Random forest-integrated analysis in AD and LATE brain transcriptome-wide data to identify disease-specific gene expression.

Alzheimer's disease (AD) is a complex neurodegenerative disorder that affects thinking, memory, and behavior. Limbic-predominant age-related TDP-43 encephalopathy (LATE) is a recently identified common neurodegenerative disease that mimics the clinical symptoms of AD. The development of drugs to prevent or treat these neurodegenerative diseases has been slow, partly because the genes associated with these diseases are incompletely understood. A notable hindrance from data analysis perspective is that, usually, the clinical samples for patients and controls are highly imbalanced, thus rendering

In silico modelling of permeation enhancement potency in Caco-2 monolayers based on molecular descriptors and random forest.

Structural traits of permeation enhancers are important determinants of their capacity to promote enhanced drug absorption. Therefore, in order to obtain a better understanding of structure-activity relationships for permeation enhancers, a Quantitative Structural Activity Relationship (QSAR) model has been developed. The random forest-QSAR model was based upon Caco-2 data for 41 surfactant-like permeation enhancers from Whitehead et al. (2008) and molecular descriptors calculated from their structure. The QSAR model was validated by two test-sets: (i) an eleven compound experimental set with

Published genetic variants in retinopathy of prematurity: random forest analysis suggests a negligible contribution to risk and severity.

PURPOSE: Our recent investigations suggested association between severe retinopathy of prematurity (ROP) and some genetic polymorphisms contributing to angiogenesis. While these findings may help to identify specific elements in ROP pathogenesis, the predictive value of these genetic variants at birth is unknown. We applied a high-dimensional nonparametric method called random forest technique (RFT) to evaluate the predictive value of genetic polymorphisms in ROP at birth. METHODS: We used published genetic (i.e., VEGF T(-460)C, G(+ 405)C, and C(-2578)A; IGF-I receptor G(+ 3174)A, angiopoieti

Predicting Pregnancy After Fixed-Time Artificial Insemination in Goats: Comparative Performance of Multivariable Logistic Regression and Random Forest Models.

Accurate prediction of pregnancy following fixed-time artificial insemination (FTAI) remains challenging under commercial goat production conditions. This study compared multivariable logistic regression and Random Forest models for pregnancy prediction and used SHAP analysis to characterize predictor contributions. Data from 920 goats subjected to FTAI across four regions of Peru were analyzed using reproductive, hormonal, animal-level, and management variables. Logistic regression was developed after univariable screening, whereas Random Forest models used 500 trees with mtry values of 2, 3,

Personalized Hearing Loss Care Using SNOMED CT-Aligned Ontology and Random Forest Machine Learning: A Hybrid Decision-Support Framework.

BACKGROUND: Hearing loss affects over 466 million individuals globally and is recognized as a major risk factor for Alzheimer's disease, yet treatment personalization remains limited due to the complexity and diversity of underlying causes. Current diagnostic and therapeutic approaches lack standardized methods to accurately predict the most appropriate intervention for individual patients. The integration of medical ontologies with machine learning offers a promising solution for enhancing diagnostic accuracy and treatment personalization. AIM: Our study aimed to (i) develop a Systematized N

Hierarchical random forest model, inflammation and oxidative stress as predictors of the atherogenic index of plasma and diabetes progression.

Type 2 diabetes mellitus (T2DM) is a chronic metabolic disease that increases the risk of cardiovascular complications. The atherogenic index of plasma (AIP) is a risk marker for T2DM and cardiovascular disease on the basis of lipid profiles. T2DM and CVD risk are also associated with nonlipid biomarkers, including oxidative stress, inflammation, and mitochondrial dysfunction, and are linked to diabetes progression. This study applies hierarchical random forest (HRF) machine learning to identify stage-specific predictors of AIP in normoglycemic, prediabetic, and diabetic individuals. Participa

Identifying environmental factors related to motorcyclist crash rates: variable selection using spatial Random Forest with network distance and barriers.

