Correlations Between Amelioration of Rotenone-Induced Parkinson's Symptoms by Amomum tsaoko Flavonoids and Gut Microbiota in Mice.

Parkinson's disease (PD) is the second most common neurodegenerative disease, but the existing therapeutic drugs for PD have limitations; thus, there is an urgent need to discover new methods of prevention and treatment. Amomum tsaoko Crevost et Lemarie (AT) is a classic traditional Chinese medicine and food. Its main pharmacological effect is the regulation of the gastrointestinal tract. To date, no studies on the use of AT or its extracts to treat PD have been reported. In this study, a rotenone-induced PD mouse model was utilized to evaluate the protective effect of Amomum tsaoko flavonoids

Neuroprotective effects of Morus alba ethanolic extract against rotenone-induced neurodegeneration and motor impairments in mouse model.

Morus alba formulations impart neuroprotective properties against dementia, oxidative stress and mitochondrial impairments. The current study aimed to evaluate the neuroprotective potential of ethanolic extract of Morus alba (EMA) on motor impairments. GC-MS characterization of extract revealed active ingredients. Adult mice were exposed to rotenone to induce neurodegeneration and motor dysfunctions. EMA reduced striatal dopamine depletion and motor impairment. Candidate genes for anti-oxidant enzymes, dopamine transmission, synaptogenesis and mitochondrial regulator were significantly modulat

Rotenone-induced cellular dysfunction in human blood platelets: Unraveling calcium dysregulation, mitochondrial impairment, oxidative stress, and apoptosis.

Rotenone, a naturally occurring pesticide and a well-established mitochondrial complex I inhibitor, disrupts electron transport chain activity, resulting in impaired energy metabolism, oxidative stress, and apoptosis. Our recent findings revealed that rotenone suppresses agonist-induced platelet functional activity; however, the molecular mechanisms underlying this effect remain largely unclear. In this study, we demonstrate that rotenone exposure induces pronounced cytotoxic effects in human platelets, evident from decreased cell viability and phosphatidylserine externalization, a hallmark of

Neuroprotective Potential of Chromium-D-Phenylalanine: Insights from Molecular Docking, Dynamics and Rotenone-Induced Parkinsonism in Rats.

This study investigates the neuroprotective potential of Chromium-D-Phenylalanine [Cr(D-Phe)₃] using molecular docking, molecular dynamics (MD) simulations and an in vivo rotenone-induced Parkinsonism model in rats. Molecular docking revealed a strong binding affinity between Cr(D-Phe)₃ and α-synuclein (PDB ID: 1XQ8), with a docking score of -8.9 kcal/mol, surpassing Levodopa (-7.7 kcal/mol). MD simulations further confirmed the stability of the Cr(D-Phe)₃-α-synuclein complex, demonstrating minimal RMSD fluctuations and sustained structural compactness over a 100-ns simulation period. Prophylactic neuroprotective propensity of Crocin, a carotenoid against rotenone induced neurotoxicity in mice: behavioural and biochemical evidence.

Previously we have demonstrated the potential neuroprotective propensity of saffron and Crocin (CR) employing a Drosophila model of Parkinsonism. Rotenone (ROT) has been extensively used as a model neurotoxin to induce Parkinson's disease (PD) like symptoms in mice. In the present study, as a proof of concept we evaluated the efficacy of CR prophylaxis (25 mg/ kg bw/d, 7d) to attenuate ROT(0.5 mg/Kg bw/d,7d) -induced neurotoxic effects in male mice focussing on neurobehavioural assessments and biochemical determinants in the striatum. CR prophylaxis significantly alleviated ROT-induced behavio

Tetramethylpyrazine Ameliorates Rotenone-Induced Parkinson's Disease in Rats: Involvement of Its Anti-Inflammatory and Anti-Apoptotic Actions.

