Characterization of cell-to-cell signaling-deficient Pseudomonas aeruginosa strains colonizing intubated patients.

Cell-to-cell signaling involving N-acyl-homoserine lactone compounds termed autoinducers (AIs) is instrumental to virulence factor production and biofilm development by Pseudomonas aeruginosa. In order to determine the importance of cell-to-cell signaling during the colonization of mechanically ventilated patients, we collected 442 P. aeruginosa pulmonary isolates from 13 patients. Phenotypic characterization showed that 81% of these isolates produced the AI-dependent virulence factors elastase, protease, and rhamnolipids. We identified nine genotypically distinct P. aeruginosa strains. Six of

Identification and functional analysis of sRNAs in Pseudomonas aeruginosa quorum sensing and virulence regulation.

Small RNAs (sRNAs) act as critical regulators of post-transcriptional activity and diverse cellular processes in bacteria. However, the entire repertoire of sRNAs and their functions has not yet been extensively studied. This study employs a novel combined computational-experimental prediction approach to identify and characterize small RNAs (sRNAs) in Pseudomonas aeruginosa PAO1. We defined the first pan-sRNome, including 1663 sRNAs across 484 P. aeruginosa strains, and classified our 9 newly discovered sRNAs as 3 core and 6 accessory elements within the regulatory network. Functional studies

Exploring the anti-quorum sensing potential of synthetic lipstick dyes against Pseudomonas aeruginosa.

Lip care products such as balms, glosses, and lipsticks are widely used to enhance lip health, appearance, and protection. Lipsticks provide aesthetic appeal, social and psychological benefits, that act as a protective barrier against environmental stressors. However, growing concerns have emerged regarding chronic lip infections associated with multidrug-resistant bacteria, particularly Pseudomonas aeruginosa. This is especially concerning in individuals with conditions such as juvenile bullous pemphigoid, hypochromic microcytic anemia, and sickle cell trait. The persistence of P. aeruginosa

The effect of quaternary ammonium compounds (QACs) on quorum sensing and resistance of P. aeruginosa in clinical settings.

Pseudomonas aeruginosa, a formidable opportunistic pathogen, is notorious for its ability to form biofilms and produce virulence factors that favor chronic infections, especially in cystic fibrosis patients. The misuse of disinfectants, combined with environmental leakage and biodegradation, has led to widespread exposure of microorganisms to sub-lethal concentrations of disinfectants, particularly quaternary ammonium compounds (QACs). This study investigates the interaction between QACs, specifically ethylbenzalkyl dimethyl ammonium chloride (EBAC), and the quorum sensing (QS) mechanisms gove

Fitness trade-off and the discovery of a novel missense mutation in the PmrB sensor kinase of a colistin-resistant Pseudomonas aeruginosa strain developed by adaptive laboratory evolution.

Pseudomonas aeruginosa is a prominent bacterial pathogen that causes several nosocomial infections and is notorious for its environmental resilience and rapid development of resistance to frontline antibiotics. A major cause of mortality and morbidity among cystic fibrosis patients, multidrug-resistant P. aeruginosa is often targeted with the antibiotic colistin as a last option. However, increasing reports of colistin resistance among P. aeruginosa is a significant concern. Though the molecular mechanisms responsible for the development of colistin resistance are well known, the evolutionary

Isoliquiritigenin targets las, rhl, and Pqs quorum sensing systems to mitigate the virulence and infection of Pseudomonas aeruginosa.

Pseudomonas aeruginosa is an opportunistic pathogen with high antibiotic resistance, often causing hard-to-treat infections. Its quorum sensing (QS) system regulates virulence and drug resistance. Targeting quorum sensing via QS inhibitors is a promising strategy to combat such infections. In this study, isoliquiritigenin, a natural chalcone compound, could inhibit QS-related gene expression (lasR, lasI, rhlR, rhlI, pqsA, pqsR, lasA, rhlA, phzA1) and reduced virulence factor production of Pseudomonas aeruginosa PAO1, including protease, elastase, pyocyanin, rhamnolipid, biofilm formation and m

Unveiling the modulation of Pseudomonas aeruginosa virulence and biofilm formation by selective histone deacetylase 6 inhibitors.

