The lateral geniculate nucleus (LGN) is a key thalamic nucleus in the primate visual system that relays visual information from the retina to cortical visual areas. While functional and microanatomical characteristics of the LGN and its layers have been extensively studied, previous investigations on its receptor architecture have been restricted to a small subset of neurotransmitter receptors. To characterise the receptor architecture of the macaque LGN in greater detail, we analysed in vitro autoradiography data to quantify the density of 15 neurotransmitter receptors at the sublayer level,

The relationship between different cell populations in monkey primary visual cortex and their role in visual function is not fully resolved. We combined high-density Neuropixels recordings across layers of macaque V1, and a state-of-the-art nonlinear dimensionality reduction approach on waveform shape to delineate nine putative cell classes: 4 narrow-spiking (NS), 4 broad-spiking (BS), and 1 triphasic (TP). Then, we performed targeted analyses of laminar organization, spike amplitude, multichannel spatial features, functional properties, and network connectivity of these cell classes. These an

A patient with complete cortical blindness after bilateral posterior cerebral artery infarctions denied her blindness. Her pretended visual experiences could frequently be traced back to synaesthetic translations of acoustic or tactile perceptions into mental visual images. Possibly, the belief to see resulted from a confusion of mental visual images with real percepts. The patient manifested preserved visual imagery also by correct responses to questions concerning the shapes of letters and the shapes and colours of objects. MRI showed an almost complete destruction of primary visual cortex w

Our previous study with functional magnetic resonance imaging (MRI) demonstrated that acupuncture stimulation of the vision-related acupoint, B1-67, activates the visual cortex of the human brain. As a further study on the effect of B1-67 acupuncture stimulation on the visual cortex, we examined c-Fos expression in binocularly deprived rat pups. Binocular deprivation significantly reduced the number of c-Fos-positive cells in the primary visual cortex, compared with that of normal control rat pups. Interestingly, acupuncture stimulation of B1-67 resulted in a significant increase in the number

Neuronal populations in primary visual cortex adapt both to stimulus contrast and to the probability of occurrence of visual patterns. Previous work showed that the magnitude of the population response follows a separable power-law function of contrast and stimulus probability, suggesting the existence of a shared gain mechanism. Here we ask whether a similar equivalence extends beyond response magnitude to the full distribution of activity across neurons within a trial. Across a wide range of adaptation states, we find that population responses are highly sparse and well described by a zero-i

A long-standing assumption in cognitive neuroscience is that visual mental imagery (VMI) is "visual perception in reverse" and, as such, requires reactivation of the primary visual cortex (V1). On this view, total destruction of V1 should abolish VMI and provoke neurological aphantasia. We tested this prediction by reviewing 55 published cases of cortical blindness (CB) in which VMI abilities were assessed. Vivid VMI is repeatedly documented in CB patients with complete or near-complete bilateral destruction, disconnection, or functional loss of V1; in impaired-VMI cases with informative anato

In primates, primary visual cortex (V1) circuits support early visual processing, whereas in the lateral prefrontal cortex (LPFC), they support higher-order computations. Here, we characterized the intrinsic electrophysiological (e)-properties and morphology of excitatory (EXC), fast-spiking inhibitory (FSI), and non-fast-spiking inhibitory (NFSI) neurons in layers I-III of V1 and LPFC, of a small primate, the common marmoset. EXC and FSI neurons exhibited broader action potentials in LPFC than in V1. Compared to V1, LPFC EXC neurons exhibited reduced excitability, whereas FSI neurons showed i

Visual input reaches the primary visual cortex (V1) at the stria of Gennari, a band of highly myelinated fibers at the level of layer IV. Damage to the visual pathways can cause visual field defects (scotomas) that prevent visual input from reaching V1. Here, we evaluate whether the stria of Gennari degrades in the absence of visual input. Using 7 Tesla MRI, we investigate the integrity of the stria of Gennari in case of acquired homonymous visual field defects, in particular hemianopia: the loss of vision in half of the visual field. These visual field defects allow us to compare stria of Gen

How early sensory experience during "critical periods" of postnatal life affects the organization of the mammalian neocortex at the resolution of neuronal cell types is poorly understood. We previously reported that the functional and molecular profiles of layer 2/3 (L2/3) cell types in the primary visual cortex (V1) are vision-dependent [S. Cheng et al., Cell 185, 311-327.e24 (2022)]. Here, we characterize the spatial organization of L2/3 cell types with and without visual experience. Spatial transcriptomic profiling based on 500 genes recapitulates the zonation of L2/3 cell types along the p

Head movements are sensed by the vestibular organs. Unlike classical senses, signals from vestibular organs are not selectively conveyed to a dedicated cortical area but are broadcast throughout the cortex. This distributed processing pattern reflects the fundamental role of vestibular information in contextual modulation across diverse cortical computations. Surprisingly, the routes taken by vestibular signals to reach the cortex are still largely uncharted. Here, we show that the primary visual cortex (V1) receives real-time head movement signals-direction, velocity, and acceleration-from th

