WO3/Pt nanoparticles are NADPH oxidase biomimetics that mimic effector cells in vitro and in vivo.

To provide a means of delivering an artificial immune effector cell-like attack on tumor cells, we report the tumoricidal ability of inorganic WO3/Pt nanoparticles that mimic a leukocyte's functional abilities. These nanoparticles route electrons from organic structures and electron carriers to form hydroxyl radicals within tumor cells. During visible light exposure, WO3/Pt nanoparticles manufacture hydroxyl radicals, degrade organic compounds, use NADPH, trigger lipid peroxidation, promote lysosomal membrane disruption, promote the loss of reduced glutathione, and activate apoptosis. In a mod

Steady state dynamics of intermediate filament networks.

We have conducted experiments to examine the dynamic exchange between subunit and polymer of vimentin intermediate filaments (IF) at steady state through the use of xrhodamine-labeled vimentin in fluorescence recovery after photobleaching (FRAP) analysis. The xrhodamine-vimentin incorporated into the endogenous vimentin IF network after microinjection into fibroblasts and could be visualized with a cooled charge-coupled device (CCD) camera and digital imaging fluorescence microscopy. Bar shaped regions were bleached in the fluorescent IF network using a beam from an argon ion laser and the cel

Clustering and mobility of voltage-dependent sodium channels during myelination.

In myelinated axons, voltage-dependent sodium channels are segregated at high density at nodes of Ranvier (Rosenbluth, 1976; Waxman and Quick, 1978; Black et al., 1990; Elmer et al., 1990), a distribution that is critical for the saltatory conduction of action potentials (Huxley and Stampfli, 1949). The factors that specifically control the organization and immobilization of sodium channels at nodes are unknown. Recently we have reported that segregation of sodium channels on axons is highly dependent on interactions with active Schwann cells and that continuing axon-glial interactions are nec

Infrared Laser Heating Reveals a Reversible Phase Transition of Phosphofructokinase 1 in Mammalian Cells.

Metabolic enzymes can operate in concerted networks, so-called metabolons. The glucosome is a multi-enzyme complex that includes enzymes of glycolysis and gluconeogenesis and regulates glucose metabolism in mammalian cells. The mechanism of how glucosomes assemble remains largely unknown. Fluorescence recovery after photobleaching (FRAP) experiments suggest that glucosomes are liquid-like, but direct evidence of a phase transition mechanism is missing. Here, we developed a novel method to measure the phase transition of condensates in living cells by combining infrared laser heating with confo

Modulation of Single-Molecule Emission at Hexagonal Boron Nitride Surfaces.

Hexagonal boron nitride (hBN) is gaining increasing attention in the field of biomolecule characterization due to its compatibility with single-molecule fluorescence imaging and real-time tracking. Embedding fluorescent molecules within hBN layers offers potential for molecular-resolution sensing devices, since these probes are highly sensitive to their surroundings. Yet, the effect of hBN surfaces on the fluorophore properties remains largely unexplored. Here, we monitor the photophysical properties of ATTO647N-ssDNA on hBN surfaces and elucidate the effects of the environment and substrate.

An Integrated Workflow for the Purification and Fluorescence Imaging of Clostridioides difficile Spores Using Lanthanide Beta-Diketone Complexes.

We present a robust protocol for both the purification and imaging of C. difficile spores. First, we describe a simplified, one-step isopycnic centrifugation method using Percoll that enables efficient separation of spores from vegetative cells. Next, we introduce a fluorescence-based imaging method using lanthanide beta-diketone complexes (primarily Eu[TTA]3) that allows for the differential visualization of dormant and germinated spores, as well as sporulating cells. These complexes exploit the unique photophysical properties of lanthanides, including sharp emission spectra and resistance to

piSTORM: programmable illumination in stochastic optical reconstruction microscopy.

