Congenital dyserythropoietic anaemia type II (CDA II) is the most common congenital dyserythropoietic anaemia. CDA II is frequently misdiagnosed as Hereditary Spherocytosis (HS) due to the presence of mild chronic haemolytic anaemia with splenomegaly, increased osmotic fragility, and presence of microspherocytes. Accurate diagnosis of CDA II is important to prevent severe iron overload. Erythrocyte and reticulocyte indices were assessed in 10 patients from six families with CDA II, 18 patients from eight families with HS, and 50 normal controls. Characteristic increases in distribution width w

Background and objectives Iron deficiency anemia (IDA) remains a major global health concern, with diagnostic challenges arising from the limitations of traditional biochemical markers. This study aimed to evaluate the diagnostic utility and reliability of reticulocyte hemoglobin (RET-Hb) in identifying IDA and assessing the early functional response, defined as any change in RET-Hb within 48 hours of intravenous iron infusion, as a marker of early marrow iron availability. Specifically, the study sought to assess the percentage change in RET-Hb from baseline following iron infusion. Furthermo

BACKGROUND: Chronic kidney disease (CKD) is a serious, long-term illness that damages kidneys and lowers glomerular filtration rate. CKD often causes anemia. Iron deficiency (ID) is common in these patients and worsens illness symptoms. Modern hematology analysers can measure reticulocyte mean cell hemoglobin (MCHr), which directly measures iron integration into erythrocyte hemoglobin. MCHr can improve iron deficiency detection in CKD patients, who have aberrant iron indicators due to chronic inflammation. This research aims to evaluate the effectiveness of MCHr as a marker for ID in patients

INTRODUCTION: Antibodies targeting the blood-stage of Plasmodium falciparum play a critical role in naturally acquired immunity to malaria by limiting blood-stage parasitemia. One mode of action of antibodies is the direct inhibition of merozoite invasion of erythrocytes through targeting invasion ligands. However, evasion of inhibitory antibodies may be mediated in P. falciparum by switching between various ligand-mediated merozoite invasion pathways. Here, we investigated the potential roles of invasion ligands PfRH1, PfRH2a and PfRH2b in immune evasion through phenotypic variation, and thei

BACKGROUND AND OBJECTIVE: Regular blood donors are at risk of developing Latent Iron Deficiency (LID), most commonly seen in female blood donors. Although female blood donors outnumber males in some populations, latent iron deficiency may limit the ability of young females to donate regularly. Current blood donor selection guidelines only screen for pre-donation hemoglobin levels, potentially missing those with LID, which leads to overt iron deficiency later. The gold standard for diagnosing LID is serum transferrin receptor level (sTfR). However, it may not be practical as a screening tool du

Diagnosis of pyruvate kinase deficiency (PKD) remains challenging in clinical practice. The pyruvate kinase (PK) to hexokinase (HK) activity ratio (PK/HK) was proposed to reduce the confounding effect of reticulocytosis on PK activity measurement. However, decreased PK activity and PK/HK ratios have also been observed in other anemias, raising doubts about their diagnostic value. We assessed the diagnostic accuracy of PK/HK ratio versus PK activity in differentiating PKD from other hereditary anemias. This study included 41 patients with molecularly confirmed PKD and 62 patients with other ane

INTRODUCTION: Iron deficiency anemia (IDA) is still a major global health problem. The prevalence was reported to be highest among children under 5 years, 39.7%, with the primary contributing cause being dietary iron deficiency (ID). Reticulocyte hemoglobin equivalent (Ret-He) assesses the amount of hemoglobin in reticulocytes. Ret-He is an easy, inexpensive, and applicable diagnostic parameter of iron deficiency. This study aims to determine the role of Ret-He as a diagnostic parameter of ID and IDA in infants 1-4 months old.
MATERIAL AND METHODS: The study was conducted prospectively at 10

