Smartphone Typing Dynamics for Assessing Hand Function in Psoriatic Arthritis: A Proof-of-Concept Study.

Psoriatic Arthritis (PsA) is a chronic inflammatory disease affecting both the peripheral and axial skeleton, significantly impacting patients' quality of life. Diagnosing and monitoring PsA remain challenging, as they often rely on subjective assessments by experienced healthcare professionals. In this study, we introduce a novel method for assessing hand function impairment, with emphasis on fine motor skills, in individuals with PsA by developing digital biomarkers (dBMs) that analyze patterns formed during natural typing interactions with touchscreen smartphones. The assessment is conducte

A non-invasive capacitive sensor to investigate the Leidenfrost phenomenon: a proof of concept study.

A volatile sessile liquid droplet or a sublimating solid manifests levitation on its own vapor when placed on a sufficiently heated surface, illustrating the Leidenfrost phenomenon. In this study, we introduce a non-invasive capacitance method for investigating this phenomenon, offering a potentially simpler alternative to existing optical techniques. The designed sensor features in-plane miniaturized electrodes forming a double-comb structure, also known as an interdigitated capacitor. Initially, the sensor's capacitance is characterized for various distances between the sensor and a dielectr

Brain Activation During Virtual Reality Symptom Provocation in Obsessive-Compulsive Disorder: Proof-of-Concept Study.

BACKGROUND: Obsessive-compulsive disorder (OCD) is a psychiatric disorder characterized by obsessions and compulsions. We previously showed that a virtual reality (VR) game can be used to provoke and measure anxiety and compulsions in patients with OCD. Here, we investigated whether this VR game activates brain regions associated with symptom provocation. OBJECTIVE: In this study, we aim to investigate the neural regions that are activated in patients with OCD when they are interactively confronted with a symptom-provoking event and when they are performing compulsive actions in VR. METHODS:

Impaired creative cognition after surgery for an IDH-mutated glioma: A proof-of-concept study.

Assessment of high cognitive functions, such as creativity, is often overlooked in medical practice. However, it is crucial to understand the impact of brain tumors, specifically low-grade gliomas, on creative cognition, as these tumors predominantly affect brain regions associated with cognitive creativity. In this study, we investigated creative cognition using the Alternative Uses Task (AUT) and the Combination of Associates Task (CAT) in a cohort of 29 patients who underwent brain surgery for a low-grade glioma, along with 27 control participants. While the group of patients did not exhibi

Surgical step recognition in laparoscopic distal gastrectomy using artificial intelligence: a proof-of-concept study.

PURPOSE: Laparoscopic distal gastrectomy (LDG) is a difficult procedure for early career surgeons. Artificial intelligence (AI)-based surgical step recognition is crucial for establishing context-aware computer-aided surgery systems. In this study, we aimed to develop an automatic recognition model for LDG using AI and evaluate its performance. METHODS: Patients who underwent LDG at our institution in 2019 were included in this study. Surgical video data were classified into the following nine steps: (1) Port insertion; (2) Lymphadenectomy on the left side of the greater curvature; (3) Lympha

Toward AI-driven neuroepigenetic imaging biomarker for alcohol use disorder: A proof-of-concept study.

Alcohol use disorder (AUD) is a disorder of clinical and public health significance requiring novel and improved therapeutic solutions. Both environmental and genetic factors play a significant role in its pathophysiology. However, the underlying epigenetic molecular mechanisms that link the gene-environment interaction in AUD remain largely unknown. In this proof-of-concept study, we showed, for the first time, the neuroepigenetic biomarker capability of non-invasive imaging of class I histone deacetylase (HDAC) epigenetic enzymes in the in vivo brain for classifying AUD patients from healthy

CArdiac and REspiratory adaptive Computed Tomography (CARE-CT): a proof-of-concept digital phantom study.

