Relative effects of systemic pH, PCO2, and bicarbonate concentration on ileal ion transport.

To determine the relative effects of systemic pH, CO2 tension (PCO2), and bicarbonate concentration on ileal electrolyte transport, states of acute metabolic acidosis and alkalosis were created in Sprague-Dawley rats by gavage feeding (NH4)2SO4 and NaHCO3, respectively. During in situ perfusion of the ileum in anesthetized animals, electrolyte transport was measured before and after respiratory compensation of the systemic pH. Acute respiratory acidosis and alkalosis also were studied by ventilating animals with 0, 3, or 8% CO2. When animals in all groups were considered, net sodium absorption

Evidence of active regulation of cerebrospinal fluid acid-base balance.

To test the passive transport hypothesis of cerebrospinal fluid (CSF) [H+] regulation, we altered the relationship between plasma [H+] and the electrical potential difference between CSF and blood (PD) by elevating plasma [K+] during 6-h systemic acid-base disturbances. In five groups of pentobarbital-anesthetized dogs, we increased plasma [K+] from 3.5 to an average of 7.8 meq/l. Hyperkalemia produced an increase in the PD of 6.3 mV by 6 h with normal plasma acid-base status (pHa 7.4), of 8.3 mV with isocapnic metabolic acidosis (pHa 7.2), of 5.3 mV with isocapnic metabolic alkalosis (pHa 7.6

Infant With Acute Bilateral Pneumothorax During Air Transport.

In rural settings, medically complicated patients may require air transport to facilities that are capable of providing higher levels of care. Extra consideration must be given to pulmonary pathologies when considering this mode of transport. Altitude changes impact both air pressure and volume as described by Boyle's law. These changes can complicate the care of these patients in several ways. We present a case of patient with respiratory failure secondary to viral infection who developed acute bilateral pneumothorax (PTX) while mechanically ventilated during a transport on a fixed-wing aircr

Factors Associated with Transport Instability and Outcome Deterioration During Interhospital Intensive Care Transfers of Patients with COVID-19 in Germany.

Background/Objectives: The COVID-19 pandemic created demand for interhospital transfers of critically ill patients to balance intensive care capacity. Evidence on the logistics and immediate physiological outcomes of such transfers in Germany remains limited. Methods: This multicenter retrospective observational cohort study included 414 interhospital intensive care transport records of adults with COVID-19 in Germany between January 2020 and June 2021. Logistic regression assessed factors associated with transport instability and deterioration, and linear regression assessed factors associate

The effect of pesticides on the mtDNA integrity and bioenergetic properties of potato mitochondria.

Potato (Solanum tuberosum L.) is one of the most common crops in the world, and it is very susceptible to a wide range of pests such as insects and fungi. The use of pesticides often results in the suppression of seed germination and plant growth, in particular, due to their effect on the respiratory chain of mitochondria. There are numerous studies of the effect of pesticides on animal mitochondria, but their interference with the electron transport in plant mitochondria is not well documented. We present the data showing that a number of pesticides inhibit electron flow, and other pesticides

Strategies for measuring airway mucus and mucins.

Mucus secretion and mucociliary transport are essential defense mechanisms of the airways. Deviations in mucus composition and secretion can impede mucociliary transport and elicit airway obstruction. As such, mucus abnormalities are hallmark features of many respiratory diseases, including asthma, cystic fibrosis and chronic obstructive pulmonary disease (COPD). Studying mucus composition and its physical properties has therefore been of significant interest both clinically and scientifically. Yet, measuring mucus production, output, composition and transport presents several challenges. Here

Respiratory toxicity of buprenorphine results from the blockage of P-glycoprotein-mediated efflux of norbuprenorphine at the blood-brain barrier in mice.

OBJECTIVES: Deaths due to asphyxia as well as following acute poisoning with severe respiratory depression have been attributed to buprenorphine in opioid abusers. However, in human and animal studies, buprenorphine exhibited ceiling respiratory effects, whereas its metabolite, norbuprenorphine, was assessed as being a potent respiratory depressor in rodents. Recently, norbuprenorphine, in contrast to buprenorphine, was shown in vitro to be a substrate of human P-glycoprotein, a drug-transporter involved in all steps of pharmacokinetics including transport at the blood-brain barrier. Our objec

Substrates respired by mitochondrial fractions of two isolates of the nematode Aphelenchus avenae and the effects of electron transport inhibitors.

Electron transport has been assayed and compared in two isolates (M and F) of the free-living (model) nematode Aphelenchus avenae. Of the substrates tested only alpha-glycerophosphate and succinate were utilised to any significant extent by both isolates. Comparative data on respiratory rates, respiratory control ratios and ADP:O ratios for various substrates are given. Succinate oxidation by isolate-F mitochondria was ca 80-90% sensitive to antimycin A while that of isolate M was almost completely refractory to antimycin A. The response to other electron transport inhibitors suggests the oper

Increased activity and sensitivity of mitochondrial respiratory enzymes to tumor necrosis factor alpha-mediated inhibition is associated with increased cytotoxicity in drug-resistant leukemic cell lines.

The drug-resistant leukemic cell lines, CEM/VLB100 and K/DAU600, are more sensitive to tumor necrosis factor alpha (TNFalpha)-mediated cytotoxicity compared with their parental cell lines, CCRF-CEM and K562 cl.6. Drug-resistant leukemic cell lines have more active mitochondrial function, which is associated with a greater susceptibility to TNFalpha-induced respiratory inhibition. TNFalpha blocked electron transfer at three sites, NADH dehydrogenase (complex I), succinate dehydrogenase (complex II), and cytochrome c oxidase (complex IV). Respiratory rate and electron transport chain enzyme acti

SLC26A9 in airways and intestine: secretion or absorption?

