Thiamin biofortification of crops.

Thiamin is essential for human health. While plants are the ultimate source of thiamin in most human diets, staple foods like white rice have low thiamin content. Therefore, populations whose diets are mainly based on low-thiamin staple crops suffer from thiamin deficiency. Biofortification of rice grain by engineering the thiamin biosynthesis pathway has recently been attempted, with up to 5-fold increase in thiamin content in unpolished seeds. However, polished seeds that retain only the starchy endosperm had similar thiamin content than that of non-engineered plants. Various factors such as

Effects of enhancing endogenous and exogenous glutathione in roots on cadmium movement in Arabidopsis thaliana.

Glutathione (GSH) is a thiol-containing compound involved in many aspects of plant metabolism. In the present study, we investigated how enhancing endogenous and exogenous GSH affects cadmium (Cd) movement and distribution in Arabidopsis plants cultured hydroponically. Transgenic Arabidopsis plants with a strong ability to synthesize GSH in roots were generated by transforming the gene encoding the bifunctional γ-glutamylcysteine synthetase-glutathione synthetase enzyme from Streptococcus thermophiles (StGCS-GS). Enhancing endogenous and exogenous GSH decreased the Cd translocation ratio in di

Control of foreign polypeptide localization in specific layers of protein body type I in rice seed.

Prolamin-GFP fusion proteins, expressed under the control of native prolamin promoters, were localized in specific layers of PB-Is. Prolamin-GFP fusion proteins were gradually digested from outside by pepsin digestion. In rice seed endosperm, protein body type I (PB-I) has a layered structure consisting of prolamin species and is the resistant to digestive juices in the intestinal tract. We propose the utilization of PB-Is as an oral vaccine carrier to induce mucosal immune response effectively. If vaccine antigens are localized in a specific layer within PB-Is, they could be protected from ga

Proteomic Techniques and Management of Flooding Tolerance in Soybean.

Climate change is considered a major threat to world agriculture and food security. To improve the agricultural productivity and sustainability, the development of high-yielding stress-tolerant, and climate-resilient crops is essential. Of the abiotic stresses, flooding stress is a very serious hazard because it markedly reduces plant growth and grain yield. Proteomic analyses indicate that the effects of flooding stress are not limited to oxygen deprivation but include many other factors. Although many flooding response mechanisms have been reported, flooding tolerance mechanisms have not bee

Strigolactones regulate protonema branching and act as a quorum sensing-like signal in the moss Physcomitrella patens.

Strigolactones are a novel class of plant hormones controlling shoot branching in seed plants. They also signal host root proximity during symbiotic and parasitic interactions. To gain a better understanding of the origin of strigolactone functions, we characterised a moss mutant strongly affected in strigolactone biosynthesis following deletion of the CAROTENOID CLEAVAGE DIOXYGENASE 8 (CCD8) gene. Here, we show that wild-type Physcomitrella patens produces and releases strigolactones into the medium where they control branching of protonemal filaments and colony extension. We further show tha

Decrease of phosphoribulokinase activity by antisense RNA in transgenic tobacco: definition of the light environment under which phosphoribulokinase is not in large excess.

To test the hypothesis that the contribution of phosphoribulokinase (PRK) to the control of photosynthesis changes depending on the light environment of the plant, the response of transgenic tobacco (Nicotiana tabacum L.) transformed with antisense PRK constructs to irradiance was determined. In plants grown under low irradiance (330 micromol m(-2) s(-1)) steady-state photosynthesis was limited in plants with decreased PRK activity upon exposure to higher irradiance, with a control coefficient of PRK for CO2 assimilation of 0.25 at and above 800 micromol m(-2) s(-1). The flux control coefficie

Multiplex CRISPR/Cas12a editing of ROC5, GS3, GW2, and LARGE2 reveals effects on yield-related traits in rice cultivar Kasalath.

Sustainable improvement of rice yield requires coordinated modification of multiple agronomic traits, yet the agronomic outcomes of multiplex genome editing remain difficult to predict. Here, we applied CRISPR/Cas12a-mediated multiplex editing in the indica rice cultivar Kasalath to target six yield-related genes, Gn1a, TAD1, ROC5, GS3, GW2, and LARGE2, using a single Agrobacterium-delivered construct. Sanger sequencing of 28 T0 plants detected edits in Gn1a (3.57%), ROC5 (7.14%), GS3 (35.71%), and GW2 (10.71%), whereas TAD1 and LARGE2 showed no detectable T0 edits. Single, double, and triple

Characterization of Transgenic Cotton Plants by GUS Histochemical Staining and GUS Fluorometric Assay.