Current respiratory 4DCT imaging for high-dose rate thoracic radiotherapy treatments are negatively affected by the complex interaction of cardiac and respiratory motion. We propose an imaging method to reduce artifacts caused by thoracic motion, CArdiac and REspiratory adaptive CT (CARE-CT), that monitors respiratory motion and ECG signals in real-time, triggering CT acquisition during combined cardiac and respiratory bins. Using a digital phantom, conventional 4DCT and CARE-CT acquisitions for nineteen patient-measured physiological traces were simulated. Ten respiratory bins were acquired f

A novel procedure to investigate social anxiety using videoconferencing software: A proof-of-concept study.

Social anxiety disorder (SAD) is very common and can be significantly disabling. New treatments are needed as the remission rate for SAD is the lowest of all the anxiety disorders. Experimental medicine models, in which features resembling a clinical disorder are experimentally induced, are a cost-effective and timely approach to explore potential novel treatments for psychiatric disorders. Following the emergence of SARS-CoV-2, there is a need to develop experimental medicine models that can be carried out remotely. We developed a novel procedure to investigate SAD (the InterneT-based Stress

Recombinant gonadotropin therapy to improve spermatogenesis in nonobstructive azoospermic patients - A proof of concept study.

PURPOSE: Nonobstructive azoospermia (NOA) associated with primary spermatogenic failure is a common cause of male infertility usually considered untreatable; however, some reports have suggested that hormonal stimulation to boost the intra-testicular testosterone level and spermatogenesis might increase the chance of achieving pregnancy using homologous sperm. MATERIALS AND METHODS: We report a series of eight NOA males who received long-term treatment with recombinant human chorionic gonadotropin twice a week for spermatogenesis stimulation. Six males received additional recombinant follicle

Reserpine Substantially Lowers Blood Pressure in Patients With Refractory Hypertension: A Proof-of-Concept Study.

BACKGROUND: Refractory hypertension (RfHTN), a phenotype of antihypertensive treatment failure, is defined as uncontrolled automated office blood pressure (AOBP) ≥130/80 mm Hg and awake ambulatory blood pressure (ABP) ≥130/80 mm Hg on ≥5 antihypertensive medications, including chlorthalidone and a mineralocorticoid receptor antagonist. Previous studies suggest that RfHTN is attributable to heightened sympathetic tone. The current study tested whether reserpine, a potent sympatholytic agent, lowers blood pressure (BP) in patients with RfHTN. METHODS: Twenty-one out of 45 consecutive patients w

Aldosterone synthase inhibition with LCI699: a proof-of-concept study in patients with primary aldosteronism.

We report the first administration of an orally active aldosterone synthase inhibitor, LCI699, to 14 patients with primary aldosteronism. After a 2-week placebo run-in, patients received oral LCI699 (0.5 mg BID) for 2 weeks, LCI699 (1.0 mg BID) for 2 weeks, and placebo for 1 week. We assessed changes in hormone concentrations, plasma potassium levels, and 24-hour ambulatory systolic blood pressure and safety. The supine plasma aldosterone concentration decreased from 540 pmol/L (95% CI: 394 to 739 pmol/L) to 171 pmol/L (95% CI: 128 to 230 pmol/L) after 0.5 mg of LCI699 (-68%; P<0.0001) and to

Using games to identify adolescents at risk for substance misuse-a proof-of-concept study.

INTRODUCTION: Early identification of adolescents at risk for substance use is critical for timely intervention. However, standard screening tools like CRAFFT (Car, Relax, Alone, Forget, Family and Friends, Trouble) and S2BI (Screening to Brief Intervention) face significant barriers, including adolescent disclosure reluctance, limited clinic privacy, and administrative challenges with paper-based forms. Digital games offer a promising alternative by generating behavioral data that may serve as digital biomarkers of substance use risk through engaging, low-burden gameplay. The objective of thi

Leveraging Immersive Virtual Reality to Prevent Child Injuries in the Home: Proof-of-Concept Study.

BACKGROUND: Injury is the leading cause of death among US children, with over 2700 deaths due to unintentional injuries among children aged <6 years in 2023. Many of these deaths occur in the home and involve addressable safety hazards. Evidence-based parenting programs (EBPPs), in which parents work with a provider to learn parenting skills, offer an infrastructure for disseminating home safety content. However, few programs have structured modules on home safety, with most home safety education coming from printed materials or websites. Immersive virtual reality (IVR) allows participants to
