Magnetic Resonance Imaging Evaluation of Pregnancy and Aging on External Anal Sphincter Volume in Squirrel Monkeys.

INTRODUCTION AND HYPOTHESIS: Both humans and squirrel monkeys are susceptible to pelvic floor injury and prolapse from pregnancy, delivery, and aging. The mechanisms for external anal sphincter (EAS) injury in squirrel monkeys have not been evaluated in detail. This study evaluates a method for measuring EAS volume using magnetic resonance imaging (MRI) in squirrel monkeys and demonstrates the feasibility of serial measurements. METHODS: Using a previously described procedure, MRI was performed on 10 squirrel monkeys prior to euthanasia. After euthanasia, tissue blocks of EAS were cryo-sectio

In vitro and in vivo characterization of novel 8-methoxy derivatives of chlortetracycline.

The in vitro activities of three new 8-methoxychlortetracyclines, Sch 36969, 33256 and 34164 were compared to tetracycline, minocycline and doxycycline. Against aerobic Gram-negative rods Sch 36969 had a geometric mean MIC (GMM) of 4.2 micrograms/ml, about 8-fold more potent than Sch 33256, and similar to all the other compounds. Sch 36969 also had good activity against methicillin-resistant (GMM, 0.21 micrograms/ml) and -susceptible Staphylococci (GMM, 0.14 micrograms/ml), Streptococci (GMM, 0.06 micrograms/ml), and most anaerobic bacteria (GMM, less than 0.5 micrograms/ml). Change in susceptibility to vestibular-visual conflict sickness in monkeys by repeated exposure.

Squirrel monkeys, susceptible to the vestibular-visual conflict sickness in pitch (with frank vomiting), were subjected to repeated exposure to pitch conflict in various modes and in a randomly mixed order (30 min daily, for 10 consecutive days). Immediately after the training, a significant decline in susceptibility was found, represented by reduced vomiting rates, reduced sickness scores, reduced salivation and improved regularity of vertical oculomotor responses. Susceptibility at the pre-training level returned when the test was repeated 10 days later. Temporary suppression of pitch confli

Effects of head and body restraint on experimental motion-induced sickness in squirrel monkeys.

Each of 16 Bolivian-phenotype squirrel monkeys of mixed sex had a machine bolt mounted on the skull with acrylic cement; 13 were provided with temporary plaster body casts allowing free movement. With eyes open, all were rotated in the horizontal plane at 30 rpm every other day until vomiting occurred or for a maximum duration of 120 min/spin. Latencies for motion-sickness signs were recorded under three experimental conditions: free movement, torso fixed to an aluminum frame and both torso and head restrained by bolting to the frame. Subsequently, 10 monkeys from this sample were rotated whil

Vestibular-visual conflict sickness in the squirrel monkey.

When combined vestibular and visual (optokinetic) stimuli were given to squirrel monkeys in a direction and phase mismatching sinusoidal mode (in yaw plane), five of six monkeys exhibited actual emesis during three repeated exposures on separate trial days. Two animals vomited on two trial days and three animals, on one day. The incidence took place in 38.9% of the total trial days. When the identical vestibular and visual stimuli were given individually, the effect was negligible, thus confirming each stimulus magnitude was not stressful enough to produce emesis when used separately.

Distribution of somatostatin-28 (1-12), calcitonin gene-related peptide, and substance P in the squirrel monkey brainstem: an immunocytochemical study.

Using an immunocytochemical technique, we have studied the distribution of fibers and cell bodies containing somatostatin-28 (1-12) [SOM-28 (1-12)], calcitonin gene-related peptide (CGRP), and substance P (SP) in the brainstem of Saimiri sciureus. The distribution of the peptidergic cell bodies was very restricted: perikarya containing SOM-28 (1-12) were only observed in the substantia grisea centralis, while no immunoreactive cell bodies containing CGRP or SP were visualized. Fibers containing SOM-28 (1-12), CGRP, or SP were widely distributed in the brainstem: immunoreactive fibers containin

Squirrel monkeys support replication of BK virus more efficiently than simian virus 40: an animal model for human BK virus infection.

We performed experiments to test the suitability of squirrel monkeys (Saimiri sciureus) as an experimental model for BK virus (BKV) and simian virus 40 (SV40) infection. Four squirrel monkeys received intravenous inoculation with BKV Gardner strain, and six squirrel monkeys received intravenous inoculation with SV40 777 strain. Eight of 10 monkeys received immunosuppression therapy, namely, cyclophosphamide subcutaneously either before or both before and after viral inoculation. The presence of viral infection was assessed by quantitative real-time PCR amplification of viral DNA from blood, ur

[Infection of New World primates with Junín virus. IV. Aotus trivirgatus].

Owl monkeys (Aotus trivirgatus) were inoculated with XJ, a pathogenic strain of Junin virus, seeking new animal models for Argentine Hemorrhagic Fever. Nine monkeys were inoculated intramuscularly with 30 or 300,000 TCID50 of junin virus. Hematological and virological studies showed no alteration in blood elements such as red cell, reticular cell and platelets, up to 28 days after inoculation. Hemoglobin and hematocrit determinations also remained constant. However, significant neutropenia was seen at day 11 and minimal viremia was detected in some animals during the second and third week post

The mechanism of labor in nonhuman primates: A look inside.

While in humans, the flexed position of the fetus and its rotating course down the birth canal are well documented, in other primates the mechanism of labor is unknown. Despite the lack of comparative data, it is commonly assumed that the human obstetric mechanism is unique, and anthropologists have disputed when and why the transition to the modern human mechanism occurred. The purpose of this study is to establish the mechanism of labor in two species of nonhuman primates. The data in this study consist of a series of radiographs taken throughout labor in unanesthetized, unrestrained, sponta

Towards A Microbead Occlusion Model of Glaucoma for a Non-Human Primate.

