Ultraviolet-B radiation effects on the structure and function of lower trophic levels of the marine planktonic food web.

The impact of UV-B radiation (UVBR; 280-320 nm) on lower levels of a natural plankton assemblage (bacteria, phytoplankton and microzooplankton) from the St. Lawrence Estuary was studied during 9 days using several immersed outdoor mesocosms. Two exposure treatments were used in triplicate mesocosms: natural UVBR (N treatment, considered as the control treatment) and lamp-enhanced UVBR (H treatment, simulating 60% depletion of the ozone layer). A phytoplankton bloom developed after day 3, but no significant differences were found between treatments during the entire experiment for phytoplankton

The impact of dissolved inorganic nitrogen and phosphorous on responses of microbial plankton to the Texas City "Y" oil spill in Galveston Bay, Texas (USA).

Ongoing bioremediation research seeks to promote naturally occurring microbial polycyclic aromatic hydrocarbon (PAH) degradation during and after oil spill events. However, complex relationships among functionally different microbial groups, nutrients and PAHs remain unconstrained. We conducted a surface water survey and corresponding nutrient amendment bioassays following the Texas City "Y" oil spill in Galveston Bay, Texas. Resident microbial groups, defined as either heterotrophic or autotrophic were enumerated by flow cytometry. Heterotrophic abundance was increased by oil regardless of nu

A Sulfur-Modified Cryptolepine-Based Type I/II AIE Photosensitizer for Efficient Eradication of Planktonic Bacteria and Biofilms.

In recent years, the application of aggregation-induced emission photosensitizers (AIE PSs) in photodynamic therapy has gained significant traction as an effective approach for eradicating bacteria and biofilms. We developed a series of AIE PSs based on structural modifications of the natural product cryptolepine. Among them, TPA-CRYS exhibited exceptional type I/II reactive oxygen species (ROS) generation capability. At a low concentration of 20 nM, TPA-CRYS exerted potent antibacterial activity, achieving over 99.7% eradication of both Staphylococcus aureus and MRSA. Notably, TPA-CRYS effect

Synergistic antibacterial activity of norfloxacin and sulfadiazine against planktonic and biofilm-forming multidrug-resistant Escherichia coli strain.

Antimicrobial resistance represents a major global health concern, particularly in countries where multidrug-resistant (MDR) pathogens are widespread. Biofilm formation further complicates therapeutic strategies. This study investigated the synergistic effects of combining norfloxacin (NOR) and sulfadiazine (SDZ) against three Escherichia coli strains: a reference, a quinolone-resistant clinical isolate, and a highly resistant extended-spectrum β-lactamase (ESBL)-producing strain. Checkerboard assays and isobolograms revealed synergistic or partially synergistic effects across all strains, wit

Long-Term Changes in Planktonic Ciliate Beta Diversity Under Contrasting Hydrological Regulation in A River-Floodplain System.

This study assessed temporal beta (β) diversity of planktonic ciliates over 14 years (2010-2024) in the Upper Paraná River floodplain, comparing a dam-impacted system (EID) and a non-impacted, protected system (ENID). We tested predictions regarding temporal variation in richness, abundance, composition, and community structure. Both systems exhibited declines in richness and density, more pronounced in EID. Temporal β diversity (TBI) was higher in the impacted system, indicating stronger compositional turnover over time. The local contribution to β diversity (LCBD) values were also elevated m

Chamber-by-Chamber Measurements of Planktonic Foraminiferal Mg, Sr, and Na to Ca Ratios With Femtosecond LA-ICP-MS.

RATIONALE: Distribution patterns of foraminifera are controlled by environmental parameters such as temperature, salinity, and nutrient concentrations in each water mass. Since trace elements to Ca ratios of marine microfossil calcite test of foraminifera record environmental and ecological habitat information, we used femtosecond (fs) LA-ICP-MS to obtain accurate chamber-by-chamber Mg/Ca, Sr/Ca, and Na/Ca of four foraminifera species to clarify the impact of foraminiferal depth migration on paleoceanographic reconstruction. The Mg/Ca and Sr/Ca ratios were measured with precision better than 5

Ecological restoration reduces greenhouse gas emissions by altering planktonic and sedimentary microbial communities in a shallow eutrophic lake.

