High prevalence of mixed Plasmodium falciparum and Plasmodium vivax co-infections in Duffy negative individuals in Ethiopia.

Malaria remains a major public health concern in Ethiopia, where both Plasmodium falciparum and Plasmodium vivax are endemic. Notably, P.vivax infections have been documented in duffy-negative individuals, who were traditionally thought to be resistant to this parasite. PvDBP gene duplication has been proposed as a mechanism for P. vivax to invade erythrocytes in these individuals, but evidence is limited. We conducted a cross-sectional study from 2020 to 2021 with1,154 febrile patients in three Ethiopian regions: Gambela, Oromia, and SNNPR. Plasmodium species were identified by PCR, Duffy gen

Treatment Efficacy of Dihydroartemisinin-Piperaquine for Uncomplicated Plasmodium falciparum and Plasmodium vivax Malaria in Timika, Papua, Indonesia.

Dihydroartemisinin-piperaquine (DP), the first-line treatment for uncomplicated malaria in Timika, Papua, Indonesia, has also been used for intermittent preventive treatment in pregnancy (IPTp-DP) since February 2022. Concerns about the potential emergence of drug resistance associated with this dual policy prompted the present study, which was conducted to assess DP efficacy in treating uncomplicated Plasmodium falciparum (P. falciparum) and Plasmodium vivax (P. vivax) malaria in the general population 15 months after IPTp-DP was introduced. Between May and December 2023, the current study re

Genetic diversity of Plasmodium vivax and Plasmodium falciparum field isolates from Honduras in the malaria elimination phase.

Malaria continues to be a major threat to public health in tropical regions, primarily affecting sub-Saharan Africa but also Asia, the Middle East, and Latin America. Malaria cases in Honduras have seen a significant decline and the country aims to eliminate the disease by 2030. This study examines the genetic diversity of Plasmodium falciparum and Plasmodium vivax in Honduras using four molecular markers (Pfama1, Pfglurp, Pvmsp3α, and Pvmsp3β), and the chloroquine resistance marker pfcrt in the context of the elimination phase. Our findings indicate that P. falciparum populations in Honduras

Genome-wide microsatellite characteristics of five human Plasmodium species, focusing on Plasmodium malariae and P. ovale curtisi.

Microsatellites can be utilized to explore genotypes, population structure, and other genomic features of eukaryotes. Systematic characterization of microsatellites has not been a focus for several species of Plasmodium, including P. malariae and P. ovale, as the majority of malaria elimination programs are focused on P. falciparum and to a lesser extent P. vivax. Here, five human malaria species (P. falciparum, P. vivax, P. malariae, P. ovale curtisi, and P. knowlesi) were investigated with the aim of conducting in-depth categorization of microsatellites for P. malariae and P. ovale curtisi.

Acute acalculous cholecystitis complicating an imported case of mixed malaria caused by Plasmodium falciparum and Plasmodium vivax.

A 40-year-old man was admitted with a 6-day history of fever and abdominal pain. His right upper quadrant was tender on palpation. A blood smear revealed trophozoite forms of Plasmodium vivax and Plasmodium falciparum. Abdominal ultrasound findings were consistent with acute acalculous cholecystitis. He was treated successfully with quinine and doxycycline and discharged in good clinical condition.

Molecular mechanisms involved in Plasmodium gametocytogenesis.

Malaria, a disease caused by protozoa of the genus Plasmodium, remains a major challenge for global public health. The persistence of disease transmission to the mosquito vector depends on the differentiation of asexual blood-stage parasites into gametocytes, a process known as gametocytogenesis. Interrupting this stage of the parasite's life cycle represents a critical strategy for malaria control and eventual eradication. This review aims to consolidate recent advances in the understanding of the complex molecular mechanisms regulating gametocytogenesis in Plasmodium, with a particular focus

Plasmodium ovale: Exploring an Atypical Presentation.

Malaria is an infection caused by five different Plasmodium species. The most common are Plasmodium falciparum and vivax. Plasmodium ovale is more rarely reported and mostly has a benign course. We present a case of a 40-year-old male with a six-day history of headaches, chills, and fever who was initially evaluated in our emergency room, from where he was discharged after a negative workup for malaria. He returned to the hospital five days later in septic shock. Now P. ovale was found in a blood smear. The patient was treated with three doses of intravenous artesunate, followed by a three-day

Plasmodium cynomolgi: What Should We Know?

Even though malaria has markedly reduced its global burden, it remains a serious threat to people living in or visiting malaria-endemic areas. The six Plasmodium species (Plasmodium falciparum, Plasmodium vivax, Plasmodium malariae, Plasmodium ovale curtisi, Plasmodium ovale wallikeri and Plasmodium knowlesi) are known to associate with human malaria by the Anopheles mosquito. Highlighting the dynamic nature of malaria transmission, the simian malaria parasite Plasmodium cynomolgi has recently been transferred to humans. The first human natural infection case of P. cynomolgi was confirmed in 2

Plasmodium vivax transcriptomics: What is new?

