CTP:phosphoethanolamine cytidylyltransferase (ET), encoded by PCYT2, is the rate-limiting enzyme for phosphatidylethanolamine synthesis via the CDP-ethanolamine pathway. Phosphatidylethanolamine is one of the most abundant membrane lipids and is particularly enriched in the brain. We identified five individuals with biallelic PCYT2 variants clinically characterized by global developmental delay with regression, spastic para- or tetraparesis, epilepsy and progressive cerebral and cerebellar atrophy. Using patient fibroblasts we demonstrated that these variants are hypomorphic, result in altered

Hippocampal cholinergic neurostimulating peptide (HCNP) is involved in the phenotype development of the septo-hippocampal system. HCNP precursor protein (HCNP-pp) is known to interact with other molecules including phosphatidylethanolamine and Raf-1 kinase, and is also known as phosphatidylethanolamine-binding protein and raf kinase-inhibitory protein. To assess whether HCNP-pp is involved in the pathogenesis of Alzheimer disease (AD), the expression levels of its mRNA in the hippocampus of autopsy brains from patients with dementia (including AD and ischemic vascular dementia) were compared w

The stimulatory effects of sphingosine and oleylamine upon the incorporation of [3H]serine into its corresponding phospholipid, phosphatidylserine, by LA-N-2 cell cultures, have been reported (Singh et al. (1992) FEBS Lett., 296, 166-168). The presence of chlorpromazine and N-(6-aminohexyl)-5-chloro-1-naphthalenesulfonamide (W-7) in cultures of these cells provoked increased phosphatidylserine synthesis but decreased protein synthesis in a concentration-dependent fashion. Several other amphiphilic cations, including dimethyldidodecylammonium bromide, verapamil, desipramine, amitriptyline, and

Plasmodium knowlesi-infected erythrocytes efficiently incorporated choline and metabolize it into phosphatidylcholine via the de novo Kennedy pathway. No formation of either betaine or acetylcholine was detected. At physiological concentrations of external choline, isotopic equilibrium between intracellular choline and phosphocholine was reached in less than 1 h, whereas labeled phosphatidylcholine accumulated constantly, until at least 210 min. During this time, intracellular CDP-choline remained quite low compared to phosphocholine, which suggests that choline-phosphate cytidylyltransferase

Lens cells can synthesize, degrade, and remodel lipids. Endogenous lipid synthesis, in conjunction with uptake of exogenous cholesterol and certain fatty acids, leads to the formation of a plasma membrane that is especially rich in sphingomyelin, cholesterol, and long-chain saturated fatty acids. As a result of this unusual lipid composition, lens membranes have very low fluidity, which is restricted even further by lipid-protein interactions. The composition and metabolism of membrane lipids may affect the formation of various types of cataracts. Diets rich in vegetable oils offer some protec

The authors studied serum phospholipid subunits in patients with anthracosilicosis associated with vibration disease. Findings are significantly higher detergent fraction of phospholipids--lysophosphatidylcholine, phosphatidylserine, sphingomyelin, lower phosphatidylcholine, phosphatidylethanolamine and phosphoinositides--that suggests increased transmissivity of biologic membranes and activated inosite mechanism of transmembrane signal transmission. Aeroionization appeared to have positive influence, normalizing serum phospholipids level.

Excessive methylation has been proposed to be involved in the pathogenesis of Parkinson's disease (PD), via mechanisms that involve phospholipid methylation. Meanwhile, 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) was found to stimulate phospholipid methylation via the oxidized metabolite, 1-methyl-4-phenyl-pyridinium (MPP+), in the rat brain and liver tissues. In the present study, we investigated the effect of MPP+ on phosphatidylethanolamine N-methyltransferases (PENMT) and the potential role of this pathway in MPP(+)-induced neurotoxicity using PC12 cells. The results obtained indic

Obeticholic acid (OCA) has been examined to treat non-alcoholic steatohepatitis (NASH), but has unsatisfactory antifibrotic effect and deficient responsive rate in recent phase III clinical trial. Using a prolonged western diet-feeding murine NASH model, we show that OCA-shaped gut microbiota induces lipid peroxidation and impairs its anti-fibrotic effect. Mechanically, Bacteroides enriched by OCA deconjugates tauro-conjugated bile acids to generate excessive chenodeoxycholic acid (CDCA), resulting in liver ROS accumulation. We further elucidate that OCA reduces triglycerides containing polyun

Dried seahorse is a precious raw food material for cooking soups. In this study, a lipidomics strategy using the techniques of solid-phase extraction (SPE) and hydrophilic interaction chromatography-tandem mass spectrometry (HILIC-QTOF/MS) was developed for extraction, visualization, and quantification of phospholipids in dried seahorses. The parameters of SPE were optimized, and 1 mL of sample and chloroform/methanol (1:2, v/v) were found to be the best loading volume and eluting solvent, respectively. Afterwards, each phospholipid class was successfully separated on a HILIC column and analyz