We implement a programmable illumination system in stochastic optical reconstruction microscopy (STORM) using holographic modulation of the excitation beam profile in the sample plane of a widefield fluorescence microscope. A binary hologram is implemented in a plane conjugate to the sample plane using a liquid crystal spatial light modulator (LCSLM). This approach allows a user-defined yet complex illumination pattern to match approximately any arbitrary region of interest (ROI) within the field of view (FOV) of the microscope, while keeping regions outside the ROI unilluminated. The proposed

Dual-emission fluorescent ZnS nanosheets for sensitive detection of carcinogenic Sudan dyes in chili powder and lipstick samples.

Sudan dyes are carcinogenic colorants illicitly added to foods and cosmetics, posing serious health risks. Herein, a rapid dual-emission fluorescent probe based on two-dimensional (2D) ZnS nanosheets is developed for sensitive detection of Sudan dyes (III/IV). Compared with conventional nanoparticles, the nanosheet structure offers high surface accessibility, excellent dispersion, and reduced aggregation, enabling reliable sensing in complex matrices. The ZnS nanosheets (NS), synthesized via the solvothermal method, exhibit stable fluorescence emissions at 521 and 557 nm with photobleaching re

Bleaching protection and axial sectioning in fluorescence nanoscopy through two-photon activation at 515 nm.

Activation of caged fluorophores in microscopy has mostly relied on the absorption of a single ultraviolet (UV) photon of ≲400 nm wavelength or on the simultaneous absorption of two near-infrared (NIR) photons >700 nm. Here, we show that two green photons (515 nm) can substitute for a single photon (~260 nm) to activate popular silicon-rhodamine (Si-R) dyes. Activation in the green range eliminates the chromatic aberrations that plague activation by UV or NIR light. Thus, in confocal fluorescence microscopy, the activation focal volume can be matched with that of confocal detection. Besides, d

Reciprocal discoidin domain receptor signaling strengthens integrin adhesion to connect adjacent tissues.

Separate tissues connect through adjoining basement membranes to carry out molecular barrier, exchange, and organ support functions. Cell adhesion at these connections must be robust and balanced to withstand independent tissue movement. Yet, how cells achieve synchronized adhesion to connect tissues is unknown. Here, we have investigated this question using the Caenorhabditis elegans utse-seam tissue connection that supports the uterus during egg-laying. Through genetics, quantitative fluorescence, and cell-specific molecular disruption, we show that type IV collagen, which fastens the linkag

Optimal inference of molecular interaction dynamics in FRET microscopy.

Intensity-based time-lapse fluorescence resonance energy transfer (FRET) microscopy has been a major tool for investigating cellular processes, converting otherwise unobservable molecular interactions into fluorescence time series. However, inferring the molecular interaction dynamics from the observables remains a challenging inverse problem, particularly when measurement noise and photobleaching are nonnegligible-a common situation in single-cell analysis. The conventional approach is to process the time-series data algebraically, but such methods inevitably accumulate the measurement noise

Variable number of TMC1-dependent mechanotransducer channels underlie tonotopic conductance gradients in the cochlea.

Functional mechanoelectrical transduction (MET) channels of cochlear hair cells require the presence of transmembrane channel-like protein isoforms TMC1 or TMC2. We show that TMCs are required for normal stereociliary bundle development and distinctively influence channel properties. TMC1-dependent channels have larger single-channel conductance and in outer hair cells (OHCs) support a tonotopic apex-to-base conductance gradient. Each MET channel complex exhibits multiple conductance states in ~50 pS increments, basal MET channels having more large-conductance levels. Using mice expressing flu

Laminin-111 and the Level of Nuclear Actin Regulate Epithelial Quiescence via Exportin-6.

Nuclear actin (N-actin) is known to participate in the regulation of gene expression. We showed previously that N-actin levels mediate the growth and quiescence of mouse epithelial cells in response to laminin-111 (LN1), a component of the mammary basement membrane (BM). We know that BM is defective in malignant cells, and we show here that it is the LN1/N-actin pathway that is aberrant in human breast cancer cells, leading to continuous growth. Photobleaching assays revealed that N-actin exit in nonmalignant cells begins as early as 30 min after LN1 treatment. LN1 attenuates the PI3K pathway

Tension Independence of Lipid Diffusion and Membrane Viscosity.