Parkinson's disease (PD) is a slowly progressive neurodegenerative movement disorder. Apoptosis, neuroinflammation, and oxidative stress are the current hypothesized mechanisms for PD pathogenesis. Tetramethylpyrazine (TMP), the major bioactive component of Ligusticum wallichii Franchat (ChuanXiong), Family Apiaceae, reportedly has anti-apoptotic, anti-inflammatory and antioxidant effects. This study investigated the role of 'TMP' in preventing rotenone-induced neurobiological and behavioral sequelae. A preliminary dose-response study was conducted where rats received TMP (10, 20, and 40 mg/kg

Rotenone Attenuates Renal Injury in Aldosterone-Infused Rats by Inhibiting Oxidative Stress, Mitochondrial Dysfunction, and Inflammasome Activation.

BACKGROUND: Reactive oxygen species (ROS) and inflammation both contribute to the progression of aldosterone-induced renal injury. To better understand the underlying mechanisms, we examined mitochondrial dysfunction and NLRP3 inflammasome activation in aldosterone-infused rats, and explored the role of rotenone in attenuating these injuries. MATERIAL AND METHODS: Sprague-Dawley rats were divided into 3 groups: vehicle-treated, aldosterone-infused, and aldosterone plus rotenone. Renal damage was evaluated using PAS staining and electron microscopy. Levels of ROS were measured from renal tissu

Sodium butyrate improves locomotor impairment and early mortality in a rotenone-induced Drosophila model of Parkinson's disease.

Parkinson's disease (PD) is a neurodegenerative disorder primarily affecting the dopaminergic neurons in the nigrastriatal pathway resulting in debilitating motor impairment in both familial and sporadic cases. Histone deacetylase (HDAC) inhibitors have been recently implicated as a therapeutic candidate because of their ability to correct the disrupted HDAC activity in PD and other neurodegenerative diseases. Sodium butyrate (SB), an HDAC inhibitor, reduces degeneration of dopaminergic neurons in a mutant alpha-synuclein Drosophila transgenic model of familial PD. Chronic exposure to the pest

The Neuroameliorative Effects of Enzymatically Modified Isoquercitrin and Sodium R-lipoate on the Rotenone rat Model of Parkinson's Disease.

Parkinson's disease (PD) is a neurodegenerative condition distinguished by both motor and non-motor signs. Currently, therapeutic interventions effectively mitigate motor symptoms; however, they fail to impede the progression of neurodegeneration. Enzymatically modified isoquercitrin (EMIQ) is a variant of isoquercitrin with enhanced bioavailability and potentially greater health benefits. Sodium R-Lipoate (NaRLA) is a modified form of lipoic acid with improved stability and efficacy. The current work assessed EMIQ and NaRLA neuroameliorative properties in a rotenone (ROT) model of PD in rats.

Rescue of a Rotenone Model of Parkinson's Disease in C. elegans by the Mitochondrial Na+/Ca2+ Exchanger Inhibitor CGP37157.

We have previously shown that the compound CGP37157, a mitochondrial Na+/Ca2+ exchanger inhibitor, increases lifespan and improves muscle and mitochondrial structure during aging in wild-type C. elegans nematodes. We used here a rotenone model of Parkinson's disease in C. elegans to test the ability of CGP37157 to rescue the alterations induced by the toxicant. Rotenone, a mitochondrial respiratory chain complex I inhibitor, reduced worm lifespan and muscle activity, measured as worm mobility, pharyngeal pumping, and defecation rate. It also increased ROS production, decreased mitochondrial me

Parkinson's disease (PD) is characterized by progressive dopaminergic neurodegeneration leading to motor deficits, yet effective neuroprotective therapies remain limited. Curcuma longa (turmeric), a traditionally cultivated spice in Southeast Asia, exhibits potent antioxidant properties that may counteract neurodegenerative processes. This study investigated the neuroprotective potential of orally administered turmeric extract (1100 mg/kg) in a rotenone-induced rat model of PD. Rats received rotenone (2.5 mg/kg) for 21 days to induce PD-like pathology, followed by concurrent turmeric treatment

Intranasal administration of rotenone in mice attenuated olfactory functions through the lesion of dopaminergic neurons in the olfactory bulb.

