State dependence of noradrenergic activity in a rapidly cycling bipolar patient.

Plasma levels of the major norepinephrine (NE) metabolite, 3-methoxy-4-hydroxyphenylglycol (MHPG), were measured in a rapidly cycling bipolar patient and her first-degree relatives. The mood state dependence and the reciprocal relationship between noradrenergic and cholinergic activity were investigated by assessing mood, thought disorder, and plasma MHPG following the infusion of physostigmine. A correlation was found between plasma MHPG and mood states, with exceedingly high levels during mania and hypomania. Levels were significantly decreased by ECT or combined lithium and chlorpromazine a

Interactions between anticholinesterase agents and neuroleptics in terms of cholinesterase inhibition in brain and other tissues of rats.

Treatment of rats with chlorpromazine (CPZ, 15 mg/kg i.p. 60 min before sacrifice) did not modify cholinesterase (ChE) activity, but considerably enhanced the inhibition of total ChE induced by physostigmine (PhS, 0.5 mg/kg i.p. 40 min after CPZ) in brain, skeletal muscle, myocardium, lung, liver, and kidney. Additional experiments also showed a prolongation of PhS inhibition by CPZ in brain. The enhanced inhibition of total ChE due to CPZ depended in most peripheral organs on the effect on pseudoChE (as measured by a spectrophotometric method), except in the case of skeletal muscle in which p

Effects of prostaglandin E2 and cholinergic drugs on intraocular pressure.

Effects of prostaglandin E2 (PGE2) and cholinergic drugs on intraocular pressure (IOP) have been studied in the rabbit. PGE2 injected intracamerally in a dose of 5 micrograms produced a long-lasting increase in IOP. Topical pretreatment with carbachol, physostigmine and pilocarpine abolished the rise of IOP after the intracameral injection of PGE2. When the IOP had already been increased by PGE2 topical application of pilocarpine and physostigmine did not change it. Carbachol even potentiated the pressor effect of PGE2. Topical application of cholinergic drugs only slightly increased protein c

Oral and Transdermal Rivastigmine for the Treatment of Anticholinergic Delirium: A Case Report.

BACKGROUND: Anticholinergic toxicity is a common cause of delirium in emergency department patients. The standard antidotal treatment for anticholinergic toxicity is physostigmine. Physostigmine functions as a reversible acetylcholinesterase inhibitor that readily crosses the blood-brain barrier. Rivastigmine is another member of this class currently approved for the treatment of Alzheimer's disease and Parkinson's disease. Rivastigmine also crosses the blood-brain barrier and has been found to be effective in the management of anticholinergic toxicity in limited case reports. CASE REPORT: A

Conservative and surgical semen retrieval in patients with spinal cord injury.

Assisted semen retrieval was applied in 219 men with spinal cord injury (212 supranuclear, seven infranuclear lesions). Vibrostimulation in supranuclear lesions was successful in 133, and in five more after physostigmine injection. Electroejaculation was successful in all seven infranuclear lesions and in four supranuclear patients failing with vibrostimulation. Eight more supranuclear patients responded to electroejaculation and physostigmine. Surgical retrieval was applied in 27 patients. Nine Wagenknecht spermatoceles showed only one success, from all 18 implanted Brindley reservoirs semen

Studies on the receptors involved in the action of the various agents in the phenylbenzoquinone analgesic assay in mice.

1. Tolerance to the activity of several narcotic analgesics (morphine, levorphanol, and methadone) and several narcotic-antagonist analgesics (pentazocine, cyclazocine, and nalorphine) was studied in the mouse phenylbenzoquinone stretching test. Virtually complete tolerance was induced by chronic treatment with each of the narcotic agents, while no apparent tolerance was induced by the narcotic antagonists.2. In morphine-tolerant mice there was a high degree of cross-tolerance to the effects of not only the other narcotic drugs but also to those of the narcotic antagonists, acetylsalicylic aci

Central anticholinergic syndrome (CAS) in anesthesia and intensive care.