Bacterial infections represent a key public health issue due to the occurrence of multidrug-resistant bacteria. Recently, the amount of data supporting the dynamic control of epigenetic pathways by environmental cues has triggered research efforts toward the clarification of their role in microbial infections. Among protein post-translational modifications, reversible acetylation is the most implicated in the feedback to environmental stimuli and in cellular homeostasis. Accordingly, the latest studies identified the histone deacetylase 6 (HDAC6) enzyme as a crucial player in the complex molec

Edible Medicinal Guava Fruit (Psidium guajava L.) Are a Source of Anti-Biofilm Compounds against Pseudomonas aeruginosa.

Psidium guajava is one of the most common edible medicinal plants frequently used in Malagasy traditional medicine to treat gastrointestinal infections. In order to evaluate their probable antibacterial activities, three organic extracts (successive extractions by hexane, dichloromethane, and ethanol) of ripe guava fruits were assessed for their bactericidal and anti-virulence properties against P. aeruginosa PAO1. Although these three extracts have shown no direct antibacterial activity (MIC of 1000 µg/mL) and, at the non-bactericidal concentration of 100 µg/mL, no impact on the production of

Plant Beneficial Features and Application of Paraburkholderia sp. NhPBG1 Isolated from Pitcher of Nepenthes hamblack.

Pitchers are the unique structures of carnivorous plants used for the trapping of insects and other small invertebrates. The digestion of captured prey here is assisted by the bacteria, which have been associated with pitchers. These bacterial communities can therefore expect to have a variety of plant beneficial functions. In this study, the bacterial isolate NhPBG1 from the pitcher of Nepenthes hamblack was screened for activity against Pythium aphanidermatum, Rhizoctonia solani, Fusarium oxysporum, and Colletotrichum accutatum and was found to have the inhibitory activity towards all the te

Efficient modification of the Pseudomonas aeruginosa toxin 2-heptyl-1-hydroxyquinolin-4-one by three Bacillus glycosyltransferases with broad substrate ranges.

Glycosylation of natural and synthetic products can alter the physical, chemical and pharmacological properties of the aglycon. Conversion of 2-heptyl-1-hydroxyquinolin-4-one (HQNO), a potent respiratory inhibitor produced by Pseudomonas aeruginosa, to the less toxic 2-heptyl-1-(β-D-glucopyranosydyl)-quinolin-4-one, was recently demonstrated for Bacillus subtilis strain 168. In this study, we compared the genomes of several Bacillus spp. to identify candidate enzymes for HQNO glucosylation. All three (putative) UDP-glycosyltransferases (GT) of B. subtilis 168 tested, YjiC, YdhE and YojK, were

Pathway-Centric Structure-Based Multi-Target Compound Screening for Anti-Virulence Drug Repurposing.

The emergence of superbugs that are resistant to last-resort antibiotics poses a serious threat to human health, and we are in a "race against time to develop new antibiotics." New approaches are urgently needed to control drug-resistant pathogens, and to reduce the emergence of new drug-resistant microbes. Targeting bacterial virulence has emerged as an important strategy for combating drug-resistant pathogens. It has been shown that pyocyanin, which is produced by the phenazine biosynthesis pathway, plays a key role in the virulence of Pseudomonas aeruginosa infection, making it an attractiv

Curcumin encapsulation in yeast glucan particles promotes its anti-inflammatory potential in vitro.

Glucan particles (GPs) from Saccharomyces cerevisiae are hollow shells that are composed mainly of β-1,3-d-glucan, which has demonstrated immunomodulatory and anti-inflammatory potential both in vitro and in vivo. Curcumin is a natural hydrophobic phenolic compound, which possesses a significant anti-inflammatory effect and is used as supportive therapy in the treatment of many inflammatory diseases. The aim of this study is to evaluate the possible synergic effect and other benefits of the co-application of GPs and curcumin in the form of pharmaceutical composites. GP/curcumin composites were

Candida albicans and Pseudomonas aeruginosa interactions: more than an opportunistic criminal association?