Ecological restoration is a promising approach to alleviate eutrophication. However, its impacts on greenhouse gas (GHG) emissions and the underlying microbial mechanisms in different habitats of lakes remain unclear. To address this knowledge gap, we measured carbon dioxide (CO2), methane (CH4) and nitrous oxide (N2O) fluxes at both water-air and sediment-water interfaces of eutrophic (Caohai) and restored area (Dapokou) of Dianchi Lake, a typical eutrophic lake in China. Meanwhile, we investigated the responses of planktonic and sedimentary bacterial and fungal communities by high-throughput

Eradicating Biofilms of Carbapenem-Resistant Enterobacteriaceae by Simultaneously Dispersing the Biomass and Killing Planktonic Bacteria with PEGylated Branched Polyethyleneimine.

Carbapenem-resistant Enterobacteriaceae (CRE) are emerging pathogens that cause variety of severe infections. CRE evade antibiotic treatments because these bacteria produce enzymes that degrade a wide range of antibiotics including carbapenems and β-lactams. The formation of biofilms aggravates CRE infections, especially in a wound environment. These difficulties lead to persistent infection and non-healing wounds. This creates the need for new compounds to overcome CRE antimicrobial resistance and disrupt biofilms. Recent studies in our lab show that 600 Da branched polyethyleneimine (BPEI) a

In vitro Antifungal Activity of a Novel Antimicrobial Peptide AMP-17 Against Planktonic Cells and Biofilms of Cryptococcus neoformans.

BACKGROUND: Cryptococcus neoformans is a common human fungal pathogen in immunocompromised people, as well as a prevalent cause of meningitis in HIV-infected individuals. With the emergence of clinical fungal resistance and the shortage of antifungal drugs, it is urgent to discover novel antifungal agents. AMP-17, a novel antimicrobial peptide from Musca domestica, has antifungal activity against C. neoformans. However, its antifungal and anti-biofilm activities remain unclear. Thus, this study aimed to evaluate the antifungal activity of AMP-17 against planktonic cells and biofilms of C. neof

Abundant and Rare Taxa of Planktonic Fungal Community Exhibit Distinct Assembly Patterns Along Coastal Eutrophication Gradient.

Revealing planktonic fungal ecology under coastal eutrophication is crucial to our understanding of microbial community shift in marine pollution background. We investigated the diversity, putative interspecies interactions, assembly processes and environmental responses of abundant and rare planktonic fungal communities along a eutrophication gradient present in the Beibu Gulf. The results showed that Dothideomycetes and Agaricomycetes were the predominant classes of abundant and rare fungi, respectively. We found that eutrophication significantly altered the planktonic fungal communities and

Hit Identification of New Potent PqsR Antagonists as Inhibitors of Quorum Sensing in Planktonic and Biofilm Grown Pseudomonas aeruginosa.

Current treatments for Pseudomonas aeruginosa infections are becoming less effective because of the increasing rates of multi-antibiotic resistance. Pharmacological targeting of virulence through inhibition of quorum sensing (QS) dependent virulence gene regulation has considerable therapeutic potential. In P. aeruginosa, the pqs QS system regulates the production of multiple virulence factors as well as biofilm maturation and is a promising approach for developing antimicrobial adjuvants for combatting drug resistance. In this work, we report the hit optimisation for a series of potent novel

The evolutionary trajectories of P. aeruginosa in biofilm and planktonic growth modes exposed to ciprofloxacin: beyond selection of antibiotic resistance.

Ciprofloxacin (CIP) is used to treat Pseudomonas aeruginosa biofilm infections. We showed that the pathways of CIP-resistance development during exposure of biofilms and planktonic P. aeruginosa populations to subinhibitory levels of CIP depend on the mode of growth. In the present study, we analyzed CIP-resistant isolates obtained from previous evolution experiments, and we report a variety of evolved phenotypic and genotypic changes that occurred in parallel with the evolution of CIP-resistance. Cross-resistance to beta-lactam antibiotics was associated with mutations in genes involved in ce

Efficacy of a ciprofloxacin/amikacin combination against planktonic and biofilm cultures of susceptible and low-level resistant Pseudomonas aeruginosa.