Motorcycle crashes remain a major global safety concern, particularly in many Asian countries where motorcycles are the primary mode of transportation. While previous studies have identified factors associated with motorcycle crashes using traditional variable selection methods, they often overlook spatial patterns and contextual constraints (such as motorcycles being prohibited from freeways), leading to lower model accuracy. To address this limitation, we introduce a novel variable selection framework, designated as Spatial Random Forest with Network Distance and Barriers (spatialRF-NDBAR),

Multi-modal predictive modeling of schizophrenia severity: Leveraging liver function indicators and cognitive scores with random forest and SVM.

Schizophrenia is a complex neuropsychiatric disorder with cognitive deficits and systemic physiological disturbances, including emerging links to hepatic dysfunction via the gut-liver-brain axis. Despite growing evidence, the integration of liver function biomarkers into predictive models for schizophrenia severity remains largely unexplored. This study proposes a multimodal machine learning framework combining hepatic indicators-Alanine Aminotransferase (ALT), Aspartate Aminotransferase (AST), Bilirubin, Albumin, and International Normalized Ratio (INR)-with cognitive assessment scores to enh

Applying a Random Forest Approach in Predicting Health Status in Patients with Carotid Artery Stenosis 30 Days Post Stenting.

BACKGROUND: Approximately 20% of ischemic strokes in the United States result from carotid artery stenosis. Carotid artery stenting (CAS) can reduce stroke risk, but variability in poststenting health outcomes and their predictors is poorly understood. We examined the 30-day post-CAS health status and derived its most important predictors. METHODS: The Stenting and Angioplasty with Protection of Patients with High Risk for Endarterectomy (SAPPHIRE) Worldwide Registry measured preprocedural and 30-day health status for patients undergoing transfemoral CAS using the 36-item Short Form Survey Me

Finding the needle in the haystack of isokinetic knee data: Random Forest modelling improves information about ACLR-related deficiencies.

The difficulties of rehabilitation after anterior cruciate ligament (ACL) injuries, subsequent return-to-sport (RTS) let alone achieving pre-injury performance, are well known. Isokinetic testing is often used to assess strength capacities during that process. The aim of the present machine learning (ML) approach was to examine which isokinetic data differentiates athletes post ACL reconstruction (ACLR) and healthy controls. Two Random Forest models were trained from data of unilateral concentric and eccentric knee flexor and extensor tests (30°/s, 150°/s) of 366 male (63 post ACLR) as well as

Diagnostic Performance of MRI Volumetry in Epilepsy Patients With Hippocampal Sclerosis Supported Through a Random Forest Automatic Classification Algorithm.

Introduction: Several methods offer free volumetry services for MR data that adequately quantify volume differences in the hippocampus and its subregions. These methods are frequently used to assist in clinical diagnosis of suspected hippocampal sclerosis in temporal lobe epilepsy. A strong association between severity of histopathological anomalies and hippocampal volumes was reported using MR volumetry with a higher diagnostic yield than visual examination alone. Interpretation of volumetry results is challenging due to inherent methodological differences and to the reported variability of h

Analysis of related factors of radiation pneumonia caused by precise radiotherapy of esophageal cancer based on random forest algorithm.

The precise radiotherapy of esophageal cancer may cause different degrees of radiation damage for lung tissues and cause radioactive pneumonia. However, the occurrence of radioactive pneumonia is related to many factors. To further clarify the correlation between the occurrence of radioactive pneumonia and related factors, a random forest model was used to build a risk prediction model for patients with esophageal cancer undergoing radiotherapy. In this study, we retrospectively reviewed 118 patients with esophageal cancer confirmed by pathology in our hospital. The health characteristics and

A random forest model based on core genome allelic profiles of MRSA for penicillin plus potassium clavulanate susceptibility prediction.

Treatment failure of methicillin-resistant Staphylococcus aureus (MRSA) infections remains problematic in clinical practice because therapeutic options are limited. Penicillin plus potassium clavulanate combination (PENC) was shown to have potential for treating some MRSA infections. We investigated the susceptibility of MRSA isolates and constructed a drug susceptibility prediction model for the phenotype of the PENC. We determined the minimum inhibitory concentration of PENC for MRSA (n=284) in a teaching hospital (SRRSH-MRSA). PENC susceptibility genotypes were analysed using a published ge