Malaria is the leading human parasitosis and is transmitted through the bite of anopheline mosquitoes infected with parasites of the genus Plasmodium spp. Among the seven species that cause malaria in humans, Plasmodium vivax is the most prevalent species in Latin America. In recent years, there have been an increasing number of reports of clinical complications caused by P. vivax infections, which were previously neglected and underestimated. P. vivax biology remains with large gaps. The emergence of next-generation sequencing technology has ensured a breakthrough in species knowledge. Couple

Symptomatic Plasmodium vivax Infection in Rwanda.

We report a Plasmodium vivax infection in a Rwandan child misdiagnosed with Plasmodium falciparum and administered artemether-lumefantrine. Antigen detection revealed an absence of P falciparum histidine-rich protein 2 (HRP2) and presence of Plasmodium vivax lactate dehydrogenase. Nested and real-time polymerase chain reactions verified that the sample only contained P vivax deoxyribonucleic acid.

Apicoplast Dynamics During Plasmodium Cell Cycle.

The deadly malaria parasite, Plasmodium falciparum, contains a unique subcellular organelle termed the apicoplast, which is a clinically-proven antimalarial drug target. The apicoplast is a plastid with essential metabolic functions that evolved via secondary endosymbiosis. As an ancient endosymbiont, the apicoplast retained its own genome and it must be inherited by daughter cells during cell division. During the asexual replication of P. falciparum inside human red blood cells, both the parasite, and the apicoplast inside it, undergo massive morphological changes, including DNA replication a

Plasmodium vivax infection compromises reticulocyte stability.

The structural integrity of the host red blood cell (RBC) is crucial for propagation of Plasmodium spp. during the disease-causing blood stage of malaria infection. To assess the stability of Plasmodium vivax-infected reticulocytes, we developed a flow cytometry-based assay to measure osmotic stability within characteristically heterogeneous reticulocyte and P. vivax-infected samples. We find that erythroid osmotic stability decreases during erythropoiesis and reticulocyte maturation. Of enucleated RBCs, young reticulocytes which are preferentially infected by P. vivax, are the most osmoticall

Plasmodium falciparum goes bananas for sex.

The sexual blood stages of the human malaria parasite Plasmodium falciparum undergo a remarkable transformation from a roughly spherical shape to an elongated crescent or "falciform" morphology from which the species gets its name. In this review, the molecular events that drive this spectacular shape change are discussed and some questions that remain regarding the mechanistic underpinnings are posed. We speculate on the role of the shape changes in promoting sequestration and release of the developing gametocyte, thereby facilitating parasite survival in the host and underpinning transmissio

Plasmodium falciparum Multidrug Resistance Proteins (pfMRPs).

The capacity of the lethal Plasmodium falciparum parasite to develop resistance against anti-malarial drugs represents a central challenge in the global control and elimination of malaria. Historically, the action of drug transporters is known to play a pivotal role in the capacity of the parasite to evade drug action. MRPs (Multidrug Resistance Protein) are known in many phylogenetically diverse groups to be related to drug resistance by being able to handle a large range of substrates, including important endogenous substances as glutathione and its conjugates. P. falciparum MRPs are associa

Stressed Out About Plasmodium falciparum Gametocytogenesis.

Blocking malaria transmission is critical to malaria control programs but remains a major challenge especially in endemic regions with high levels of asymptomatic infections. New strategies targeting the transmissible sexual stages of the parasite, called gametocytes, are needed. This review focuses on P. falciparum gametocytogenesis in vivo and in vitro. Highlighting advances made elucidating genes required for gametocyte production and identifying key questions that remain unanswered such as the factors and regulatory mechanisms that contribute to gametocyte induction, and the mechanism of s

Human Plasmodium vivax mosquito experimental transmission.

Plasmodium vivax bench research greatly lags behind Plasmodium falciparum because of an inability to culture in vitro. A century ago, intentionally inducing a malaria infection was a strategy commonly used to cure late-stage syphilis. These controlled human malaria infections were used with expertise and persisted to the end of World War II. While controlled malaria liver-stage infection has been achieved for both P. vivax and P. falciparum, controlled human transmission to mosquitoes falls short for both species. In this issue of the JCI, Collins et al. present groundbreaking work that establ

Severe Plasmodium vivax infection in Korea.

BACKGROUND: Although severe malaria by Plasmodium vivax has been increasingly reported, there are marked variations in the type and rate of the complications by geographic area. This is possibly because of the presence of concurrent falciparum malaria or bacteraemia, and of differences in underlying immune status among the infected subjects. Furthermore, published studies on P. vivax in temperate regions are limited. The present study investigated severe vivax malaria in Korea, where only vivax malaria occurs. Hence, other compounding factors are rare. Additionally, most of the patients are po

Geographical variation in Plasmodium vivax relapse.

BACKGROUND: Plasmodium vivax has the widest geographic distribution of the human malaria parasites and nearly 2.5 billion people live at risk of infection. The control of P. vivax in individuals and populations is complicated by its ability to relapse weeks to months after initial infection. Strains of P. vivax from different geographical areas are thought to exhibit varied relapse timings. In tropical regions strains relapse quickly (three to six weeks), whereas those in temperate regions do so more slowly (six to twelve months), but no comprehensive assessment of evidence has been conducted.
