The vectorial capacity of Rhipicephalus sanguineus in the transmission of canine visceral leishmaniasis has been evaluated through a laboratory-controlled experiment. One healthy Leishmania-free dog and two dogs naturally infected with Leishmania were infested with R. sanguineus in various stages of development. Engorged larvae, unfed nymphs, engorged nymphs, unfed adults, engorged female adults and fed male adults were collected from the experimental animals and examined for Leishmania infection by optical microscopy, polymerase chain reaction (PCR) and parasite culture. Leishmania forms were

Rabbits developed resistance to Rhipicephalus appendiculatus instars following repetitive infestations. Rejection was accompanied by elevated IgM and IgG titres. Extracts of salivary gland, mouth-part cement and whole ticks induced a dose-related lymphocyte transformation. Skin-provocation tests with tick extracts elicited an immediate type-I hypersensitivity reaction with a delayed time-course which was influenced by antihistamines. Passive-transfer studies indicated that resistance was partially transferred with serum. A comparative histological study of the attachment sites of larvae on res

Climate change is altering the environmental conditions that regulate vector persistence, yet attribution remains difficult where long-term surveillance data are limited. Here, we developed a trait-based mechanistic framework to quantify climate-attributable changes in the environmental persistence potential of Rhipicephalus sanguineus s.l. using a host-delay persistence metric, RH,τ. The framework integrates temperature- and humidity-dependent development, mortality, fecundity, and dog-host availability within a factual-counterfactual climate design. Seasonal dynamics were retained by computi

Ehrlichia minasensis is an emerging tick-borne bacterium associated with cattle and Rhipicephalus microplus. It has been reported in South America and, more recently, in Panama, but sequence-confirmed evidence from Costa Rica has not been documented. In this short communication, we screened 19 adult R. microplus ticks collected opportunistically from four cattle at three locations in Costa Rica, together with two available corresponding EDTA blood samples. DNA from ticks and blood was tested by conventional PCR targeting the dsb and groEL genes of Ehrlichia spp. Amplicons compatible with Ehrli

Arthropod vectors are responsible for a multitude of human and animal diseases affecting poor communities in sub-Saharan Africa. Their control still relies on chemical agents, despite growing evidence of insecticide resistance and environmental health concerns. Biorational agents, such as the entomopathogenic fungus Metarhizium anisopliae, might be an alternative for vector control. Recently, the M. anisopliae isolate ICIPE 7 has been developed into a commercial product in Kenya for control of ticks on cattle. We were interested in assessing the potential of controlling not only ticks but also

To minimize the damage caused by synthetic acaricides to non-target organisms, the use of bioactives compounds extracted from plants has been considered. In this study, semi-engorged females of Rhipicephalus linnaei ticks were exposed to Cannabis sativa flower extract (submersion for 5 minutes in different concentrations diluted in ethanol 30 %) evaluated alterations in the integument and salivary glands. The results demonstrated that lower concentrations of the extract caused damage to both the integument and salivary glands, affecting the cuticular epithelial cells responsible for synthesizi

Rhipicephalus microplus is an ixodid tick with a pantropical distribution that represents a serious threat to livestock. West Africa was free of this tick until 2007, when its introduction into Benin was reported. Shortly thereafter, further invasion of this tick species into other West African countries was identified. In this paper, we describe the first detection of R. microplus in Guinea and list the vector-borne haemoparasites that were detected in the invading and indigenous Boophilus species. In 2018, we conducted a small-scale survey of ticks infesting cattle in three administrative re

The tick Rhipicephalus microplus is responsible for the transmission of Anaplasma marginale, which causes hemolytic anemia, abortion, decreased production, and mortality in cattle in Brazil. However, A. marginale can also persist in cattle herds without any clinical signs. This study investigated the relationship between the number of ticks present on each cattle and the circulating number of A. marginale msp1β gene copies in the blood of Brangus and Nellore cattle reared in the Brazilian Cerrado through a year period. Twenty-three animals (11 Brangus and 12 Nellore) were raised for 12 months

Babesia bovis, an intra-erythrocytic tick-borne apicomplexan protozoan, is one of the causative agents of bovine babesiosis. Its life cycle includes sexual reproduction within cattle fever ticks, Rhipicephalus spp. Six B. bovis 6-Cys gene superfamily members were previously identified (A, B, C, D, E, F) where their orthologues in Plasmodium parasite have been shown to encode for proteins required for the development of sexual stages. The current study identified four additional 6-Cys genes (G, H, I, J) in the B. bovis genome. These four genes are described in the context of the complete ten 6-

At present, chemical-based tick control strategies are still the most efficient and widely used methods in control of ticks and tick-borne diseases. In this study, the efficacies of lambda-cyhalothrin, beta-cypermethrin, emamectin benzoate, spirotetramat and hexaflumuron in vitro were evaluated against Hyalomma asiaticum, Haemaphysalis longicornis and Rhipicephalus sanguineus that are widespread and able to transmit a variety of human and animal diseases in China. The results showed that the LC (lethal concentration) 50 of lambda-cyhalothrin, beta-cypermethrin, emamectin benzoate, spirotetrama