Transgenic cotton plants are responsible of a greater breakthrough in the field of agricultural biotechnology which focuses on improving plant characteristics such as pest resistance, drought tolerance, and fiber quality. The precise molecular characterization of these genetically transformed plants is fundamental in assessing the insertion, inheritance, and expression of transgenes. The β-glucuronidase (GUS) assay is among the conventional tools to test transgenic cotton that offer a consistent and productive process for determining gene expression. This chapter offers the exploration of the

Differential effects of arsenite and arsenate on rice (Oryza sativa) plants differing in glutathione S-transferase gene expression.

Contamination of paddy soils with arsenic (As) can cause phytotoxicity in rice and increase the accumulation of arsenic in grains. The uptake and accumulation of As in rice depends on the different As species present in the soil. Plants detoxify As by conjugating and sequestering xenobiotic compounds into vacuoles using various enzymes. However, the severity of damage induced by arsenite (As(III)) and arsenate (As(V)), as well as the roles of glutathione S-transferase in detoxifying these As species in rice, are not fully understood. In this study, we developed plant materials overexpressing a

Identification and characterization of a new geminivirus from soybean plants and determination of V2 as a pathogenicity factor and silencing suppressor.

BACKGROUND: Soybean is one of the four major crops in China. The occurrence of viruses in soybean causes significant economic losses. RESULTS: In this study, the soybean leaves from stay-green plants showing crinkle were collected for metatranscriptomic sequencing. A novel geminivirus, tentatively named soybean geminivirus A (SGVA), was identified in soybean stay-green plants. Sequence analysis of the full-length SGVA genome revealed a genome of 2762 nucleotides that contain six open reading frames. Phylogenetic analyses revealed that SGVA was located adjacent to the clade of begomoviruses in

Overexpression of AspAT alleviates the inhibitory effects of ammonium on root development in Populus tomentosa.

Ammonium toxicity, resulting from prolonged use of ammonium as the sole nitrogen source, can lead to physiological and morphological disorders, ultimately stunting plant growth. Enhancing ammonium assimilation efficiency has been extensively explored as a strategy to mitigate ammonium toxicity. However, the role of AspAT, a key enzyme in nitrogen assimilation, remains underexplored. This study investigates the function of AspAT in alleviating ammonium toxicity and uncovers the underlying physiological mechanisms. The results show that two Populus AspAT genes, AspAT13 and AspAT15, exhibit the h

GmSTOP1-3 Increases Soybean Manganese Accumulation Under Phosphorus Deficiency by Regulating GmMATE2/13 and GmZIP6/GmIREG3.

Mineral nutrient deficiencies and metal ion toxicities coexist on acid soils. Phosphorus (P) deficiency in plants is generally accompanied with significant levels of leaf manganese (Mn) accumulation. However, the molecular regulatory mechanisms underpinning remain unclear. The present study found that P-deficient soybean plants accumulated more Mn compared to P-sufficient ones on acid soils in both field and greenhouse experiments. Meanwhile, both P deficiency and Mn toxicity enhanced the expression of GmSTOP1-3. Over-expressing GmSTOP1-3 enhanced Mn accumulation in transgenic soybean hairy ro

Armadillo repeat only gene VlARO confers resistance to Botrytis cinerea by promoting guaijaverin accumulation in grapevine.

The ARM repeat only gene VlARO positively regulates grape resistance to gray mold by promoting the guaijaverin accumulation, which directly suppresses the mycelial growth of Botrytis cinerea. Gray mold, caused by the necrotrophic fungus Botrytis cinerea, severely threatens global grapevine production. Here, we identified VlARO, a nuclear-localized gene from the highly resistant grapevine variety 'Beta', as a positive regulator of gray mold resistance. Although VlARO contains Armadillo repeats, it lacks a canonical U-box domain, thus distinguishing it from typical plant U-box E3 ubiquitin ligas

Identification and Functional Analysis of WOX Genes in Macadamia spp. Reveal WOX1 and WOX4 Homologs Involved in Shoot Regeneration.

