Cathepsin L potentiates autoimmunity by inhibiting lysosome-mediated STING degradation.

The stimulator of interferon genes (STING) orchestrates type I interferon (IFN) production in response to cytosolic DNA and plays essential roles in antiviral defense and autoimmune pathogenesis. The stability of STING determines the activation intensity of the pathway. Therefore, identifying proteins that govern its homeostatic regulation is needed. Here, we uncover the lysosomal protease cathepsin L (CTSL) as a critical stabilizer of STING in cells. CTSL deficiency selectively impairs STING-induced IFN responses without compromising overall lysosomal digestive function. Mechanistically, CTSL

Unravelling the interplay between ER stress, UPR and the cGAS-STING pathway: Implications for osteoarthritis pathogenesis and treatment strategy.

Osteoarthritis (OA) is a debilitating chronic degenerative disease affecting the whole joint organ leading to pain and disability. Cellular stress and injuries trigger inflammation and the onset of pathophysiological changes ensue after irreparable damage and inability to resolve inflammation, impeding the completion of the healing process. Extracellular matrix (ECM) degradation leads to dysregulated joint tissue metabolism. The reparative effort induces the proliferation of hypertrophic chondrocytes and matrix protein synthesis. Aberrant protein synthesis leads to endoplasmic reticulum (ER) s

Suppression of STING by Acacetin attenuates renal fibrosis via balancing mitophagy and protective nucleoid-phagy.

ETHNOPHARMACOLOGICAL RELEVANCE: Acacetin (5,7-dihydroxy-4'-methoxyflavone) is a primary active flavonoid extracted from traditional medicinal plants such as Agastache rugosa and Saussurea involucrata. Historically, these herbs have been utilized in Traditional Chinese Medicine to clear heat, eliminate dampness, and treat inflammatory and kidney-related ailments. Despite its potent anti-inflammatory properties, the direct molecular targets of Acacetin and its specific mechanisms regarding mitochondrial quality control (MQC) in the progression of renal fibrosis (RF) remain undefined. Hepatitis B Virus Might Be Sensed by STING-Dependent DNA Sensors and Attenuates the Response of STING-Dependent DNA Sensing Pathway in Humans with Acute and Chronic Hepatitis B Virus Infection.

DNA-dependent activator of interferon regulatory factors (DAIs), interferon gamma inducible protein 16 (IFI16), DEAD-box polypeptide 41 (DDX41), DNA-dependent protein kinase (DNA-PK), meiotic recombination 11 homolog A (MRE11), and cyclic GMP-AMP synthase (cGAS) have been identified as intracellular STING-dependent DNA sensors in recent years. Studies have shown that the DNA sensor-STING-interferon (IFN)-β pathway plays an important role in the defense against intracellular invasion of many DNA viruses. However, the intracellular recognition of hepatitis B virus (HBV) DNA by DNA sensors is sti

DNA damage-triggered activation of cGAS-STING pathway induces apoptosis in human keratinocyte HaCaT cells.

Exposure to ultraviolet B (UVB) from sunlight causes DNA damage, serious cellular inflammation and aging, and even cell death in the skin, commonly known as sunburn, leading to cutaneous tissue disorders. DNA damage can be sensed as a danger-associated molecular pattern (DAMP) by the innate immune system. It has not been studied, however, whether cGAS-STING activation is involved in the apoptosis induced by UVB irradiation or by cisplatin treatment. Here we report the findings that within hours of DNA damages keratinocytes show an innate immune response, which involves the activation of cGAS-S

Dumbbell-shape DNA structure activates cGAS-STING pathway for enhanced checkpoint immunotherapy in colorectal cancer.

Colorectal cancer (CRC), a prevalent malignancy of the digestive tract with poor responsiveness to existing immunotherapies, presents an urgent need for strategies to overcome therapeutic resistance. The cyclic GMP-AMP synthase (cGAS)- stimulator of interferon genes (STING) pathway, a key mediator of innate antitumor immunity through DNA sensing and type I interferon (IFN-I) signaling, offers a promising target for intervention. However, conventional STING agonists are limited by insufficient tumor targeting and systemic adverse effects. To address these challenges, this study developed a tumo

Antiviral functional divergence between two duplicated sting homeologs in amphitriploid gibel carp.