Measures to control the cattle tick, Rhipicephalus (Boophilus) microplus, based only on chemical products are becoming unsustainable, mainly because of the development of resistance. The objective of this study was to test the effect of the aqueous extract of pineapple skin (AEPS) and bromelain extracted from the stem (Sigma-Aldrich®, B4882) on engorged females and larvae of R. (B.) microplus in vitro. These substances were diluted in water and evaluated at eight concentrations. Engorged females were collected and distributed in groups of 10, with three repetitions for each treatment. After im

Anti-ectoparasite vaccines offer attractive alternatives to the use of chemical pesticides, especially if they also control the pathogens that ectoparasites transmit. However, selection of suitable antigens is a major constraint on vaccine development. The recombinant tick cement protein, 64TRP, derived from the African brown ear tick, Rhipicephalus appendiculatus, acts as a transmission-blocking vaccine in a mouse model of tick-borne encephalitis virus (TBEV) transmission, protecting immunised mice against lethal challenge with TBEV after exposure to infected ticks. 64TRP acts as a dual actio

The brown dog tick, Rhipicephalus sanguineus plays a significant role in transmitting pathogens to humans and animals, necessitating control measures. Given the drawbacks of synthetic chemicals, plant-based alternatives are a promising option for tick management. The present study aimed to evaluate the acaricidal potential of thyme oil (TO) and nano-formulations {thyme oil nano-emulsion (TNE), silver nanoparticles (AgNPs), and thyme nano-emulsion containing silver nanoparticles (TNE- AgNPs)} against R. sanguineus using an adult immersion bioassay. Rhipicephalus microplus is a critical cattle ectoparasite and a vector of pathogens, including bacteria and protozoan parasites, that cause diseases such as bovine anaplasmosis and babesiosis. To better understand the pathogen profile of this tick in China, we collected 592 tick specimens from seven provinces. Using conventional or nested PCR, we detected a range of pathogens. Six Anaplasma species were identified (prevalence: 3 - 21%), with the highest detection rate in Hunan Province (24%). Spotted fever group rickettsiae, including R. raoultii (21%), Candidatus R. jingxinensis (12%), R. slo

The increasing prevalence of tick-borne arboviral infections worldwide necessitates advanced control strategies, particularly those targeting vectors, to mitigate the disease burden. However, the cellular interactions between arboviruses and ticks, especially for negative-strand RNA viruses, remain largely unexplored. Here, we employ a proteomics informed by transcriptomics approach to elucidate the cellular response of the Rhipicephalus microplus-derived BME/CTVM6 cell line to severe fever with thrombocytopenia syndrome virus (SFTSV) infection. We generate the de novo transcriptomes and prote

BACKGROUND: The first isoxazoline-based acaricide (fluralaner) for the control of Rhipicephalus microplus was introduced onto the market in 2022, initially in Brazil, followed by other Latin American countries. Therefore, it is important to establish laboratory methods to monitor the susceptibility of populations of R. microplus to this molecule and to determine the relationship between the results of laboratory tests and those from field trials.
METHODS: A larval immersion test (LIT) was performed on 18 populations of R. microplus. The lethal concentration 50 (concentration causing 50% morta

BACKGROUND: For the diagnosis of tick sensitivity against different acaricides, there are in vitro and in vivo methods. The main in vivo method, the stable test, is considered a defining methodology. In Uruguay, the Rhipicephalus microplus (R. microplus) strain Mozo is used as the standard susceptible strain by the regulatory authorities. In vitro techniques applied both on adult and larvae stages are validated by FAO and can serve as an orientation diagnosis of the resistance profile developed in field conditions. An alternative was proposed as a modification of the larval immersion test (LIT

BACKGROUND: There has been a long-standing debate over the taxonomic status of Rhipicephalus sanguineus sensu lato. Different studies worldwide have reported the occurrence of different well-defined lineages, in addition to Rhipicephalus sanguineus sensu stricto. To date, there are very few studies examining the diverse aspects of this tick in Colombia. We assessed the population structure and genetic diversity of R. sanguineus s.l. in eight departmental regions across Colombia.
METHODS: A total of 170 ticks were collected from dogs in different departments of Colombia. All specimens were mor

The bovine tick Rhipicephalus (Boophilus) microplus is found in several tropical and subtropical regions of the world. This parasite transmits pathogens that cause disease, such as babesiosis (Babesia bovis and B. bigemina) and anaplasmosis (Anaplasma marginale). Tick infestations cause enormous livestock losses, and controlling tick infestations and the transmission of tick-borne diseases remains a challenge for the livestock industry. Because the currently available commercial vaccines offer only partial protection against R. (B.) microplus, there is a need for more efficient vaccines. Sever

The cosmopolitan species Rhipicephalus sanguineus s.l. is one of the most widely distributed ticks all over the world. These ectoparasites are vectors of several pathogens and cause significant direct damage to their hosts. The biological success of these ectoparasites has been attributed to their ovaries and salivary glands, organs that ensure their survival in various environmental conditions. The importance of the ovaries in ticks is that, after mating, the individuals are able to lay approximately three thousand eggs. The present study had the objective to demonstrate the effects of deltam