Glaucoma is a group of optic neuropathies associated with aging and sensitivity to intraocular pressure (IOP). The disease causes vision loss through the degeneration of retinal ganglion cell neurons and their axons in the optic nerve. Using an inducible model of glaucoma, we elevated IOP in the squirrel monkey (Saimiri boliviensis) using intracameral injection of 35 μm polystyrene microbeads and measured common pathogenic outcomes in the optic projection. A 42% elevation in IOP over 28 weeks reduced anterograde transport of fluorescently-labeled cholera toxin beta from retina to the lateral g

Amyloid in the brains of aged squirrel monkeys.

In this immunocytochemical study, the brains of nine squirrel monkeys (Saimiri sciureus), ranging from 8 to 27 years of age, were examined for the presence and distribution of beta/A4 amyloid, a 4-kilodalton peptide. In aged squirrel monkeys, amyloid is associated primarily with intracerebral and meningeal capillaries and arterioles and occurs to a lesser degree as small and/or diffuse deposits in the neural parenchyma and in the dense cores of senile plaques. Cerebrovascular amyloid is found primarily in neocortex, amygdala, and septum verum and is rare or nonexistent in other subcortical gra

Characterization of a chemokine receptor-related gene in human herpesvirus 8 and its expression in Kaposi's sarcoma.

Human herpesvirus 8 (HHV-8) is a recently discovered, virus that is highly associated with Kaposi's sarcoma (KS) and AIDS-associated body cavity lymphomas, although it is also found in some normal individuals. HHV-8 is related by nucleotide sequence homology to herpesvirus saimiri (HVS), which causes T cell lymphomas in some New World monkeys, and to Epstein-Barr virus (EBV), a human herpesvirus linked etiologically with Burkitt's lymphoma and nasopharyngeal carcinoma. We report that, like HVS but unlike EBV, HHV-8 contains a gene (ORF74) with significant sequence homology to the high-affinity

Cortical Morphology and White Matter Tractography of Three Phylogenetically Distant Primates: Evidence for a Simian Elaboration.

Comparative neuroimaging has been used to identify changes in white matter architecture across primate species phylogenetically close to humans, but few have compared the phylogenetically distant species. Here, we acquired postmortem diffusion imaging data from ring-tailed lemurs (Lemur catta), black-capped squirrel monkeys (Saimiri boliviensis), and rhesus macaques (Macaca mulatta). We were able to establish templates and surfaces allowing us to investigate sulcal, cortical, and white matter anatomy. The results demonstrate an expansion of the frontal projections of the superior longitudinal

Pterygodermatites nycticebi (Nematoda: Rictulariidae): accidental detection of encapsulated third-stage larvae in the tissue of a white-fronted marmoset.

Twin, white-fronted marmosets (Callithrix geoffroyi) born and raised in a zoo in Japan died at 7 mo of age. Several encapsulated nematode larvae were detected in the intestinal wall, as well as a few in the mesenteric lymph nodes of 1 of the twins. In the other marmoset, no encapsulated nematode larva was detected in the organs, but many adult Pterygodermatites nycticebi were found in the intestinal lumen. In the past 5 yr, 5 primates kept in the same zoo, i.e., 1 squirrel monkey (Saimiri sciureus), 2 Pygmy marmosets (Cebuella pygmaea), 1 Senegal galago (Galago senegalensis), and 1 cotton-top

Caveola-vesicle complexes of Plasmodium vivax and Plasmodium cynomolgi: large-scale aggregation and structure of PHIST-positive vesicles in late schizont-infected red blood cells.

BACKGROUND: Numerous caveola-vesicle complexes (CVCs) form around the surface of Plasmodium vivax and Plasmodium cynomolgi infected erythrocytes during the asexual cycle. They include a 95 kDa protein in both species, the Plasmodium helical interspersed subtelomeric (PHIST) protein (PHIST/CVC-8195) located at the cytoplasmic face of CVC vesicles and tubules. The functions and detailed structure of CVCs are poorly understood, although they are essential for parasite survival. In this study, electron and immuno-electron microscopy were used to explore structural changes to CVCs during the final

Chronic Δ9-tetrahydrocannabinol exposure in adolescent nonhuman primates: persistent abnormalities in economic demand and brain functional connectivity.

Although chronic cannabis use during adolescence can alter brain function and impair complex behavioral processes, it is unclear whether such deficits persist into adulthood. Using a coordinated awake neuroimaging and behavioral approach in nonhuman primates, we addressed this issue by examining the impact of chronic adolescent exposure to Δ9-tetrahydrocannabinol (THC) on brain functional connectivity and motivational processes during early adulthood. Female and male squirrel monkeys (n = 23) were treated daily for 6 months during adolescence with vehicle or either a low (0.32 mg/kg) or high d

Functional imaging and anatomical connections in squirrel monkeys reveal parietal-frontal circuits underlying eye movements.

The posterior parietal cortex (PPC) of squirrel monkeys contains subregions where long trains of intracortical microstimulation evoke complex, behaviorally meaningful movements. Recently, we showed that such stimulation of a part of the PPC in the caudal lateral sulcus (LS) elicits eye movements in these monkeys. Here, we studied the functional and anatomical connections of this oculomotor region we call parietal eye field (PEF) with frontal eye field (FEF) and other cortical regions in 2 squirrel monkeys. We demonstrated these connections with intrinsic optical imaging and injections of anato
