mutations in the basement membrane zone affect the human periodontium? Review with special reference to epidermolysis bullosa.

Epidermolysis bullosa (EB) is a group of diseases characterized by the development of blisters or erosions following minor trauma to the skin. Oral findings that have been associated with EB include keratin-filled cysts; blistering of the mucosa, tongue, and lips; perioral carcinomas; ankyloglossia; lingual papilla atrophy; caries; enamel hypoplasia; rapid attrition of the teeth; and obliteration of the oral vestibule. Defects at the basal cell/basement membrane/connective tissue levels correspond to the mutations in basal cell keratins, hemidesmosome components, and type VII collagen, respect

[Salvage of a tooth with necrotised periodontium, caused by endodontic use of radiosurgery. Long-term results. Case report].

Electrosurgery and radiosurgery bear certain popularity in contemporary dentistry. Nevertheless, the inadequate treatment allocation may lead to tissue necrosis. A young female patient was referred to our department following root canal treatment (RCT) by radiosurgical device. Necrotised alveolar bone, lack of keratinized gingiva and increased tooth mobility were noticed. After considering all the treatment options, we attempted to keep the tooth. Therefore, the necrotised part of the alveolar process was removed and the gingival fringe was rejuvenated alongside root surface debridement. The t

Orthodontically stressed periodontium of transgenic mouse as a model for studying mechanical response in bone: The effect on the number of osteoblasts.

A better understanding of cellular and molecular mechanisms involved in response to mechanical stress is a prerequisite for future improvements in orthodontic treatment. To expand the application of molecular biology techniques in this area of research, we developed and characterized a mouse tooth movement model. The aim of this study was to biomechanically characterize this model and to evaluate the effect of orthodontic stress on the proliferation of periodontal osteoblasts. We used an orthodontic coil spring appliance with a low force/deflection rate, which produced an average force of 10-1

[Experimental study of periodontal tissue regeneration. Changes in the periodontium after tooth implantation with and without periodontal ligament].

The role of the periodontal ligament in periodontal tissue regeneration was evaluated. The materials included bilateral upper 1st premolars and lower 2nd, 3rd, and 4th premolars of 11 adult Beagle dogs, aged from 3 to 6 years. Before implantation, the right teeth were extracted and kept for a period of more than 3 months in "a solution for reserving teeth". The left teeth on the other hand, were extracted and soon implanted thus retaining the periodontal ligament into the newly created bone cavities prepared in the right edentulous areas. At the same time, the right reserved teeth were implant

Brain-Derived Neurotrophic Factor Systemic Response in the Periodontium, Trigeminal Nucleus Caudalis, and Hippocampus Induced by Occlusal Trauma.

Occlusal trauma (OT), by causing periodontal tissue damage, can activate and enhance the activity of the peripheral and central nervous system (CNS) neuropeptides. The brain-derived neurotrophic factor (BDNF) gene is activity-dependent and exhibits marked alterations, characterized by protection against injury and repair. Our results show the possible molecular mechanism through which noxious environmental stimuli induce alterations in BDNF activity in the local periodontal tissue, the primary sensory neurons-Vc, and the hippocampus, suggesting systemic impairment. BDNF serves a more positive