Liquid chromatography-mass spectrometry (LC-MS)-based lipidomics has been a subject of dramatic developments over the past decade. This review focuses on state of the art in LC-MS-based lipidomics, covering all the steps of global lipidomic profiling. On the basis of review of 185 original papers and application notes, we can conclude that typical LC-MS-based lipidomics methods involve: (1) extraction using chloroform/MeOH or MTBE/MeOH protocols, both with addition of internal standards covering each lipid class; (2) separation of lipids using short microbore columns with sub-2-μm or 2.6-2.8-μ

Hypoxic ischemic encephalopathy (HIE) due to insufficient oxygen or blood flow to the brain can result in inflammation, impaired neurodevelopment, or death. Changes to microdomain lipid composition may contribute to altered structure and function of key regulatory processes; however, knowledge regarding changes to membrane lipid composition after HIE is incomplete. Here, we describe the application of untargeted lipidomics to investigate the impact of hypoxia-ischemia on detergent-resistant membrane (DRM) and detergent-soluble membrane (DSM) domains fractionated from the cerebellum in a rat mo

As textured pea protein (TPP) emerges as a sustainable strategy for plant-based meat analogues, its influence on flavor precursors and the resulting volatile profile and sensory attributes remains underexplored. This study investigated physicochemical properties, lipid composition, volatiles, and sensory characteristics of dry-fermented sausages formulated with varying levels of TPP substitution. TPP substitution yielded significant decrease in pH, water activity, glossiness and yellowness intensity, and increased weight loss. A 25 % TPP substitution substantially diminished free fatty acids c

The development of chemosensors that can selectively detect phosphatidylcholines (PCs) in biological samples is of medical relevance considering the importance of these phospholipids in cell growth and survival. Their selective sensing over phosphatidylethanolamines (PEs) is however a challenging task. We report here on the chemosensing capacities of calix[6]tris-pyrenylurea 1, which is able to selectively interact with phosphatidylcholine-type lipids in organic media. Host 1 also binds them in a biphasic chloroform/water solution, opening the way to the design of selective chemosensors for th

Studies of conjunctival microcirculation, thromboelastographic coagulation (as assessed by 8 parameters) and plasma phospholipid spectrum (with 8 fractions obtained), carried out in 136 patients with acute phases of chronic bronchitis, have demonstrated considerable disorders of microcirculation, hypercoagulation and shifts of blood phospholipid spectrum. Marked perivascular disorders were matched by increased levels of plasma lysophosphatidylcholines, which promote increased capillary permeability. Intravascular changes (erythrocyte aggregation and sludging, intermittent flow and stasis of bl

To accurately quantify the phospholipids in infant formula by subclass, we developed an analytical method using phosphorus-31 nuclear magnetic resonance spectroscopy (31P NMR). We performed heated extraction method using a mixture of ethanol and water to extract phospholipids from infant formula and replace the highly toxic chloroform traditionally used for extraction. In the 31P NMR measurement, we also avoided using chloroform by dispersing the extracts in surfactants with a strong affinity for phospholipids. Although polar lipids in milk are characterized by a high content of sphingomyelin,

Gonadal white adipose tissue (gWAT) can regulate gametogenesis via modulation of neuroendocrine signaling. However, the effect of gWAT on the local microenvironment of the gonad was largely unknown. Herein, we ruled out that gWAT had a neuroendocrine effect on gonad function through a unilateral lipectomy strategy, in which cutting off epididymal white adipose tissue could reduce seminiferous tubule thickness and decrease sperm counts only in the adjacent testis and epididymis of the affected gonad. Consistent with the results in males, in females, ovary mass was similarly decreased by lipecto

Lipid peroxidation is a process of oxidative degradation of cellular lipids that is increasingly recognized as an important factor in the pathogenesis of neurodegenerative diseases. We were therefore interested in the manifestation of lipid peroxidation in synucleinopathies, a group of neurodegenerative diseases characterized by the central pathology of α-synuclein aggregates, including Parkinson's disease, multiple system atrophy, dementia with Lewy bodies and Alzheimer's disease with Lewy bodies. We assessed lipid peroxidation products, lipid aldehydes, in the amygdala, a common disease-affe

Membrane lipid composition is altered in the brain during the pathogenesis of several age-related neurodegenerative diseases, including Alzheimer's disease. The entorhinal cortex is one of the first regions of the brain to display the neuropathology typical of Alzheimer's disease, yet little is known about the changes that occur in membrane lipids within this brain region during normal aging (i.e., in the absence of dementia). In the present study, the phospholipid composition of mitochondrial and microsomal membranes from human entorhinal cortex was examined for any changes over the adult lif

Myelin is the lipid-rich membrane that surrounds neuronal axons and is essential for neurological function in vertebrates. The development of therapeutics that stimulate myelin repair to treat demyelinating disorders such as multiple sclerosis is hampered by the inability to distinguish newly synthesised from pre-existing myelin. This study aimed to develop a method to quantify and localise new myelin lipid synthesis in the mouse brain. Deuterium oxide was administered for two weeks in the drinking water of mice fed normal chow, chow containing the demyelinating toxin cuprizone, or during spon