Gibel carp (Carassius gibelio) is an economically important aquaculture species, and the infection with cyprinid herpesvirus 2 (CyHV-2) can cause severe disease outbreaks and significant economic losses; however, the antiviral innate immune mechanisms in this species remain poorly understood. The stimulator of interferon (IFN) genes (STING) is a key adaptor in antiviral innate immunity and plays a central role in the induction of IFN responses. In this study, we identified and characterized two sting homeologs, sting-A and sting-B, in amphitriploid gibel carp. Although both STING proteins loca

Loss of cGAS facilitates angiogenesis in diabetic foot ulcer healing by suppressing the STING/ferroptosis pathway.

Diabetic foot ulcers (DFUs) are a major complication associated with diabetes and frequently result in serious health consequences. The cGAS-STING pathway has been linked to various disorders through its modulation of inflammation and ferroptosis, yet its specific function in the context of DFUs remains poorly understood. This work sought to explore the impact of targeting cGAS to inhibit STING activation on DFU healing, as well as its regulatory role in angiogenesis and endothelial cell ferroptosis. We observed elevated levels of cGAS and STING in wound samples from DFU subjects and animal mo

Paclitaxel induces trained immunity via the GPR183-STING axis to enhance host defense against MRSA infection.

Methicillin-resistant Staphylococcus aureus (MRSA) remains a major global health threat with limited prophylactic options. Trained immunity, characterized by nonspecific functional reprogramming of innate immune cells, offers a promising strategy for infection control. Here, we identify paclitaxel (PTX), a microtubule-stabilizing agent widely used in cancer therapy, as a novel inducer of trained immunity in macrophages. Unlike the microtubule-destabilizing agent nocodazole (Noco), PTX enhanced macrophage proinflammatory responses, phagocytosis, and bacterial killing upon secondary stimulation.

E3 ubiquitin ligase RNF5 attenuates pathological cardiac hypertrophy through STING.

Ring-finger protein 5 (RNF5) is an E3 ubiquitin ligase which is expressed in a variety of human tissues. RNF5 is involved in the regulation of endoplasmic reticulum stress, inflammation, and innate immunity and plays an important role in the occurrence and development of various tumors. However, the role of RNF5 in cardiac hypertrophy has not been reported. In this study, we found the expression of RNF5 was increased in the hearts of mice with pathological cardiac hypertrophy. The loss-of-function research demonstrated that RNF5 deficiency exacerbated cardiac hypertrophy, whereas gain-of-funct

Senkyunolide I Inhibits mtDNA-cGAS-STING Signaling in Macrophages via Targeting VDAC1 Oligomerization to Attenuate Ulcerative Colitis.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease with limited therapeutic options. The cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) pathway, activated by cytosolic mitochondrial DNA (mtDNA), has been increasingly implicated in UC. Through proteomics and artificial intelligence modeling, this study identified for the first time that senkyunolide I (SEI), a primary bioactive phthalide from Ligusticum striatum DC. (L. striatum), inhibits experimental colitis via the cGAS-STING pathway, with mechanistic validation performed in both intestinal tissues and cul

Lovastatin-Induced Mitochondrial Oxidative Stress Leads to the Release of mtDNA to Promote Apoptosis by Activating cGAS-STING Pathway in Human Colorectal Cancer Cells.

Statins are 3-hydroxy-3-methylglutaryl coenzyme-A (HMG-CoA) reductase inhibitors widely used in the treatment of hyperlipidemia. The inhibition of HMG-CoA reductase in the mevalonate pathway leads to the suppression of cell proliferation and induction of apoptosis. The cyclic GMP-AMP synthase (cGAS) stimulator of the interferon genes (STING) signaling pathway has been suggested to not only facilitate inflammatory responses and the production of type I interferons (IFN), but also activate other cellular processes, such as apoptosis. It has not been studied, however, whether cGAS-STING activatio

The multifaceted functions of STING in CD4+ T cell biology.

The stimulator of interferon genes (STING) protein has centered on its role as a transmembrane receptor in professional innate immune cells, mediating type I interferon (IFN-I) signaling. However, recent advancements have significantly revealed its marked expression and functional importance in adaptive immune cells, particularly CD4+ T lymphocytes. STING signaling intersects with crucial T cell pathways, influencing development, activation, survival, and differentiation. This review synthesizes current knowledge on the dynamic regulation and multifaceted roles of STING in CD4+ T cell biology.