Anemia, iron deficiency and other hematinic deficiencies are a major cause of perioperative transfusion needs and are associated with increased morbidity and mortality. Anemia can be caused either by decreased production of hemoglobin or red blood cells or by increased consumption and blood loss. Decreased production can involve anything from erythropoietin or vitamin B12 insufficiency to absolute or functional lack of iron. Thus, to achieve the goal of patient blood management, anemia must be addressed by addressing its causes. The traditional parameters to diagnose anemia, despite offering e

BACKGROUND: The integrated evaluation of traditional and innovative red blood cell (RBC) and reticulocyte parameters is a rapid, inexpensive and non-invasive diagnostic tools for differential diagnosis and follow-up of anemia and other pathological conditions needing bone marrow erythropoiesis assessment. Therefore, estimating the biological variation (BV) of these parameters is essential for evaluating the analytical performance of hematological analyzers, and for enabling accurate data interpretation and appropriate clinical management. This study aims to define short- and medium-term BV est

INTRODUCTION: Development of additional parameters for complete blood count has emerged in recent hematology analyzers, leading to many publications. However, few studies have been conducted on advanced RBC parameters and hemolytic anemias. We investigated the interest of Sysmex unique parameters, MicroR and HypoHe, as well as the immature fraction of reticulocytes (IRF) in combination with complete blood and reticulocyte count, for screening hereditary spherocytosis (HS) and pyruvate kinase deficiency.
METHODS: We analyzed 182 samples using Sysmex XE-5000 analyzers from a cohort of red cell

The aim of this study was to evaluate sensitivity and specificity for reticulocyte hemoglobin content (CHr) compared to other hematimetric and biochemical iron parameters, in particular, mean corpuscular hemoglobin (MCH), when screening for iron deficiency in elderly anemic patients. Bone marrow staining negative for iron was used as the gold standard criterion for iron deficiency anemia (IDA). Sensitivity and specificity for CHr, soluble transferrin receptor (sTfR), soluble transferrin receptor/log ferritin (TfR-F index), ferritin, MCH, and transferrin saturation were determined. The best cut

Malarial merozoites use an array of ligands, including members of the Reticulocyte Binding Like (RBL) super-family of invasion proteins, to identify and invade erythrocytes. RBL family members are large Type I membrane anchored proteins expressed at the invasive end of merozoites that share homology with the Reticulocyte Binding Proteins 1 and 2 (PvRBP1 and 2) of Plasmodium vivax. Plasmodium species vary widely both in the number and sequence of their RBL genes, with the recently completed Plasmodium falciparum genome containing five RBL genes. Of these, three encode proteins shown to be invol

Studies on baboons and preliminary observations in three patients with sickle cell anemia (SS) suggested that high doses of pulse administered recombinant human erythropoietin (rHuEPO) stimulate F-reticulocyte production. We now report on the administration of rHuEPO in a double-blind format to ascertain frequency of response and potential precipitation of side effects. Ten patients were enrolled, but one was discontinued due to the indication of a blood transfusion. Of the other nine, five received rHuEPO in escalating doses (from 400 to 1,500 U per kg twice daily [BID] per week), alternating

Inflammation suppresses erythropoiesis, leading to non-regenerative anemia in dogs. Currently, clinical evidence of this inhibitory effect remains limited. This study provides important clinical insight by demonstrating a negative correlation between C-reactive protein (CRP) concentration and reticulocyte count over time in dogs with inflammatory anemia. This study aimed to leverage inflammatory anemia as a model to examine the longitudinal relationship between CRP levels and reticulocyte counts in dogs. Medical records of 23 dogs diagnosed with various inflammatory conditions and persistently

Plasmodium falciparum Reticulocyte Binding Protein Homologue (RH5), a leading malaria vaccine candidate, is essential for erythrocyte invasion by the parasite, interacting with the human host receptor, basigin. RH5 has a small number of polymorphisms relative to other blood-stage antigens, and in vitro studies have shown that vaccine-induced antibodies raised against RH5 are strain-transcending, however most studies investigating RH5 diversity have been done in Africa. Understanding the genetic diversity and evolution of malaria antigens in other regions is important for their validation as va