The diffusion of biomolecules at lipid membranes is governed by the viscosity of the underlying two-dimensionally fluid lipid bilayer. For common three-dimensional fluids, viscosity can be modulated by hydrostatic pressure, and pressure-viscosity data have been measured for decades. Remarkably, the two-dimensional analogue of this relationship, the dependence of molecular mobility on tension, has to the best of our knowledge never been measured for lipid bilayers, limiting our understanding of cellular mechanotransduction as well as the fundamental fluid mechanics of membranes. Here we report

A self-quenching-resistant carbon nanodot powder with multicolored solid-state fluorescence for ultra-fast staining of various representative bacterial species within one minute.

In this study, we prepared self-quenching-resistant solid-state fluorescent carbon nanodots (SFCDs) without any other solid matrices. The SFCDs were prepared using a one-step microwave synthesis method through precise control of the heating power and time. The resulting SFCD powder showed excitation-dependent emission behavior with a maximum fluorescence quantum yield of 40%. The multicolored SFCDs were successfully used as fluorescent agents for rapid staining of 14 representative bacterial species, including Gram-negative, Gram-positive, and acid-fast bacteria. Moreover, some pathogenic bact

Proinflammatory cytokine-induced tight junction remodeling through dynamic self-assembly of claudins.

Tight junctions (TJs) are dynamic, multiprotein intercellular adhesive contacts that provide a vital barrier function in epithelial tissues. TJs are remodeled during physiological development and pathological mucosal inflammation, and differential expression of the claudin family of TJ proteins determines epithelial barrier properties. However, the molecular mechanisms involved in TJ remodeling are incompletely understood. Using acGFP-claudin 4 as a biosensor of TJ remodeling, we observed increased claudin 4 fluorescence recovery after photobleaching (FRAP) dynamics in response to inflammatory

Suppressed blinking and auger recombination in near-infrared type-II InP/CdS nanocrystal quantum dots.

Nonblinking excitonic emission from near-infrared and type-II nanocrystal quantum dots (NQDs) is reported for the first time. To realize this unusual degree of stability at the single-dot level, novel InP/CdS core/shell NQDs were synthesized for a range of shell thicknesses (~1-11 monolayers of CdS). Ensemble spectroscopy measurements (photoluminescence peak position and radiative lifetimes) and electronic structure calculations established the transition from type-I to type-II band alignment in these heterostructured NQDs. More significantly, single-NQD studies revealed clear evidence for bli

Basement membrane sliding and targeted adhesion remodels tissue boundaries during uterine-vulval attachment in Caenorhabditis elegans.

Large gaps in basement membrane occur at sites of cell invasion and tissue remodelling in development and cancer. Though never followed directly in vivo, basement membrane dissolution or reduced synthesis have been postulated to create these gaps. Using landmark photobleaching and optical highlighting of laminin and type IV collagen, we find that a new mechanism, basement membrane sliding, underlies basement membrane gap enlargement during uterine-vulval attachment in Caenorhabditis elegans. Laser ablation and mutant analysis reveal that the invaginating vulval cells promote basement membrane

Pax6 localizes to chromatin-rich territories and displays a slow nuclear mobility altered by disease mutations.

The transcription factor Pax6 is crucial for the embryogenesis of multiple organs, including the eyes, parts of the brain and the pancreas. Mutations in one allele of PAX6 lead to eye diseases including Peter's anomaly and aniridia. Here, we use fluorescence recovery after photobleaching to show that Pax6 and also other Pax family proteins display a strikingly low nuclear mobility compared to other transcriptional regulators. For Pax6, the slow mobility is largely due to the presence of two DNA-binding domains, but protein-protein interactions also contribute. Consistently, the subnuclear loca