Many environmental chemicals are thought to affect brain function. It was reported that chemicals in the nasal cavity directly reach the brain through the connection between olfactory neurons and the olfactory bulb (OB). In this 'olfactory transport,' xenobiotics absorbed at the nasal mucosa reach the brain by bypassing some physical barriers and defenses, and thus olfactory transport is suspected to be a vulnerable mechanism of the brain against invasion threats of environmental chemicals. In this study, we focused on the neuronal toxicity of rotenone administered intranasally to mice. The re

N-acetylcysteine prevents rotenone-induced Parkinson's disease in rat: An investigation into the interaction of parkin and Drp1 proteins.

There are convincing evidences that oxidative stress has an important role in both the initiation and progression of Parkinson's disease. N-acetylcysteine (NAC) is shown to have antioxidant properties via fortifying glutathione which is one of the main endogenous antioxidant systems. Therefore our study was aimed to evaluate the effect of NAC in management of Parkinson's disease. To this aim, male Wistar rats (10-12 months) received rotenone 2.5mg/kg/48 h intraperitoneally (ip) to induce a Parkinson's disease model. Pretreatment with NAC (25 and 50mg/kg/48 h ip) was administered 1h before the

Regulation of HSP90AA1/NRF2 by BDNF Contributes to the Attenuation of Rotenone-Induced Oxidative Stress in Parkinson's Disease Models.

Parkinson's disease, a neurodegenerative disorder, is characterized by the degeneration of dopaminergic neurons and the accumulation of α-synuclein, both of which are aggravated by oxidative stress. This study utilized rotenone-treated SH-SY5Y cells to assess cell viability, ROS levels, and mitochondrial function. RNA-seq, mass spectrometry, and Co-IP analyses identified BDNF-regulated proteins linked to oxidative stress. In rotenone-induced PD mice, evaluations were made of motor performance, neuronal degeneration, and protein expression. Results showed that a 36-h exposure to 0.5 µM rotenone

Rotenone Induces Morphological Damage and Reduction in Catecholamine Levels in the Central Nervous System of the Ascidian Styela plicata.

Ascidians are invertebrates that occupy a key phylogenetic position as a sister group of the vertebrates. The organization of their central nervous system (CNS), with cortex and medulla, indicates a plesiomorphic character of the gray and white matter of vertebrates, making this group of animals useful for studies of neurodegenerative events. Rotenone, a natural pesticide extracted from the roots of specific plants, can be used as a neurotoxin to promote parkinsonism in various organism models, leading to degeneration of the dopaminergic neurons, mitochondrial dysfunction, and locomotor defici

The Effect of Neuronal CoQ10 Deficiency and Mitochondrial Dysfunction on a Rotenone-Induced Neuronal Cell Model of Parkinson's Disease.

Parkinson's disease (PD) is the second most prevalent neurodegenerative disorder currently affecting the ageing population. Although the aetiology of PD has yet to be fully elucidated, environmental factors such as exposure to the naturally occurring neurotoxin rotenone has been associated with an increased risk of developing PD. Rotenone inhibits mitochondrial respiratory chain (MRC) complex I activity as well as induces dopaminergic neuronal death. The aim of the present study was to investigate the underlying mechanisms of rotenone-induced mitochondrial dysfunction and oxidative stress in a

The effects of rotenone on TH, BDNF and BDNF-related proteins in the brain and periphery: Relevance to early Parkinson's disease.

Loss of dopaminergic neurons in the substantia nigra (SN) is one of the pathological hallmarks in Parkinson's disease (PD). This neuron loss is accompanied by reduced protein and activity levels of tyrosine hydroxylase (TH), the rate-limiting enzyme of catecholamine synthesis. Reduced nigral brain-derived neurotrophic factor (BDNF) has been postulated to contribute to the loss of nigral dopaminergic neurons in PD by causing a lack of trophic support. Prior to this nigral cell loss many patients develop non-motor symptoms such as hyposmia, constipation and orthostatic hypotension. We investigat
