Fast-conducting mechanonociceptors uniquely engage reflexive and affective pain circuitry to drive protective responses.

Nociceptors detect damaging stimuli and evoke pain in healthy animals. We conducted an optogenetic activation screen to identify genetically defined nociceptor populations that elicit place aversion and nocifensive behaviors in response to stimulation. Smr2 Cre - and Bmpr1b Cre -labeled Aδ high-threshold mechanoreceptors (HTMRs) emerged as two of the few nociceptor populations, and we focused on investigating their physiological, morphological, functional, and synaptic properties. These neurons densely innervate skin and other organs, are activated only by intense, potentially damaging stimuli

[Clinico-electroneuromyographic characteristics of myelodysplasia in children].

Myelodysplasia is a topical problem of childhood neurology. The early diagnosis and treatment of myelodysplasia prevents the development of severe neurological symptomatology. Twenty patients with myelodysplasia were investigated using clinical, electroneuromyographic, and thermographic methods of examination. Electroneuromyography included determination of the bulbocavernous reflex which served as an objective criterion of the status of the lumbosacral portion of the spinal cord. Early diagnosis of myelodysplasia makes it possible to conduct preventive therapy including electrostimulation of

Vagal sensory neurones directly synapse onto parasympathetic motor (DMV) neurones.

Autonomic reflexes, controlling unconscious processes such as heart rate and digestion, are essential for homeostasis. Vago-vagal reflexes describe one category of autonomic reflexes in which the sensory input and motor output are both mediated through the vagus nerve. These reflexes are widely considered to rely on the nucleus of the solitary tract (NTS) for the integration of sensory information before regulating the motor output from the dorsal motor nucleus of the vagus (DMV). Here, we electrically activated the solitary tract to evoke sensory input simultaneous to whole cell recordings of

Transcutaneous spinal direct current stimulation increases corticospinal transmission and enhances voluntary motor output in humans.

Optimization of motor performance is of importance in daily life, in relation to recovery following injury as well as for elite sports performance. The present study investigated whether transcutaneous spinal direct current stimulation (tsDCS) may enhance voluntary ballistic activation of ankle muscles and descending activation of spinal motor neurons in able-bodied adults. Forty-one adults (21 men; 24.0 ± 3.2 years) participated in the study. The effect of tsDCS on ballistic motor performance and plantar flexor muscle activation was assessed in a double-blinded sham-controlled cross-over expe

The H reflex recovery curve reinvestigated: low-intensity conditioning stimulation and nerve compression disclose differential effects of presumed group Ia fibres in man.

The recovery curve of the soleus H reflex, evoked by stimulation of the tibial nerve in the popliteal fossa, was studied by applying an electrical conditioning stimulus to the inferior soleus nerve. Under these conditions a long-latency facilitatory phase could be superimposed on a long-lasting inhibition, the excitability cycle being therefore similar to that obtained by means of a paired shock to the tibial nerve. In order to identify the afferent fibres responsible for the effects observed, various conditioning stimulus strengths and nerve compression were used. A low-intensity stimulus ind

Central trigeminal and posterior eighth nerve projections in the turtle Chrysemys picta studied in vitro.

Recent electrophysiological studies in the turtle Chrysemys picta have suggested that a neural correlate of the eye-blink reflex can be evoked in an in vitro brain-stem-cerebellum preparation by electrical rather than natural stimulation of the cranial nerves. Discharge recorded in the abducens nerve, which is similar to EMG recordings from extraocular muscles during eye retraction, is triggered by a brief electrical stimulus applied to the ipsilateral trigeminal nerve. Evidence also suggests that pairing a one-second electrical stimulus applied to the posterior eighth nerve immediately prior

Electrophysiological studies in alcoholism.

Using a range of electrophysiological techniques, it has been possible to demonstrate impaired function in smaller calibre motor fibres and in distal large cutaneous sensory nerve fibres in both alcoholic patients without neuropathy and in those alcoholics with clinical manifestations of peripheral nerve disease. Evidence of more proximal involvement of Ia sensory fibres was obtained, but in the majority of our patients, large motor fibres functioned normally. The nature of the underlying pathological process is discussed.

Facilitation of transmission in heteronymous group II pathways in spastic hemiplegic patients.

OBJECTIVE: A potent heteronymous group II excitation of quadriceps motor neurons has been recently demonstrated in normal subjects. The present study was undertaken to investigate whether this heteronymous group II excitation also contributes to spasticity in hemiplegic patients. METHOD: The early and late facilitations of the quadriceps H reflex elicited by a conditioning volley to the common peroneal nerve at three times motor threshold, attributed to non-monosynaptic group I and group II excitations respectively, were investigated. The comparison was drawn between results obtained in 20 pa

Conduction velocity along human nociceptive reflex afferent nerve fibres.