BACKGROUND: Eradicating bacterial biofilm without mechanical dispersion remains a challenge. Combination therapy has been suggested as a suitable strategy to eradicate biofilm. OBJECTIVES: To evaluate the efficacy of a ciprofloxacin/amikacin combination in a model of in vitro Pseudomonas aeruginosa biofilm. METHODS: The antibacterial activity of ciprofloxacin and amikacin (alone, in combination and successively) was evaluated by planktonic and biofilm time-kill assays against five P. aeruginosa strains: PAO1, a WT clinical strain and three clinical strains overexpressing the efflux pumps Mex

Clearance of Staphylococcus aureus from In Vivo Models of Chronic Infection by Immunization Requires Both Planktonic and Biofilm Antigens.

Staphylococcus aureus is a causative agent of chronic biofilm-associated infections that are recalcitrant to resolution by the immune system or antibiotics. To combat these infections, an antistaphylococcal, biofilm-specific quadrivalent vaccine against an osteomyelitis model in rabbits has previously been developed and shown to be effective at eliminating biofilm-embedded bacterial populations. However, the addition of antibiotics was required to eradicate remaining planktonic populations. In this study, a planktonic upregulated antigen was combined with the quadrivalent vaccine to remove the

Planktonic Growth of Pseudomonas aeruginosa around a Dual-Species Biofilm Supports the Growth of Fusobacterium nucleatum within That Biofilm.

PURPOSE: The goal of this study was to understand the potential interaction between Pseudomonas aeruginosa and Fusobacterium nucleatum within the middle ear. METHODS: We examined the microbiota of ear fluid and tympanostomy tubes (TTs) obtained from patients with posttympanostomy tube otorrhea. We also examined biofilms formed by P. aeruginosa and F. nucleatum, singly or together, under aerobic or anaerobic conditions. RESULTS: While the facultative anaerobe P. aeruginosa dominated the bacterial population within the ear fluid, strict anaerobes, including F. nucleatum, dominated bacterial po

Do orally administered antibiotics reach concentrations in the middle ear sufficient to eradicate planktonic and biofilm bacteria? A review.

INTRODUCTION: Infectious conditions of the middle ear are a common and significant cause of morbidity and mortality worldwide. Systemic antibiotics are frequently used, but their effectiveness will depend on whether an adequate antibiotic concentration is achieved in the middle ear; this is especially important in biofilm infections such as otitis media with effusion (OME), where high antibiotic concentrations are typically required for effective treatment. OBJECTIVE: This review examines what antibiotic levels can be reached in the middle ear with oral administration, as a means of guiding r

Comparative proteomic analysis of Streptococcus suis biofilms and planktonic cells that identified biofilm infection-related immunogenic proteins.

Streptococcus suis (SS) is a zoonotic pathogen that causes severe disease symptoms in pigs and humans. Biofilms of SS bind to extracellular matrix proteins in both endothelial and epithelial cells and cause persistent infections. In this study, the differences in the protein expression profiles of SS grown either as planktonic cells or biofilms were identified using comparative proteomic analysis. The results revealed the existence of 13 proteins of varying amounts, among which six were upregulated and seven were downregulated in the Streptococcus biofilm compared with the planktonic controls.

Green-synthesized silver nanoparticles against Streptococcus mutans: antibacterial activity and transcriptomic insights into planktonic and biofilm states.

Dental caries, one of the most common infectious diseases worldwide, is closely associated with Streptococcus mutans (S. mutans) biofilms that exhibit strong resistance to conventional antimicrobial agents. Herein, a green synthesis of silver nanoparticles (AgNPs) is reported to use extracellular metabolic products derived from Pseudomonas putida KT2440 as natural reducing, capping, and stabilizing agents for antibacterial therapy. The resulting AgNPs possess nanoscale size, negative surface charge, and excellent colloidal stability. These green AgNPs display potent antibacterial and antibiofi