SLC26A9 is one out of 11 proteins that belong to the SLC26A family of anion transporters. Apart from expression in the gastrointestinal tract, SLC26A9 is also found in the respiratory system, in male tissues and in the skin. SLC26A9 has gained attention because of its modifier role in the gastrointestinal manifestation of cystic fibrosis (CF). SLC26A9 appears to have an impact on the extent of intestinal obstruction caused by meconium ileus. SLC26A9 supports duodenal bicarbonate secretion, but was assumed to provide a basal Cl- secretory pathway in airways. However, recent results show that ba

Captopril versus hydralazine in primary pulmonary hypertension.

Captopril was tested as the treatment for a patient with primary pulmonary hypertension (PPH) and its effects were compared with those of hydralazine. Captopril induced a rise in pulmonary pressures and in intrapulmonary shunt; hydralazine lowered pulmonary resistances and increased paO2 and blood O2 transport. Prospective studies in PPH treated with captopril are recommended and evaluation of all drugs not only by hemodynamic but also respiratory and O2 transport measurements.

Skin-to-skin stabilisation and uninterrupted respiratory support for preterm infants after birth: feasibility of a new and simplified rPAP system.

BACKGROUND: The rPAP respiratory support system, used for delivery room stabilisation with nasal prongs, has been shown to reduce the need for intubation in extremely preterm infants. A simplified version of the system has been developed. The purpose of this study was to determine the feasibility of providing uninterrupted respiratory support with the simplified rPAP from birth up to 4 hours of life and to assess ease of use for skin-to skin stabilisation. METHODS: This was a non-randomised feasibility study conducted at Karolinska University Hospital, Sweden. Respiratory support with continu

Graduate Student Literature Review: Reducing mortality and morbidity in transported preweaning dairy calves: Colostrum management and pretransport nonsteroidal anti-inflammatory drug administration.

Currently, mortality and morbidity rates for preweaning calves in the US dairy industry are high, with the major cause being digestive disorders and respiratory diseases. One of the most important management practices that can reduce calf mortalities and morbidities is the feeding of colostrum, provided its quantity, quality, and cleanliness, and timing of feeding are according to recommendations. However, other management practices similar to transportation, can also compromise calf health and production performance. When preweaning calves are transported, stressors similar to physical restra

Breakthroughs in the development of antibiotics, antifungals and antiparasitics targeting the pathogens' respiratory chain.

The aerobic respiratory chain is vital to bacterial and eukaryotic cell energy transformation. Embedded in the mitochondrial inner membrane and the bacterial plasma membrane, the respiratory chain couples sequential redox reactions with ion pumping, thereby generating the motive force that is used to drive ATP synthesis. Due to the essential role of oxidative phosphorylation in cellular life, the electron transport chain proteins, their cofactors, and ATP synthase components serve as a target for antibacterial, antifungal, and antiparasitic drugs. Whether by (1) inhibition of electron flow thr

Interhospital transportation of a COVID-19 patient undergoing veno-venous extracorporeal membrane oxygenation by helicopter.

Some coronavirus disease 2019 (COVID-19) patients develop rapidly progressive acute respiratory distress syndrome and require veno-venous extracorporeal membrane oxygenation (V-V ECMO). A previous study recommended the transfer of ECMO patients to ECMO centers. However, because of the pandemic, a limited number of ECMO centers are available for patient transfer. The safe long-distance interhospital transport of these patients is a concern. To minimize transportation time, helicopter use is a suitable choice. We report the first case of a COVID-19 patient on V-V ECMO, transferred to our ECMO ce

Air transport of patients with severe lung injury: development and utilization of the Acute Lung Rescue Team.

BACKGROUND: Critical Care Air Transport Teams (CCATTs) are an integral component of modern casualty care, allowing early transport of critically ill and injured patients. Aeromedical evacuation of patients with significant pulmonary impairment is sometimes beyond the scope of CCATT because of limitations of the transport ventilator and potential for further respiratory deterioration in flight. The Acute Lung Rescue Team (ALRT) was developed to facilitate transport of these patients out of the combat theater. METHODS: The United States TRANSCOM Regulation and Command/Control Evacuation System

Respiratory patterns in human brain tumors.

It has been recognized for some time that malignant cells have a diminished respiratory rate coupled with an excessive rate of aerobic glycolysis. Subsequent studies indicated, however, that this pattern was neither unique to malignant tumors nor an essential characteristic of all varieties of cancer. In attempting to synthesize the data on oxygen consumption of human brain tumors, it is difficult to relate activity to malignancy, and the integrity of the mitochondrial electron transport chain has not been systematically examined. In this study, oxygen consumption was measured using a whole ce

SARS-CoV-2 may hijack GPCR signaling pathways to dysregulate lung ion and fluid transport.

The tropism of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), a virus responsible for the ongoing coronavirus disease 2019 (COVID-19) pandemic, toward the host cells is determined, at least in part, by the expression and distribution of its cell surface receptor, angiotensin-converting enzyme 2 (ACE2). The virus further exploits the host cellular machinery to gain access into the cells; its spike protein is cleaved by a host cell surface transmembrane serine protease 2 (TMPRSS2) shortly after binding ACE2, followed by its proteolytic activation at a furin cleavage site. The viru