Macadamia spp., high-value forest crops, face considerable challenges in increasing yields due to inefficient clonal propagation of superior varieties, which severely limits industrial development. This study focused on the WUSCHEL-related homeobox (WOX) family, which includes key regulators of plant regeneration. We systematically identified the members of the WOX family in four cultivated varieties of the "smooth-shelled" type and in one species of the "rough-shelled" type. We demonstrated the presence of 19 WOX family members in M. integrifolia "GR1." Expression analysis revealed that the W

Characterization, expression, and functional analysis of the pathogenesis-related gene PtDIR11 in transgenic poplar.

Lignins and lignans are important for plant resistance to pathogens. Dirigent (DIR) proteins control the regio- and stereo-selectivity of coniferyl alcohol in lignan and lignin biosynthesis. DIR genes have been implicated in defense-related responses in several plant species, but their role in poplar immunity is unclear. We cloned PtDIR11 from Populus trichocarpa; we found that overexpression of PtDIR11 in poplar improved the lignan biosynthesis and enhanced the resistance of poplar to Septotis populiperda. PtDIR11 has a typical DIR domain; it belongs to the DIR-b/d family and is expressed in

FAST-TRACE: delineating tissue-specific cis-regulatory activities in the cadmium/zinc hyperaccumulator Sedum plumbizincicola.

FAST-TRACE is a transient expression system for rapid homologous promoter analysis in Sedum plumbizincicola, enabling identification of critical regulatory regions and a MYB transcription factor, SpTRB4, that activates the SpHMA3 promoter. Investigating the SpHMA3 promoter in Sedum plumbizincicola is essential to elucidate the molecular regulatory mechanisms underlying the above-ground metal detoxification. Given that generating stably transformed plants is time-consuming and costly, an efficient, low-cost method for the preliminary promoter activity assessment is needed. Here, we present FAST

Visualization of auxin responses during vine twining in Ipomoea nil and the functional role of the subapical hook in this process.

This study suggests that hook-shaped bending associated with properly regulated auxin response patterns in the subapical region may contribute to the maintenance of twining in morning glory vines. The twining behavior demonstrated by vine plants represents an important morphogenetic adaptation essential to their survival; however, the physiological mechanisms driving this response remain largely unknown. To elucidate the role of auxin in vine twining in morning glory (Ipomoea nil), we generated and investigated transgenic morning glory carrying the GUS reporter gene under an auxin-responsive p

BSMV-mediated genome editing exhibits host-specific heritability: germline transmission in barley and somatic edits in Nicotiana benthamiana.

BACKGROUND: Plant RNA virus-mediated guide RNA (gRNA) delivery represents a transformative advance in genome editing technologies. Unlike conventional transformation methods that rely on labor-intensive tissue culture and regeneration for each individual gRNA delivery, viral vectors can rapidly and systemically transmit gRNAs into pre-established Cas-expressing plants, providing an accelerated route for functional genomics and trait discovery directly in planta. However, key design parameters, including subgenomic promoter choice, transcript architecture, and their effects on viral fitness and

Pest resistance function of BtACO8 gene from Balsas teosinte.

The peach aphid (Myzus persicae) is a major agricultural pest that causes significant yield losses and transmits numerous plant viruses. We profiled BtACO8 expression in Balsas teosinte and found that aphid infestation induced its transcripts 12.6-fold at 12 h post-infestation relative to non-infested controls. We therefore cloned BtACO8, encoding 1-aminocyclopropane-1-carboxylate oxidase, and investigated its role in aphid resistance by heterologous overexpression in Arabidopsis thaliana. Three homozygous BtACO8-OE lines were obtained and used for functional characterization. Compared with wi

A rapid and efficient in planta assay for assessment of RNA editing factor performance.

Land plant mitochondria and plastids are endosymbiotic organelles that possess their own, reduced genomes. Gene expression is largely regulated at the transcript level, predominantly by nuclear-encoded proteins. Notably, up to thousands of cytidines in organellar RNAs are specifically edited to uridines, mostly restoring conserved genetic information. Key players in this process are nuclear-encoded pentatricopeptide repeat (PPR) proteins, which bind target RNAs in a one-PPR:one-nucleotide manner, following a two-amino-acid-based code. Cytidine deamination is catalyzed by intrinsic or trans-act