The conduction velocity along the nociceptive flexor reflex afferent nerve fibres was investigated in human subjects. The posterior tibal nerve was stimulated at two sites by single painful electrical shocks of 1.0 ms duration and with adequate intensity and the reflex EMG discharges were recorded from the short head of the biceps femoris muscle. The fastest reflex conduction velocity along the posterior tibial nerve between the ankle and the popliteal fossa was about 10-25 m/s. Thus the fastest flexor reflex afferent fibres associated with a painful sensation were thought to be included in th

Differential motor and electrophysiological outcome in rats with mid-thoracic or high lumbar incomplete spinal cord injuries.

We have investigated the motor changes in rats subjected to a moderate photochemical injury on mid-thoracic (T8) or high lumbar (L2) spinal cord segments. Fourteen days after surgery, L2 injured animals presented gross locomotor deficits (scored 10+/-2.8 in the BBB scale), decreased amplitude of motor-evoked potentials (MEPs) recorded on tibialis anterior (TA) and plantar (PL) muscles (24% and 6% of the preoperative mean values, respectively), reduced M wave amplitudes (75%, 62%), and also facilitated monosynaptic reflexes evidenced by an increase of the H/M amplitude ratio (158% and 563%). Immediate electrical stimulation enhances regeneration and reinnervation and modulates spinal plastic changes after sciatic nerve injury and repair.

We have studied whether electrical stimulation immediately after nerve injury may enhance axonal regeneration and modulate plastic changes at the spinal cord level underlying the appearance of hyperreflexia. Two groups of adult rats were subjected to sciatic nerve section followed by suture repair. One group (ES) received electrical stimulation (3 V, 0.1 ms at 20 Hz) for 1 h after injury. A second group served as control (C). Nerve conduction, H reflex, motor evoked potentials, and algesimetry tests were performed at 1, 3, 5, 7 and 9 weeks after surgery, to assess muscle reinnervation and chan

Development of twitching in sleeping infant mice depends on sensory experience.

Myoclonic twitches are jerky movements that occur exclusively and abundantly during active (or REM) sleep in mammals, especially in early development [1-4]. In rat pups, limb twitches exhibit a complex spatiotemporal structure that changes across early development [5]. However, it is not known whether this developmental change is influenced by sensory experience, which is a prerequisite to the notion that sensory feedback from twitches not only activates sensorimotor circuits but modifies them [4]. Here, we investigated the contributions of proprioception to twitching in newborn ErbB2 conditio

Reciprocal inhibition between forearm muscles in patients with writer's cramp and other occupational cramps, symptomatic hemidystonia and hemiparesis due to stroke.

Reciprocal inhibition of H reflexes in the forearm flexor muscles was examined in a group of 16 patients with writer's and other occupational cramps. The early disynaptic phase of reciprocal inhibition was normal. However, there was a reduction in the amount of later, presynaptic inhibition, when compared with age-matched normal subjects. Similar findings were seen in 2 patients with symptomatic hemidystonia in whom structural brain lesions were present. However, this reduction in presynaptic inhibition was not specific to patients with dystonia. In a further group of 13 patients with hemipare

The late facilitation in H-reflex recovery cycles in different pyramidal lesions.

H-reflex amplitudes were recorded after stimulation of the tibial nerve and different electrical stimuli in 18 normal persons and 26 patients showing pyramidal spasticity (8 spastic spinal paralysis, 6 spastic hemiparesis, 12 spinal lesions). A just subthreshold stimulus of the tibial nerve facilitated the H-reflex in spastic patients slightly after about 300 ms (up to 113%), following an early strong facilitation (10 ms) and a longer lasting depression (20-200ms). Similar postinhibitory facilitation was obtained in spastic patients after ipsilateral stimulation of the plantar surface and afte

[Functional state of spinal centers in patients with cerebral circulatory disorders according to the results of stimulation electromyography].

In 53 patients in different periods of brain strokes the authors studied the functional state of spinal centers by means of registration of H-reflex and F-wave of the skeleton muscles. In spite of the stroke character a marked increase of excitation of the spinal centers was observed, it being especially significant in the subacute period of the disease. The increase of H-reflex amplitude was maximal in extremities, contralateral to the focus of the lesion, but it was observed on the lesion side as well. The patients with cerebral coma in the acute stage of the stroke demonstrated a descrease

The effects of naloxone, morphine and methionine enkephalinamide on Ia afferent terminations in the cat spinal cord.

Naloxone, morphine, Met5-enkephalinamide (MENKA) and procaine were administered microelectrophoretically near extracellularly stimulated extensor muscle group Ia afferent fibres and terminations in the lumbar spinal cord of cats anaesthetized with pentobarbitone sodium. Observations were made of effects on the electrical threshold, on the depolarizing action of GABA or piperidine-4-sulphonate (P4S), and on bicuculline-sensitive primary afferent depolarization (PAD) generated by tetanic stimulation of flexor muscle low threshold afferents. All 4 agents reversibly elevated the threshold of Ia fi