Many of the drugs used in anesthesia and intensive care may cause blockade of the central cholinergic neurotransmission. Acetylcholine is of significance in modulation of the interaction among most other central transmitters. The clinical picture of the central cholinergic blockade, known as the central anticholinergic syndrome (CAS), is identical with the central symptoms of atropine intoxication. This behaviour consists of agitation including seizures, restlessness, hallucinations, disorientation or signs of depression such as stupor, coma and respiratory depression. Such disturbances may be

Severity of manchineel fruit (Hippomane mancinella) poisoning: A retrospective case series of 97 patients from French Poison Control Centers.

In this retrospective series of 97 cases of manchineel fruit ingestion reported to French Poison Control Centers between 2009 and 2017, we investigated cases of poisoning due to manchineel fruit (from the Hippomane mancinella tree). This fruit is known to be responsible for oropharyngeal and gastrointestinal tract lesions and possibly hypotension and bradycardia (previously attributed to the presence of physostigmine). The most commonly observed clinical signs were oropharyngeal pain, abdominal pain, diarrhea and oropharyngeal irritation. No major gastrointestinal tract lesions were observed i

A quantitative study of the effects of cholinergic drugs on carotid chemoreceptors in the cat.

1. Conflicting qualitative evidence exists concerning the effects on chemoreceptor activity of some drugs which influence the cholinergic system. Quantitative evidence has been obtained in the present study which should resolve the conflict. 2. Experiments were performed in pentobarbitone-anaesthetized cats in which the activity of chemoreceptor units in the sinus nerve was used to assess chemoreceptor responses. The effects of drugs on responses to I.A. ACH and NaCN were determined from dose-response data obtained from several animals and expressed as mean dose ratios 3. The chemoreceptor res

Nicotine affects the occipital theta rhythm after lesion of the pedunculopontine tegmental nucleus in rats.

The pedunculopontine tegmental nucleus (PPTg) is assumed to be one of the brainstem sites for eliciting the theta rhythm that is expressed in rat hippocampal and occipital EEG in high arousal states. Unilateral lesions of the PPTg in rats induced a hemispheric asymmetry in the expression of occipital theta waves during exploratory sniffing with a significantly lower mean power in the lesioned hemisphere. Acetylcholine was found to be the main neurotransmitter used by the PPTg neurones. Therefore, the present study was conducted to determine if nicotine was effective in counteracting the lesion

Drugs influencing the GABAergic neurotransmission have no effect on the non-epileptic myoclonus of baboons.

In Papio papio baboons benzodiazepines can facilitate the appearance of a naturally occurring non-epileptic myoclonus, suggesting a possible role of GABAergic transmission in their physiopathology. Nevertheless, as this myoclonus is blocked by physostigmine, the effect of benzodiazepines is probably due to their indirect action on the cholinergic system. Therefore, in this study, we report the effects on the non-epileptic myoclonus of drugs influencing GABAergic transmission. Systemic injections of progabide (GABA precursor), baclofen (GABAB receptor agonist) and allylglycine (glutamic acid de

Effects of anti-cholinergic and cholinergic drugs on habituation to motion in rats.

The effects of the anti-cholinergic drug scopolamine, an anti-motion sickness drug, and of the cholinergics physostigmine and neostigmine on habituation to motion sickness in rats were examined using pica, measured as eating of kaolin, as a behavioural index of motion sickness in rats. Rats were rotated around two axes for 1 h once a day for 10 or 11 days. Rotation-induced kaolin intake of control rats gradually decreased from day 9 of daily rotation. Test rats were not treated for the first 3 days, given drugs on days 4-7 of rotation and then again given no drugs for the next 3 or 4 days. Rot

[Aminophylline antagonism of the sedation caused by meperidine and diazepam].