PRÉCIS: The area corresponding to the peripheral visual field in the primary visual cortex of the brain is associated with ophthalmologic parameters.
PURPOSE: Reproducing the relationship of the visual field with the primary visual cortex is known as retinotopic mapping. The primary visual cortex is known to be atrophic in glaucoma patients. However, there are few reports on which areas most reflect glaucomatous changes. In this study, we used magnetic resonance imaging to evaluate the relationship between various ophthalmic parameters and overall and regional structural changes in the primar

Damage to the primary visual cortex (V1) causes field defects in conscious vision, yet stimuli in the affected field may still guide saccades and other visuomotor responses ("blindsight"). Chronic blindsight is well documented, but early postlesional saccade profiles to scotoma stimuli remain poorly characterized. We mapped visuomotor functions in two macaques 3 to 4 weeks after a unilateral V1 lesion using a perimetry-like saccade assay up to 35° eccentricity, with analyses of saccade direction and catch-trial baselines. Targets had either unpredictable onset, as in clinical perimetry, or tem

OBJECTIVE: The primary visual cortex (V1) supports both rapid feedforward coding of local features and contextual processing shaped by recurrent and top-down feedback. While fear learning is known to prioritize threat-predictive signals in V1, it remains unclear whether this prioritization uniformly enhances all stages of cortical processing or selectively biases certain computations. We investigated how fear conditioning modulates V1 population coding for simple gratings versus global contours, which rely on distinct temporal stages of integration.
METHODS: Monkeys were trained in a visual f

To move through the world, animals extract visual features from objects. Although the dorsomedial striatum (DMS) contains visual processing circuits, visual perception is considered a cortical attribute. Here, we asked whether neurons from the DMS encode visual features in the absence of behavioral contingencies contributing to visual perception. Using electrophysiological recordings of spiny projection neurons (SPNs) located in a target region of the primary visual cortex, we identified visually responsive neurons. A low percentage of SPNs were orientation selective. However, different orient

The intrinsically photosensitive retinal ganglion cells (ipRGCs) are the third class of photoreceptors apart from rods and cones, containing the photopigment melanopsin and are characterized into different subtypes (M1-M6) that support conscious visual perception. They transmit the light information to the brain through complex circuits serving both non-image-forming and image-forming functions. Of the total population, there are four main ipRGCs, such as M2, M4, M5 and M6 that project substantially to the image-forming visual pathway and target the dorsal lateral geniculate nucleus (dLGN). Using functional magnetic resonance imaging (fMRI), the authors examined visual cortex function in Parkinson's disease patients who did and did not experience visual hallucinations. Patients with visual hallucinations demonstrated increased activation in the visual association cortex and deficits in the primary visual cortex, suggesting that visual hallucinations are associated with an abnormality of visual-cortex function.

Motion perception is crucial to animal survival and effective environmental interactions. In mammals, detection of movement begins in the retina. Directionally selective (DS) retinal ganglion cells were first discovered in the rabbit eye,1 and they have since been found in mouse,2,3 cat,4 and monkey.5,6 These DS retinal neurons contact a small population of neurons in the visual thalamus (dorsal lateral geniculate nucleus [LGN]) that are highly DS.7,8,9,10 The primary visual cortex (V1) also contains DS neurons, but whether directional selectivity in V1 emerges de novo11,12,13 or is inherited

The mammalian neocortex, organized into six cellular layers or laminae, forms a cortical network within layers. Layer-specific computations are crucial for sensory processing of visual stimuli within the primary visual cortex. Laminar recordings of local field potentials (LFPs) are a powerful tool to study neural activity within cortical layers. Electric brain stimulation is widely used in basic neuroscience and in a large range of clinical applications. However, the layer-specific effects of electric stimulation on LFPs remain unclear. To address this gap, we recorded laminar LFP from capuchi

Cerebral visual impairment (CVI) is associated with a wide range of visual perceptual deficits including global motion processing. However, the underlying neurophysiological basis for these impairments remain poorly understood. We investigated global motion processing abilities in individuals with CVI compared to neurotypical controls using a combined behavioral and multi-modal neuroimaging approach. We found that CVI participants had a significantly higher mean motion coherence threshold (determined using a random dot kinematogram pattern simulating optic flow motion) compared to controls. Us

Cross-modal responses are common in primary sensory cortex, yet a recent study showed that signals in primary visual cortex (V1) once labeled multisensory were instead more closely related to face movements. To what extent can multisensory processing in other modalities also be explained by face movements? We measured face movements and neural activity in awake mouse primary auditory cortex (A1) and V1 in response to visual and auditory stimulation as well as during spontaneous activity. We find that visual stimuli rarely evoke face movements, A1 responses to visual input remain robust even in