Pseudomonas aeruginosa and Candida albicans are frequently coexisting opportunistic pathogens, responsible for colonization and infection in predisposed patients. They share a virulence specificity relying on auto-inducing, cell density-dependent molecules named quorum-sensing (QS). C. albicans virulence depends on its QS that influences morphological switch from yeast to filamentous form. Similarly, the production of P. aeruginosa virulence factors depends partly on QS molecules. Interactions have been investigated and demonstrated in vitro. P. aeruginosa may kill C. albicans either by produc

Reduced virulence of a hfq mutant of Pseudomonas aeruginosa O1.

The Sm-like protein Hfq has been implicated in the regulation of sigmaS-dependent and sigmaS-independent genes in E. coli and in the regulation of virulence factors in both, Yersinia enterocolitica and Brucella abortus. Here, we have studied the effect of Hfq on virulence and stress response of Pseudomonas aeruginosa (PAO1). We have constructed a PAO1hfq- mutant and a PAO1hfq-rpoS- double mutant to permit distinction between direct and indirect effects of Hfq. When compared to the wild-type and the rpoS- strains, the hfq knock out strain showed a reduced growth rate and was unable to utilize g

Quorum quenching by endophytic Bacillus cereus AL1: a lactonase-based anti-virulence strategy against Pseudomonas aeruginosa.

BACKGROUND: Pseudomonas aeruginosa infections are often challenging to treat due to multiple drug resistance, besides the development of biofilms and a plethora of virulence factors regulated by quorum sensing. Quorum-quenching enzymes, such as N-acyl homoserine lactonases, represent a promising anti-virulence strategy by disrupting this signaling mechanism without exerting selective pressure, leading to resistance. This study aimed to screen endophyte and epiphyte isolates for lactonase activity and evaluate their potential to inhibit virulence in Pseudomonas aeruginosa. RESULTS: Fifty-two b

PA4933 Mediates Dual Regulation of Oxidative Resistance and Virulence in Pseudomonas aeruginosa Pulmonary Infection.

Pseudomonas aeruginosa is a critical opportunistic pathogen requiring rigorous prevention and control, which triggers severe infections in immunocompromised populations. Bacterial membrane proteins serve as core regulatory molecules coordinating stress tolerance and virulence homeostasis during host colonization. Nevertheless, the physiological functions of numerous structurally homologous yet unannotated membrane proteins remain largely elusive. In this study, we identified a significantly upregulated hypothetical gene, PA4933, from published transcriptomic datasets of P. aeruginosa PA14 infe

Synthesis of novel quinazolinone-based heterocyclic acetamides as P. aeruginosa PqsR inhibitors.

Antimicrobial resistance (AMR) has become a major global concern for public health and economics and there is an urgent need to develop novel classes of antimicrobial drugs. Quorum Sensing (QS) has emerged as a promising target to counter AMR in P. aeruginosa as it controls the virulence factors and biofilm formation, causing infections in host and resistance against antibiotics. Pseudomonas quinolone signal (PQS) is one of the major signalling systems that includes PQS and HHQ as autoinducers which binds to transcription regulator PqsR (MvfR). Targeting PqsR to inhibit QS signalling could be

Influence of nonpolar aliphatic amino acids on Pseudomonas aeruginosa for virulence factors production.

Multidrug-resistant (MDR) microorganisms are widely recognized as a public health concern due to the antibiotic resistance, which is a major risk to global human health. Amino acids have been utilized to produce antimicrobial peptides as they are extremely effective and less likely to build microbial resistance. This study evaluates the efficacy of six aliphatic amino acids, isoleucine, methionine, valine, glycine, alanine, and proline against Pseudomonas aeruginosa. It is a Gram-negative MDR pathogen that utilizes N-acyl-homoserine lactones as signaling molecules to regulate the quorum sensin