Recent reports showed that aminophylline antagonizes the sedation caused by diazepam and narcotic agents. This study examined the antagonism of diazepam and meperidine in extracorporeal shock wave lithotripsy (ESWL) anesthesia by low dose aminophylline. Eighty patients, aged 20-60 year-old, ASA I-II were randomly selected. No premedication was given. Anesthesia was induced and maintained with diazepam (0.2-0.5 mg/kg) and meperidine (2-5 mg/kg). The patients were divided into two groups. Group A, the control group, was given physostigmine 1 mg iv and naloxone 0.4 mg im as antagonists for diazep

Validation of a photobeam system for assessment of motor activity in rats.

To evaluate the validity and sensitivity of test procedures for routine assessment of chemically induced changes in motor activity in rats, studies were carried out with a commercially available photocell cage system. Testing of single, untreated rats showed that motor activity decreased within 10 min to a steady level. Overall activity was the same when testing was repeated at weekly intervals for 6 weeks. In groups of each 10 rats tested immediately after i.p. injection, chlorpromazine HCl (6 or 2 mg/kg) and physostigmine salicylate (0.5 mg/kg) decreased activity. d-Amphetamine sulfate (1.0

Hyperuricemic effects of cholinergic agents in rats.

The hyperuricemic effect of cholinergic agents was studied in rats given potassium oxonate which inhibits urate oxidase and in functionally hepatectomized rats. Acetylcholine slightly increased plasma uric acid in the oxonate-treated animals, and the effect was markedly enhanced by pretreatment with physostigmine. Carbachol alone showed the hyperuricemic effect at a smaller dose than acetylcholine. The hyperuricemic effects of physostigmine plus acetylcholine and carbachol alone were also observed in eviscerated rats with nonfunctioning livers. Nephrectomy markedly potentiated cholinergic agen

Neuroeffector mechanisms in the voiding cycle of the guinea-pig urinary bladder.

1. The effect of drugs expected to modify cholinergic (atropine, physostigmine) or GABA-ergic (GABA, picrotoxin, bicuculline methiodide, BMI) neurotransmission were investigated on the voiding cycle of the urinary bladder in urethane-anaesthetized guinea-pigs. 2. Transvesical saline infusion elicited a series of regular voiding cycles which were abolished by topical tetrodotoxin as well as by topical or intravenous (i.v.) hexamethonium, thus indicating their reflex neurogenic origin. 3. Either i.v. or topical atropine decreased amplitude and duration of micturition contraction, reduced efficie

Clinical pharmacology of lorazepam.

As a preanesthetic medication, lorazepam is available for oral, intravenous, or intramuscular administration. A parenteral dose of 0.04 to 0.06 mg per kg has been shown to be most effective as a preanesthetic medication in terms of antianxiety and antirecall effect (Table 1). Lorazepam has as its predominant advantage over other benzodiazepines the ability to produce anterograde amnesia reliably and for a relatively long duration. From an anesthesia standpoint, the drug finds its major usage as a premedicant or adjuvant (administered in the peri-induction period) to minimize the possibility of

Voltage clamp study of fast excitatory synaptic currents in bullfrog sympathetic ganglion cells.

Excitatory postsynaptic currents (EPSCs) have been studied in voltage-clamped bullfrog sympathetic ganglion B cells. The EPSC was small, rose to a peak within 1-3 ms, and then decayed exponentially over most of its time-course. For 36 cells at --50 mV (21-23 degrees C), peak EPSC size was --6.5 +/- 3.5 nA (mean +/- SD), and the mean decay time constant tau was 5.3 +/- 0.9 ms. tau showed a small negative voltage dependence, which appeared independent of temperature, over the range --90 to --30 mV; the coefficient of voltage dependence was --0.0039 +/-0.0014 mV-1 (n = 29). Cholinergic approaches to the treatment of progressive supranuclear palsy.

In spite of the severe loss of cholinergic neurons in the brains of patients with progressive supranuclear palsy (PSP), marginal or null benefits are seen in clinical trials after the administration of physostigmine, a cholinesterase inhibitor, or RS-86, a cholinergic agonist. The possible role of cholinergic therapy in PSP is reevaluated.
